Circadian-related heteromerization of adrenergic and dopamine D₄ receptors modulates melatonin synthesis and release in the pineal gland.

The role of the pineal gland is to translate the rhythmic cycles of night and day encoded by the retina into hormonal signals that are transmitted to the rest of the neuronal system in the form of serotonin and melatonin synthesis and release. Here we describe that the production of both melatonin and serotonin by the pineal gland is regulated by a circadian-related heteromerization of adrenergic and dopamine D₄ receptors. Through α(₁B)-D₄ and β₁-D₄ receptor heteromers dopamine inhibits adrenergic receptor signaling and blocks the synthesis of melatonin induced by adrenergic receptor ligands.

Leptin regulates bone formation via the sympathetic nervous system.

We previously showed that leptin inhibits bone formation by an undefined mechanism. Here, we show that hypothalamic leptin-dependent antiosteogenic and anorexigenic networks differ, and that the peripheral mediators of leptin antiosteogenic function appear to be neuronal. Neuropeptides mediating leptin anorexigenic function do not affect bone formation. Leptin deficiency results in low sympathetic tone, and genetic or pharmacological ablation of adrenergic signaling leads to a leptin-resistant high bone mass. beta-adrenergic receptors on osteoblasts regulate their proliferation, and a beta-adr

Effects of caerulein on the gastric motility of rats.

The effects of caerulein on gastric motility in urethane-anesthetized rats were studied. Caerulein administered into the lateral cerebral ventricle (i.c.v.) and jugular vein (i.v.) caused predominantly an inhibitory effect on gastric motility but sometimes an excitatory or a biphasic effect. The inhibitory response was reduced after vagotomy and/or splanchnicotomy, or after guanethidine. The remaining inhibitory response was abolished by tetrodotoxin, but was resistant to atropine and guanethidine. The excitatory response was abolished by atropine. Discharges of the gastric branch of the vagus

Determination of infundibular innervation and amine receptor content in cyanotic and acyanotic myocardium: relation to clinical events in tetralogy of Fallot.

Right ventricular myocardium was assessed for cholinergic and adrenergic innervation, as well as alpha-adrenergic, beta-adrenergic, and muscarinic receptors, in 18 cyanotic patients with tetralogy of Fallot (TOF) and four acyanotic control patients with ventricular septal defect, each of whom underwent a cardiac repair from June through December 1987. Neurons containing acetylcholine (ACH), neuron-specific enolase (NSE), S-100 protein, neuropeptide-Y (NPY), dopamine-beta-hydroxylase (DBH), and calcitonin gene-related polypeptide (CGRP) were detected surrounding arterioles and myocytes in all s

Effect of sialectomy on the superior cervical ganglion sympathetic neurons in young adult and aged mice.

The dependence of superior cervical ganglion (SCG) adrenergic neurons on their target organ submandibular salivary gland containing high concentrations of nerve growth factor was studied in adult and aged male mice. The submandibular salivary glands were removed (sialectomy) either uni- or bilaterally, and the SCG were studied by fluorescence microscopy and histochemically. Catecholamine fluorescence and tyrosine hydroxylase immunoreactivity decreased after sialectomy, suggesting reduced noradrenaline production. Neuronal density was lower in the aged controls than in the young controls. In bo

Effect of serotonin on brown adipose tissue and on its sympathetic neurons.

Previously we have shown that there are two types of sympathetic neurons in the rat interscapular brown fat-pad: long adrenergic neurons with cell bodies located in the paraspinal ganglia and short adrenergic neurons with cell bodies located within the pad itself. The present studies examine 1) the uptake of [3H]norepinephrine by these neurons, 2) the effect of serotonin on release of the catecholamine, and 3) the metabolic action of serotonin on brown fat. [3H]norepinephrine uptake from the medium in which brown fat slices are incubated is linear with time up to 90 min. This uptake is blocked

Structural and functional impairment of adrenergic input to intraocular cerebellar grafts during thyroid hormone deficiency.

