Measurement of acetaldehyde: what levels occur naturally and in response to alcohol?

The aim of the present paper is to update the status regarding human acetaldehyde levels in blood, breath and saliva during normal ethanol oxidation, i.e. without deficiency in, or inhibition of, aldehyde dehydrogenase activity. The previous conclusion according to which no detectable (<0.5 microM), adequately determined 'free and/or loosely bound' acetaldehyde has not yet been found in venous blood, more or less, still holds. The only new findings within this context consist of low venous blood acetaldehyde levels (1-3 microM on average) observed in some women during the use of oral contracep

Toxicity of beta-carotene and its exacerbation by acetaldehyde in HepG2 cells.

In rats and baboons, the hepatotoxicity of chronic ethanol consumption is exacerbated by beta-carotene feeding, but the mechanism of this adverse effect is unknown. In this study, the toxicity of beta-carotene and acetaldehyde was documented by the MTT test (an assay of reduction of tetrazolium to formazan) and by lactate dehydrogenase (LDH) leakage. In HepG2 cells, beta-carotene or acetaldehyde inhibited mitochondrial reduction function as indicated by a decrease of the MTT test. beta-Carotene was inhibitory at very low concentration, in a dose-dependent manner. The combination of these two c

The role of acetaldehyde in pregnancy outcome after prenatal alcohol exposure.

It is not known why some heavy-drinking women give birth to children with alcohol-related birth defects (ARBD) whereas others do not. The objective of this study was to determine whether the frequency of elevated maternal blood acetaldehyde levels among alcoholics is in the range of ARBD among alcoholic women. MEDLINE was searched from 1980 to 2000 using the key words acetaldehyde, pharmacokinetics, and alcoholism for controlled trials reporting blood or breath acetaldehyde levels in alcoholics and nonalcoholics. Separately, using the key words fetal alcohol syndrome, epidemiology, prevalence,

N-Terminal dipeptides of D(-)-penicillamine as sequestration agents for acetaldehyde.

Since acetaldehyde (AcH), a toxic oxidation product of ethanol, may play an etiologic role in the initiation of alcoholic liver disease, we had earlier pioneered the development of beta, beta-disubstituted-beta-mercapto-alpha-amino acids as AcH-sequestering agents. We now report the synthesis of a series of N-terminal dipeptides of D(-)-penicillamine, prepared from the synthon 3-formyl-2,2,5,5-tetramethylthiazolidine-4S-carboxylic acid (3), a cyclized N-protected derivative of D(-)-penicillamine. These dipeptides were equally or more effective than penicillamine in trapping AcH in a cell-free

Detection of antibodies to acetaldehyde-albumin conjugates in alcoholic liver disease.

The principal metabolite of ethanol, acetaldehyde, conjugates with various proteins that form antibody-inducing neo-antigens. We have analysed sera from patients with biopsy-proven alcoholic liver disease (hepatitis = 10, cirrhosis = 11, steatosis = 3) and controls = 19 (normal teetotallers and 6 non-alcoholic liver disease). Sera were examined with an enzyme-linked immunoabsorbent assay (ELISA) for antibodies binding preferentially to an acetaldehyde-albumin conjugate. Reactive sera from alcohol misusers were then purified using an amino-hexyl Sepharose affinity column. Antibodies binding to

Effects of naloxone and buprenorphine on intravenous acetaldehyde self-injection in rats.

Rats can be induced to self-inject acetaldehyde under an appropriate operant conditioning schedule. The narcotic antagonist naloxone (30 mg/kg) is shown to produce a decrease in schedule-induced acetaldehyde self-injection, but was without effect on both barpress responding and spontaneous activity in rats tested individually for fine, gross and total activity. On the other hand buprenorphine (0.3 and 3 mg/kg), the mixed agonist-antagonist derived from the opium alkaloid thebaine, also produced a significant decrease in acetaldehyde self-injection. However, a significant effect of buprenorphin

Relationship between acetaldehyde concentration in mouth air and tongue coating volume.

OBJECTIVE: Acetaldehyde is the first metabolite of ethanol and is produced in the epithelium by mucosal ALDH, while higher levels are derived from microbial oxidation of ethanol by oral microflora such as Candida species. However, it is uncertain whether acetaldehyde concentration in human breath is related to oral condition or local production of acetaldehyde by oral microflora. The aim of this pilot study was to investigate the relationship between physiological acetaldehyde concentration and oral condition in healthy volunteers. MATERIAL AND METHODS: Sixty-five volunteers (51 males and 14

Human blood acetaldehyde levels: with improved methods, a clearer picture emerges.

New reliable methods for the determination of acetaldehyde in human blood, either from separated plasma or from acid-precipitated whole blood, demonstrate that the blood of healthy Caucasians contains at most only extremely small amounts of acetaldehyde (less than 1 microM) after moderate alcohol intoxication. On the other hand, among about 50% of the Japanese population ethanol ingestion results in elevated blood acetaldehyde levels (10-50 microM) with consequent unpleasant cardiovascular responses such as facial flushing and tachycardia, apparently because of a lack of one of the acetaldehyd

L-cysteine improves blood fluidity impaired by acetaldehyde: In vitro evaluation.

Blood fluidity is reportedly influenced by the volume and function of blood cells and plasma and is a predictor of primary cardiovascular events in patients with traditional cardiovascular risk factors. Heavy alcohol consumption was shown to be associated with a higher risk for cardiovascular diseases. Acetaldehyde (ACD), an oxidizing substance formed from ethanol, reportedly stimulates monocyte adhesion, causes abnormalities in the red blood cell (RBC) membrane, and decreases RBC deformability. In addition, it was reported that blood ACD levels are reduced in mice pretreated with L-cysteine.

