Production of anti-inflammatory and antidiabetic oligosaccharides from okra mucilage through one-step microbial fermentation.

Plant-based natural products are increasingly being explored as safer and more sustainable alternatives than synthetic drugs for controlling globally rising chronic health conditions, including inflammation and diabetes. Microbial fermentation has long been used to enhance the value and bioactivity of natural products. In this study, we produced bioactive oligosaccharides from okra (Abelmoschus esculentus) mucilage polysaccharides using a sustainable one-step Pichia kudriavzevii-mediated fermentation approach. The oligosaccharide fractions separated by size-exclusion chromatography primarily c

Supplementation of okra seed oil ameliorates ethanol-induced liver injury and modulates gut microbiota dysbiosis in mice.

This aim of this study is to assess the possible effects of dietary okra seed oil (OSO) consumption on attenuation of alcohol-induced liver damage and gut microbiota dysbiosis, and associated mechanisms in mice. Mice were orally administered alcohol alone or in combination with OSO at 400 and 800 mg per kg bw for 8 weeks. OSO caused a strong inhibition of abnormal weight loss and liver fat accumulation in alcohol-administered mice. Malonaldehyde production was also effectively antagonized, and glutathione peroxidase and superoxide dismutase activities were elevated by OSO treatment in ethanol-

Rapid, high-throughput, and sensitive screening of herbal product metabolites in blood plasma using 2D carbon microfiber fractionation coupled with quadrupole time-of-flight high-resolution mass spectrometry: Huangkui capsule insights.

Drug metabolite characterization from complex endogenous matrices such as plasma is a challenging task, especially for herbal product compounds. The analyses are generally carried out by flow-injection mass spectrometry (FI-MS) or combined with liquid chromatography (LC-MS), but the first one suffers from significant matrix effects, while the second is time-consuming and unsuitable for high-throughput analysis. In this study, a rapid method combining 2D-carbon microfiber fractionation (2DµCFs) with quadrupole time-of-flight high-resolution mass spectrometry (QTOF-HRMS) is proposed for the comp

Addressing GI Health Through the Bidirectional Modulation of the Gut-Brain Axis With Herbal Extracts: A Narrative Review.

Functional gastrointestinal disorders (FGIDs) refer to a group of disorders with chronic symptoms, such as abdominal pain, dysphagia, dyspepsia, diarrhea, constipation, and bloating. Among these, functional constipation significantly impacts the quality of life and is linked with comorbidities, such as anxiety and depression. The exact pathophysiology remains unclear despite the widespread occurrence. Research suggests that the gut-brain axis plays a role in FGIDs. Disruptions in the bidirectional communication between the brain and gastrointestinal (GI) tract contribute to GI symptoms and moo

Clinical evidence for fat-binding-mediated fecal fat excretion by RODLTM (Red Okra-Diospyros lotus botanical combination) under a prescribed restricted diet: a randomized, double-blind, placebo-controlled trial.

Preclinical studies have demonstrated that a botanical combination of red okra (Abelmoschus esculentus) and Diospyros lotus (RODLTM) exerts dual anti-obesity actions by both inhibiting intestinal fat absorption and stimulating adipocyte lipid metabolism. However, its clinical efficacy in promoting fecal fat excretion under controlled dietary conditions has not been validated in humans. A randomized, double-blind, placebo-controlled clinical trial was conducted in 70 healthy adults under a prescribed restricted diet containing 130-150 g/day of fat. Participants were randomly assigned to receive

The fungal alkaloid Okaramine-B activates an L-glutamate-gated chloride channel from Ixodes scapularis, a tick vector of Lyme disease.

A novel L-glutamate-gated anion channel (IscaGluCl1) has been cloned from the black-legged tick, Ixodes scapularis, which transmits multiple pathogens including the agents of Lyme disease and human granulocytic anaplasmosis. When mRNA encoding IscaGluCl1 was expressed in Xenopus laevis oocytes, we detected robust 50-400 nA currents in response to 100 μM L-glutamate. Responses to L-glutamate were concentration-dependent (pEC50 3.64 ± 0.11). Ibotenate was a partial agonist on IscaGluCl1. We detected no response to 100 μM aspartate, quisqualate, kainate, AMPA or NMDA. Ivermectin at 1 μM activated

Formulation and in-vitro characterization of okra mucilage-based controlled release hydrogels.

BACKGROUND: Certain drugs at therapeutic doses can cause gastrointestinal toxicity, which controlled-release formulations help to address. Natural polymers are widely used in drug delivery due to their non-toxic, biodegradable, and biocompatible properties. Okra mucilage powder (OMP) is especially promising because of its strong gelling ability and pH-sensitive behavior. OBJECTIVES: This study aimed to develop OMP-based hydrogels for the controlled release of methotrexate (MTX) over extended periods via free-radical polymerization. METHODS: Acrylic acid (AA) was used as the monomer with OMP

Hyperoside stabilizes ACAT1 to promote fatty acid oxidation and attenuate kidney fibrosis via the L-carnitine-SIRT3 axis.

BACKGROUND: Kidney fibrosis represents a key pathological process driving the progression of chronic kidney disease (CKD) and is closely associated with mitochondrial impairment and altered lipid metabolism. Hyperoside, a major flavonoid glycoside from Abelmoschus manihot, has shown anti-fibrotic activity, yet its mechanistic role in renal fibrosis remains unclear. METHODS: Two murine models, folic acid-induced nephropathy and unilateral ureteral obstruction, were employed to assess the renoprotective actions of hyperoside. Mitochondrial function, lipid metabolic remodeling, and fibrotic prog
