Genome sequence analysis of Pseudomonas extremaustralis provides new insights into environmental adaptability and extreme conditions resistance.

The genome of the Antarctic bacterium Pseudomonas extremaustralis was analyzed searching for genes involved in environmental adaptability focusing on anaerobic metabolism, osmoregulation, cold adaptation, exopolysaccharide production and degradation of complex compounds. Experimental evidences demonstrated the functionality of several of these pathways, including arginine and pyruvate fermentation, alginate production and growth under cold conditions. Phylogenetic analysis along with genomic island prediction allowed the detection of genes with probable foreign origin such as those coding for

Metabolic and Gene Expression Profile Reveal Genetic Basis of Reproductive Stage Drought Tolerance in a Drought-Defying Rice Mutant.

Mutation breeding provides a powerful route to generate novel genetic variation and to uncover genes underlying stress tolerance. Identifying mechanisms that sustain rice productivity under water-limited conditions is a central challenge in plant biology. We developed a gamma-ray-mutagenised population of Oryza sativa L. cv. IR64 and systematically screened the M2, M3 and M4 generations for drought tolerance at the seedling, pre-flowering and post-flowering stages. Through this multi-stage, multi-generational, and multi-environment phenotypic selection, we identified a drought-defying (DD) mut

The SigD regulon of Mycobacterium abscessus determines cell envelope composition and antibiotic susceptibility.

A major determinant of the exceptional intrinsic resistance of M. abscessus is the lipid-rich cell envelope, yet the regulatory systems that remodel envelope-associated pathways remain poorly defined. Here, we determine the σD regulon in M. abscessus and establish its role in cell envelope homeostasis and intrinsic resistance to hydrophobic antibiotics. RNA-Seq analysis of a MabΔsigD mutant identified 447 differentially expressed genes, while ChIP-Seq mapped 72 σD binding sites and defined a conserved promoter motif (GTAACA/G-N16-CGAT). Using a combination of σD binding, motif orientation and

Polypeptide Sequence Effects on Virus Thermostability in Complex Coacervate Formulations.

Current vaccine formulations heavily rely on cold chains to avoid degradation during transportation and storage. Vaccines typically degrade when exposed to temperatures outside the 2-8 °C range, leading to waste and logistical challenges, particularly in rural areas. This study investigates the ability of poly(lysine)- and poly(glutamate)-based peptide coacervates to improve the thermal stability of porcine parvovirus (PPV), a model nonenveloped viral vaccine. We hypothesized that both the length and specific amino acid sequence of the peptides forming the coacervates would influence the stabi

[Effects of vacuum freeze-drying based on different lyoprotectants on the stability of foot-and-mouth disease virus-like particles].

Vaccination is a crucial strategy for the prevention and control of infectious diseases. Virus-like particles (VLPs), composed of structural proteins, have garnered significant attention as a novel type of vaccine due to their excellent safety and immunogenicity. However, similar to most vaccine antigens, VLPs exhibit insufficient thermal stability, which not only restricts the widespread application of vaccines but also increases the risk of vaccine inactivation. This study aims to enhance the stability and shelf life of VLPs derived from type A foot-and-mouth disease virus (FMDV) by employin

Amplification, purification, and lyophilization of mycobacteriophages for therapeutic use.

Infections with nontuberculous mycobacterium (NTM) strains are frequently nonresponsive to antibiotic regimens, even after multiple drugs are used for extensive periods of months or years. Therapy with bacteriophages specific for NTM clinical isolates shows considerable promise in compassionate-use single-patient cases but, like antibiotics, must be administered over long periods of time and typically intravenously. It is thus important that the phage preparations are highly purified and stable to avoid adverse reactions or decay of viability. Here, we describe methods for purification of myco

Fab-conjugated PLGA nanoparticles effectively target cancer cells expressing human CD44v6.

Targeting of CD44 isoforms containing exon v6 (CD44v6) represents a viable strategy for the therapy and/or early diagnosis of metastatic cancers of the epithelium (e.g. gastric and colorectal cancer). We developed and characterized poly(lactic-co-glycolic acid) (PLGA)-based nanoparticles (NPs) modified with polyethylene glycol (PEG) and engrafted, by site-directed conjugation, with an engineered human Fab that specifically target human CD44v6 (v6 Fab-PLGA NPs). The v6 Fab-PLGA NPs displayed spherical morphology around 300 nm and were negatively charged. They strongly bound to a CD44v6-derived

Analysis of the potency of various low molecular weight chemical chaperones to prevent protein aggregation.

