De Novo Designed Hexadecapeptides Synergize Glycopeptide Antibiotics Vancomycin and Teicoplanin against Pathogenic Klebsiella pneumoniae via Disruption of Cell Permeability and Potential.

Given the worldwide prevalence of pathogenic drug-resistant bacteria and the slow pace of new antibacterial development, discovering new uses for approved drugs that are outside the scope of the original indication is increasingly becoming an attractive proposition. In this work, seven linear cationic hexadecapeptides were designed, synthesized, and characterized. These amphiphilic peptides are able to transform from the random coil structure in water to α-helix in SDS solution and have only modest bioactivity to limited bacterial strains when used alone. Surprisingly, one of them, namely, zp1

Antifungal Triazole Posaconazole Targets an Early Stage of the Parechovirus A3 Life Cycle.

Viruses in species Parechovirus A (Picornaviridae) are associated with a wide variety of clinical manifestations. Parechovirus A3 (PeV-A3) is known to cause sepsis-like illness, meningitis, and encephalitis in infants and young children. To date, no specific therapies are available to treat PeV-A3-infected children. We had previously identified two FDA-cleared antifungal drugs, itraconazole (ITC) and posaconazole (POS), with potent and specific antiviral activity against PeV-A3. Time-of-addition and synchronized infection assays revealed that POS targets an early stage of the PeV-A3 life cycle

Rapid fluorescent focus inhibition test optimization and validation: Improved detection of neutralizing antibodies to rabies virus.

The rabies rapid fluorescent focus inhibition test (RFFIT) is the most widely used cell-based assay for detecting and quantitating rabies virus neutralizing antibodies (RVNA) in human serum. However, it is a complex, labor intensive, and somewhat subjective manual assay, the performance of which may be affected by a number of factors including the quality of cells and virus, variability of assay reagents and the skill and expertise of analysts. This study sought to identify and evaluate conditions that may impact RFFIT performance and RVNA detection by evaluating assay parameters including: di

Genetic stability of genome-scale deoptimized RNA virus vaccine candidates under selective pressure.

Recoding viral genomes by numerous synonymous but suboptimal substitutions provides live attenuated vaccine candidates. These vaccine candidates should have a low risk of deattenuation because of the many changes involved. However, their genetic stability under selective pressure is largely unknown. We evaluated phenotypic reversion of deoptimized human respiratory syncytial virus (RSV) vaccine candidates in the context of strong selective pressure. Codon pair deoptimized (CPD) versions of RSV were attenuated and temperature-sensitive. During serial passage at progressively increasing temperat

Molecularly cloned SHIV-1157ipd3N4: a highly replication- competent, mucosally transmissible R5 simian-human immunodeficiency virus encoding HIV clade C Env.

Human immunodeficiency virus type 1 (HIV-1) clade C causes >50% of all HIV infections worldwide, and an estimated 90% of all transmissions occur mucosally with R5 strains. A pathogenic R5 simian-human immunodeficiency virus (SHIV) encoding HIV clade C env is highly desirable to evaluate candidate AIDS vaccines in nonhuman primates. To this end, we generated SHIV-1157i, a molecular clone from a Zambian infant isolate that carries HIV clade C env. SHIV-1157i was adapted by serial passage in five monkeys, three of which developed peripheral CD4(+) T-cell depletion. After the first inoculated monk

Selection of the same mutation in the U69 protein kinase gene of human herpesvirus-6 after prolonged exposure to ganciclovir in vitro and in vivo.

After serial passage in the presence of increasing concentrations of ganciclovir (GCV) in vitro, a human herpesvirus-6 (HHV-6) mutant exhibiting a decreased sensitivity to the drug was isolated. Analysis of drug susceptibility showed that the IC(50) of this mutant was 24-, 52- and 3-fold higher than that of the wild-type (wt) IC(50) in the case of GCV, cidofovir and foscarnet, respectively. Genotypic analysis showed two single nucleotide changes as compared to the wild-type: an A-->G substitution of the U69 protein kinase (PK) gene resulted in an M(318)V amino acid substitution and the other c

[Establishment and analysis of cell culture infected persistently with JC virus].

