Induction of CD8+ T-cell responses against subunit antigens by the novel cationic liposomal CAF09 adjuvant.

Vaccines inducing cytotoxic T-cell responses are required to achieve protection against cancers and intracellular infections such as HIV and Hepatitis C virus. Induction of CD8+ T cell responses in animal models can be achieved by the use of viral vectors or DNA vaccines but so far without much clinical success. Here we describe the novel CD8+ T-cell inducing adjuvant, cationic adjuvant formulation (CAF) 09, consisting of dimethyldioctadecylammonium (DDA)-liposomes stabilized with monomycoloyl glycerol (MMG)-1 and combined with the TLR3 ligand, Poly(I:C). Different antigens from tuberculosis (

Improved Recovery of Hepatocytes Isolated From Warm Ischemic Rat Liver by Citrate Phosphate Dextrose (CPD)-Supplemented Euro-Collins Solution.

Demand for human primary hepatocytes is increasing, particularly for clinical trials of hepatocyte transplantation. However, due to the severe shortage of organ transplant donors, the source of cells for these endeavors is restricted to untransplantable livers, such as those from non-heart-beating donors and surgically resected liver tissues. To improve cell recovery from such sources after warm ischemia, we evaluated the efficacy of applying perfusion solutions, focusing on improvement of hepatocyte recovery. Warm ischemia was induced by clamping both portal vein and hepatic artery for 10 or

Comparative evaluation of fungal, tubercular, and pyogenic brain abscesses with conventional and diffusion MR imaging and proton MR spectroscopy.

BACKGROUND AND PURPOSE: It is difficult to differentiate the cause of brain abscesses with the use of CT and MR imaging. We did a comparative evaluation of pyogenic, tubercular, and fungal brain abscesses by using conventional, diffusion-weighted imaging (DWI), and proton MR spectroscopy (PMRS) with an aim to define the unique features that may differentiate among the pyogenic, tubercular, and fungal brain abscesses. MATERIALS AND METHODS: We performed a retrospective analysis on 110 patients with surgically proved brain abscesses. Imaging studies included T2, T1, postcontrast T1, DWI, and PM

A comparative study of cationic liposome and niosome-based adjuvant systems for protein subunit vaccines: characterisation, environmental scanning electron microscopy and immunisation studies in mice.

Vesicular adjuvant systems composing dimethyldioctadecylammonium (DDA) can promote both cell-mediated and humoral immune responses to the tuberculosis vaccine fusion protein in mice. However, these DDA preparations were found to be physically unstable, forming aggregates under ambient storage conditions. Therefore there is a need to improve the stability of such systems without undermining their potent adjuvanticity. To this end, the effect of incorporating non-ionic surfactants, such as 1-monopalmitoyl glycerol (MP), in addition to cholesterol (Chol) and trehalose 6,6'-dibehenate (TDB), on th

A preliminary study on epigenetic changes during boar spermatozoa cryopreservation.

Artificial insemination (AI) with cryopreserved boar semen is limited to no more than 1% of the total number of inseminations due to low conception rates and litter sizes. Cryopreservation causes a dramatic decrease in the viability, motility and fertility of spermatozoa, but the underlying mechanism remains unknown. In this study, mRNA expression and protein levels of epigenetic-related genes (Dnmt3a, Dnmt3b, Jhdm2a, Kat8, Prm1, Prm2 and IGF2) in fresh and cryopreserved boar spermatozoa were evaluated using qRT-PCR and ELISA. The results showed that cryopreservation or freezing, which drastic

Improvement of biosurfactant production by Rhodococcus ruber IEGM 231 grown on waste cooking oil, their emulsifying activity and antioxidant potential.

In recent years, the production of microbial biosurfactants, increasingly used in various industries and agriculture, from low-cost renewable substrates is intensively studied. Large volumes of food waste, in particular waste cooking oil, are promising substrates for the production of biosurfactants on an industrial scale. However, studies on the ability of Rhodococcus bacteria, well-known biosurfactant producers, to grow on this substrate and the functional characteristics of synthesized biosurfactants are still scarce. In this study, the possibility of improving biosurfactant production by R

Protein Kinase A and High-Osmolarity Glycerol Response Pathways Cooperatively Control Cell Wall Carbohydrate Mobilization in Aspergillus fumigatus.

