The pathogenesis of African swine fever virus (ASFV) infection in Ornithodoros porcinus porcinus was examined in nymphal ticks infected with the ASFV isolate Chiredzi/83/1. At times postinfection (p.i.) ranging from 6 h to 290 days, ticks or dissected tick tissues were titrated for virus and examined ultrastructurally for evidence of virus replication. The ASFV infection rate in ticks was 100% in these experiments, and virus infection was not associated with a significant increase in tick mortality. Initial ASFV replication occurred in phagocytic digestive cells of the midgut epithelium. Subse

Thirteen sows between 38 and 92 days of pregnancy were exposed oronasally to African swine fever virus (1979 Dominican Republic isolate) to determine whether infection would affect their reproductive performance. Abortions occurred in sows in all stages of pregnancy. Of 11 sows, 7 aborted 5 to 8 (6.1 +/- 0.90) days after inoculation or 0 to 3 (1.4 +/- 0.98) days after fever development. Abortions occurred soon after clinical signs of infection first developed and viremia titers peaked. Swelling of the vulva and treading preceded abortion in some sows. Ecchymotic placental and petechial cutaneo

African swine fever virus (ASFV) relies on a virus-encoded transcription machinery to initiate gene expression within cytoplasmic viral factories. The viral early transcription factor (VETF), a heterodimer composed of pD1133L and pG1340L, is required for early transcription and viral replication, yet its structural organization and regulation remain poorly understood. Here, we present cryo-EM structures of ASFV VETF in five distinct conformations. The complex adopts an unusual chair-like architecture stabilized by an extensive buried interfacial area. The small subunit, pD1133L, contains a con

Since African swine fever virus (ASFV) genotype II reached Europe in 2007 and has widely spread, causing important economic losses to the pig production sector. To guide policy and management actions, robust quantitative evidence about possible explanatory variables associated with ASF in domestic pigs and Eurasian wild boar (Sus scrofa) is needed. To this aim, a systematic literature review of the scientific evidence available on variables analysed through quantitative methods investigating their possible association with ASF occurrence was carried out in 2021 and updated in 2024. The swine industry across the globe is recently facing a devastating situation imparted by a highly contagious and deadly viral disease, African swine fever. The disease is caused by a DNA virus, the African swine fever virus (ASFV) of the genus Asfivirus. ASFV affects both wild boars and domestic pigs resulting in an acute form of hemorrhagic fever. Since the first report in 1921, the disease remains endemic in some of the African countries. However, the recent occurrence of ASF outbreaks in Asia led to a fresh and formidable challenge to the global swine production industry. Culling of the i

African swine fever virus (ASFV), a highly virulent double-stranded DNA virus, poses a significant threat to global pig farming, with mortality rates in domestic pigs reaching up to 100%. Originating in Kenya in 1921, ASFV has since proliferated to Western Europe, Latin America, Eastern Europe, and most recently China in 2018, resulting in substantial global agricultural losses. Antigenic epitopes, recognized by the immune system's T cells and B cells, are pivotal in antiviral immune responses. The identification and characterization of these antigenic epitopes can offer invaluable insights in

A common rule in many countries for mitigating the damage caused by African swine fever (ASF) is to eradicate the virus at the outbreak in order to prevent its dispersal and the associated social costs of depopulating infected domestic pigs. The economic performance of this practice, as measured by five different evaluation criteria (net present value, benefit-cost ratio, rate of return, internal rate of return, and payback time), depends on the type of control cost and the spatial and dynamic allocation of benefits, i.e. avoided losses from infected domestic pig farms. The present paper calcu

Due to the current unavailability of vaccines or treatments for African swine fever (ASF), which is caused by African swine fever virus (ASFV), rapid and reliable detection of the virus is essential for timely implementation of emergency control measures and differentiation of ASF from other swine diseases with similar clinical presentations. Here, an improved PCR assay was developed and evaluated for sensitive and universal detection of ASFV. Primers specific for ASFV were designed based on the highly conserved region of the vp72 gene sequences of all ASFV strains available in GenBank, and th

Toxoplasma gondii is widespread worldwide and can infect swine. This study evaluated the seroprevalence of T. gondii in swine from central China after an outbreak of African swine fever (ASF). A total of 2683 swine serum samples were collected from farms in four provinces. Of the serum samples, 1.42% (38/2683) (95% CI, 1.03-1.94) tested positive for T. gondii IgG antibody by a modified agglutination test (MAT) (cut-off: 1:25). Comparing with the results of previous studies, specifically our survey from before the outbreak, the seroprevalence of T. gondii in swine from central China was signifi

