Association of insulin resistance with hyperandrogenia in women.

In humans, the skin is a target tissue for androgen action; hair growth and sebum secretion are under active androgen control. An increased production or metabolism of testosterone, the main active androgen, shows up clinically in dermatological symptoms such as hirsutism, hyperseborrheic acne and alopecia. Polycystic ovary syndrome (PCOS) is the most frequent androgen disorder of ovarian function. PCOS patients have amenorrhea or severe oligomenorrhea, increased testosterone levels and most often enlarged polycystic ovaries on ultrasound examination. In addition, many PCOS patients have a ten

Acne Vulgaris: Integrating Pathophysiology with Conventional, Herbal, and Nanocarrier-Based Therapeutic Strategies.

Acne vulgaris is a chronic inflammatory disease of the pilosebaceous unit, affecting 9.4% of the population globally, and is one of the most prevalent dermatoses globally. It is most common in adolescents but can continue through adulthood, causing physical and emotional health deterioration. Acne is a multifactorial process involving genetic factors, hormonal factors, sebum production, hyperkeratinization of the follicular lumen, colonization by Cutibacterium acnes bacteria and inflammation [Ma2.1]. Inflammation is now recognised as a critical process from microcomedone formation to the devel

Scalp Microbiota Dysbiosis in Seborrheic Alopecia and Restoration Following Herbal Extract Shampoo Intervention.

Seborrheic alopecia (SA) is one of the most common forms of hair loss with a complex pathogenesis involving multiple etiological factors. Although the scalp microbiome has been implicated in various scalp disorders, its specific role in the development and progression of SA remains incompletely understood. To characterize the scalp microbiome in SA, we performed high-throughput sequencing of the 16S rRNA gene and ITS region on scalp samples from 41 Chinese SA participants before and after a 12-week intervention with a shampoo containing herbal extracts (ginger root, Polygonum multiflorum, and

Sebaceous immunobiology - skin homeostasis, pathophysiology, coordination of innate immunity and inflammatory response and disease associations.

This review presents several aspects of the innovative concept of sebaceous immunobiology, which summarizes the numerous activities of the sebaceous gland including its classical physiological and pathophysiological tasks, namely sebum production and the development of seborrhea and acne. Sebaceous lipids, which represent 90% of the skin surface lipids in adolescents and adults, are markedly involved in the skin barrier function and perifollicular and dermal innate immune processes, leading to inflammatory skin diseases. Innovative experimental techniques using stem cell and sebocyte models ha

Individual shedder status and the origin of touch DNA.

Due to improved laboratory techniques, touched surfaces and items are increasingly employed as sources of forensic DNA evidence. This has urged a need to better understand the mechanisms of DNA transfer between individuals. Shedder status (i.e. the propensity to leave DNA behind) has been identified as one major factor regulating DNA transfer. It is known that some individuals tend to shed more DNA than others, but the mechanisms behind shedder status are largely unknown. By comparing the amounts of DNA deposited from active hands (i.e. used "as usual") and inactive hands (i.e. not allowed to

Findings of illicit drugs in hair of children at different ages.

Hair is a preferred material to detect exposure or use of illegal drugs in children. In the present study, we investigated a total of 387 hair samples for commonly applied illegal drugs of children up to 16 years. Analysis was by liquid chromatography/mass spectrometry with LOQs of 0.01 ng/mg hair for all analytes except tetrahydrocannabinol carboxylic acid with an LOQ of 0.1 pg/mg hair. Results were firstly compared with our in-house statics on results from adults' hair, and secondly to literature data. We started from the assumption that drug concentrations decrease with increasing age.Resul

A comparative study of biological and metabolic biomarkers between healthy individuals and patients with acne vulgaris: A cross-sectional study protocol.

Acne is a multifactorial dermatosis, which is influenced not only by hormones but also by the biochemical relationship between them and the pilosebaceous unit. Inflammatory cytokines, chemokines, active oxygen, and zinc are known to be associated with the development of acne. Further, steroid metabolism is known as one of the important factors related to sebum secretion and comedone formation in acne. However, there is a lack of studies comparing these human biomarkers between healthy individuals and patients with acne. In particular, no study has investigated the relationship between human bi

ADT-G as a promising biomarker for peripheral hyperandrogenism in adult female acne.

