BACKGROUND: Endocannabinoids such as anandamide are thought to have adverse effects on pregnancy and embryonic development. The activity of the degradative enzyme anandamide hydrolase may therefore be crucial for prevention of excessive concentrations of anandamide in the uterus, and thus prevention of pregnancy failure or female infertility. We tested this hypothesis in a preliminary study, and then used the results to find out whether anandamide hydrolase activity could predict miscarriage in a group of pregnant women.
METHODS: We assessed anandamide hydrolase activity in peripheral lymphoc

OBJECTIVE: This study investigated the role of the cannabinoid receptor type 2 (CB2R) in the spinal cord dorsal horn (SCDH) using a spared nerve injury (SNI) model of neuropathic pain (NP). The aim was to clarify the analgesic effects of different electroacupuncture (EA) frequencies in SNI rats and to explore whether the higher frequency exerts analgesic effects by modulating the CB2R signaling pathway.
METHODS: An SNI rat model was initially established. Mechanical allodynia was assessed using the paw withdrawal threshold (PWT), and motor function was assessed using a gait analysis system. E

BACKGROUND: Pulmonary arterial hypertension (PAH) is a severe multifactorial disease associated with impaired pulmonary hemodynamics, leading to right ventricular (RV) hypertrophy and failure. Induction of inducible nitric oxide synthase (iNOS) and/or activation of cannabinoid CB1 receptor (CB1R) is associated with pro-inflammatory, pro-fibrotic, and pro-hypertrophic effects. Therefore, we tested the effects of chronic simultaneous iNOS/CB1R blockade in a rat model of monocrotaline-induced pulmonary hypertension (MCT-PH).
METHODS: Rats were injected with monocrotaline or saline (control) and

Cerebral ischemia-reperfusion injury arises predominantly from excessive oxidative stress and sustained neuroinflammation. The selective cannabinoid receptor type 2 (CB2R) agonist AM1241 has shown potential to alleviate both pathological processes, yet its therapeutic efficacy is severely hampered by poor blood-brain barrier (BBB) permeability and inadequate intracerebral accumulation. To elevate the intracerebral therapeutic efficacy of AM1241, we constructed polydopamine-modified mesoporous silica nanoparticles (PMSNs) as nanocarriers to encapsulate AM1241, generating PMSNs@AM1241. Comprehen

Bipolar disorder (BD) is a severe psychiatric condition characterized by recurrent mood episodes and progressive neurobiological alterations associated with oxidative stress, mitochondrial dysfunction, and neuronal damage. Current pharmacological treatments remain limited by incomplete efficacy and significant adverse effects, highlighting the need for novel therapeutic strategies. The present study investigated the neuroprotective effects of beta-caryophyllene (BCP), a natural sesquiterpene and selective cannabinoid receptor type 2 (CB2R) agonist, in a rat model of mania induced by intracereb

The endocannabinoid (eCB) system comprises cannabinoid receptors; endogenous ligands, including anandamide (AEA), 2-arachidonoylglycerol (2-AG); and related N -acylethanolamines (NAEs), N- palmitoylethanolamide (PEA), and N- oleoylethanolamide (OEA); and metabolizing enzymes (eg, fatty acid amide hydrolase [FAAH]). The eCB system modulates nociceptive circuits in rodents. In humans, the FAAH C385A polymorphism is associated with reduced pain sensitivity, suggesting eCB tone contributes to individual pain differences, but this has yet to be tested. Here, we determined whether the eCB system is

IN BRIEF: The cervix plays a crucial role not only in the maintenance of pregnancy but also during delivery, when it undergoes extensive changes. This study highlights the involvement of the endocannabinoidome in cervical remodeling, emphasizing its relevance in the shift from a nonpregnant to pregnant state and its potential contribution to preterm delivery in inflammatory contexts.
ABSTRACT: During pregnancy, the main role of the cervix is to isolate the fetus from outside pathogens and maintain the relatively closed system of uterine gestation. Conversely, toward the end of pregnancy, the

The cardiac baroreflex is an autonomic neural mechanism involved in the modulation of the cardiovascular system. It influences the heart rate and peripheral vascular resistance to preserve arterial blood pressure within a narrow variation range. This mechanism is mainly controlled by medullary nuclei located in the brain stem. However, supramedullary areas, such as the ventral portion of medial prefrontal cortex (vMPFC), are also involved. Particularly, the glutamatergic NMDA/NO pathway in the vMPFC can facilitate baroreflex bradycardic and tachycardic responses. In addition, cannabinoid recep

Symptoms of depressive disorders such as anhedonia and despair can be a product of an aberrant adaptation to stress conditions. Chronic unpredictable stress model (CUS) can generate an increase in the activity of the hypothalamic-pituitary-adrenal axis (HPA) and induce a reduction of neurotrophin signaling and the proliferation of neural progenitors in the adult dentate gyrus, together with increased oxidative stress. Levels of the endocannabinoid anandamide (AEA) seem to affect these depression-by-stress-related features and could be modulated by fatty acid amide hydrolase (FAAH). We aimed to

The organized, tightly regulated signaling relays engaged by the cannabinoid receptors (CBs) and their ligands, G proteins and other effectors, together constitute the endocannabinoid system (ECS). This system governs many biological functions including cell proliferation, regulation of ion transport and neuronal messaging. This review will firstly examine the physiology of the ECS, briefly discussing some anomalies in the relay of the ECS signaling as these are consequently linked to maladies of global concern including neurological disorders, cardiovascular disease and cancer. While endogeno

