Mechanistic insights into compatibility-based detoxification of Fengshi Gutong Capsules through network toxicology and in vivo validation.

ETHNOPHARMACOLOGICAL RELEVANCE: Aconitum is a crucial medicinal herb widely used in the treatment of rheumatic diseases; however, its clinical application is restricted due to its inherent toxicity. In traditional Chinese medicine, the efficacy of Aconitum is enhanced and its toxicity reduced through the principle of "herbal compatibility." Fengshi Goutong Capsules (FSGTC) represent a classical prescription exemplifying this compatibility-based detoxification strategy. Although FSGTC has demonstrated significant therapeutic efficacy with minimal adverse effects, the underlying mechanisms respo

The role of efflux transporters on the transport of highly toxic aconitine, mesaconitine, hypaconitine, and their hydrolysates, as determined in cultured Caco-2 and transfected MDCKII cells.

Aconitum alkaloids including aconitine (AC), mesaconitine (MA), hypaconitine (HA), are highly toxic. Their hydrolysates, such as benzoylaconine (BAC), benzoylmesaconine (BMA), benzoylhypaconine (BHA), aconine, and mesaconine, are considerably less toxic. Efflux transporters, including P-glycoprotein (P-gp), breast cancer resistance protein (BCRP), and multidrug resistance-associated protein isoform 2 (MRP2), act as a first line of defence and play key roles in toxicity prevention. The aim of the present study was to determine the role of efflux transporters in the transport of Aconitum alkaloi

Applications of Higenamine in pharmacology and medicine.

ETHNOPHARMACOLOGICAL RELEVANCE: Aconitum has been used as local and traditional medicines in many asian regions for the treatment of various diseases such as collapse, syncope, painful joints, oedema, bronchial asthma et al. Higenamine, a plant-based alkaloid, was initially isolated from Aconitum and identified as the active cardiotonic component of Aconitum. It has been tested as a candidate of pharmacologic stress agent in the detection of coronary artery diseases (CADs) and now researchers have just accomplished the phase III clinical studies successfully in China. Besides, a large number o

Aconitine outperforms mesaconitine and hypaconitine in triggering excessive mitophagy via lysosomal two-pore channels disruption in SH-SY5Y cells and zebrafish.

The genus Aconitum L. is widely used in the treatment of rheumatoid arthritis, tumors, and cardiovascular diseases due to its prominent pharmacological properties. However, increasing scientific attention has been directed toward its neurotoxicity. Diester-diterpenoid alkaloids (DDAs), such as aconitine (AC), mesaconitine (MA), and hypaconitine (HA), have been identified as the principal toxic constituents of Aconitum. Although disruption of calcium homeostasis has been demonstrated to mediate DDAs-induced neurotoxicity, the key neurotoxic components and their underlying molecular mechanisms r

Inhibition of the INa/K and the activation of peak INa contribute to the arrhythmogenic effects of aconitine and mesaconitine in guinea pigs.

Aconitine (ACO), a main active ingredient of Aconitum, is well-known for its cardiotoxicity. However, the mechanisms of toxic action of ACO remain unclear. In the current study, we investigated the cardiac effects of ACO and mesaconitine (MACO), a structurally related analog of ACO identified in Aconitum with undocumented cardiotoxicity in guinea pigs. We showed that intravenous administration of ACO or MACO (25 μg/kg) to guinea pigs caused various types of arrhythmias in electrocardiogram (ECG) recording, including ventricular premature beats (VPB), atrioventricular blockade (AVB), ventricula

Poisonous Piperidine Plants and the Biodiversity of Norditerpenoid Alkaloids for Leads in Drug Discovery: Experimental Aspects.

There are famous examples of simple (e.g., hemlock, Conium maculatum L.) and complex (e.g., opium poppy, Papaver somniferum L., Papaveraceae) piperidine-alkaloid-containing plants. Many of these are highly poisonous, whilst pepper is well-known gastronomically, and several substituted piperidine alkaloids are therapeutically beneficial as a function of dose and mode of action. This review covers the taxonomy of the genera Aconitum, Delphinium, and the controversial Consolida. As part of studying the biodiversity of norditerpenoid alkaloids (NDAS), the majority of which possess an N-ethyl group

Effect of herbal extracts on peripheral nerve regeneration after microsurgery of the sciatic nerve in rats.

