Differential gut microbiota modulation associated with anti-Parkinsonian effects of Ophiopogon japonicus across geographical origins and extraction methods.

INTRODUCTION: Ophiopogon japonicus (OJ) is a medicine-food homologous plant widely consumed in East Asia and has attracted attention for its potential anti-Parkinsonian properties. However, the influence of geographical origin and extraction methods on its bioactive profile, gut microbiota modulation, and anti-Parkinsonian effects remain insufficiently understood. METHODS: OJ samples were collected from two major producing regions (Sichuan and Zhejiang) and prepared using two extraction methods (water and ethanol). Chemical profiles were characterized by UHPLC-Q-TOF-MS. Anti-Parkinsonian effe

Attenuation of neuro-inflammation improves survival and neurodegeneration in a mouse model of severe neonatal hyperbilirubinemia.

All pre-term newborns and a high proportion of term newborns develop neonatal jaundice. Neonatal jaundice is usually a benign condition and self-resolves within few days after birth. However, a combination of unfavorable complications may lead to acute hyperbilirubinemia. Excessive hyperbilirubinemia may be toxic for the developing nervous system leading to severe neurological damage and death by kernicterus. Survivors show irreversible neurological deficits such as motor, sensitive and cognitive abnormalities. Current therapies rely on the use of phototherapy and, in unresponsive cases, excha

Focused ultrasound enhances targeted curcumin delivery and alleviates behavioral and neuropathological deficits in a Parkinson's disease mouse model.

INTRODUCTION: Curcumin exhibits potent neuroprotective properties but is limited by poor blood-brain barrier (BBB) permeability. This study aimed to evaluate whether focused ultrasound (FUS)-mediated BBB opening enhances curcumin delivery and therapeutic efficacy in a 6-hydroxydopamine (6-OHDA) mouse model of Parkinson's disease (PD). METHODS: Male C57BL/6 mice with unilateral 6-OHDA lesions received intravenous curcumin with or without FUS targeted to the lesioned striatum for 4 weeks. Behavioral performance was assessed using rotarod and open-field tests. Postmortem analyses included tyrosi

Delayed onset, immunomodulation, and lifespan improvement of SOD1G93A mice after intravenous injection of human mesenchymal stem cells derived from adipose tissue.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by the selective and progressive loss of motor neurons from the spinal cord, brain stem, and motor cortex. Although the hallmark of ALS is motor neuron degeneration, astrocytes, microglia, and T cells actively participate. Pharmacological treatment with riluzole has little effect on the lifespan of the patient. Thus, the development of new therapeutic strategies is of utmost importance. The objective of this study was to verify whether human mesenchymal stem cells (hMSCs) from adipose tissue have therapeutic poten

The nuclear factor erythroid 2-like 2 activator, tert-butylhydroquinone, improves cognitive performance in mice after mild traumatic brain injury.

Traumatic Brain injury affects at least 1.7 million people in the United States alone each year. The majority of injuries are categorized as mild but these still produce lasting symptoms that plague the patient and the medical field. Currently treatments are aimed at reducing a patient's symptoms, but there is no effective method to combat the source of the problem, neuronal loss. We tested a mild, closed head traumatic brain injury model for the effects of modulation of the antioxidant transcription factor Nrf2 by the chemical activator, tert-butylhydroquinone (tBHQ). We found that post-injur

A critical window of CAG repeat-length correlates with phenotype severity in the R6/2 mouse model of Huntington's disease.

The R6/2 mouse is the most frequently used model for experimental and preclinical drug trials in Huntington's disease (HD). When the R6/2 mouse was first developed, it carried exon 1 of the huntingtin gene with ~150 cytosine-adenine-guanine (CAG) repeats. The model presented with a rapid and aggressive phenotype that shared many features with the human condition and was particularly similar to juvenile HD. However, instability in the CAG repeat length due to different breeding practices has led to both decreases and increases in average CAG repeat lengths among colonies. Given the inverse rela

Harpagoside attenuates MPTP/MPP⁺ induced dopaminergic neurodegeneration and movement disorder via elevating glial cell line-derived neurotrophic factor.

Parkinson's disease is a chronic neurodegenerative movement disorder characterized by the loss of dopaminergic neurons in the substantia nigra pars compacta. New therapeutic approaches aiming at delaying or reversing the neurodegenerative process are under active investigations. In this work, we found that harpagoside, an iridoid purified from the Chinese medicinal herb Scrophularia ningpoensis, could not only prevent but also rescue the dopaminergic neurodegeneration in MPTP/MPP(+) intoxication with promising efficacy. Firstly, in cultured mesencephalic neurons, harpagoside significantly atte

Serofendic acid prevents 6-hydroxydopamine-induced nigral neurodegeneration and drug-induced rotational asymmetry in hemi-parkinsonian rats.

Serofendic acid was recently identified as a neuroprotective factor from fetal calf serum. This study was designed to evaluate the neuroprotective effects of an intranigral microinjection of serofendic acid based on behavioral, neurochemical and histochemical studies in hemi-parkinsonian rats using 6-hydroxydopamine (6-OHDA). Rats were injected with 6-OHDA in the presence or absence of serofendic acid, or were treated with serofendic acid on the same lateral side, at 12, 24 or 72 h after 6-OHDA lesion. Intranigral injection of 6-OHDA alone induced a massive loss of tyrosine hydroxylase (TH)-im

A novel mouse model of cerebral microbleeds by targeted Col4a1 editing in adult brain microvessels.