Arachidonoylethanolamide (anandamide) is an endogenous ligand for cannabinoid receptors. We demonstrated previously that the periimplantation mouse uterus has high levels of anandamide and can synthesize and hydrolyse anandamide. In the present investigation, we examined the expression of the recently identified fatty-acid amide hydrolase (FAAH) gene, which is involved in hydrolyzing anandamide to arachidonic acid and ethanolamine, in the periimplantation mouse embryo and uterus. As previously reported, Northern blot hybridization detected a transcript of approximately 2.5 kilobases of FAAH mR

BACKGROUND: The endocannabinoid (eCB) 2-arachidonoylglycerol (2-AG) is a known modulator of inflammation and ligand to both, pro-inflammatory cannabinoid receptor 1 (CB1) and anti-inflammatory CB2. While the role of both receptors in atherogenesis has been studied extensively, the significance of 2-AG for atherogenesis is less well characterized.
METHODS: The impact of 2-AG on atherogenesis was studied in two treatment groups of ApoE-/- mice. One group received the monoacylglycerol lipase (MAGL)-inhibitor JZL184 [5 mg/kg i.p.], which impairs 2-AG degradation and thus causes elevated 2-AG leve

Osteoporosis is a prevalent skeletal disorder characterized by imbalanced bone remodeling, leading to excessive bone loss. Cannabinoid receptor 2 (CB2) has emerged as a promising therapeutic target in bone metabolism due to its peripheral expression and dual regulation of osteoblast and osteoclast activity. Desmethoxyyangonin (DMY) has demonstrated CB2 agonist activity, yet its therapeutic efficacy and molecular mechanisms in bone remain incompletely understood. Here, we investigated the anti-osteoporotic effects of DMY and its mechanistic insight on CB2 signaling. An ovariectomized (OVX) mous

BACKGROUND: Radiation-induced intestinal injury (RIII) represents a significant dose-limiting complication of radiotherapy, characterized by substantial loss of intestinal epithelial cells (IECs). While the activation of cannabinoid receptor 2 (CB2R) is protective in immune‑mediated colitis, the intrinsic role of CB2R in IECs and its potential therapeutic relevance in RIII have not been defined.
METHODS: An RIII mouse model was established in wild-type and CB2R-/- mice. Small-molecule CB2R agonists were screened for radioprotective efficacy, followed by pharmacological and siRNA-based interro

17β-estradiol (E2) enhances the cerebellar molecular layer interneurons (MLIs)-Purkinje cells (PCs) synaptic transmission via activation of the Erβ in vivo in mice. Whether E2 regulates cerebellar MLI-PC synaptic plasticity is unknown. To investigate the mechanism of E2, we evaluated the modulation of facial stimulation-evoked MLI-PC long-term plasticity in mice. Cell-attached recordings from PCs of Crus II were performed using an Axopatch-700B patch-clamp amplifier. The MLI-PC synaptic transmission was evoked by facial stimulation. Immunohistochemistry was used to detect the expression of ERβ

Since the pioneering work of Gadea-Ciria (Gadea-Ciria M, Stadler H, Lloyd KG, Bartholini G. Acetylcholine release within the cat striatum during the sleep-wakefulness cycle. Nature 1973; 243:518-519) indicating pointing to the involvement of acetylcholine and basal ganglia in sleep regulation; extensive literature has suggested that this brain complex participates in the control of the sleep-waking cycle (SWC). On the other hand, it has been demonstrated that the endocannabinoid system (eCBS) is prominently involved in the regulation of the SWC, mood and its related disorders. Since cannabinoi

In adult rodents, endocannabinoids (eCBs) regulate fast glucocorticoid (GC) feedback in the hypothalamus-pituitary adrenal (HPA) axis, acting as retrograde messengers that bind to cannabinoid receptors (CB1R) and inhibit glutamate release from presynaptic CRH neurons in the paraventricular nucleus of the hypothalamus (PVN). During the first two weeks of life, rat pups exhibit significant CRH and ACTH responses to stress although the adrenal GC output remains reduced. At the same time, pups also display increased sensitivity to GC feedback, but it is unclear whether eCBs play a role in mediatin

Background: Periodontitis is a chronic inflammatory disease characterized by dysregulated host immune responses that drive connective tissue destruction and alveolar bone loss. Human gingival fibroblasts (HGFs) are central regulators of periodontal inflammation through their production of cytokines, chemokines, matrix-remodeling enzymes, and other inflammatory mediators. Although cannabinoid receptor 2 (CB2) activation has demonstrated anti-inflammatory properties, its coordinated effects on multiple inflammatory pathways in gingival fibroblasts remain poorly understood. Gastric cancer (GC) has poor survival in advanced stages, with limited treatment options. Paclitaxel (PTX) is commonly used, but resistance often arises, highlighting the need for targeted therapies. Cannabinoid receptor type 2 (CB2R) is overexpressed in several cancers and its activation has been associated with reduced tumor growth and metastasis. This study evaluated the antitumor activity of selected CB2R agonists with dual activity (CC48 and Fi9) compared to single-target compounds (ASF151), a reference agonist (compound 1), and an antagonist (AM630). The compounds' cytotoxicity was deter