Aspergillus fumigatus mitogen-activated protein kinases (MAPKs) are involved in maintaining the normal morphology of the cell wall and providing resistance against cell wall-damaging agents. Upon cell wall stress, cell wall-related sugars need to be synthesized from carbohydrate storage compounds. Here we show that this process is dependent on cAMP-dependent protein kinase A (PKA) activity and regulated by the high-osmolarity glycerol response (HOG) MAPKs SakA and MpkC. These protein kinases are necessary for normal accumulation/degradation of trehalose and glycogen, and the lack of these gene

Physical correlates of the ultrasonic reflectivity of lipid dispersions suitable as diagnostic contrast agents.

The objective of this study was to determine the physical basis of ultrasound (US) reflectivity of echogenic lipid dispersions. These dispersions were made using a process previously described involving sonication of the lipid in water, addition of mannitol, freezing, lyophilization and rehydration. The component lipids were egg phosphatidylcholine, dipalmitoylphosphatidylethanolamine, dipalmitoylphosphatidylglycerol and cholesterol in a molar ratio of 69:8:8:15. Ultrasound reflectivity, as assessed with a 20-MHz intravascular US catheter and analyzed using computer-assisted videodensitometry,

Investigation of regioselectivity and thermostability of free and immobilized Pleurotus citrinopileatus lipase.

A fungal lipase from the Basidiomycete Pleurotus citrinopileatus (PCI_Lip) has been investigated for its regioselectivity. Therefore, the conversion products of a triglyceride were analyzed using 1H-NMR to determine the position of hydrolysis within the substrate. PCI_Lip is a 1,3-selective lipase, as it preferentially hydrolyzes the fatty acids at the outer positions of the triglyceride. Furthermore, this study aimed to gain deeper insights into the stability of the lipase. The melting point of the enzyme was determined to be at 63 °C in ultrapure water, which could be increased to 71 °C by d

Biochemistry of Reactivation of Dormant Mycobacteria.

An important aspect of medical microbiology is identification of the causes and mechanisms of reactivation (resuscitation) of dormant non-sporulating bacteria. In particular, dormant Mycobacterium tuberculosis (Mtb) can cause latent tuberculosis (TB), which could be reactivated in the human organism to the active form of the disease. Analysis of experimental data suggested that reactivation of mycobacteria and reversion of the growth processes include several stages. The initial stage is associated with breakdown of the storage substances like trehalose upon the action of trehalase and with pe

Evaluation of the effects of different formulations of protectants on the preservation of the microbiota in fecal microbiota transplantation.

BACKGROUND: With the increasing indications for fecal microbiota transplantation for the treatment of diseases, there is a growing demand for the preparation of frozen or lyophilized fecal microbiota products that are viable and can stably colonize the recipient. The addition of protective agents plays an important role in the preparation. However, there has been no systematic evaluation of the protective agents used in fecal microbiota sample transplantation preparation for transplantation. METHODS: We were used the donor bacterial flora containing 10 different formulations of protective age

Pleiotropic Effects on Tachyzoite and Host Cell Proteomes in Knock-Out Clones of the Open Reading Frames 297720 and 319730 Constitutively Expressed in T. gondii ShSp1 Tachyzoites.

Toxoplasma gondii, the causative agent of toxoplasmosis widespread in animals and humans, is an intracellular apicomplexan protozoan parasite infecting a variety of host cells. Gene editing using CRISPR-Cas9 has become a standard tool to investigate the molecular genetics of this interaction. With respect to gene knock-out (KO) studies, the general paradigm implies that the gene of interest is expressed in the wildtype and that only the gene of interest is affected by the knock-out. Consequently, the observed phenotype depends on the presence or absence of genes of interest. To challenge this

MmpL3, Wag31, and PlrA are involved in coordinating polar growth with peptidoglycan metabolism and nutrient availability.

UNLABELLED: Cell growth in mycobacteria involves cell wall expansion that is restricted to the cell poles. The DivIVA homolog Wag31 is required for this process, but the molecular mechanism and protein partners of Wag31 have not been described. In this study of Mycobacterium smegmatis, we identify a connection between wag31 and trehalose monomycolate (TMM) transporter mmpl3 in a suppressor screen and show that Wag31 and polar regulator PlrA are required for MmpL3's polar localization. In addition, the localization of PlrA and MmpL3 is responsive to nutrient and energy deprivation and inhibitio

A compromised developmental trajectory of the infant gut microbiome and metabolome in atopic eczema.

