Gene diversity and genetic variation in lung flukes (genus Paragonimus).

Paragonimiasis caused by lung flukes (genus Paragonimus) is a neglected disease occurring in Asia, Africa and the Americas. The genus is species-rich, ancient and widespread. Genetic diversity is likely to be considerable, but investigation of this remains confined to a few populations of a few species. In recent years, studies of genetic diversity have moved from isoenzyme analysis to molecular phylogenetic analysis based on selected DNA sequences. The former offered better resolution of questions relating to allelic diversity and gene flow, whereas the latter is more suitable for questions r

Development of an immunodiagnosis method using recombinant PsCP for detection of Paragonimus skrjabini infection in human.

Paragonimiasis skrjabini is a kind of zoonosis and prevalent in 16 provinces in China, such as Chongqing, Fujian, Sichuan, and Yunnan. However, sensitive and efficient diagnostic methods for the infection are limited. In order to provide a more convenient and simple method for serologic diagnosis, the recombinant P. skrjabini cysteine protease (PsCP) was expressed, purified, and then used to develop an indirect enzyme-linked immunosorbent assay (ELISA) for detecting anti-PsCP antibodies in human. Given the positive/negative cutoff value as 0.606, the maximum dilution of human sera in which ant

The drugs we have and the drugs we need against major helminth infections.

Parasitic worms (helminths) have accompanied humans for thousands of years and, still today, they are pervasive where poverty persists, including large parts of Southeast Asia and the Western Pacific Region. The global strategy for the control of helminth infections is morbidity control and elimination as a public health problem. Regular administration of anthelminthic drugs to at-risk populations (e.g. school-aged children) serves as the backbone of interventions in areas where helminth infections are highly endemic. In this review, we focus on soil-transmitted helminthiasis (ascariasis, hook

Tropical infection and the lung.

The term "tropical lung" has been used to describe the lungs that are vulnerable to those indigenous diseases that occur with particular prevalence in tropical countries. International travel, student and cultural exchanges and changing immigration patterns are insidiously transforming the face of medicine in Europe and other developed countries. The climates of tropical countries provide an ideal environment for pathogenic organisms, and their vectors and intermediate hosts, to flourish. In an average out-patient department, 20-40% of patients have been seen with respiratory complaints, and 2

Gray foxes (Urocyon cinereoargenteus) as natural definitive hosts of Paragonimus mexicanus.

Paragonimus mexicanus is a trematode that causes pulmonary and extrapulmonary infections in humans, characterized by chest pain, dyspnea, fever, and weight loss. The detection of Paragonimus spp. is primarily achieved through the microscopic observation of eggs in feces, sputum, and pleural fluid. Paragonimus mexicanus has been found in various wild animals, including dogs, cats, raccoons, and opossums. Although the reservoirs of P. mexicanus in Costa Rica are unknown, this study analyzed fecal samples from gray foxes (Urocyon cinereoargenteus) using microscopic and molecular methods. In the m

Experimental transfer of Paragonimus westermani from rodents to rodents following subcutaneous and intraperitoneal routes.

In order to investigate the experimental transfer of Paragonimus westermani from rodents to rodents following subcutaneous and intraperitoneal routes, 13 rats and 23 mice were inoculated with a total of 115 (1 mature and 114 immature) worms of P. westermani subcutaneously and intraperitoneally. The age of worms before transfer was 25-193 days. The transfer was performed immediately after worm collection from rodents which were killed at various intervals from 4 to 144 days after infection. The location, development and size of worms were recorded. An infection rate of 58% (or 21/36) was demons

The natural first intermediate host of Paragonimus siamensis (Miyazaki and Wykoff, 1965) in Thailand.

The first intermediate host of six-known the Paragonimus species in Thailand had not been found until the Filopaludina (Siamopaludina) martensi martensi snail was discovered to maintain the cercariae of a Paragonimus species. An extensive study examined cercarial development through to adult worms by infecting 3 genera of 7 crab species with penetration of cercariae and feeding of snails containing such cercariae. These crabs provided many metacercariae which were fed to cats and bandicoots. The animals gave many Paragonimus adult worms which were characterized as Paragonimus siamensis by the

The life cycle of Paragonimus kellicotti in cats.

Migration and development of Paragonimus kellicotti were studied in cats killed between 1 and 265 days after feeding 15--20 metacercariae from the hearts of naturally infected crayfish. Metacercariae excysted in the intestine and appeared in the peritoneal cavity within 24 hr. Some young flukes penetrated the diaphragm and migrated to the pleural cavity during the first week, but the majority penetrated the pleura between 10 and 14 days. No flukes were found in the pleural cavity after 23 days postinfection. By 4 weeks postinfection, the flukes were well established within lung parenchyma, mai

Infection of a group of boar-hunting dogs with Paragonimus westermani in Miyazaki Prefecture, Japan.

