[Effects of Thap6 gene knockout on emotional and social behaviors in mice].

OBJECTIVES: THAP domain-containing protein 6 (THAP6) is a member of the THAP family and contains a conserved THAP zinc-finger domain, allowing it to regulate gene expression as a transcription factor. Previous studies have shown that THAP6 may be involved in macrophage activation and transcriptional regulation related to social stimuli; however, whether THAP6 affects emotional behavior and brain tissue structure has not yet been reported. This study aims to clarify the effects of Thap6 gene deletion on emotional behaviors in mice, preliminarily explore the effects of Thap6 gene knockout on mou

Near-complete adaptation of the PRiMA knockout to the lack of central acetylcholinesterase.

Acetylcholinesterase (AChE) rapidly hydrolyzes acetylcholine. At the neuromuscular junction, AChE is mainly anchored in the extracellular matrix by the collagen Q, whereas in the brain, AChE is tethered by the proline-rich membrane anchor (PRiMA). The AChE-deficient mice, in which AChE has been deleted from all tissues, have severe handicaps. Surprisingly, PRiMA KO mice in which AChE is mostly eliminated from the brain show very few deficits. We now report that most of the changes observed in the brain of AChE-deficient mice, and in particular the high levels of ambient extracellular acetylcho

Parkin-mediated protection of dopaminergic neurons in a chronic MPTP-minipump mouse model of Parkinson disease.

Loss-of-function mutations in the ubiquitin ligase parkin are the major cause of recessively inherited early-onset Parkinson disease (PD). Impairment of parkin activity caused by nitrosative or dopamine-related modifications may also be responsible for the loss of dopaminergic (DA) neurons in sporadic PD. Previous studies have shown that viral vector-mediated delivery of parkin prevented DA neurodegeneration in several animal models, but little is known about the neuroprotective actions of parkin in vivo. Here, we investigated mechanisms of neuroprotection of overexpressed parkin in a modified

Grafts of adult subependymal zone neuronal progenitor cells rescue hemiparkinsonian behavioral decline.

Neuronal progenitor cells (NPCs) residing in the adult subependymal zone (SEZ) are a potential source of expandable cells for autologous transplantation to treat Parkinson's Disease and other types of brain injury. We have previously demonstrated the capacity of transplanted adult SEZ NPCs for heterotypic differentiation in the hippocampus. To further characterize the therapeutic potential of these cells, NPCs expanded from the adult rat SEZ were grafted to the striatum of normal and 6-OHDA lesioned adult rats. Grafted cells were assessed for neuronal differentiation, and lesioned animals were

Hypothalamic expression of huntingtin causes distinct metabolic changes in Huntington's disease mice.

OBJECTIVE: In Huntington's disease (HD), the disease-causing huntingtin (HTT) protein is ubiquitously expressed and causes both central and peripheral pathology. In clinical HD, a higher body mass index has been associated with slower disease progression, indicating the role of metabolic changes in disease pathogenesis. Underlying mechanisms of metabolic changes in HD remain poorly understood, but recent studies suggest the involvement of hypothalamic dysfunction. The present study aimed to investigate whether modulation of hypothalamic HTT levels would affect metabolic phenotype and disease f

LEP Knockdown Improves Functional Recovery After Spinal Cord Injury and Potentially Regulates Microglial Polarization via JAK-STAT Signaling Pathway.

Neuroinflammation following spinal cord injury (SCI) is primarily driven by abnormal microglial activation and represents a major barrier to neurological recovery. This study aimed to investigate the regulatory role of leptin (LEP) in SCI, with a particular focus on its effects on the JAK-STAT signaling pathway, microglial polarization, and neuronal injury. Transcriptome sequencing of spinal cord tissues was performed to identify differentially expressed genes (DEGs), followed by enrichment, immune cell infiltration, and protein-protein interaction (PPI) analyses to determine key pathways invo

Salidroside derivative SHPL-49 enhances synaptic remodeling in BCCAO rats via the CDK5/p35/p25 signaling pathway.

BACKGROUND: In recent years, significant attention has been directed toward the development of therapeutic agents targeting cognitive and motor dysfunction following ischemic stroke. Our research team synthesized SHPL-49, a salidroside derivative, which has demonstrated neuroprotective effects in rat models of ischemic stroke in prior studies. However, the underlying mechanisms remain poorly understood. This study aims to evaluate the therapeutic potential of orally administered SHPL-49 over a 28-day period in rats with bilateral common carotid artery occlusion (BCCAO), specifically focusing o

Identification of a nanomolar, selective MAO-B inhibitor from piperidine-based chalcones with anti-Parkinson potential.

