Immune responses elicited in rainbow trout through the administration of infectious pancreatic necrosis virus-like particles.

Virus like particles (VLPs) against viral pathogens not only constitute a novel approach for the development of antiviral vaccines for an specific virus, but also for the creation of multivalent vaccines in which antigens from other pathogens may be expressed on the surface of these VLPs. Despite positive results on protection for many of these VLPs in both fish and mammals, not many studies have focused on the immune response triggered by these particles; studies that may provide hints for the identification of immune mechanisms responsible for antiviral protection, which are mostly unknown i

Uptake of waterborne tributyltin in the brain of fish: axonal transport as a proposed mechanism.

In previous studies, it was shown that waterborne Hg(II), Cd(II), and Mn(II) enter nerves innervating water-exposed sensory organs of fish and are transported to the brain by axonal transport. However, it is not known if organometals, such as tributyltin (TBT), can reach the brain of fish via the same route. In this work, we exposed rainbow trout (Oncorhynchus mykiss) to waterborne [113Sn]-TBT (4.2 kBq/L). Three fish were sampled after a 2-week exposure, and three others were sampled after a 2-week depuration period. Another group of four fish received an intravenous injection of [113Sn]-TBT a

Volumetric and ionic responses of goldfish hepatocytes to anisotonic exposure and energetic limitation.

The relationship between cell volume and K(+) transmembrane fluxes of goldfish (Carassius auratus) hepatocytes exposed to anisotonic conditions or energetic limitation was studied and compared with the response of hepatocytes from trout (Oncorhynchus mykiss) and rat (Rattus rattus). Cell volume was studied by video- and fluorescence microscopy, while K(+) fluxes were assessed by measuring unidirectional (86)Rb(+) fluxes. In trout and rat hepatocytes, hyposmotic (180 mosmoll(-1)) exposure at pH 7.45 caused cell swelling followed by a regulatory volume decrease (RVD), a response reported to be m

Regulation of glucose production in rainbow trout: role of epinephrine in vivo and in isolated hepatocytes.

The rate of hepatic glucose production (R(a) glucose) of rainbow trout (Oncorhynchus mykiss) was measured in vivo by continuous infusion of [6-(3)H]glucose and in vitro on isolated hepatocytes to examine the role of epinephrine (Epi) in its regulation. By elevating Epi concentration and/or blocking beta-adrenoreceptors with propranolol (Prop), our goals were to investigate the mechanism for Epi-induced hyperglycemia to determine the possible role played by basal Epi concentration in maintaining resting R(a) glucose and to assess indirect effects of Epi in the intact animal. In vivo infusion of

Assessing a semi-validated quantitative PCR for high-throughput surveillance of Ichthyophonus sp. in Yukon River Chinook Salmon.

OBJECTIVE: We sought to determine the suitability of a semi-validated quantitative PCR (qPCR) assay for assessing the prevalence and density of Ichthyophonus sp. infections in Yukon River Chinook Salmon Oncorhynchus tshawytscha, representing a new species and location for the assay's application. Ichthyophonus sp. causes severe disease that has been implicated in the en route prespawning mortality of this Chinook Salmon stock. The reduced survival of these fish has caused many hardships for the people who rely on them. METHODS: We assessed a modified qPCR assay with stage 1 and partial stage

Effect of Supplementation of Antioxidant Lipids Synthetized by Enzymatic Acidolysis with EPA/DHA Concentrate and Maqui (Aristotelia chilensis (Mol.) Stuntz) Seed Oil for Mitigating High-Fat Diet-Induced Obesity and Metabolic Disorders in Mice.

Bioactive compounds have shown significant potential in the management of obesity and metabolic syndrome (MetS). This study investigates the effects of antioxidant lipids (ALω-3), synthetized through enzymatic acidolysis using non-specific lipase B from Candida antarctica under supercritical CO2 conditions. These lipids were derived from a concentrate of rainbow trout (Oncorhynchus mykiss) belly oil, rich in long-chain polyunsaturated omega-3 fatty acids (LCPUFAn-3), and cold-pressed maqui seed oil (MO, Aristotelia chilensis (Mol.) Stuntz). Their effects were then evaluated in a murine high-fa

Mitigating tire wear particles and tire additive chemicals in stormwater with permeable pavements.