Embryonic cerebellae were transplanted to the anterior eye chamber of normal and thyroidectomized adult rats and were left in the eye to mature for 5 weeks. The cerebellar grafts received adrenergic innervation from the sympathetic ground plexus of the host iris. The density of adrenergic fibers innervating the grafts was reduced by approximately 50% in all thyroidectomized groups, without any effect on the fluorescence intensity or the morphology of individual nerve fibers. The reduction in adrenergic ingrowth was entirely prevented in grafts raised in similarly thyroidectomized recipients, w

Insulin-responsive autonomic neurons in rat medulla oblongata.

Low blood glucose activates brainstem adrenergic and cholinergic neurons, driving adrenaline secretion from the adrenal medulla and glucagon release from the pancreas. Despite their roles in maintaining glucose homeostasis, the distributions of insulin-responsive adrenergic and cholinergic neurons in the medulla are unknown. We fasted rats overnight and gave them insulin (10 U/kg i.p.) or saline after 2 weeks of handling. Blood samples were collected before injection and before perfusion at 90 min. We immunoperoxidase-stained transverse sections of perfused medulla to show Fos plus either phen

Cholinergic suppression of heart rate acceleration during intrinsic optogenetic activation of sympathetic cardiac neurons in perfused hearts.

Cardiac autonomic pathways can be interrogated using optogenetics via selective expression of channelrhodopsin (ChR2) within intrinsic cardiac neurons. We tested the hypothesis that cholinergic activation via exogenous acetylcholine (ACh) would suppress the β-adrenergic heart rate (HR) acceleration induced by tyrosine hydroxylase (TH) neuron stimulation. HR responses were measured for wild-type (WT) mice at increasing norepinephrine (NE) concentrations and for transgenic mice that expressed ChR2 in TH neurons while illuminating the right atrium using a 465-nm microLED. Light pulse duty cycles

Uncoupling between noradrenergic and serotonergic neurons as a molecular basis of stable changes in behavior induced by repeated drugs of abuse.

A challenge in drug dependence is to delineate long-term behavioral and neurochemical modifications induced by drugs of abuse. In rodents, drugs of abuse induce locomotor hyperactivity, and repeating injections enhance this response. This effect, called behavioral sensitization, persists many months after the last administration, thus mimicking long-term sensitivity to drugs observed in human addicts. Although addictive properties of drugs of abuse are generally considered to be mediated by an increased release of dopamine in the ventral striatum, recent pharmacological and genetic experiments

Norepinephrine Protects against Amyloid-β Toxicity via TrkB.

The locus coeruleus (LC), the brainstem noradrenergic nucleus that is the sole source of norepinephrine (NE) in the forebrain, is one of the first structures affected in Alzheimer's disease (AD). Experimental ablation of the LC exacerbates, while increasing NE abates, AD-like neuropathology and cognitive deficits in animal models of the disease. Some neuroprotective effects of NE appear to be mediated by tropomyosin-related kinase B (TrkB), the canonical receptor for brain-derived neurotrophic factor (BDNF). Here, we report that NE dose-dependently protected primary cortical and LC neurons fro

Anterograde axonal transport of horseradish peroxidase in the rat postganglionic sympathetic nerves.

Seven minutes after initiating the experiment, horseradish peroxidase (HRP)-labeled pinocytotic vesicles were observed on the plasma membranes of the postganglionic sympathetic neurons in the left superior cervical ganglia (SCG). HRP reaction product was also demonstrated in the vesicles of the smooth endoplasmic reticulum (sER) and multivesicular bodies in the neuronal perikarya. The HRP-positive vesicles of sER and multivesicular bodies were also detected in the adrenergic sympathetic nerve endings in the pineal gland 7 minutes after initiation of the experiment. This suggests that the HRP i

Cancer-induced Nerve Injury Unveils a Sympathetic-to-Sensory Nerve Axis in Head and Neck Cancer.