A library of twenty-seven piperidine-based chalcones (PBCs) was designed, synthesised via microwave-assisted methodology, and subsequently evaluated for their inhibitory activity against monoamine oxidases (MAO-A/B), acetylcholinesterase (AChE), and butyrylcholinesterase (BChE). The PBCs exhibited potent and selective inhibition of MAO-B, with IC50 values ranging from 0.014 to >40 μM, while activity towards MAO-A was negligible (>40 μM). Among them, PBC21 was the most active (IC50 = 0.014 ± 0.0026 μM), whereas PBC1, PBC2, and PBC5 showed comparable potencies around 0.5-0.6 μM. Kinetic and reve

L-Serine Attenuates Metabolic and Behavioural Features of Diabetic Neuropathy with Dose-Dependent Central Proteomic Correlates in a Rat Model.

Diabetic neuropathy (DN) is a multifactorial complication of diabetes mellitus driven by chronic hyperglycemia, insulin resistance, and disturbed metabolic homeostasis, leading to progressive injury of both the peripheral and central nervous systems. This study investigated whether L-serine supplementation could attenuate DN through dose-dependent metabolic and neuroprotective mechanisms in a high-fat diet (HFD) plus streptozotocin (STZ)-induced diabetic rat model. Male Wistar rats (n = 8 per group) were allocated to five groups: normal control (NC), diabetic control (DC), pioglitazone (PIO; 1

Neural-induced human adipose tissue-derived stem cell secretome exerts neuroprotection against rotenone-induced Parkinson's disease in rats.

BACKGROUND: Parkinson's disease (PD) is a multifactorial disease that involves genetic and environmental factors, which play an essential role in the pathogenesis of PD. Mesenchymal stem cells release a set of bioactive molecules called "secretome" that regulates intercellular communication and cargo transfer in signaling pathways for PD treatment. Thus, this study aimed to evaluate the neuroprotective effects of neural-induced human adipose tissue-derived stem cell (NI-hADSC)-conditioned medium (NI-hADSC-CM) and its exosomes (NI-hADSC-Exo) in a rotenone (ROT)-induced model of PD in rats. Neural Stem/Progenitor Cell Therapy in Patients and Animals with Amyotrophic Lateral Sclerosis: A Systematic Review and Meta-analysis.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative malady that causes progressive degeneration and loss of motor neuron function in the brain and spinal cord, eventually resulting in muscular atrophy, paralysis, and death. Neural stem/progenitor cell (NSPC) transplantation can improve bodily function in animals and delay disease progression in patients with ALS. This paper summarizes and analyzes the efficacy and safety of neural stem/progenitor cell (NSPC) transplantation as a treatment for ALS, aiming to improve function and delay disease progression in patients. We present a summary

Neuroprotective and neuro-rehabilitative effects of acute purinergic receptor P2X4 (P2X4R) blockade after ischemic stroke.

Stroke remains a leading cause of disability in the United States. Despite recent advances, interventions to reduce damage and enhance recovery after stroke are lacking. P2X4R, a receptor for adenosine triphosphate (ATP), regulates activation of myeloid immune cells (infiltrating monocytes/macrophages and brain-resident microglia) after stroke injury. However, over-stimulation of P2X4Rs due to excessive ATP release from dying or damaged neuronal cells can contribute to ischemic injury. Therefore, we pharmacologically inhibited P2X4R to limit the over-stimulated myeloid cell immune response and

Sex-specific metabolic and microbial remodeling in a rotenone-induced rat model of Parkinson's disease following nicotine administration.

BACKGROUND: Parkinson's disease (PD) is a neurodegenerative disorder with established sex differences in incidence and progression. Epidemiological evidence suggests nicotine may confer protection against PD, but its mechanisms, particularly regarding sex-specific effects, remain unclear. This study investigated the neuroprotective mechanisms of nicotine in a rotenone-induced PD rat model, with a specific focus on evaluating sex-dependent modulation across behavioral, pathological, and gut-related outcomes. METHODS: Male and female Sprague-Dawley rats were administered rotenone (2 mg/kg/day,

Impact of Transchalcone on Neurological Outcomes in Cerebral Ischemia-reperfusion Injury in Rat: Role of AMP-activated Protein Kinase-mitochondrial Signaling Pathways.