JC polyomavirus (JCV) is the causative agent of a demyelinating disease in the central nervous system, known as progressive multifocal leukoencephalopathy. Since it is difficult to propagate JCV in vitro, many laboratories have attempted to develop cell lines permissive for replication of JCV. It was recently reported that serial passage of JCV (strain Mad-1) in a human neuroblastoma cell line (IMR-32 cells) generated JCV variants (M1-IMRa, -IMRb, -IMRc) adapted to the growth in IMR-32 cells. We obtained a persistently-infected cell culture, designated JCI cells, by continued subcultivation of

An immunocompetent model of MCPyV-driven Merkel cell carcinoma reveals tumor evolution under immune selection.

Merkel cell carcinoma (MCC) is a neuroendocrine skin tumor that is frequently driven by integration of Merkel cell polyomavirus (MCPyV). In MCC, the MCPyV genome is truncated, but expression of the viral tumor antigens, truncated large tumor antigen (LTT) and small tumor antigen (sT), is maintained and drives uncontrolled proliferation. We introduced constitutive expression of the MCPyV T antigens (TAs) into primary mouse dermal fibroblasts (MDFs) to determine whether these cells are susceptible to MCPyV-driven transformation. TA expression alone in MDFs induced key MCC markers, cytokeratin-20

Development of a Novel Live Attenuated QX-Like Infectious Bronchitis Virus Vaccine and Its Efficacy Against Recent GI-19 Subgroup Variants.

The emergence of novel nephropathogenic QX-like infectious bronchitis virus (IBV) strains is a major problem across multiple continents. Despite extensive vaccination efforts, the emergence of novel QX-like variant strains that lack serologic cross-reactivity with commercially available vaccines complicates the control of IBV. In the present study, a live attenuated IBV vaccine strain was developed by serial passages with heat adaptation on the basis of the currently circulating IBV strain K29/18 of the QX-like III subgroup, and its safety, immunogenicity, and cross-protective efficacy against

A new recombinant avian metapneumovirus vaccine candidate provides complete protection against the novel variant infectious bursal disease virus in chickens.

Infectious bursal disease (IBD) is an acute, highly contagious, and immunosuppressive condition in chickens, caused by the infectious bursal disease virus (IBDV). The recent emergence of novel variant IBDV (nVarIBDV) poses a significant threat to the global poultry industry. However, currently available vaccines provide only limited protection against nVarIBDV strains. In this study, a recombinant avian metapneumovirus subtype B (aMPV/B) expressing the nVarIBDV VP2 gene (rLN16A-nVarVP2) was successfully rescued by inserting the gene between the G and L genes of the attenuated aMPV/B strain LN1

Engineering a Non-Aphid-Transmissible Mild Mutant of Zucchini Yellow Mosaic Virus for Cross Protection.

Zucchini yellow mosaic virus (ZYMV; genus Potyvirus), an aphid-transmitted pathogen, has become a major threat to melon (Cucumis melo) production in China, severely reducing both crop yield and fruit quality. Cross protection using attenuated virus strains is a key strategy for controlling plant viral diseases. However, few ZYMV vaccine candidates possess both multiple attenuating mutations and impaired aphid transmissibility. In this study, we identified that the conserved residues in helper component-protease (HC-Pro), glycine at position 322 (G322) and lysine at position 360 (K360), played

Isolation and Characterization of a Naturally Occurring Brevundimonas vesicularis Strain Exhibiting High Phytoene Accumulation.

Phytoene, a colorless precursor of carotenoids, has attracted increasing attention because of its favorable bioavailability, antioxidant activity, and potential applications in functional foods, nutraceuticals, and animal nutrition. However, its industrial utilization remains limited by low natural abundance and the dependence of current production strategies on genetic engineering or metabolic pathway manipulation. In this study, we identified and characterized a naturally occurring Brevundimonas vesicularis strain (Bv-xms2024) exhibiting pronounced phytoene accumulation without genetic modif

Novel organophosphorus-quinolone hybrids as potential antimicrobial agents: design, synthesis, biological evaluation, and in vivo antibacterial effect in bacteria-infected zebrafish model.

