Unravelling the function of prdm16 in human tumours: A comparative analysis of haematologic and solid tumours.

Extensive research has shown that PR domain 16 (PRDM16) plays a critical role in adipose tissue metabolism, including processes such as browning and thermogenesis of adipocytes, beigeing of adipocytes, and adipogenic differentiation of myoblasts. These functions have been associated with diseases such as obesity and diabetes. Additionally, PRDM16 has been correlated with various other conditions, including migraines, heterochromatin abnormalities, metabolic syndrome, cardiomyopathy, sarcopenia, nonsyndromic cleft lip, and essential hypertension, among others. However, there is currently no sys

Meta-Analysis of Porcine Beige Adipogenesis with Cross-Species CD137-CD137L Molecular Docking: Potential Implications for Pig-to-Human Xenotransplantation.

Breakthroughs in gene-editing technologies have brought pig-to-human xenotransplantation to the clinical forefront. Nevertheless, the micro-scale impacts of the complex pathophysiological milieu within recipients on grafts have long been overlooked by the academic community. Focusing on stromal vascular fraction (SVF) cells and preadipocytes widely present in donor organs, this study explores the potential mechanism underlying beige adipogenesis induced by post-operative anti-inflammatory drugs (e.g., dexamethasone) and surgical stress (norepinephrine), as well as its possible relationship wit

The AdipoChaser mouse: A model tracking adipogenesis in vivo.

White adipose tissue is considered to have high plasticity. Dynamic, abnormal expansion of white adipose tissue leads to obesity and associated metabolic disorders. Due to technical limitations, the life cycle and turnover rates of different white adipose depots during development and under various physiological conditions and environmental challenges has been assessed only through highly indirect approaches. We have recently described a system for the inducible, permanent labeling of mature adipocytes, the "AdipoChaser" mouse. Utilizing this AdipoChaser mouse model, we found that epididymal f

The Rosmarinus Bioactive Compound Carnosic Acid Is a Novel PPAR Antagonist That Inhibits the Browning of White Adipocytes.

Thermogenic brown and brite adipocytes convert chemical energy from nutrients into heat. Therapeutics that regulate brown adipocyte recruitment and activity represent interesting strategies to control fat mass such as in obesity or cachexia. The peroxisome proliferator-activated receptor (PPAR) family plays key roles in the maintenance of adipose tissue and in the regulation of thermogenic activity. Activation of these receptors induce browning of white adipocyte. The purpose of this work was to characterize the role of carnosic acid (CA), a compound used in traditional medicine, in the contro

Regulatory microRNAs in Brown, Brite and White Adipose Tissue.

MicroRNAs (miRNAs) constitute a class of short noncoding RNAs which regulate gene expression by targeting messenger RNA, inducing translational repression and messenger RNA degradation. This regulation of gene expression by miRNAs in adipose tissue (AT) can impact on the regulation of metabolism and energy homeostasis, particularly considering the different types of adipocytes which exist in mammals, i.e., white adipocytes (white AT; WAT), brown adipocytes (brown AT; BAT), and inducible brown adipocytes in WAT (beige or brite or brown-in-white adipocytes). Indeed, an increasing number of miRNA

Magnolol promotes thermogenesis and attenuates oxidative stress in 3T3-L1 adipocytes.

OBJECTIVE: The aim of this study was to explore the browning and antioxidative effects of magnolol in 3T3-L1 adipocytes, as recruitment of beige-like adipocytes (browning) by natural compounds is being considered as a promising strategy to fight against obesity. METHODS: Magnolol-induced browning effect was evaluated by determining the expression levels of specific marker genes and proteins using real-time polymerase chain reaction and immunoblotting, respectively. Induction of thermogenesis and suppression of oxidative stress in 3T3-L1 adipocytes were further validated by immunofluorescence.

UCP1-Dependent Thermogenic Adipose Tissue in Human Disease: Adipose-Centered Mechanisms, Biomarker Limitations, and Translational Perspectives.