Cerebral ischemia-reperfusion (CIR) injury results in significant secondary brain damage after ischemic stroke due to oxidative stress, mitochondrial dysfunction, and neuroinflammation. Transchalcone (TCH), a polyphenolic compound, exhibits antioxidant and anti-inflammatory properties that may contribute to neuroprotection. The present study investigated the potential protective effects of TCH in a rat model of CIR, focusing on its impact on the activation of AMP-activated protein kinase (AMPK) pathway, mitochondrial function, and inflammatory mediators. Sixty adult Sprague-Dawley rats were ra

Mitofusin 2 plays a critical role in maintaining the functional integrity of the neuromuscular-skeletal axis.

PURPOSE: Mitofusin 2 (Mfn2) is one of two mitofusins involved in regulating mitochondrial size, shape and function, including mitophagy, an important cellular mechanism to limit oxidative stress. Reduced expression of Mfn2 has been associated with impaired osteoblast differentiation and function and a reduction in the number of viable osteocytes in bone. We hypothesized that the genetic absence of Mfn2 in these cells would increase their susceptibility to aging-associated metabolic stress, leading to a progressive impairment in skeletal homeostasis over time. METHODS: Mfn2 was selectively del

Lack of riluzole efficacy in the progression of the neurodegenerative phenotype in a new conditional mouse model of striatal degeneration.

BACKGROUND: Huntington's disease (HD) is a rare familial autosomal dominant neurodegenerative disorder characterized by progressive degeneration of medium spiny neurons (MSNs) located in the striatum. Currently available treatments of HD are only limited to alleviating symptoms; therefore, high expectations for an effective therapy are associated with potential replacement of lost neurons through stimulation of postnatal neurogenesis. One of the drugs of potential interest for the treatment of HD is riluzole, which may act as a positive modulator of adult neurogenesis, promoting replacement of

Electroacupuncture Inhibits Cartilage Degeneration in a Rat Knee Osteoarthritis (KOA) Model by Suppressing ADAMTS5 Expression.

Background Knee osteoarthritis (KOA) is characterized by cartilage degradation, osteophyte formation, and synovitis. Cartilage degradation in KOA begins with the loss of aggrecan, primarily due to A Disintegrin and Metalloproteinase with Thrombospondin Motif 5 (ADAMTS5), which is produced by chondrocytes and synovial cells and a key target for therapeutic intervention. Current treatments for KOA primarily focus on pain relief, as disease-modifying osteoarthritis drugs (DMOADs) remain unavailable. Electroacupuncture (EA), applying electrical stimulation to acupoints, has been investigated for i

Impact of hydroxytyrosol on stroke: tracking therapy response on neuroinflammation and cerebrovascular parameters using PET-MR imaging and on functional outcomes.

Immune cells have been implicated in influencing stroke outcomes depending on their temporal dynamics, number, and spatial distribution after ischemia. Depending on their activation status, immune cells can have detrimental and beneficial properties on tissue outcome after stroke, highlighting the need to modulate inflammation towards beneficial and restorative immune responses. Novel dietary therapies may promote modulation of pro- and anti-inflammatory immune cell functions. Among the dietary interventions inspired by the Mediterranean diet, hydroxytyrosol (HT), the main phenolic component o

Natural killer cells aggravate neuroinflammation through microglial type I interferon pathway activation after traumatic brain injury.

BACKGROUND: Traumatic brain injury (TBI) compromises blood-brain barrier integrity, facilitating infiltration of peripheral immune cells into cerebral tissue. While natural killer (NK) cells - key innate lymphocytes - have been established as critical mediators of secondary neuroinflammation in acute brain injuries including ischemic stroke and intracerebral hemorrhage, their precise involvement in TBI pathophysiology remains undetermined. This study systematically elucidates the role of NK cells in TBI progression and proposes novel immunomodulatory therapeutic strategies for post-TBI interve

CaMKIIα inhibition facilitates functional recovery after preterm hypoxic-ischemic brain injury by disrupting microglial GPNMB signaling.

BACKGROUND: Preterm brain injury involves white matter damage and aberrant synaptic development, yet the mechanisms linking these pathologies remain unclear. Our previous work demonstrated that hypoxia-ischemia (HI) upregulates microglial glycoprotein non-metastatic melanoma protein B (GPNMB), which is released and correlates with abnormal synaptogenesis and neurobehavioral deficits. However, the downstream signaling pathways by which GPNMB impairs synaptic function are unknown. Given the central role of hippocampal Ca2⁺/calmodulin-dependent protein kinase II (CaMKII) and its substrate, the gl
