Augmentation and suppression of action potentials by estradiol in the myometrium of pregnant rat.

The purpose of this study was to investigate the actions of estradiol on spontaneous and evoked action potentials in the isolated longitudinal smooth muscle cells of the pregnant rat. Single cells were obtained by enzymatic digestion from pregnant rat longitudinal myometrium. Action potentials and currents were recorded by whole-cell current-clamp and voltage-clamp methods, respectively. The acute effects of 17beta-estradiol on action potentials and inward and outward currents were investigated. The following results were obtained. The average resting membrane potential of single myometrial ce

Action potentials and relations to the theta rhythm of medial septal neurons in vivo.

The influence of the medial septal nucleus and the nucleus of the diagonal band of Broca (MS-DB) on the hippocampal theta rhythm includes both cholinergic and gamma-aminobutyric acid (GABAergic) components. To understand the intrinsic septal interactions and the separate contributions of the cholinergic and GABAergic septohippocampal neurons to the theta rhythm in behaving animals, it is essential to be able to identify these two classes from extracellular recordings. Here the durations of extracellularly recorded action potentials are compared with the other characteristics of the neurons. Ex

Orthodromic sensory action potentials from palmar stimulation in the diagnosis of carpal tunnel syndrome.

It is known that the major slowing of nerve conduction in the carpal tunnel syndrome occurs in the palm to wrist segment. A technique exploiting this fact and easily applicable in the routine laboratory would be useful in diagnosing early examples of the condition. To this end, orthodromic sensory potentials from stimulation of digits II and V, and of the palmar branches of the median and ulnar nerves were recorded at the wrist in 53 healthy hands, 72 hands with a tentative diagnosis of carpal tunnel syndrome and in 20 hands of patients with unrelated neurological conditions. Using conventiona

The characteristics of spontaneous and evoked action potentials recorded from the rabbit's uterine nerves.

1. A method is described for recording in vivo the action potentials of afferent and efferent fibres in whole nerves supplying the rabbit's uterus.2. Examination of these nerves under the light microscope and the electron microscope showed them to be composed almost entirely of non-myelinated fibres.3. Two types of spontaneous action potential were observed; one travelled at about 4 m/sec and probably came from the myelinated fibres, the other travelled at 0.4-1.4 m/sec and certainly came from non-myelinated fibres.4. The efferent fibre spikes were shown to be faster and higher than the spikes

Acoustically Evoked Compound Action Potentials Recorded From Cochlear Implant Users With Preserved Acoustic Hearing.

OBJECTIVES: Less traumatic intracochlear electrode design and the introduction of the soft surgery technique allow for the preservation of low-frequency acoustic hearing in many cochlear implant (CI) users. Recently, new electrophysiologic methods have also been developed that allow acoustically evoked peripheral responses to be measured in vivo from an intracochlear electrode. These recordings provide clues to the status of peripheral auditory structures. Unfortunately, responses generated from the auditory nerve (auditory nerve neurophonic [ANN]) are somewhat difficult to record because they

Scorpion venom increases acetylcholine release by prolonging the duration of somatic nerve action potentials.

Scorpionism is frequently accompanied by a massive release of catecholamines and acetylcholine from peripheral nerves caused by neurotoxic peptides present in these venoms, which have high specificity and affinity for ion channels. Tityus bahiensis is the second most medically important scorpion species in Brazil but, despite this, its venom remains scarcely studied, especially with regard to its pharmacology on the peripheral (somatic and autonomic) nervous system. Here, we evaluated the activity of T. bahiensis venom on somatic neurotransmission using myographic (chick and mouse neuromuscula

Action-associated modulation of visual event-related potentials evoked by abstract and ecological stimuli.

This study investigated the influence of action-associated predictive processes on visual ERPs. In two experiments, we sought evidence for sensory attenuation (SA) indexed by ERP amplitude reductions for self-induced stimuli when compared to passive viewing of the same images. We assessed if SA is (a) present for both ecological and abstract stimuli (pictures depicting hands or checkerboards), (b) modulated by the degree of stimulus predictability (certain or uncertain action-effect contingencies), and (c) sensitive to laterality of hand movements (dominant or subdominant hand actions). We fou

The Venus Flytrap Dionaea muscipula Counts Prey-Induced Action Potentials to Induce Sodium Uptake.