Uncoupling protein 1 (UCP1) is a mitochondrial inner-membrane carrier classically recognized as the molecular effector of non-shivering thermogenesis in brown adipose tissue. By dissipating the proton-motive force generated by oxidative phosphorylation, UCP1 converts stored chemical energy into heat and enables adaptive thermogenesis during cold exposure. The rediscovery of metabolically active brown adipose tissue (BAT) and inducible beige adipocytes in adult humans has renewed interest in UCP1-positive thermogenic adipose tissue as a regulator of systemic metabolism and a potential target fo

Adaptation of White Adipocytes to Cooler Temperatures: Impacts on Energy Metabolism and Protein Acetylation.

Adipocytes throughout the body reside in distinct thermal environments. Visceral adipocytes within the body core are maintained near 37°C, whereas those in bone marrow, subcutaneous, and dermal depots occupy cooler regions within the peripheral shell. Although brown and beige adipocyte responses to cold stress are well characterized, much less is known about how white adipocytes adapt to moderately reduced temperatures below 37°C. Our recent work revealed that cultured adipocytes exposed to 31°C, a temperature representative of distal adipose regions, exhibit enhanced mitochondrial function, i

Mice lacking triglyceride synthesis enzymes in adipose tissue are resistant to diet-induced obesity.

Triglycerides (TGs) in adipocytes provide the major stores of metabolic energy in the body. Optimal amounts of TG stores are desirable as insufficient capacity to store TG, as in lipodystrophy, or exceeding the capacity for storage, as in obesity, results in metabolic disease. We hypothesized that mice lacking TG storage in adipocytes would result in excess TG storage in cell types other than adipocytes and severe lipotoxicity accompanied by metabolic disease. To test this hypothesis, we selectively deleted both TG synthesis enzymes, DGAT1 and DGAT2, in adipocytes (ADGAT DKO mice). As expected

Circular RNA circINSR regulates bovine intramuscular adipocyte development and lipid accumulation through insulin receptor signaling.

Intramuscular fat, deposited as marbling in beef, is a key economic indicator of meat quality in cattle production. A series of circular RNAs (circRNAs) have emerged as critical regulators of lipogenesis. In this study, we identified 373 differentially expressed circRNAs between calves and adult cattle, among which circINSR was markedly upregulated in adults. Single-nucleus RNA sequencing (snRNA-seq) analysis revealed the developmental trajectory of intramuscular adipocytes, demonstrating the differentiation of preadipocytes into both white and beige adipocytes. Along the preadipocyte-to-white

The pro-tumorigenic functions of cancer-associated adipocytes are dependent on the mitochondrial chaperone tumor necrosis factor receptor-associated protein 1.

Adipocytes are essential stromal components of the tumor microenvironment (TME) in breast cancer that play pivotal roles in cancer progression and chemoresistance. In close proximity to tumor cells, they undergo phenotypic reprogramming into cancer-associated adipocytes (CAAs), characterized by multilocular lipid droplets, increased mitochondrial content, and elevated expression of uncoupling protein 1 (UCP1). Although these features superficially resemble those of beige adipocytes, they do not recapitulate classical thermogenic programming, reflecting a unique metabolic adaptation driven by t

Clock Modulation by Naringenin Suppresses Lipogenesis and Promotes Adipose Tissue Browning.

The circadian clock orchestrates adipocyte development and lipid remodeling, with its disruption leading to the development of obesity and insulin resistance. Here we demonstrate that the flavonoid compound naringenin displays clock modulatory activity that suppresses adipocyte lipid storage while promoting browning. In adipogenic progenitors, naringenin activates RORα with induction of clock gene expression to promote circadian clock oscillation with protective effect against cytokine-induced dampening. The clock-enhancing properties of naringenin suppressed lipogenesis in mature adipocytes t

Latest Advances in Obesity Treatment by Regulating Adipocyte Thermogenesis.

Obesity is driven by chronic energy imbalance and is associated with multiple metabolic diseases. As adipocyte thermogenesis contributes to energy expenditure, it is being actively investigated as a possible target for obesity treatment. This review summarizes recent progress in the molecular regulation of adipocyte thermogenesis, focusing mainly on preclinical mechanistic evidence from cellular and animal studies, while discussing clinical and human data only where available. We review emerging regulators within the cAMP pathway, the PRDM16-PPARγ axis, and mitochondrial quality control and me

HuR-dependent expression of RyR2 contributes to calcium-mediated thermogenesis in murine brown adipocytes.