A novel series of organophosphorus–quinolone hybrids were designed and synthesized as potential antimicrobial agents through efficient phospho-Mannich reactions. The most potent compound, 5d, demonstrated exceptional activity against methicillin-resistant Staphylococcus aureus (MRSA) with an MIC of 0.5 μg/mL, representing a 16-fold and twofold improvement over norfloxacin and ciprofloxacin, respectively. Structure–activity relationship studies revealed the critical influence of substituent electronic properties and steric bulk, with electron-donating groups enhancing potency. Mechanistic studi

Biological characterization and genome analysis of Bacillus thuringiensis GX0003935 with biocontrol activity against Meloidogyne enterolobii.

Meloidogyne enterolobii is a highly aggressive root-knot nematode, and reduced availability of chemical nematicides increases the need for effective biocontrol alternatives. We characterized Bacillus thuringiensis GX0003935 in terms of nematicidal activity, stability, biocontrol efficacy, and genome features. The culture broth and filtrate caused more than 97% corrected mortality of second-stage juveniles within 48 h, whereas bacterial suspension showed limited activity, suggesting that extracellular factors substantially contribute to nematicidal activity. The culture filtrate retained high n

Hydrazine Derivative-Based Carbon Dots for Potent Antibacterial Activity Against Multidrug-Resistant Bacterial.

Bacterial infections, particularly those caused by multidrug-resistant strains, remain a significant global public health challenge. The growing resistance to traditional antibiotics highlights the urgent need for novel antibacterial strategies. Herein, we successfully synthesized three types of nitrogen-doped carbon dots (tBuCz-CDs, HAH-CDs, and EC-CDs) via hydrothermal method using tert-butyl carbazate, hydroxyacetic acid hydrazide, and ethyl carbazate as precursors. tBuCz-CDs, HAH-CDs, and EC-CDs exhibited potent antibacterial activity against methicillin-resistant Staphylococcus aureus (MR

Injectable ε-Polylysine/Hyaluronic Acid Hydrogels with Resistance-Preventing Antibacterial Activity for Treating Wound Infections.

The growing threat of antimicrobial resistance has created an urgent demand for nonantibiotic biomaterials capable of preventing infections without promoting bacterial resistance. In this study, we developed injectable, covalently cross-linked hydrogels composed of ε-poly-l-lysine (ε-PL) and hyaluronic acid (HA) for localized wound infection treatment. These hydrogels combine the inherent antibacterial properties of ε-PL with the biocompatibility of HA, forming a shear-thinning, self-recovering system suitable for syringe-based administration. We first evaluated the antibacterial activity of p

Fitness cost and compensatory evolution of penicillin-induced resistant Staphylococcus aureus.

Staphylococcus aureus is a widespread pathogen in nature, with staphylococcal enterotoxins being a major cause of foodborne illness. The extensive use of antibiotics on farms has contributed to the spread of antibiotic-resistant S. aureus. Understanding the fitness cost and compensatory evolution of antibiotic-resistant isolates is crucial for assessing the consequences of resistance acquisition and predicting the potential spread of resistant mutants in various environments. In this study, penicillin (PEN) was used to induce resistance in antibiotic-sensitive S. aureus, resulting in PEN-resis

Establishment and evaluation of a patient-derived organoids-based xenograft model of primary aldosteronism using [18F]AlF-NOTA-pentixather PET/CT: bridging preclinical and clinical imaging.

BACKGROUND: Primary aldosteronism (PA) is a leading cause of secondary hypertension, but the lack of representative preclinical models hampers translational research and imaging evaluation. We aimed to establish a patient-derived organoids-based xenograft (PDOX) model of PA and to assess its imaging characteristics using [18F]AlF-NOTA-pentixather PET/CT, with validation in clinical patients. METHODS: Adrenal tissues from patients with PA were used to generate adrenal organoids in vitro. After serial passaging, these organoids were implanted subcutaneously into immunodeficient mice to establis

Infectious cDNA Clone of Chikungunya Virus 181/25 for Antiviral Assay Development.

Chikungunya is a mosquito-borne viral disease in humans and remains a significant threat in many parts of the world. Although Chikungunya fever is self-limiting, it often progresses to a chronic phase marked by debilitating arthralgia and joint pain that can persist for months or even years. The Chikungunya virus (CHIKV) vaccine strain 181/25 is a prototypical strain of the virus that has been extensively used to develop second-generation vaccine candidates. In this study, we report the construction of an infectious cDNA clone of CHIKV-181/25 and a GFP-expressing variant designed to facilitate