Carnivorous plants, such as the Venus flytrap (Dionaea muscipula), depend on an animal diet when grown in nutrient-poor soils. When an insect visits the trap and tilts the mechanosensors on the inner surface, action potentials (APs) are fired. After a moving object elicits two APs, the trap snaps shut, encaging the victim. Panicking preys repeatedly touch the trigger hairs over the subsequent hours, leading to a hermetically closed trap, which via the gland-based endocrine system is flooded by a prey-decomposing acidic enzyme cocktail. Here, we asked the question as to how many times trigger h

The effects of anticholinesterases on the latencies of action potentials in mouse skeletal muscles.

1. The purpose of this investigation was to determine the long-term effects of a single dose of persistent anticholinesterases on muscle action potentials evoked by nerve stimulation. 2. Action potentials (APs), elicited by stimulation of the phrenic nerve, were recorded intracellularly in muscle fibres of mouse diaphragm. The time between stimulus and AP was measured and the variability of this latency was calculated during trains of APs. At the beginning of trains of APs there was an increase in latency, and this delay was also measured. 3. Within 3 h of subcutaneous injection, a single dose

Nerve conduction velocity distributions: a method for estimation based upon two compound action potentials.

A model was presented describing the nerve-bundle CAP in terms of its constituent SFAPs. The model has a general structure capable of accommodating a large variety of specific assumptions regarding the nature of the CAP. Using this model, several techniques were presented for estimating the DCV of a nerve bundle. One method, refered to as the 2CAP method, was presented in detail. This method does not require explicit knowledge of the SFAP waveshapes in order to yield an estimate of the DCV, but does require that the CAP be recorded at two sites along the course of the nerve. In order to evalua

Digital nerve action potentials in healthy subjects, and in carpal tunnel and diabetic patients.

A technique is described for stimulating and recording from nerves in the finger using surface electrodes. A decrease in amplitude and velocity was found with increasing age. In control subjects the digital potential was approximately one and a half times larger than the potential recorded at the wrist. In patients with carpal tunnel syndrome there was some reduction in amplitude and velocity of the digital potential, but the changes were more marked at the wrist. In diabetic patients more uniform changes were found in the two segments. The technique was particularly useful in enabling conduct

Antiprogestins: mechanism of action and contraceptive potential.

Antiprogestins are characterized by substitutions at the 11 beta and 17 alpha positions of the steroid ring system and bind strongly to both progesterone and glucocorticoid receptors. Although they function predominantly as antiprogestins and antiglucocorticoids, on occasion they display progestin agonistic and even antiestrogenic properties. The most common clinical use of the antiprogestin mifepristone is to induce a medical abortion in the early stages of pregnancy. Progesterone maintains the endometrium, transforming it from a proliferative to a secretory state. It also facilitates the lut

Mechanism of action and thrombolytic potential of staphylokinase.

Staphylokinase is a profibrinolytic agent that forms a 1:1 stoichiometric complex with plasminogen that, after conversion to plasmin, activates other plasminogen molecules to plasmin. The plasmin.staphylokinase complex, unlike the plasmin.streptokinase complex, is rapidly inhibited by alpha2-antiplasmin. In a plasma milieu, staphylokinase is able to dissolve fibrin clots without associated fibrinogen degradation. This fibrin-specificity of staphylokinase is the result of reduced inhibition by alpha2-antiplasmin of plasmin.staphylokinase complex bound to fibrin, recycling of staphylokinase from

Potentiation of the antidepressant action of clomipramine by tryptophan.

In a double-blind study of 24 patients with endogenous depressiona group treated with clomipramine hydrochloride (chlorimipramine) plus tryptophan was compared with a group treated with clomipramine plus placebo. The sum of the ratings for depressed mood, suicidal intent, depressive thought content, and anxiety showed a more rapid improvement in the former group, the difference being already significant after 12 days of treatment. On the other hand, the ratings for retardation decreased about equally in both groups during the three-week treatment period. Side-effect ratings showed no significa