Newly translated proteins must undergo proper folding to ensure their function. To enter a low energy state, misfolded proteins form aggregates, which are associated with many degenerative diseases, such as Huntington's disease and chronic kidney disease (CKD). Recent studies have shown the use of low molecular weight chemical chaperones to be an effective method of reducing protein aggregation in various cell types. This study demonstrates a novel non-biased assay to assess the molecular efficacy of these compounds at preventing protein misfolding and/or aggregation. This assay utilizes a thi

Regulation of GacA in Pseudomonas chlororaphis Strains Shows a Niche Specificity.

The GacS/GacA two-component system plays a central role in the regulation of a broad range of biological functions in many bacteria. In the biocontrol organism Pseudomonas chlororaphis, the Gac system has been shown to positively control quorum sensing, biofilm formation, and phenazine production, but has an overall negative impact on motility. These studies have been performed with strains originated from the rhizosphere predominantly. To investigate the level of conservation between the GacA regulation of biocontrol-related traits in P. chlororaphis isolates from different habitats, the stud

Differences between reversible (self-association) and irreversible aggregation of rHuG-CSF in carbohydrate and polyol formulations.

Severe immunogenic and other debilitating human disorders potentially induced by protein aggregates have brought this phenomenon into the focus of biopharmaceutical science over the past decade. Depending on its driving forces, the process induced in the model protein rHuG-CSF may be either reversible or irreversible, resulting in the assembly of self-associated protein species or irreversible aggregates of various final morphologies. The aim of our work was to investigate the correlation between irreversible and reversible aggregation and the protective effect of non-specific formulation stab

[Riddles in human tuberculous infection].

Tuberculosis is indeed an infectious disease caused by Mycobacterium tuberculosis. However, only a small percentage of individuals infected develops overt disease, tuberculosis whereas the infected bacilli persist alive years long within the vast majority of persons infected but remained healthy. There are several riddles or enigmas in the natural history of M. tuberculosis infection in humans. Some of them are as follows: 1. What is the virulence of M. tuberculosis? 2. How does M. tuberculosis persist dormant within the host? 3. What determines the development of disease from remaining health

Mincle exacerbates neuroinflammation and dopaminergic neurodegeneration via synergistic crosstalk with TLR4.

Microglial activation and subsequent neuroinflammation are critically implicated in the pathogenesis and progression of Parkinson's disease (PD). Macrophage-inducible C-type lectin (Mincle) is a pivotal pattern recognition receptor that participates in the regulation of innate immune responses. Nevertheless, the functional role of Mincle in mediating dopaminergic neuronal injury in PD remains poorly defined. In the present study, we observed a marked upregulation of Mincle expression in microglia in PD mouse models. Pharmacological inhibition of Mincle via Anti-Mincle antibody treatment effect

Comparative In Vitro Analysis of Five Starch Sources: Nutrient Release Patterns and Functional Effects in a Simulated Gastrointestinal Environment.

Corn, paddy, wheat, sorghum, and cassava serve as the primary energy sources in both human and animal diets. This study aimed to evaluate their nutrient release patterns in a simulated gastrointestinal environment and to assess the in vitro biological activity of the metabolites produced during digestion. The results showed that wheat exhibited the highest dry matter degradation in the stomach-jejunum-ileum digestion stage, while wheat and paddy showed the highest crude protein degradation compared with the other starch sources. In addition, wheat had a higher total free sugar concentration th

Phylogenetic analysis, metabolic profiling, and environmental adaptation of strain LCG007: a novel Rhodobacteraceae isolated from the East China Sea intertidal zone.

Strain LCG007, isolated from Lu Chao Harbor's intertidal water, phylogenetically represents a novel genus within the family Rhodobacteraceae. Metabolically, it possesses a wide array of amino acid metabolic genes that enable it to thrive on both amino acids or peptides. Also, it could hydrolyze peptides containing D-amino acids, highlighting its potential role in the cycling of refractory organic matter. Moreover, strain LCG007 could utilize various carbohydrates, including mannopine and D-apiose-compounds primarily derived from terrestrial plants-demonstrating its capacity to degrade terrestr

Immunogenic profiling of Mycobacterium tuberculosis Rv1513 reveals its ability to switch on Th1 based immunity.

Tuberculosis (TB) is one of the infectious diseases caused by the pathogen Mycobacterium tuberculosis that continuously threatens the global human health. Bacillus Calmette-Guérin (BCG) vaccine is the only vaccine that has been used clinically to prevent tuberculosis in recent centuries, but its limitations in preventing latent infection and reactivation of tuberculosis do not provide full protection. In this study, we selected the membrane-associated antigen Rv1513 of Mycobacterium. In order to achieve stable expression and function of the target gene, the prokaryotic expression recombinant v