Cerebral small vessel disease is a leading cause of cognitive decline and stroke in the elderly, with cerebral microbleeds (CMBs) serving as a key imaging biomarker. Despite their clinical significance, the pathophysiological mechanisms underlying cerebral small vessel disease remain poorly understood owing to a lack of appropriate animal models. We performed targeted deletion of Col4a1 in brain microvessels of adult mice using brain endothelium-specific adeno-associated virus (AAV)-BR1 vectors with clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9 (CRISPR/C

Concentration-dependent effects of bacterial melanin on new superoxide-producing associates in rat tissues: a rotenone neurotoxic model of parkinson's disease.

BACKGROUND: Oxidative stress plays a central role in the degeneration of dopaminergic neurons and is a key factor in the pathogenesis of Parkinson's disease (PD). Therefore, antioxidant therapy represents a promising therapeutic strategy. This process involves numerous enzymes and signaling pathways that contribute to disease progression. In the present study, bacterial melanin (BM), known for its high biological activity and biostimulatory effects, was applied as a potential antioxidant. We examined quantitative and qualitative alterations in total fractions of biomembrane-associated componen

Pharmacological potential of aqueous extract of Ononis natrix in mitigating early-life lead toxicity: behavioral, biochemical, histological, and immunohistochemical evaluation.

BACKGROUND: Chronic lead (Pb) exposure is a significant public health concern due to its long-term neurobehavioral and physiological effects. Decoctions and infusions of Ononis natrix have been widely used in traditional medicine for their therapeutic effects. While some studies have highlighted its rich bioactive composition and antioxidant properties, there remains a lack of comprehensive scientific research investigating its pharmacological effects. We have previously demonstrated the ameliorative effects of O. natrix against Pb toxicity. However, its protective effects during the juvenile

Purmorphamine mitigates 3-nitropropionic acid-induced neurodegeneration via enhancing the brain-derived neurotrophic factor/protein kinase B/glycogen synthase kinase-3β signaling and attenuation of oxidative/nitrosative stress, neuroinflammation, and apoptosis.

Huntington disease (HD) is a progressive neurodegenerative condition marked by characteristic motor deficits, cognitive decline, and psychiatric disturbances. The mitochondrial toxin 3-nitropropionic acid (3-NP) reliably replicates the HD pathological features by inducing selective striatal and cortical neurodegeneration. Increasing evidence indicates that activation of the Sonic Hedgehog (Shh) signaling could enhance neurotrophic and prosurvival signaling, particularly through modulation of the brain-derived neurotrophic factor (BDNF)/protein kinase B (Akt)/glycogen synthase kinase-3β (GSK-3β

GSDME-N-induced mitochondrial neurotoxicity in early neurodegeneration was suppressed by nicotine via enhancing autophagic flux.

BACKGROUND: Neurodegeneration is a chronic, progressive process initiated by early neurite retraction, a pathological hallmark preceding neuronal death in neurodegenerative disorders such as Parkinson’s disease (PD). Cleavage of gasdermin E (GSDME) releases its N-terminal domain (GSDME-N), which has been shown to mediate mitochondrial dysfunction and the onset of neurodegeneration. However, the therapeutic potential of targeting GSDME-N for early intervention in PD, and whether nicotine, a tobacco component with neuroprotective properties, acts through GSDME modulation remain unexplored. H₂O₂ preconditioning enhances the transplantation-mediated therapeutic effect of bone marrow-derived mesenchymal stem cells after traumatic brain injury.

BACKGROUND: Bone marrow mesenchymal stem cell (BMSC) transplantation has shown therapeutic potential in the treatment of traumatic brain injury (TBI). However, its efficacy is limited by the low survival rate of transplanted BMSCs within the brain microenvironment characterized by persistent inflammation and oxidative stress. Preconditioning BMSCs with low concentrations of hydrogen peroxide (H₂O₂) has been reported to enhance cell viability under oxidative stress. Nevertheless, the mechanisms by which H₂O₂ pretreatment enhances BMSC survival and the role of pretreated BMSCs in promoting funct

The herbicides glyphosate and glufosinate and the cyanotoxin β-N-methylamino-l-alanine induce long-term motor disorders following postnatal exposure: the importance of prior asymptomatic maternal inflammatory sensitization.

BACKGROUND: Prenatal maternal immune activation (MIA) and/or perinatal exposure to various xenobiotics have been identified as risk factors for neurological disorders, including neurodegenerative diseases. Epidemiological data suggest an association between early multi-exposures to various insults and neuropathologies. The "multiple-hit hypothesis" assumes that prenatal inflammation makes the brain more susceptible to subsequent exposure to several kinds of neurotoxins. To explore this hypothesis and its pathological consequences, a behavioral longitudinal procedure was performed after prenata

Germline nuclear-predominant Pten murine model exhibits impaired social and perseverative behavior, microglial activation, and increased oxytocinergic activity.

BACKGROUND: Autism spectrum disorder (ASD) has a strong genetic etiology. Germline mutation in the tumor suppressor gene PTEN is one of the best described monogenic risk cases for ASD. Animal modeling of cell-specific Pten loss or mutation has provided insight into how disruptions to the function of PTEN affect neurodevelopment, neurobiology, and social behavior. As such, there is a growing need to understand more about how various aspects of PTEN activity and cell-compartment-specific functions, contribute to certain neurological or behavior phenotypes.