Evidence is accumulating that the establishment of the gut microbiome in early life influences the development of atopic eczema. In this longitudinal study, we used integrated multi-omics analyses to infer functional mechanisms by which the microbiome modulates atopic eczema risk. We measured the functionality of the gut microbiome and metabolome of 63 infants between ages 3 weeks and 12 months with well-defined eczema cases and controls in a sub-cohort from the Growing Up in Singapore Toward healthy Outcomes (GUSTO) mother-offspring cohort. At 3 weeks, the microbiome and metabolome of allerge

Purification and partial biochemical characterization of a membrane-bound type II-like α-glucosidase from the yeast morphotype of Sporothrix schenckii.

The early steps of glycoprotein biosynthesis involve processing of the N-glycan core by endoplasmic reticulum α-glucosidases I and II which sequentially trim the outermost α1,2-linked and the two more internal α1,3-linked glucose units, respectively. We have demonstrated the presence of some components of the enzymic machinery required for glycoprotein synthesis in Sporothrix schenckii, the etiological agent of human and animal sporotrichosis. However, information on this process is still very limited. Here, a distribution analysis of α-glucosidase revealed that 38 and 50% of total enzyme acti

Mycobacterium cosmeticum sp. nov., a novel rapidly growing species isolated from a cosmetic infection and from a nail salon.

Four isolates of a rapidly growing Mycobacterium species had a mycolic acid pattern similar to that of Mycobacterium smegmatis, as determined by HPLC analyses. Three of the isolates were from footbath drains and a sink at a nail salon located in Atlanta, GA, USA; the fourth was obtained from a granulomatous subdermal lesion of a female patient in Venezuela who was undergoing mesotherapy. By random amplified polymorphic DNA electrophoresis and PFGE of large restriction fragments, the three isolates from the nail salon were shown to be the same strain but different from the strain from the patie

Deoxygenation of carbohydrates by thiol-catalysed radical-chain redox rearrangement of the derived benzylidene acetals.

Five- or six-membered cyclic benzylidene acetals, derived from 1,2- or 1,3-diol functionality in carbohydrates, undergo an efficient thiol-catalysed radical-chain redox rearrangement resulting in deoxygenation at one of the diol termini and formation of a benzoate ester function at the other. The role of the thiol is to act as a protic polarity-reversal catalyst to promote the overall abstraction of the acetal hydrogen atom by a nucleophilic alkyl radical. The redox rearrangement is carried out in refluxing octane and/or chlorobenzene as solvent at ca. 130 degrees C and is initiated by thermal

Bevacizumab-Functionalized Exosome-Associated Chitosan Nanocomposite for Temozolomide Delivery and Apoptosis-Related Transcriptional Modulation in Experimental Glioma.

INTRODUCTION: Glioblastoma (GBM) has a poor prognosis, and temozolomide (TMZ) therapy is limited by a short half-life and insufficient intratumoral availability. We developed a bevacizumab (BVZ)-functionalized, exosome-associated chitosan nanocomposite loaded with TMZ (BVZ-Exo@CS-TMZ) as a multifunctional biohybrid platform for experimental glioma therapy. METHODS: CS-TMZ nanoparticles were prepared by ionotropic gelation, non-covalently associated with adipose-derived stromal cell exosomes (Exo), and functionalized with BVZ via EDC/NHS conjugation to accessible surface carboxyl groups. Role of primary protectors of plant cells in salinity tolerance: molecular mechanisms and adaptive strategies.

Salinity stress is among the most pervasive abiotic factors limiting plant growth and agricultural productivity worldwide, currently affecting more than 20% of irrigated croplands. Elevated salt concentrations induce osmotic imbalance, ionic toxicity, and oxidative stress, leading to disruptions in photosynthesis, metabolism, and plant development. To counteract these deleterious effects, plants have evolved intricate, highly coordinated defense systems comprising structural and functional cellular protectors. At the structural level, the cell wall, plasma membrane, vacuole, and peroxisome pla
