Recent advances in the use of adapalene 0.1%/benzoyl peroxide 2.5% to treat patients with moderate to severe acne.

The central role of inflammation in acne is now more clearly understood. Adapalene, a third-generation topical retinoid, down-regulates toll-like receptor 2 expression and inhibits activator protein-1 activity. In a fixed-dose combination, adapalene and benzoyl peroxide (BPO) act synergistically on inflammatory patterns through regulation of innate immunity. In addition to reducing inflammatory and non-inflammatory lesions, adapalene/BPO helps prevent lesion and microcomedone formation. The combination of a topical retinoid and antimicrobial agent remains the preferred approach for almost all

In Silico, In Vitro, and In Vivo Investigations on Adapalene as Repurposed Third Generation Retinoid against Multiple Myeloma and Leukemia.

The majority of hematopoietic cancers in adults are incurable and exhibit unpredictable remitting-relapsing patterns in response to various therapies. The proto-oncogene c-MYC has been associated with tumorigenesis, especially in hematological neoplasms. Therefore, targeting c-MYC is crucial to find effective, novel treatments for blood malignancies. To date, there are no clinically approved c-MYC inhibitors. In this study, we virtually screened 1578 Food and Drug Administration (FDA)-approved drugs from the ZINC15 database against c-MYC. The top 117 compounds from PyRx-based screening with th

Clindamycin Phosphate 1.2%/Adapalene 0.15%/ Benzoyl Peroxide 3.1% for Acne: Results From A 6-Month Open-Label Study.

BACKGROUND: Treatment of acne may require many months of treatment before maximal benefits are observed, and acne sequelae (eg, scarring, dyspigmentation) can persist long after lesion resolution. In 12-week clinical trials, triple-combination clindamycin phosphate 1.2%/adapalene 0.15%/benzoyl peroxide 3.1% gel (CAB) demonstrated efficacy and tolerability in the treatment of moderate to severe acne. This study assessed CAB long-term efficacy/tolerability and reductions in acne scarring/dyspigmentation. METHODS: This 24-week, single-center, open-label study assessed once-daily CAB in participa

Dendrobium polysaccharide-based microneedles loaded with Celosia cristata flavonoids and adapalene nanoparticles for efficacious treatment of acne vulgaris.

Acne vulgaris is a common chronic inflammatory skin disorder primarily caused by the overgrowth of Propionibacterium acnes (P. acnes). However, the therapeutic efficacy of traditional drugs is often limited due to their inability to penetrate the stratum corneum. Microneedles (MNs) are designed to penetrate the stratum corneum, enabling direct drug delivery to the epidermis. In this study, a new Dendrobium polysaccharide-based composite microneedle (DOP/CCF/PLGA@Adap-MN) delivery system was developed. Dendrobium polysaccharide (DOP) was utilized as the microneedle matrix, loaded with the Celos

Assessing Cutibacterium Acnes Susceptibility With Clindamycin Phosphate 1.2%/Adapalene 0.15%/Benzoyl Peroxide 3.1% Gel Use: Six-Month Analysis.

BACKGROUND: The only approved triple-combination acne treatment – clindamycin phosphate 1.2%/adapalene 0.15%/benzoyl peroxide 3.1% (CAB) gel - demonstrated efficacy/safety in 12-week clinical trials. However, real-world treatment may require 6 months for maximum benefits in some cases. Since long-term antibiotic use can lead to resistance in the causative Cutibacterium acnes (C. acnes), this analysis evaluated the effect of long-term CAB use on C. acnes. METHODS: Pooled data from 2 identical, 24-week, single-center, open-label studies evaluated once-daily CAB in participants aged ≥12

Antifungal Monotherapy in Acne: Topical Ketoconazole 2% Cream Versus Adapalene Gel in Treating Acne Vulgaris: A Comparative Study.

BACKGROUND: Acne is a widely prevalent disorder, with a largely negative impact on the patient's quality of life. Bacterial etiology and the role of Cutibacterium acne have always been incriminated in in acne pathogenesis. However, in the last decade, deeper understanding of skin microbiome postulates a possible role of other pathogens like Malassezia spp. ketoconazole has shown efficacy to both pathogens. OBJECTIVES: To study the efficacy of topical ketoconazole 2% in the treatment of acne vulgaris (AV) compared to the standard treatment with adapalene. METHODS: Fifty-eight acne patients in