UNLABELLED: African swine fever virus (ASFV) poses a severe threat to the global pig industry, yet the precise mechanisms of its entry into host cells are not fully understood. In this study, we identified the host ribosomal protein SA (RPSA), a protein located in lipid rafts, as a critical viral receptor that facilitates ASFV entry. Immunoprecipitation-mass spectrometry (IP-MS) screening results identified RPSA as a binding partner of the ASFV capsid protein pE120R, which was further confirmed by co-immunoprecipitation (Co-IP) and immunofluorescence assays (IFA) in ASFV-infected cells. This review explores the biological basis and epidemiological implications of natural resistance to ASFV. It dispels the misconception that African wild suids are persistent virus reservoirs and clarifies the role of Ornithodoros ticks in virus maintenance and transmission. Resistance mechanisms appear multifactorial, potentially involving host genetics and immune responses. Genomic studies have begun identifying genes potentially linked to ASFV resistance, including in pigs from areas with more recent ASF incursions. Understanding and harnessing natural resistance is a promising avenue to dev

African swine fever (ASF) is a highly contagious and lethal viral disease affecting domestic pigs and wild boars, with profound implications for global swine production and food security. Caused by the ASF virus (ASFV), a complex double-stranded DNA virus of the Asfarviridae family, the disease exhibits diverse clinical outcomes - from peracute death to chronic infection - depending on viral genotype and host immunity. ASFV primarily targets monocytes and macrophages, leading to severe lymphoid depletion, systemic inflammation, and vascular pathology mediated by cytokine storms. The virus demo

African swine fever virus (ASFV) devastates swine herds and lacks widely available antivirals. We show that the ASFV DNA polymerase beta (Pol β; gene G1211R) localizes to viral replication factories and is required for viral replication. Cryo-electron microscopy structures of Pol β in apo and double-stranded DNA (dsDNA)-bound states reveal pronounced conformational flexibility that enables transitions between functional states. Binding to dsDNA promotes higher-order oligomerization that is essential for catalytic activity, as demonstrated by structure-guided mutagenesis and biochemical assays.

African swine fever virus (ASFV), the aetiological agent of a devastating swine disease, has developed several strategies to replicate in porcine macrophages, its main target cells. In this work, we investigated the expression of 84 antiviral genes in macrophages infected with the virulent strain 26544/OG10 or the attenuated strain NH/P68. Infection with both strains caused an early activation of antiviral defenses, with up-regulation of RNA-sensing molecules and interferon-stimulating genes. However, as viral replication progresses, down-regulation of key inflammatory genes was observed, espe

African swine fever virus (ASFV), a highly lethal pathogen, poses a catastrophic threat to global pork production. While some commercial vaccines exist, their availability remains regional limitations. This review investigates the mechanistic interplay between ASFV and antigen-presenting cells (APCs), with a particular focus on viral evasion strategies that disrupt antigen presentation pathways. Although current understanding of ASFV immunology has largely focused on monocyte/macrophage infection, we provide a systematic analysis of ASFV-mediated immunosuppression across various APCs, includin

African Swine Fever (ASF) is a highly contagious disease affecting both domestic pigs and wild boars. In domestic pigs, ASF is a rapidly-progressing disease with a mortality rate reaching 100 %, causing tremendous economic loss in affected areas. ASFV is caused by African Swine Fever Virus (ASFV), which is a large, enveloped double-stranded DNA virus belonging to the Asfarviridae family. ASFV has a remarkably large genome size that encodes more than 150 open reading frames. Among the virally encoded enzymes, thymidine kinase (ASFV-TK) has been shown to be critical for the efficient replication

The African swine fever virus (ASFV) is the cause of a recent pandemic that is threatening the global pig industry. The virus infects domestic and wild pigs and manifests with a variety of clinical symptoms, depending on the strain. No commercial vaccine is currently available to protect animals from this virus, but some attenuated and recombinant live vaccine candidates might be effective against the disease. This article describes the immunobiological characteristics of one such candidate-the laboratory-attenuated ASFV strain, Katanga-350-which belongs to genotype I. In this study, we assess

The rapid spread of the African swine fever virus (ASFV), causing severe disease with often high fatality rates in Eurasian suids, prevails as a threat for pig populations and dependent industries worldwide. Although advancing scientific progress continually enhances our understanding of ASFV pathogenesis, alternative transmission routes for ASFV have yet to be assessed. Here, we demonstrate that ASFV can efficiently be transferred from infected boars to naïve recipient gilts through artificial insemination (AI). In modern pig production, semen from boar studs often supplies many sow herds. Th

African swine fever virus (ASF) has circulated in Vietnam since 2018, causing significant losses to the pig industry. Quick, accurate diagnosis of African swine fever virus (ASFV) infection is crucial for controlling the disease. The detection of the virus in piglets with congenital tremors is described in this paper. ASFV was detected in brain tissues by polymerase chain reaction (PCR) and immunohistochemistry. Classical swine fever virus, porcine parvovirus, porcine reproductive and respiratory syndrome virus, and pseudorabies virus were not detected by PCR, suggesting that the ASFV was the

African swine fever (ASF) has spread across the globe and has reached closer to North America since being reported in the Dominican Republic and Haiti. As a result, surveillance measures have been heightened and the utility of alternative samples for herd-level monitoring and dead pig sampling have been investigated. Passive surveillance based on the investigation of dead pigs, both domestic and wild, plays a pivotal role in the early detection of an ASF incursion. The World Organization for Animal Health (OIE)-recommended samples for dead pigs are spleen, lymph nodes, bone marrow, lung, tonsi
