Enhanced rAAV Production in HEK293 Cells by Promoting Helper Plasmid Nuclear Import Under HDAC6 Inhibition.

Recombinant adeno-associated virus (rAAV) vectors have emerged as leading platforms for gene therapy owing to their safety, long-term transgene expression, and broad tissue tropism. However, rAAV production in HEK293 cells via triple-plasmid transfection remains limited by low yield, partly due to inefficient nuclear import of plasmids. In this study, we enhanced rAAV production by promoting plasmid nuclear delivery. DNA-targeting sequence (DTS) insertion alone did not improve plasmid nuclear import or rAAV yield, whereas combining DTS insertion with tubacin, an HDAC6 inhibitor, significantly

Compartmentalised Function of TaHRC-R Orchestrates ROS Production and Nuclear Condensate Dynamics.

Fhb1 is the most widely used locus for Fusarium head blight (FHB) resistance in wheat, yet the mechanistic basis of its candidate gene, TaHRC, remains elusive. Here, we demonstrate that the protein from the resistant allele TaHRC-R localises to both the nucleus and cytoplasm, whereas the susceptible protein TaHRC-S is confined to the nucleus. Remarkably, only TaHRC-R triggered a reactive oxygen species (ROS) burst in planta, dependent on its extranuclear localisation. The N-terminal 21 amino acids that distinguish TaHRC-R from TaHRC-S were essential for its nuclear export and ROS induction. Wi

The nuclear import receptor importin-7 targets HPV from the Golgi to the nucleus to promote infection.

During human papillomavirus (HPV) entry, the virus exploits COPI-dependent retrograde transport to cross the Golgi apparatus before reaching the nucleus to cause infection. How HPV enters the nucleus after exiting the Golgi is unclear, although mitotic nuclear envelope breakdown (NEB) appears important. Here, we show that importin-7 (IPO7), a nuclear pore import receptor, is present at the Golgi and promotes HPV infection. IPO7 knockdown inhibits infection and causes HPV to accumulate in the Golgi without reaching mitotic chromosomes, demonstrating that IPO7 promotes Golgi-to-nucleus transport

The varicella-zoster virus ORF16 protein promotes both the nuclear transport and the protein abundance of the viral DNA polymerase subunit ORF28.

Although all herpesviruses utilize a highly conserved replication machinery to amplify their viral genomes, different members may have unique strategies to modulate the assembly of their replication components. Herein, we characterize the subcellular localization of seven essential replication proteins of varicella-zoster virus (VZV) and show that several viral replication enzymes such as the DNA polymerase subunit ORF28, when expressed alone, are localized in the cytoplasm. The nuclear import of ORF28 can be mediated by the viral DNA polymerase processivity factor ORF16. Besides, ORF16 could

RETRACTED: Regions of Bovine Adenovirus-3 Protein VII Involved in Interactions with Viral and Cellular Proteins.

Источник пометил публикацию как отозванную

The L 1 region of bovine adenovirus (BAdV)-3 encodes a multifunctional protein named protein VII. Anti-protein VII sera detected a protein of 26 kDa in transfected or BAdV-3-infected cells, which localizes to nucleus and nucleolus of infected/transfected cells. Analysis of mutant protein VII identified four redundant overlapping nuclear/nucleolar localization signals as deletion of all four potential nuclear/nucleolar localization signals localizes protein VII predominantly to the cytoplasm. The nuclear import of protein VII appears to use importin α (α-1), importin-β (β-1) and transportin-3 n

Reversible mislocalization of a disease-associated MRE11 splice variant product.

Ataxia-telangiectasia (AT) and related disorders feature cancer predisposition, neurodegeneration, and immunodeficiency resulting from failure to respond to DNA damage. Hypomorphic mutations in MRE11 cause an AT-like disorder (ATLD) with variable clinical presentation. We have sought to understand how diverse MRE11 mutations may provide unique therapeutic opportunities, and potentially correlate with clinical variability. Here we have undertaken studies of an MRE11 splice site mutation that was found in two ATLD siblings that died of pulmonary adenocarcinoma at the young ages of 9 and 16. The

SUN5, a testis-specific nuclear membrane protein, participates in recruitment and export of nuclear mRNA in spermatogenesis.

