Enterobacter hormaechei, a new species of the family Enterobacteriaceae formerly known as enteric group 75.

The name Enterobacter hormaechei is proposed for a new species of the family Enterobacteriaceae, formerly called Enteric Group 75, which consists of 23 strains, 22 of which were isolated from humans. DNAs from 12 E. hormaechei strains tested were highly related to the type strain (ATCC 49162) by DNA hybridization, using the hydroxyapatite method (80 to 97% in 60 degrees C reactions; 80 to 90% in 75 degrees C reactions). The strains were most closely related (50 to 63%) to Enterobacter cloacae, Enterobacter dissolvens, Enterobacter taylorae, and Enterobacter nimipressuralis. E. hormaechei strai

Magnetic Field-Driven Regulation of Bioactive Metabolites and Metabolic Enzyme Inhibition in Sanghuangporus vaninii.

The effects of static magnetic field (SMF) treatment on the solid-state culture of Sanghuangporus vaninii (SV) were investigated to enhance metabolite production and bioactivity. SMF parameters including intensity, exposure duration, and temperature were optimized, and treatment at 4 mT for 2 h per day produced the most pronounced effects, increasing total flavonoid (TFC), polyphenol (TPC), and triterpenoid (TTC) contents by 61-438% compared with the control. Ultrasonic extraction and semi-preparative chromatography enabled the isolation of three key compounds: D-(+)-trehalose (1), 5,7-dihydro

Integrated transcriptomic and metabolomic analyses reveal the molecular mechanisms of sugar accumulation and yield trade-off in sugar beet.

BACKGROUND: The negative correlation between taproot yield and sugar content remains a persistent challenge in sugar beet (Beta vulgaris L.) breeding. This trade-off reflects the competitive partitioning of finite assimilated carbon between structural growth and vacuolar storage. However, the transcriptional and metabolic mechanisms underlying this shift remain poorly characterized. METHODS: We integrated transcriptomic and widely-targeted metabolomic data across five developmental stages of taproots from two lines with contrasting phenotypes: a high-sugar/low-yield line (Group K) and a low-s

Establishment of a field-adapted biotechnological protocol for Duck semen cryopreservation.

Cryopreservation of duck semen is a strategic tool for safeguarding genetic resources and maintaining breeding continuity, particularly in the context of sanitary crises such as highly pathogenic avian influenza outbreaks. However, progress has been limited by the intrinsic low cryotolerance of avian spermatozoa and the practical constraints of industrial artificial insemination. The study aimed to develop and validate cryopreservation protocols for Pekin (Anas platyrhynchos) and Muscovy (Cairina moschata) duck semen using permeating cryoprotectants compatible with direct insemination without

Descriptive Ultrastructural Characterization of Cryopreserved Murine Liver Tissue across Different Freezing/Cooling Methods and Cryoprotective Agents.

INTRODUCTION: Cryopreservation of single cells is well established; however, complex tissues remain challenging due to cellular heterogeneity and ice crystal-induced damage. Alterations in tissue architecture, for example, ice crystal formation, can compromise diagnostic usability, especially when subtle ultrastructural features are essential for interpretation. OBJECTIVES: To assess the impact of different freezing protocols, with or without cryoprotective agents (CPAs), on tissue (ultra)structure and nucleic acid integrity. METHODS: Setup A: murine livers were preserved using combinations

Disaccharidase Inhibitory Activity of Thai Plant Extracts.

Background/Objectives: Intestinal α-glucosidases, including maltase, sucrase, and trehalase, are key enzymes responsible for the final steps of carbohydrate digestion. Although Thai medicinal plants possess diverse bioactivities, most previous studies on plant-derived α-glucosidase inhibitors have focused on single-enzyme assays, primarily maltase, and lack systematic comparison of the three major intestinal disaccharidases-maltase, sucrase, and trehalase. This study aimed to determine the kinetic properties of rat intestinal α-glucosidases and evaluate the inhibitory potential of selected Tha

Hybrid genome assembly and phenotypic assays reveal carbohydrate metabolism diversity in Lacticaseibacillus strains.