Acne vulgaris is a chronic inflammatory disease that affects the pilosebaceous unit. Recent studies have shown an increasing number of cases of acne in adult women. These cases are predominantly normoandrogenic and present some clinical differences compared to adolescent acne. Local glandular metabolism turns some weak hormonal precursors into more active substances that increase the production of sebum, leaving these areas more prone to an increasing the colonization by Propionibacterium acnes (P. acnes). Our objective was to evaluate the usefulness of an androgenic metabolite as an adult fem

11β-hydroxysteroid dehydrogenase type 1 is expressed in human sebaceous glands and regulates glucocorticoid-induced lipid synthesis and toll-like receptor 2 expression in SZ95 sebocytes.

BACKGROUND: Glucocorticoids (GCs) affect the pathophysiology of sebaceous glands, causing development or exacerbation of acne. The availability of GCs is regulated by isoenzymes of 11β-hydroxysteroid dehydrogenase (11βHSD) at tissue-specific levels. 11βHSD type 1 (HSD11β1) is a reductase, catalysing the conversion of cortisone to active cortisol, and is highly expressed in liver and adipose tissue. Recently, HSD11β1 was observed in human skin in keratinocytes and fibroblasts. OBJECTIVES: To investigate the expression of HSD11β1 in sebaceous glands of normal and acne-involved skin, and to exam

Identification of Propionibacterium acnes by polymerase chain reaction for amplification of 16S ribosomal RNA and lipase genes.

Propionibacterium acnes belongs to the cutaneous flora and is present in sebaceous follicles. The fatty acids that are released from sebum triglycerides by the action of this bacterial lipase play an important role in the pathogenesis of acne vulgaris. P. acnes is also involved in postoperative disorders and opportunistic infections in immunosuppressed hosts. Recently, it has been proposed that P. acnes causes sarcoidosis. Therefore, rapid isolation and identification of P. acnes is important. This study evaluated the polymerase chain reaction (PCR) for the detection of the 16S rRNA and lipase

Round Table Discussion: Aesthetic Treatment Considerations for the Perimenopausal & Menopausal Patient.

BACKGROUND: Menopause is a normal feature of the female aging process that commonly occurs during the 6th decade of life. During perimenopause, many negative effects begin to occur affecting the skin that are associated with decreasing production of estrogen, progesterone, and testosterone. Estrogen supports the extracellular matrix, collagen production, and dermal blood supply, while progesterone promotes extracellular matrix formation, dermal blood supply, and dermal water retention. Decreased testosterone impacts cutaneous blood supply and sebum production, causing skin dryness. AIMS: The

Human exposure to polycyclic aromatic hydrocarbons and metal(oid)s from artificial turf and synthetic/recycled rubber materials: Insights from dermal bioaccessibility and risk assessment.

Synthetic and recycled rubber materials and artificial turf are widely used in public recreational settings, raising concerns about human exposure to polycyclic aromatic hydrocarbons (PAHs) and metal(oid)s. In this study, these contaminants were analysed in 37 samples, including rubber-based playground and athletic track flooring, as well as artificial turf plastic fibres and crumb rubber infill for football pitches, comprising both in-use and new materials. As dermal contact is the main exposure pathway, dermal bioaccessibility was assessed using an in vitro physiologically based extraction t

Dihalogenated indoles with antimicrobial activity against C. acnes and polymicrobial biofilms.

Acne vulgaris is a chronic inflammatory skin disorder in which Cutibacterium acnes contributes to disease persistence through biofilm formation, lipid metabolism, and production of inflammatory metabolites within the pilosebaceous unit. Targeting bacterial physiological pathways that sustain these processes represents a potential therapeutic strategy beyond conventional antibiotic approaches. In this study, we evaluated a panel of halogenated indole derivatives and identified 6-bromo-4-iodoindole as a potent inhibitor of C. acnes growth and biofilm formation. The compound exhibited a minimum i

Analysis of blood untargeted metabolomic characteristics of different subtypes of rosacea.

BACKGROUND: Rosacea is a chronic inflammatory skin disease characterized by vascular and neurological dysregulation, presenting with diverse clinical subtypes whose pathological mechanisms remain incompletely elucidated. Recent studies suggest that metabolic dysregulation may play a key role in disease onset and progression; however, systematic metabolomic studies targeting different subtypes remain limited. OBJECTIVE: This study employed untargeted metabolomic analysis to systematically compare plasma metabolic characteristic differences between patients with erythematotelangiectatic rosacea