SUN5, a testis-specific gene, is associated with acephalic spermatozoa syndrome (ASS). Here, we demonstrate that SUN5 is involved in mRNA export. In Sun5-knockout mice ( Sun5 -/-), poly(A) + RNA accumulates in the nuclei of germ cells, leading to reduced sperm counts, decreased sperm motility and disrupted sperm head-to-tail junctions. Additionally, in the GC-2 germ cell line with RNA interference of Sun5, heterogeneous nuclear ribonucleoproteins (hnRNPs) and poly (A) + RNA (mainly mRNA) are retained in the nucleus. Further mechanistic studies reveal that SUN5 interacts with Nxf1 (nuclear RNA

Subcellular Localization of MxB Determines Its Antiviral Potential against Influenza A Virus.

Mx proteins are interferon (IFN) type I (α/β)- and type III (λ)-induced effector proteins with intrinsic antiviral activity. Mammalian Mx proteins show different subcellular localizations and distinct yet partially overlapping viral specificities. However, the precise mechanism(s) of antiviral action are still unresolved. Human MxA accumulates in the cytoplasm and inhibits a wide variety of RNA and DNA viruses, among them influenza A virus (IAV). In contrast, MxB, the second human Mx protein, localizes via its amino (N) terminus to the outer nuclear membrane at or near nuclear pores and inhibi

Impaired Nuclear Export of Polyglutamine-Expanded Androgen Receptor in Spinal and Bulbar Muscular Atrophy.

Spinal and bulbar muscular atrophy (SBMA) is a neuromuscular disease caused by polyglutamine (polyQ) expansion in the androgen receptor (AR). Prior studies have highlighted the importance of AR nuclear localization in SBMA pathogenesis; therefore, in this study, we sought to determine the role of AR nuclear export in the pathological manifestations of SBMA. We demonstrate here that the nuclear export of polyQ-expanded AR is impaired, even prior to the formation of intranuclear inclusions of aggregated AR. Additionally, we find that promoting AR export with an exogenous nuclear export signal su

Nucleolar and coiled-body phosphoprotein 1 enhances the induction of CYP3A4 by promoting nuclear translocation of pregnane X receptor.

Pregnane X receptor (PXR) is a xenobiotic-sensing nuclear receptor that plays a central role in the regulation of drug-metabolizing enzymes and transporters. Under basal conditions, PXR is predominantly localized in the cytoplasm. Upon activation by ligands such as rifampicin, PXR translocates into the nucleus and induces the transcription of its target genes, including CYP3A4, a major drug-metabolizing enzyme in the human liver. From our previous proteomic analysis of coimmunoprecipitated proteins obtained from lysates of rifampicin-treated ShP51 cells (a HepG2 cell line stably expressing hum

Compensative shuttling of merlin to phosphorylation on serine 518 in vestibular schwannoma.

BACKGROUND: Vestibular schwannoma (VS) is a benign tumor with malignant biological consequence because of its special location, and its occurrence is highly related to merlin. Cell culture studies have demonstrated that merlin acts as a molecular linker between the cytoskeleton and specific membrane proteins and is linked to cell cycle control. Therefore, we sought to detect the expression of endogenous merlin and its subcellular distribution using cyclin D1 as a cell cycle marker in VS. METHODS: To confirm endogenous merlin protein expression in abiogenous VS cells, in situ hybridization, im

NXT2 is a key component of the RNA nuclear export factor complex in the human testis and essential for spermatogenesis.

In eukaryotes, the nucleocytoplasmic export of bulk poly(A)+-mRNAs through the nuclear pore complex is mediated by the ubiquitously expressed NXT1-NXF1 heterodimer. In humans, NXT1 has an X-chromosomal paralog, NXT2, which exhibits testis-enriched expression, suggesting a role in spermatogenesis. Here, we report the in vivo interaction of NXT2 with crucial components of the nuclear export machinery, including NXF1, the testis-specific NXF1 paralogs NXF2 and NXF3, and nuclear pore complex proteins. Binding to NXF2 and NXF3 is mediated by the NTF2-like domain of NXT2. By identifying infertile me

Molecular mechanism of activation and nuclear translocation of the mineralocorticoid receptor upon binding of pregnanesteroids.

The mineralocorticoid receptor (MR) is primarily localized in the cytoplasm of the cell in the absence of ligand. The first step in the genomic-dependent mechanism of action of mineralocorticoids is the binding of steroid to the MR, which in turn triggers MR nuclear translocation. The regulation of hormone-binding to MR is complex and involves a multifactorial mechanism, making it difficult to determine the optimal structure of a steroid for activating the MR and promoting its nuclear translocation. Here we review the structure-activity relationship for several pregnanesteroids that possess va

Spatiotemporal regulation of DNA repair proteins between Golgi and nucleus maintains genome stability.