Investigation of carbohydrate metabolism in lactic acid bacteria is essential for the rational selection of strains for fermentation processes, particularly in emerging applications involving non-conventional substrates or building of synthetic microbial consortia. However, establishing robust genotype-phenotype relationships remains challenging, as gene presence alone often fails to explain observed metabolic traits without considering the genomic context and regulatory architecture. In the present study, we combined hybrid genome assembly (Illumina and Oxford Nanopore) with high-throughput p

Modulation of NF-κB in tuberculosis - understanding host-pathogen interactions and therapeutic opportunities.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (M. tb), remains a major global health challenge and a leading cause of infectious mortality. Central to TB pathogenesis is the nuclear factor-kappa B (NF-κB) signaling network, a master regulator of inflammation, immune activation, and host defense. This review highlights the complex molecular interplay between M. tb virulence determinants and host NF-κB pathways, emphasizing how the pathogen strategically manipulates transcriptional responses to promote survival, persistence, and transmission. Early during infection, cell wall-derived m

Virtual Screening of Cathelicidin-Derived Anticancer Peptides and Validation of Their Production in the Probiotic Limosilactobacillus fermentum KUB-D18 Using Genome-Scale Metabolic Modeling and Experimental Approaches.

The development of anticancer peptides (ACPs) has emerged as a promising strategy in targeted cancer therapy due to their high specificity and therapeutic potential. Cathelicidin-derived antimicrobial peptides represent a particularly attractive class of ACPs, yet systematic evaluation of their anticancer activity remains limited. In this study, we conducted virtual screening of eight cathelicidin-derived peptides (AL-38, LL-37, RK-31, KS-30, KR-20, FK-16, FK-13, and KR-12) to assess their potential against colon cancer. Among these, LL-37 and FK-16 were identified as the most promising candid

Dexamethasone-loaded platelet-inspired nanoparticles improve intracortical microelectrode recording performance.

Long-term robust intracortical microelectrode (IME) neural recording quality is negatively affected by the neuroinflammatory response following microelectrode insertion. This adversely impacts brain-machine interface (BMI) performance for patients with neurological disorders or amputations. Recent studies suggest that the leakage of blood-brain barrier (BBB) and microhemorrhage caused by the IME insertions lead to the increased neuroinflammation and reduced neural recording performance. Additionally, a sustained presence of activated platelets and coagulation factors is found near the insertio

Optimizing riboflavin delivery with co-crystal and in situ hydrogel formulations for management of keratoconus: A comprehensive investigation with in vitro, ex vivo and in vivo studies.

Keratoconus is a progressive disease characterized by corneal thinning and conical deformation. Corneal cross-linking, a common treatment, strengthens collagen fibers using vitamin B2 (riboflavin) and UVA light. However, the surgical removal of the corneal epithelium (Epi-Off) required for riboflavin penetration causes complications and may affect treatment success. To address this, research has focused on delivering riboflavin without removing the epithelium (Epi-On). In this study, riboflavin-based hydrogel and co-crystal formulations were developed and evaluated through in vitro, ex vivo, a

Liposome-based dry powder vaccine immunization targeting the lungs induces broad protection against pneumococcus.

Streptococcus pneumoniae is an important human pathogen. Currently used conjugate vaccines are effective against invasive disease, but protection is restricted to serotypes included in the formulation, leading to serotype replacement. Furthermore, protection against non-invasive disease is reported to be considerably lower. The development of a serotype-independent vaccine is thus important and Pneumococcal surface protein A (PspA) is a promising vaccine candidate. PspA shows some diversity and can be classified in 6 clades and 3 families, with families 1 and 2 being the most frequent in clini

The performance of tongue swabs for detection of pulmonary tuberculosis.