Oral squamous cell carcinoma (OSCC) is one of the most painful cancers, with patients frequently reporting spontaneous, neuropathic-like pain. While sympathetic and sensory nerves have been individually implicated in cancer progression, whether and how these systems interact to drive pain and tumor growth has remained unclear. Here, we integrate prospective human data with reverse-translational mouse models to reveal that cancer-induced nerve injury unveils crosstalk between sympathetic postganglionic neurons and trigeminal sensory afferents in the tumor microenvironment. Adrenergic nerves regulate intestinal regeneration through IL-22 signaling from type 3 innate lymphoid cells.

The intestinal epithelium has high intrinsic turnover rate, and the precise renewal of the epithelium is dependent on the microenvironment. The intestine is innervated by a dense network of peripheral nerves that controls various aspects of intestinal physiology. However, the role of neurons in regulating epithelial cell regeneration remains largely unknown. Here, we investigated the effects of gut-innervating adrenergic nerves on epithelial cell repair following irradiation (IR)-induced injury. We observed that adrenergic nerve density in the small intestine increased post IR, while chemical

Clonidine in the central nervous system: site and mechanism of hypotensive action.

The data presented in this paper must be considered in conjunction with the fact that clonidine is both a partial alpha-adrenergic agonist and a partial alpha-adrenergic antagonist. In the anterior hypothalamus, clonidine, acting as an alpha-agonist, excites a pathway that inhibits excitatory cardiovascular neurons. Thus, the effect of neurons from the nucleus tractus solitarii (NTS) in inhibiting sympathetic outflow from the vasomotor center is effectively increased. In the posterior hypothalmus, clonidine, acting as an alpha-antagonist, decreases excitation of excitatory cardiovascular neuro

Infralimbic prefrontal cortical projections to the autonomic brainstem: quantification of inputs to cholinergic and adrenergic/noradrenergic nuclei.

The ventromedial prefrontal cortex regulates both emotional and physiological processes. The infralimbic cortex (IL), a prefrontal subregion in rodents, integrates behavioral, neuroendocrine, and autonomic responses to stress. However, the organization of cortical inputs to brainstem nuclei that regulate homeostatic responses are not well defined. We hypothesized that IL projections differentially target pre-ganglionic parasympathetic neurons and adrenergic/noradrenergic nuclei. To quantify IL projections to autonomic brainstem nuclei in male rats, we utilized viral-mediated gene transfer to e

Activation of α1-adrenoceptors facilitates excitatory inputs to medullary airway vagal preganglionic neurons.

In mammals, the neural control of airway smooth muscle is dominated by a subset of airway vagal preganglionic neurons in the ventrolateral medulla. These neurons are physiologically modulated by adrenergic/noradrenergic projections, and weakened α₂-adrenergic inhibition of them is indicated to participate in the pathogenesis and exacerbation of asthma. This study tests whether these neurons are modulated by α₁-adrenoceptors, and if so, how. In anesthetized adult rats, microinjection of the α₁A-adrenoceptor agonist A61603 (1 pmol) unilaterally into the medullary region containing these neurons

Hirschsprung's disease--immunohistochemical findings.

Hirschsprung's disease (HSCR) is characterized by a non-propulsive distal intestinal segment (usually colon) leading to a functional obstruction. An absence of ganglia in the affected segment explains the synonymous term "aganglionosis coli". The lack of peristalsis is partly due to a deficient intestinal smooth muscle relaxation based on an absence of non-adrenergic, non-cholinergic (NANC) inhibitory innervation. Morphological studies using conventional microscopy, immunohistochemistry and immunochemistry against general neuronal markers and neuropeptides have been used to characterize the di

Histochemistry of sympathetic neurons allotransplanted from young and aged mice to the submandibular gland.

Sympathetic ganglion tissue of young (3 months) and aged (24 months) NMRI mice was allotransplanted into the submandibular gland to study the influence of aging on the survival of grafted neurons. The submandibular gland (SMG) was chosen as a host tissue because of its high concentration of NGF and good blood supply. Four weeks postgrafting the viability of transplants was evaluated using the formaldehyde-induced fluorescence technique, tyrosine hydroxylase (TH) immunohistochemistry, and morphometry. The density of neurons, catecholamine fluorescence and TH immunoreactivity (TH-IR) appeared to