The Golgi complex serves as a critical hub for cellular homeostasis, yet its communication with the nucleus remains largely unexplored. By analyzing and siRNA-validating localization data from the Human Protein Atlas, we uncovered substantial proteome interconnectivity between the Golgi and nucleus, including an unexpected enrichment for DNA repair factors. We identify a cluster of DNA damage response (DDR) proteins occupying distinct sub-Golgi compartments that redistribute dynamically between the Golgi and nucleus in response to genotoxic stress, with the type of DNA lesion shaping the direc

Kap-centric Nsp1-mediated nuclear transport at full amino acid resolution.

Recent studies of nuclear pore complexes (NPCs) have provided detailed descriptions of the core scaffold structures, yet fall short in resolving the dynamic FG meshwork with similar precision. Here, we present an amino acid resolution model that enables full-scale simulations of selective transport through the NPC. We map the spatial distributions and nanosecond dynamics of individual FG-Nups in the central transporter, revealing that the bimodal architecture of Nsp1 forms a dynamic central meshwork essential for regulating both passive and active transport. Incorporating nuclear transport rec

Visualizing developmental dynamics of nuclear morphology and transport machinery in Drosophila.

Communication between the cytoplasm and the nucleus requires a continuous exchange of molecules across the nuclear envelope (NE). The nuclear pore complex (NPC) is the gateway embedded in the NE through which cargo moves, while transport receptors mediate the passage of macromolecules through the NPC. Although their essential role as the components of the nuclear transport machinery has been extensively studied, how these factors respond to developmental and environmental cues has been underexplored. Here we tag the nucleoporin Nup96 and the transport receptor Impβ with mEGFP and mScarlet-I at

Regulation of glypican 6-mediated Wnt activation maintains TDP-43 nuclear localization in neurons.

Abnormalities in TDP-43 (Transactive response DNA-binding protein 43 kDa) localization and function span multiple neurodegenerative diseases and are implicated in driving neuronal degeneration and loss. Nuclear pore complex (NPC) abnormalities and disrupted nucleocytoplasmic trafficking (NCT) contribute to TDP-43 mislocalization, but how these cellular changes are initiated in disease is unclear. Glycerophosphodiester phosphodiesterase 2 (GDE2) is a surface glycosylphosphatidylinositol (GPI)-anchor cleaving enzyme that encodes a physiological pathway that ensures NPC integrity, appropriate NCT

Nuclear localization of mouse Mx1 protein is necessary for inhibition of influenza virus.

The interferon-induced Mx1 protein of mice confers selective resistance to influenza virus. It inhibits viral mRNA synthesis in the nucleus of influenza virus-infected cells. The related human MxA protein is localized in the cytoplasm and can inhibit influenza virus and vesicular stomatitis virus but not other viruses. MxA blocks a poorly defined cytoplasmic multiplication step of influenza virus that follows primary transcription of the viral genome. We previously showed that nuclear variants of MxA that carry an artificial nuclear translocation signal were also active against influenza virus

Organization of projections from the raphe nuclei to the vestibular nuclei in rats.

Previous anatomic and electrophysiological evidence suggests that serotonin modulates processing in the vestibular nuclei. This study examined the organization of projections from serotonergic raphe nuclei to the vestibular nuclei in rats. The distribution of serotonergic axons in the vestibular nuclei was visualized immunohistochemically in rat brain slices using antisera directed against the serotonin transporter. The density of serotonin transporter-immunopositive fibers is greatest in the superior vestibular nucleus and the medial vestibular nucleus, especially along the border of the four

Piwi Is a Key Regulator of Both Somatic and Germline Stem Cells in the Drosophila Testis.

The Piwi-piRNA pathway is well known for its germline function, yet its somatic role remains elusive. We show here that Piwi is required autonomously not only for germline stem cell (GSC) but also for somatic cyst stem cell (CySC) maintenance in the Drosophila testis. Reducing Piwi activity in the testis caused defects in CySC differentiation. Accompanying this, GSC daughters expanded beyond the vicinity of the hub but failed to differentiate further. Moreover, Piwi deficient in nuclear localization caused similar defects in somatic and germ cell differentiation, which was rescued by somatic P