INTRODUCTION: Oral and/or tongue swabs have demonstrated ability to detect Mycobacterium tuberculosis (Mtb) in adults with pulmonary tuberculosis (TB). Swabs provide useful alternative specimens for diagnosis of TB using molecular assays however, the diagnostic pickup by culture requires further improvement and development. Several studies identified the presence of differentially culturable tubercle bacilli (DCTB) populations in a variety of clinical specimens. These organisms do not grow in routine laboratory media and require growth factors in the form of culture filtrate (CF) from logarith

The Role of Macrophage-Inducible C-Type Lectin in Different Stages of Chronic Liver Disease.

The macrophage-inducible C-type lectin (mincle) is part of the innate immune system and acts as a pattern recognition receptor for pathogen-associated molecular patterns (PAMPS) and damage-associated molecular patterns (DAMPs). Ligand binding induces mincle activation which consequently interacts with the signaling adapter Fc receptor, SYK, and NF-kappa-B. There is also evidence that mincle expressed on macrophages promotes intestinal barrier integrity. However, little is known about the role of mincle in hepatic fibrosis, especially in more advanced disease stages. Mincle expression was measu

Energy disorders caused by mitochondrial dysfunction contribute to α-amatoxin-induced liver function damage and liver failure.

Mushroom toxicity is the main branch of foodborne poisoning, and liver damage caused by amatoxin poisoning accounts for more than 90 % of deaths due to mushroom poisoning. Alpha-amatoxin (α-AMA) has been considered the primary toxin from amatoxin-containing mushrooms, which is responsible for hepatotoxicity and death. However, the mechanism underlying liver failure due to α-AMA remains unclear. This study constructed animal and cell models. In the animal experiments, we investigated liver injury in BALB/c mice at different time points after α-AMA treatment, and explored the process of inflamma

Mincle and STING-Stimulating Adjuvants Elicit Robust Cellular Immunity and Drive Long-Lasting Memory Responses in a Foot-and-Mouth Disease Vaccine.

Conventional foot-and-mouth disease (FMD) vaccines exhibit several limitations, such as the slow induction of antibodies, short-term persistence of antibody titers, as well as low vaccine efficacy and safety, in pigs. Despite the importance of cellular immune response in host defense at the early stages of foot-and-mouth disease virus (FMDV) infection, most FMD vaccines focus on humoral immune response. Antibody response alone is insufficient to provide full protection against FMDV infection; cellular immunity is also required. Therefore, it is necessary to design a strategy for developing a n

Self-assembled insulin and nanogels polyelectrolyte complex (Ins/NGs-PEC) for oral insulin delivery: characterization, lyophilization and in-vivo evaluation.

Introduction: Insulin is given by injection, because when administered orally, it would be destroyed by enzymes in the digestive system, hence only about 0.1% reaches blood circulation. The purpose of the present study was to use pH sensitive polyelectrolyte methyl methacrylate (MMA)/itaconic acid (IA) nanogels as carriers in an attempt to improve absorption of insulin administered orally. Methods: Insulin (Ins) was incorporated into the MMA/IA nanogels (NGs) using the polyelectrolyte complexation (PEC) method to form Ins/NGs-PEC. Several parameters, including Ins:NGs ratio, pH, incubation tim

Cationic DDA/TDB liposome as a mucosal vaccine adjuvant for uptake by dendritic cells in vitro induces potent humoural immunity.

The cationic dimethyldioctadecylammonium/trehalose 6,6,9-dibehenate (DDA/TDB) liposome is as a strong adjuvant system for vaccines, with remarkable immunostimulatory activity. The mucosal administration of vaccines is a potential strategy for inducing earlier and stronger mucosal immune responses to infectious diseases. In this study, we assessed whether the intranasal administration of cationic DDA/TDB liposomes combined with influenza antigen A (H3N2) can be used as a highly efficacious vaccine to induce mucosal and systemic antibody responses.
