Drug-Induced Fatty Liver Disease (DIFLD): A Comprehensive Analysis of Clinical, Biochemical, and Histopathological Data for Mechanisms Identification and Consistency with Current Adverse Outcome Pathways.

Drug induced fatty liver disease (DIFLD) is a form of drug-induced liver injury (DILI), which can also be included in the more general metabolic dysfunction-associated steatotic liver disease (MASLD), which specifically refers to the accumulation of fat in the liver unrelated to alcohol intake. A bi-directional relationship between DILI and MASLD is likely to exist: while certain drugs can cause MASLD by acting as pro-steatogenic factors, MASLD may make hepatocytes more vulnerable to drugs. Having a pre-existing MASLD significantly heightens the likelihood of experiencing DILI from certain med

Acute Oral, Subacute, and Developmental Toxicity Profiling of Naphthalene 2-Yl, 2-Chloro, 5-Nitrobenzoate: Assessment Based on Stress Response, Toxicity, and Adverse Outcome Pathways.

The U.S. National Research Council (NRC) introduced new approaches to report toxicity studies. The NRC vision is to explore the toxicity pathways leading to the adverse effects in intact organisms by the exposure of the chemicals. This study examines the toxicity profiling of the naphthalene-2-yl 2-chloro-5-dinitrobenzoate (SF5) by adopting the vision of NRC that moves from traditional animal studies to the cellular pathways. Acute, subacute, and developmental toxicity studies were assayed according to the Organization for Economic Cooperation and Development (OECD) guidelines. The stress resp

A multi‑target adverse outcome pathway network for sub-chronic ketamine‑induced cognitive impairment in adult zebrafish.

Ketamine (KET), an emerging contaminant, is increasingly detected in aquatic environments, yet its sub-chronic neurocognitive effects at environmentally relevant concentrations remain largely unknown. We exposed adult zebrafish to KET for 40 days and integrated network toxicology, transcriptomics, behavioral assays, and molecular simulations to investigate these effects. Sub-chronic KET exposure impaired spatial learning and memory at 0.4 μg/L, while anxiety‑like and social behavior were altered only at 40 μg/L. Intersecting transcriptomics with network toxicology yielded 9 candidate genes and

Validation of the Electrophilic Allergen Screening Assay for Detection of Key Event 1 of the Skin Sensitization Adverse Outcome Pathway.

The electrophilic allergen screening assay (EASA) uses small-molecule probes as surrogates for skin proteins to measure hapten protein carrier complex formation, Key Event (KE) 1 in the adverse outcome pathway for skin sensitization. Although multiple assays are accepted for this purpose, the EASA has higher throughput and needs less specialized equipment than other commonly used KE1 assays. This validation study assessed transferability of the EASA to naïve laboratories and within- and between-laboratory reproducibility. The predictive capacity of the assay in relation to reference data both

Quantitative adverse outcome pathway modeling of cisplatin-induced nephrotoxicity: developing in vitro and in vivo models for predictive extrapolation.

Nephrotoxicity is a major concern in the safety assessment of chemicals and drugs. Computational modeling, particularly the use of quantitative adverse outcome pathways (qAOPs), offers a promising strategy to improve the translation from in vitro to in vivo, thereby facilitating reliable predictions of in vivo adverse outcomes and potentially reducing the need for animal testing. Platinum-based drugs are widely used in chemotherapy, yet their clinical application is frequently constrained by nephrotoxic effects. Here, we focus on the development of ordinary differential equation (ODE)-based qA

An explorable model of an adverse outcome pathway of cytokine release syndrome related to the administration of immunomodulatory biotherapeutics and cellular therapies.

INTRODUCTION: Cytokine release syndrome (CRS) is a potentially severe systemic inflammatory condition triggered by various immunomodulatory therapies, making understanding its pathogenesis critical for improving patient outcomes. RESULTS/METHODS: By combining immunotoxicology and systems biology approaches, we offer a novel and integrative conceptual model of CRS as an adverse outcome (AO), induced by five different immunomodulatory biotherapies: 1) chimeric antigen receptor (CAR) T cells, 2) checkpoint inhibitors, 3) T cell engaging bispecific modalities, 4) monoclonal antibodies targeting a

Cyclosporin A toxicity on endothelial cells differentiated from induced pluripotent stem cells: Assembling an adverse outcome pathway.

Cyclosporin A (CSA) is a potent immunosuppressive agent in pharmacologic studies. However, there is evidence for side effects, specifically regarding vascular dysfunction. Its mode of action inducing endothelial cell toxicity is partially unclear, and a connection with an adverse outcome pathway (AOP) is not established yet. Therefore, we designed this study to get deeper insights into the mechanistic toxicology of CSA on angiogenesis. Stem cells, especially induced pluripotent stem cells (iPSCs) with the ability of differentiation to all organs of the body, are considered a promising in vitro

Adverse outcome pathway-based approach to reveal the mechanisms of skin sensitization and long-term aging effects of chlorothalonil.

Chlorothalonil (CHT) is a widely used antifungal agent and is reported to be a sensitizer that can cause allergic contact dermatitis (ACD). ACD initiation is associated with various innate immune cell contributions and is usually accompanied by persistent inflammation, which is a potential contributing factor to skin damage. However, detailed information on the mechanisms by which CHT induces skin sensitization and damage is still insufficient. This study focused on investigating the possible sensitization process and mechanism of CHT and the adverse effects of repeated CHT exposure. CHT activ

Unraveling Structural Alerts in Marketed Drugs for Improving Adverse Outcome Pathway Framework of Drug-Induced QT Prolongation.