A 5-year-old male mixed-bred boar-hunting dog with a Plott hound background weighing 23 kg was brought to the Veterinary Teaching Hospital, University of Miyazaki, in October 2002. The dog was diagnosed with active infection with the lung fluke Paragonimus westermani by serological testing and also by detection of parasite eggs in his feces. Subsequent examination of four other dogs working with this dog as a boar-hunting team revealed that all five dogs were infected with P. westermani.

Primary lung cancer occurring concomitantly with the cicatrized and calcified ova of a parasite: report of a case.

We report herein a rare case of primary lung cancer that occurred concomitantly with the calcified ova of a parasite. A 58-year-old man was referred to our department after a pulmonary abnormal shadow had been seen on a chest X-ray done at mass screening. A transbronchial lung biopsy (TBLB) revealed the calcified ova of a parasite. Because the possibility of concomitant lung cancer could not be ruled out, a lung biopsy was taken via video-assisted thoracic surgery (VATS). The pathological diagnosis was squamous cell carcinoma, and a left upper lobectomy was serially performed through a postero

Evolutionary relationships between trematodes and snails emphasizing schistosomes and paragonimids.

Snails and digeneans have been associated for at least 200 million years. Their inter-relationships over such a time-span must have been complex and varied. Few studies have attempted to explore these relationships in the light of knowledge of the phylogeny of both host and parasite groups. Here we focus on two important families of digeneans, the Schistosomatidae and the Paragonimidae, for which molecular phylogenies are available. We investigate the types of evolutionary relationships between host and parasite, operating at different phylogenetic depths, that might explain current host speci

Variation of antigenic proteins of eggs and developmental stages of Paragonimus westermani.

Diagnosis of early paragonimiasis is difficult because parasitological evidence is not easily obtained. Antibody tests have been proposed as a good substitute for classical diagnostic techniques. Using the crude extracts of Paragonimus westermani eggs, metacercariae, 4- and 7-week juveniles, and 16-week adults as antigens, we observed the early antibody responses. Sera were obtained from 4 experimental cats, fed 50 metacercariae each, at intervals until 13 weeks post-infection. Antibody (IgG) responses were identified by ELISA using extracts of 4-week juveniles, followed by those of 7- and 16-

Experimental infection of Paragonimus westermani in mice and rats.

To determine the infectivity and maturity of metacercariae of Paragonimus westermani after keeping at low temperature for a long period, 45 mice and 45 rats were each infected with 20 metacercariae which were kept at 4 degrees C for 8 to 234 days. The worm recovery in mice increased with age of worm and reached a peak of 32% at 41-50 days and then decreased with age. The rate in rats first decreased to a lowest point of 6% at 71-100 days and then increased with age. In 42 infected mice and 41 infected rats, 187 immature worms (183 tiny and 4 juvenile ones) and 190 worms (164 tiny, 19 juvenile

Studies on host specificity in Paragonimus westermani: ultrastructural study of inflammatory reactions to the parasite in rat muscles.

Inflammatory reactions to the lung fluke, Paragonimus westermani, in an abnormal host, the rat, were studied by electron microscopy. Inflammatory cells accumulated around the worms in rat muscles, but the reaction varied significantly in intensity as well as distribution of inflammatory cells from worm to worm in the same and different hosts. In an accumulation of inflammatory infiltrates around the worm, eosinophils were more commonly seen, and neutrophils and macrophages were fewer in number. Mast cells appeared rarely. Where inflammatory cells touched to the worms, the tegument of the worms

Further studies on the effect of cyclosporin A on the course of Paragonimus infection in rats.

Paragonimus ohirai-infected rats were treated with cyclosporin A (CyA) at different times during the course of infection. CyA (5 x 80 mg/kg) affected the worm recovery, growth and maturation rates of P. ohirai with respect to control values. This tendency was most remarkable in animals treated 15 days and more after infection with CyA (groups B, +15 to +19 days; C, +25 to +29; D, +35 to +39 and E, +45 to +49). In group A (0 to +4), however, the drug did not affect markedly the growth and maturation of worms, although it significantly lowered worm recovery rates. CyA administration also affecte

Induction of secondary IgE antibody response in rats immunized with X-irradiated metacercariae of Paragonimus ohirai.

Rats reinfected with Paragonimus ohirai (P.o.) elicited little secondary IgE response to adult P.o. antigen. In contrast, rats immunized with X-irradiated (2-10 krad) metacercariae elicited not only a marked primary IgE response comparable to that in normally infected rats, but also a marked secondary IgE response after challenge infection. Rats immunized with 2 krad X-irradiated metacercariae yielded a higher secondary IgE response than with 5 or 10 krad irradiated ones, and elicited a secondary response lasting at least 1 year. An IgE response to antigen of newly excysted juvenile parasites