Several thermogenic pathways have been described in adipose tissue, including calcium-mediated thermogenesis in beige adipocytes. We have previously shown that adipocyte-specific deletion of the RNA binding protein human antigen R (HuR; Elavl1) results in thermogenic dysfunction. RNA sequencing revealed the downregulation of several calcium transport genes upon HuR deletion. The goal of this work was to define the HuR-dependent mechanisms of calcium driven thermogenesis in brown adipocytes. Brown adipose tissue (BAT)-specific HuR-deletion (BAT-HuR-/-) mice have no changes in their body weight,

Adipose-specific ATGL ablation reduces burn injury-induced metabolic derangements in mice.

Hypermetabolism following severe burn injuries is associated with adipocyte dysfunction, elevated beige adipocyte formation, and increased energy expenditure. The resulting catabolism of adipose leads to detrimental sequelae such as fatty liver, increased risk of infections, sepsis, and even death. While the phenomenon of pathological white adipose tissue (WAT) browning is well-documented in cachexia and burn models, the molecular mechanisms are essentially unknown. Here, we report that adipose triglyceride lipase (ATGL) plays a central role in burn-induced WAT dysfunction and systemic outcome

Energy's long game: thermogenic adipocytes and the metabolic diseases of modern life.

The ability to generate, store, and mobilize energy is fundamental to life, and disruptions in these processes underlie metabolic disease. This review examines how these mechanisms evolved to rely profoundly on lipids, a large family of molecules that serve an unusually wide range of biological functions. From early evolutionary times, lipids have enabled chemiosmotic energy transduction, organized the endomembrane systems that synthesize and package neutral lipids, served as concentrated reservoirs of metabolic energy, and functioned as signaling molecules that coordinate cellular and systemi

Molecular and Physiological Effects of Browning Agents on White Adipocytes from Bone Marrow Mesenchymal Stromal Cells.

Two different types of adipose depots can be observed in mammals: white adipose tissue (WAT) and brown adipose tissue (BAT). The primary role of WAT is to deposit surplus energy in the form of triglycerides, along with many metabolic and hormonal activities; as thermogenic tissue, BAT has the distinct characteristic of using energy and glucose consumption as a strategy to maintain the core body temperature. Under specific stimuli-such as exercise, cold exposure, and drug treatment-white adipocytes can utilize their extraordinary flexibility to transdifferentiate into brown-like cells, called b

Ablation of LGR4 promotes energy expenditure by driving white-to-brown fat switch.

Obesity occurs when excess energy accumulates in white adipose tissue (WAT), whereas brown adipose tissue (BAT), specialized for energy expenditure through thermogenesis, potently counteracts obesity. Factors that induce brown adipocyte commitment and energy expenditure would be a promising defence against adiposity. Here, we show that Lgr4 homozygous mutant (Lgr4(m/m)) mice show reduced adiposity and resist dietary and leptin mutant-induced obesity with improved glucose metabolism. Lgr4(m/m) mice show a striking increase in energy expenditure, and exhibit brown-like adipocytes in WAT depots w

Hematopoietic stem cell-derived adipocytes suppress leptin production, and attenuate ovariectomy-induced inhibition of physical activity and insulin sensitivity in female mice.

A subpopulation of adipocytes in mice and humans is produced from haematopoietic stem cells rather than mesenchymal progenitors; the source of conventional white and brown/beige adipocytes. The abundance of these haematopoietic stem cell-derived adipocytes (HSCDAs) is elevated in female mice by ovariectomy (OVX) or oestrogen receptor alpha (ERα) knockdown, suggesting that they may be involved in the metabolic and inflammatory pathology that accompany the loss of oestrogen signalling. However, we previously demonstrated that ablation of HSCDAs elevated circulating leptin levels while suppressin

Bone Marrow Adipocytes as Novel Regulators of Metabolic Homeostasis: Clinical Consequences of Bone Marrow Adiposity.

PURPOSE OF REVIEW: Bone marrow adipose tissue is a distinctive fat depot located within the skeleton, with the potential to influence both local and systemic metabolic processes. Although significant strides have been made in understanding bone marrow adipose tissue over the past decade, many questions remain regarding their precise lineage and functional roles. RECENT FINDINGS: Recent studies have highlighted bone marrow adipose tissue's involvement in continuous cross-talk with other organs and systems, exerting both endocrine and paracrine functions that play a crucial role in metabolic ho