BACKGROUND: Recent experimental studies using herbal extracts have shown the possibility of peripheral nerve regeneration. This study aimed to investigate the effects of herbal extracts on peripheral nerve regeneration in a rat sciatic nerve injury model. METHODS: A total of 53 rats were randomly assigned to a control group or one of four experimental groups. In all rats, the sciatic nerve was completely severed and microscopic epineural end-to-end neurorrhaphy was performed. Normal saline (2 mL) was topically applied to the site of nerve repair in the control group, whereas four different he

[Aconitine poisoning due to confusion of aconite leaves with lovage].

Due to a mix-up an older couple (69 years and 71 years) ate a meal with herbs from their garden that contained leaves of monkshood (Aconitum napellus). The monkshood plants grew close to the herbs in their garden. Both patients developed the typical symptoms of aconitine poisoning with paresthesia, hypotension and bradycardia. Over the course of time both developed cardiac arrhythmia with ventricular extrasystoles and required monitoring on the intensive care unit (ICU). The husband was more severely affected and needed catecholamines for successful treatment of hypotension (70/40 mmHg) and br

Quality and safety of Chinese herbal medicines guided by a systems biology perspective.

Chinese herbal medicines, often referred as Chinese materia medica (CMM), are comprised of a complex multicomponent nature. The activities are aimed at the system level via interactions with a multitude of targets in the human body. This review aims at the toxicity aspects of CMM and its preparations at the different steps of production; harvesting, processing and the final formulation. The historic perspective and today's issues of the safety of CMM are introduced briefly, followed by the descriptions of the toxic CMM in the current Chinese Pharmacopoeia (2005). Subsequently, several aspects

What kind of poison was used in "The Name of the Rose"?

In the novel "The Name of the Rose" by Umberto Eco, a fanatical monk laced the pages of a book with a poison, which led to the death of several monks in a medieval monastery. Based on modern toxicological knowledge, an attempt can be made to determine whether there exists a substance meeting the criteria of the poison described in the novel. To this end, toxicological literature on the lethal doses of plant-derived poisons and their duration of action was reviewed. Cowbane (Cicuta virosa), a plant used for preparing poisonous potions, contains cicutoxin which kills at a dose of 500 mg. Similar

A minimal four-enzyme suite enables biosynthesis of the C20 diterpenoid alkaloid atisinium.

Diterpenoid alkaloids (DAs) are pharmaceutically important metabolites and mainly occur in the Ranunculaceae. Despite their structural complexity, all DAs originate from C20 diterpenoid scaffolds, making the elucidation of C20-DA biosynthesis pivotal for understanding and engineering this entire alkaloid class. However, the molecular basis for DA core assembly has remained unresolved despite decades of research. Here, we define a minimal four-enzyme suite comprising three cytochrome P450 monooxygenases (CYP701A216, CYP71FH3, and CYP729G3) and one neo-functionalized short-chain dehydrogenase/re

Identification of antagonistic fungi and their antifungal activities against aconite root rot pathogens.

Root rot is one of the main diseases affecting Aconitum carmichaelii Debx. during cultivation, seriously limiting yields of this herb. Currently, there is no effective control measure for aconite root rot. The antifungal activities of antagonistic strains against aconite root rot pathogens (Fusarium proliferatum, Fusarium solani, and Fusarium oxysporum) were investigated in this study. Three antagonistic strains, JKT7, JKT28 and JKT39, were screened and identified as Trichoderma asperellum, Trichoderma hamatum and Trichoderma virens, respectively. Dual culture tests showed that the inhibition

Quantitative analysis of aconitine in body fluids in a case of aconitine poisoning.

Aconitine belongs to the Aconitum alkaloids and is a natural toxic substance. Aconitine has been used as a traditional medicine in East Asian culture. Today, aconitine is still in use with or without a prescription, in the Republic of Korea. Here we present a case report of accidental death due to acute aconitine poisoning. An 81-year-old woman ingested liquid that had been heat extracted from the root of the Aconitum plant; she presented to the emergency room 1 h after ingestion. Her electrocardiogram showed irregular ventricular arrhythmias including ventricular tachycardia; she progressed t