6PPD-quinone (6PPDQ) is a recently discovered chemical that is acutely toxic to coho salmon (Oncorhynchus kisutch) and can form via environmental exposure of 6PPD, a compound found extensively in tire wear particles (TWPs). TWPs deposited on roads are transported to aquatic ecosystems via stormwater, contributing to microplastic pollution and organic contaminant loads. However, little is known about the fate of TWPs and their leachable contaminants in these systems. We conducted three experiments at a high school in Tacoma, Washington, to quantify the treatment performance of permeable pavemen

Dietary exposure to environmentally relevant pesticide mixtures impairs swimming performance and lipid homeostatic gene expression in Juvenile Chinook salmon at elevated water temperatures.

Aquatic organisms are exposed to complex mixtures of pesticides in the environment, but traditional risk assessment approaches typically only consider individual compounds. In conjunction with exposure to pesticide mixtures, global climate change is anticipated to alter thermal regimes of waterways, leading to potential co-exposure of biota to elevated temperatures and contaminants. Furthermore, most studies utilize aqueous exposures, whereas the dietary route of exposure may be more important for fish owing to the hydrophobicity of many pesticides. Consequently, the current study aimed to det

Methemoglobin concentrations in three salmonid species following exposure to benzocaine or tricaine methanesulfonate.

Methemoglobin is hemoglobin containing ferric iron rather than ferrous iron which renders it incapable of binding to oxygen. Blood sampling of fish is done under sedation or general anesthesia. Tricaine methanesulfonate (TMS) or benzocaine is commonly used but both can cause oxidation of hemoglobin to methemoglobin. Our objective was to determine if methemoglobin concentrations in healthy rainbow trout (Oncorhynchus mykiss), brook trout (Salvelinus fontinalis), or Atlantic salmon (Salmo salar) increase during sedation with 25 mg/L of a 10% benzocaine solution or with repeated short anesthetiza

Modulation of hypothalamic-pituitary-interrenal axis function by social status in rainbow trout.

Juvenile rainbow trout (Oncorhynchus mykiss) form stable dominance hierarchies when confined in pairs. These hierarchies are driven by aggressive competition over limited resources and result in one fish becoming dominant over the other. An important indicator of low social status is sustained elevation of circulating cortisol levels as a result of chronic activation of the hypothalamic-pituitary-interrenal (HPI) axis. In the present study it was hypothesized that social status modulates the expression of key proteins involved in the functioning of the HPI axis. Cortisol treatment and fasting

Approaches towards DNA vaccination against a skin ciliate parasite in fish.

Rainbow trout (Oncorhynchus mykiss) were immunized with plasmid DNA vaccine constructs encoding selected antigens from the parasite Ichthyophthirius multifiliis. Two immobilization antigens (I-ags) and one cysteine protease were tested as genetic vaccine antigen candidates. Antigenicity was evaluated by immunostaining of transfected fish cells using I-ag specific mono- and polyclonal antibodies. I. multifiliis specific antibody production, regulation of immune-relevant genes and/or protection in terms of parasite burden or mortality was measured to evaluate the induced immune response in vacci

Pacific salmon extinctions: quantifying lost and remaining diversity.

Widespread population extirpations and the consequent loss of ecological, genetic, and life-history diversity can lead to extinction of evolutionarily significant units (ESUs) and species. We attempted to systematically enumerate extinct Pacific salmon populations and characterize lost ecological, life history, and genetic diversity types among six species of Pacific salmon (Chinook [Oncorhynchus tshawytscha], sockeye [O. nerka], coho [O. kisutch], chum [O. keta], and pink salmon [O. gorbuscha] and steelhead trout [O. mykiss]) from the western contiguous United States. We estimated that, colle

Dietary and tissue residue analysis and contaminant intake estimations in rats consuming diets composed of Great Lakes salmon: a multigeneration study.

To further characterize the toxicological risk associated with chemical contaminants in Great Lakes fish, a multigeneration rat reproduction study was designed. Mature chinook salmon (Oncorhynchus tsawytscha), collected during the Fall 1991 spawning runs from Sydenham River, Lake Huron, and Credit River, Lake Ontario, were filleted, lyophilized, and incorporated into standard rat diets at 25% (w/w) or 100% (w/w) of the normal protein compliment [casein, 20% (w/w)]. This resulted in diets composed of 5 or 20% (w/w) lyophilized fish and estimated daily fish intakes by the rats at levels approxim

Minor environmental concentrations of antibiotics can modify bacterial virulence in co-infection with a non-targeted parasite.