Pueraria lobata (Kudzu root) hangover remedies and acetaldehyde-associated neoplasm risk.

Recent introduction of several commercial Kudzu root (Pueraria lobata) containing hangover remedies has occurred in western countries. The available data is reviewed to assess if there are any potential concerns in relationship to the development of neoplasm if these products are used chronically. The herb Pueraria has two components that are used as traditional therapies; Pueraria lobata, the root based herb and Pueraria flos, the flower based herb. Both of these herbal components have different traditional claims and constituents. Pueraria flos, which enhances acetaldehyde removal, is the tr

Peptide leukotrienes mediate acetaldehyde-induced bronchial hyper-responsiveness in guinea-pigs.

BACKGROUND: We previously reported that inhaled acetaldehyde, a metabolite of ethanol and a main factor in alcohol-induced asthma, causes bronchial hyper-responsiveness (BHR) in asthmatics. However, the mechanisms are unclear. OBJECTIVE: The purpose of this study was to investigate a role of a peptide leukotriene (LT) in acetaldehyde-induced BHR. METHODS: Effects of LT antagonists, ONO-1078 (0.1-1.0 mg/kg) and ICI-198, 615 (0.03-0.3 mg/kg), on acetaldehyde-induced bronchoconstriction and BHR to inhaled methacholine were examined using a modified Konzett-Rössler method in guinea pigs. Hemoglobin-acetaldehyde adducts are elevated in women carrying alcohol-damaged fetuses.

Alcohol use exceeding 3 to 5 daily drinks is associated with a 30% to 50% risk of delivering a child with fetal alcohol effects (FAE). FAE could be prevented if women were counseled and if they decreased their drinking. Therefore, we need sensitive and specific markers to detect alcohol abuse during pregnancy. We investigated whether acetaldehyde-hemoglobin adducts (Hb-Ach) could be such a marker. Using an antiserum specific for acetaldehyde-generated epitopes in proteins, we measured the levels of Hb-Ach from the red cells of 19 women visiting the outpatient department of pregnant alcohol abu

Presence and Dermal Exposure to Benzene and Acetaldehyde in Hand Sanitizers Available in Taiwan.

The widespread use of alcohol-based hand sanitizers (ABHS) during and after the COVID-19 pandemic has raised concerns about potential exposure to harmful volatile organic compounds (VOCs), such as benzene, acetaldehyde, and other impurities, which may pose health risks. This study investigated the concentrations of ethanol, isopropanol, and 12 impurities, including benzene, acetaldehyde, and methanol, in 85 commercially available ABHS products in Taiwan using gas chromatography-mass spectrometry (GC-MS). The results revealed that 12 samples contained alcohol concentrations below the recommende

Effects of Acute Ethanol Administration on Brain Oxidative Status: The Role of Acetaldehyde.

BACKGROUND: Ethanol (EtOH), one of the most widely consumed substances of abuse, can induce brain damage and neurodegeneration. EtOH is centrally metabolized into acetaldehyde, which has been shown to be responsible for some of the neurophysiological and cellular effects of EtOH. Although some of the consequences of chronic EtOH administration on cell oxidative status have been described, the mechanisms by which acute EtOH administration affects the brain's cellular oxidative status and the role of acetaldehyde remain to be elucidated in detail. METHODS: Swiss CD-I mice were pretreated with t

Interactions between Heterogeneous Uptake and Adsorption of Sulfur Dioxide and Acetaldehyde on Hematite.

Sulfur dioxide and organic aldehydes in the atmosphere are ubiquitous and often correlated with mineral dust aerosols. Heterogeneous uptake and adsorption of one of these species on mineral aerosols can potentially change the properties of the particles and further affect the subsequent heterogeneous reactions of the other species on the coating particles. In this study, the interactions between heterogeneous uptake and adsorption of sulfur dioxide and acetaldehyde on hematite are investigated by using in situ diffuse-reflectance infrared Fourier-transform spectroscopy (DRIFTS) at room tempera

The influence of B-complex vitamins upon the prolongation of prothrombin time by acetaldehyde.

Alcoholism plays a major role in the insufficient utilization or deficiency of the vitamin B-complex molecules, and the pathologies resulting therefrom. Thiamine, pyridoxamine, and folic acid, each contain primary amine functional groups, whereas nicotinamide and vitamin B12 contain amide groups, each of which are potential reactants with acetaldehyde (AcH), the primary intermediate in the metabolism of ethanol. In this current study, it is reported that prothrombin time (PT), which is prolonged in a fraction of the alcoholic population, can be modified (in the laboratory) when several B-compl

Locally-generated acetaldehyde is involved in ethanol-mediated LTP inhibition in the hippocampus.

Consistent with the ability of severe alcohol intoxication to impair memory, high concentrations of ethanol (60mM) acutely inhibit long-term potentiation (LTP) in the CA1 region of rat hippocampal slices. To account for this, we hypothesized that local metabolism to acetaldehyde may contribute to the effects of high ethanol on synaptic function. However, sodium azide, a catalase inhibitor, and allyl sulfide, an inhibitor of cytochrome P450 2E1 (CYP2E1), failed to overcome LTP inhibition by 60mM ethanol. In contrast, LTP was successfully induced in the presence of ethanol plus 4-methylpyrazole