In pharmaceutical treatment, many non-cardiac drugs carry the risk of prolonging the QT interval, which can lead to fatal cardiac complications such as torsades de points (TdP). Although the unexpected blockade of ion channels has been widely considered to be one of the main reasons for affecting the repolarization phase of the cardiac action potential and leading to QT interval prolongation, the lack of knowledge regarding chemical structures in drugs that may induce the prolongation of the QT interval remains a barrier to further understanding the underlying mechanism and developing an effec

Potential adverse outcome pathway (AOP) of emamectin benzoate mediated cardiovascular toxicity in zebrafish larvae (Danio rerio).

Emamectin benzoate (EMB) is an efficient insecticide which widely used as an anthelmintic drug additive in aquaculture fish. However, its extensive use has resulted in widespread pollution in the aquatic environment. Previous studies have identified the potential developmental and neurotoxic effects of EMB, however, systematic studies pertaining to the cardiovascular toxic effects of EMB on fish are scarce. In this study, zebrafish embryos were exposed to EMB at concentrations of 0, 0.1, 0.25, 0.5, 1, 2, 4, and 8 mg/L for 3 days, aiming to investigate the cardiovascular toxic effects of EMB vi

The connection between Bayesian networks and adverse outcome pathway (AOP) networks and how to use it for predicting drug toxicity.

There is significant interest in combining adverse outcome pathways (AOPs) with Bayesian networks (BNs) because of their shared representation using directed acyclic graphs (DAGs). However, it has not been verified empirically whether AOP networks are mathematically congruent with BNs. Furthermore, important properties for BNs, such as Markov blankets, have not been emphasized, which is a missed opportunity for simplifying and optimizing the model. Here, we summarize the connection between AOP networks and BNs and explore the implications for toxicity modeling. We also present a case study in

Using transcriptomics data and Adverse Outcome Pathway networks to explore endocrine disrupting properties of Cadmium and PCB-126.

Omics-technologies such as transcriptomics offer valuable insights into toxicity mechanisms. However, integrating this type of data into regulatory frameworks remains challenging due to uncertainties regarding toxicological relevance and links to adverse outcomes. Furthermore, current assessments of endocrine disruptors (EDs) relevant to human health require substantial amounts of data, and primarily rely on standardized animal studies. Identifying EDs is a high priority in the EU, but so are efforts to replace and reduce animal testing. Alternative methods to investigate EDs are needed, and s

Bioinformatics enrichment analysis of genes and pathways related to maternal type 1 diabetes associated with adverse fetal outcomes.

Maternal type 1 diabetes mellitus (T1DM) may affect fetal development by altering the gene expression profile of the umbilical cord. The present study aimed to explore the T1DM-induced gene expression changes in the fetal umbilical cord. The raw gene expression profiles (ID: GSE51546) of umbilical cord tissue obtained from six normal mothers (non-diabetic) and six type 1 diabetic mothers were used to identify the differentially expressed genes. Genes that correspond to official gene symbols were selected for protein-protein interaction (PPI) and sub-network construction (combined score > 0.4).

Alternative Toxicity Testing: Analyses on Skin Sensitization, ToxCast Phases I and II, and Carcinogenicity Provide Indications on How to Model Mechanisms Linked to Adverse Outcome Pathways.

This article studies alternative toxicological approaches, with new (skin sensitization, ToxCast) and previous (carcinogenicity) analyses. Quantitative modeling of rate-limiting steps in skin sensitization and carcinogenicity predicts the majority of toxicants. Similarly, successful (Quantitative) Structure-Activity Relationships models exploit the quantification of only one, or few rate-limiting steps. High-throughput assays within ToxCast point to promising associations with endocrine disruption, whereas markers for pathways intermediate events have limited correlation with most endpoints. S

Mapping DEHP-induced metabolic dysregulation through multi-omics and systems biology: an adverse outcome pathway network approach.

Di(2-ethylhexyl) phthalate (DEHP) is a widespread endocrine-disrupting plasticizer associated with metabolic disorders. To elucidate their mechanistic basis, we developed an Adverse Outcome Pathway Network (AOPN) informed by DEHP-induced perturbations, by integrating multi-omics data, systems biology modeling, and targeted literature evidence. Multi-omics data were generated in 3D human hepatic cells, pancreatic β-cells, and zebrafish larvae exposed to DEHP across biologically relevant concentration ranges. Transcriptomic and metabolomic analyses revealed perturbations in lipid metabolism, mit

Exposure to responsible components of PM2.5 induces hepatic steatosis: An in silico, in vitro, and in vivo study based on adverse outcome pathway.

Mounting evidence suggests that exposure to PM2.5 is closely associated with hepatic steatosis. However, due to the complex composition of PM2.5, evidence regarding the effects of its specific chemical components on hepatic steatosis remains insufficient. This study aims to expand our previous research by investigating whether direct exposure to PM2.5-bound responsible components induces hepatic steatosis. Additionally, we seek to elucidate the molecular mechanisms based on an adverse outcome pathway (AOP) strategy. Initially, molecular dynamics simulations and dual-luciferase assays identifie

Inhibition of Fin Regeneration in Fathead Minnow (Pimephales promelas) by a Potent Synthetic Glucocorticoid and Development of Adverse Outcome Pathway 334.

Despite structural and functional conservation across vertebrate species, the glucocorticoid receptor has been minimally studied in comparison to other biological targets for endocrine-disrupting compounds in aquatic systems. Because prolonged use of pharmaceutical glucocorticoids in humans has been linked to osteoporosis and impaired bone growth, we hypothesized that the ability of teleost fish to regenerate fins following damage may be inhibited by exposure to synthetic glucocorticoids in the environment. In the present study, we examined fin regeneration following a 7 days waterborne exposu