Leakage of medical residues into the environment can significantly impact natural communities. For example, antibiotic contamination from agriculture and aquaculture can directly influence targeted pathogens, but also other non-targeted taxa of commensals and parasites that regularly co-occur and co-infect the same host. Consequently, antibiotics could significantly alter interspecific interactions and epidemiology of the co-infecting parasite community. We studied how minor environmental concentrations of antibiotic affects the co-infection of two parasites, the bacterium Flavobacterium colum

Molecular tracing confirms that infection with infectious pancreatic necrosis virus follows the smolt from hatchery to grow-out farm.

Infectious pancreatic necrosis (IPN) is an important restraint to production of salmonids in aquaculture globally. In order to implement efficacious mitigation strategies for control of this disease, it is important to understand infection routes under current production systems. IPN virus has been shown to be transmitted vertically in Rainbow trout, from broodstock to fingerlings in hatcheries, and there is circumstantial evidence suggesting that vertical transmission can also occur in Atlantic salmon, in addition to horizontal transmission between grow-out fish in farms. In this study, we sh

Ammonia sensing by neuroepithelial cells and ventilatory responses to ammonia in rainbow trout.

Ammonia, the third respiratory gas in teleost fish, acts as an acute stimulant to ventilation in ammoniotelic rainbow trout. We investigated whether this sensitivity is maintained in trout chronically exposed (1+ months) to high environmental ammonia [HEA, 250 μmol l(-1) (NH(4))(2)SO(4)] in the water, and whether gill neuroepithelial cells (NECs) are involved in ammonia sensing. Hyperventilation was induced both by acute external (NH(4))(2)SO(4) exposure [250 or 500 μmol l(-1) (NH(4))(2)SO(4)] and by intra-arterial (NH(4))(2)SO(4) injection (580 μmol kg(-1) of ammonia) in control trout, but th

Caligus elongatus Nordmann genotypes on wild and farmed fish.

Two mitochondrial genotypes have been described for Caligus elongatus Nordmann in Norway. This article reports on the distribution of C. elongatus mitochondrial cytochrome C oxidase 1 genotypes from wild fish hosts from the SE Norwegian coast. For comparison, lice from areas with fish farming were included in the study. The genotype distribution of 841 lice from wild coastal (n = 535), wild North Sea pelagic (n = 26), farmed (n = 160) and wild hosts in areas of fish farming (n = 89) is presented. The genotype frequencies of C. elongatus on wild coastal hosts varied significantly between spring

Peripherally administered growth hormone increases brain dopaminergic activity and swimming in rainbow trout.

There is increasing evidence that growth hormone (GH) has important behavioral effects in fish, but the underlying mechanisms are not well understood. To investigate if peripherally administered GH influences the monoaminergic activity of the brain, and how this is correlated to behavior, juvenile rainbow trout were implanted intraperitoneally with ovine GH. Fish were either kept isolated or in groups of five. The physical activity and food intake of the isolated fish were observed after 1 and 7 days, when brains were also sampled. The content of serotonin, dopamine, and noradrenaline and thei

Androgenesis in rainbow trout using cryopreserved spermatozoa: the effect of processing and biological factors.

In addition to producing homozygous lines for biomedical and genomic research and monosex stocks for commercial purposes, androgenesis is the biotechnology considered most promising and reliable for recovering complete nuclear genome information from cryopreserved fish cells. That is because procedures of cryopreserving spermatozoa, contrary to procedures for oocytes or entire eggs, are being well developed. Application of androgenesis in genome banking programs addresses the needs of both the aquaculture industry (safeguard for valuable strains and lines) and natural resource conservation (in

Evidence for a mineralocorticoid-like receptor linked to branchial chloride cell proliferation in freshwater rainbow trout.

Fish acclimated to ion-deficient water exhibit proliferation of branchial chloride cells. The objective of the present study was to investigate the role of cortisol in this response using the corticosteroid receptor antagonists RU486 and spironolactone. RU486 is a potent antagonist of the glucocorticoid actions of cortisol, whereas spironolactone exhibits high-affinity binding to mineralocorticoid receptors, with a resulting blockade of mineralocorticoid properties in mammals. Untreated rainbow trout, as well as rainbow trout given a single intraperitoneal implant of coconut oil alone, coconut
