Characterization of PFOS toxicity on in-vivo and ex-vivo mouse pancreatic islets.

Considerable human data have shown that the exposure to perfluorooctane sulfonate (PFOS) correlates to the risk of metabolic diseases, however the underlying effects are not clearly elucidated. In this study, we investigated the impacts of PFOS treatment, using in-vivo, ex-vivo and in-vitro approaches, on pancreatic β-cell functions. Mice were oral-gavage with 1 and 5 μg PFOS/g body weight/day for 21 days. The animals showed a significant increase in liver triglycerides, accompanied by a reduction of triglycerides in blood sera and glycogen in livers and muscles. Histological examination of pan

Assessing the human health risks of per- and polyfluoroalkyl substances: A need for greater focus on their interactions as mixtures.

Humans are exposed to complex mixtures of per- and polyfluoroalkyl substances (PFAS). However, human health risk assessment of PFAS currently relies on animal toxicity data derived from individual substance exposure, which may not adequately predict the risk from combined exposure due to possible interactions that can influence the overall risk. Long-chain perfluoroalkyl acids (PFAAs), particularly perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) are recognised as global emerging contaminants of concern due to their ubiquitous distribution in all environmental media, wildlife

Are perfluorooctane sulfonate alternatives safer? New insights from a birth cohort study.

BACKGROUND: Experimental studies show that chlorinated polyfluorinated ether sulfonic acids (Cl-PFESA 6:2 and 8:2), one of perfluoroalkyl substances (PFAS) used as perfluorooctane sulfonate (PFOS) alternatives, are reproductive toxicants in vivo and in vitro. However, the associations between gestational exposure to Cl-PFESAs and birth outcomes are unknown. OBJECTIVES: We investigated associations between 6:2 Cl-PFESA and 8:2 Cl-PFESA in maternal serum and birth outcomes. METHODS: We measured four PFAS, including 6:2 Cl-PFESA, 8:2 Cl-PFESA, PFOS, and perfluorooctanoic acid (PFOA) in third-tr

Occurrence and incidence of 18 per- and polyfluoroalkyl compounds in edible oils commonly consumed in Guiyang, China.

A systematic examination was conducted for the first time of a wide carbon-chain range of per- and polyfluoroalkyl compounds (PFASs) in 83 samples of edible vegetable and animal oils from Guiyang, China. The occurrence and levels of 18 PFASs in seven types of edible oil were analysed. Analytes were determined based on a simplified, sensitive and reliable solvent extraction and liquid chromatography-tandem mass spectrometry method. Ten target PFASs were found and the most frequently detected compounds with maximum concentration were PFOS (88%, 1.93 ng g-1), PFNA (55.4%; 6.76 ng g-1), PFHxS (39.

Toxicological effects of acute and subacute perfluorooctane sulfonic acid exposure on the bivalve Ruditapes decussatus.

Perfluorooctane sulfonic acid (PFOS), a persistent and bioaccumulative per-and polyfluoroalkyl substance (PFAS), is widely distributed in aquatic environments and is known for its toxic effects. This study investigated the toxicological mechanisms of PFOS in the bivalve Ruditapes decussatus under acute and subacute exposure scenarios, with particular emphasis on concentration-dependent and organ-specific responses in gills and digestive glands (DGs). Clams were exposed to nominal PFOS concentrations under acute conditions (1-100 µg/ml for 5 days) and subacute conditions (1, 10, and 20 µg/ml fo

Impact of PFOS Exposure on Murine Fetal Hematopoietic Stem Cells, Associated with Intrauterine Metabolic Perturbation.

This study hypothesized that perfluorooctanesulfonate (PFOS) exposure disrupts maternal-fetal metabolism, affecting fetal liver hematopoietic stem cell (FL-HSC) development. Pregnant mice received PFOS (0.3 and 3 μg/g bw) and were sacrificed on gestation day 14.5. Metabolomic analysis of maternal plasma revealed disruptions in steroid hormone, purine, carbohydrate, and amino acid metabolism, which aligned with the enriched pathways in amniotic fluid (AF). FL analysis indicated increased purine metabolism and disrupted glucose and amino acid metabolism. FL exhibited higher levels of polyunsatur

Impact of Variable Water Chemistry on PFOS-Goethite Interactions: Experimental Evidence and Surface Complexation Modeling.

Perfluorooctanesulfonate (PFOS) has become a major concern due to its widespread occurrence in the environment and severe toxic effects. In this study, we investigate PFOS sorption on goethite surfaces under different water chemistry conditions to understand the impact of variable groundwater chemistry. Our investigation is based on multiple lines of evidence, including (i) a series of sorption experiments with varying pH, ionic strength, and PFOS initial concentration, (ii) IR spectroscopy analysis, and (iii) surface complexation modeling. PFOS was found to bind to goethite through a strong h

Validation of the Brazilian version of the Short Inventory of Grazing (SIG).

OBJECTIVE: Grazing is a disturbed eating pattern that has been associated with eating disorders and obesity. One of the new measures to investigate this eating behavior is the Short Inventory of Grazing (SIG), a two-item questionnaire that assesses grazing in general and grazing associated with the feeling of loss of control over eating (LOC grazing). However, the psychometric properties of the SIG have not been assessed in the Brazilian population. The present study aimed to cross-culturally adapt and validate a Brazilian version of the SIG. METHODS: The SIG was adapted to the Brazilian cont

Efficient workflow for suspect screening analysis to characterize novel and legacy per- and polyfluoroalkyl substances (PFAS) in biosolids.

Land application of treated sewage sludge (also known as biosolids) is considered a sustainable route of disposal because it reduces waste loading into landfills while improving soil health. However, this waste management practice can introduce contaminants from biosolids, such as per- and polyfluoroalkyl substances (PFAS), into the environment. PFAS have been observed to be taken up by plants, accumulate in humans and animals, and have been linked to various negative health effects. There is limited information on the nature and amounts of PFAS introduced from biosolids that have undergone di

Humin and biochar accelerated microbial reductive dechlorination of 2,4,6-trichlorophenol under weak electrical stimulation.

The extracellular electron transfer (EET) is regarded as one of the crucial factors that limit the application of the bioelectrochemical system (BES). In this study, two different solid-phase redox mediators (RMs), biochar (1.2 g/L, T-B) and humin (1.2 g/L, T-H) were used for boosting the microorganisms accessing the electrons required for 2,4,6-TCP dechlorination under weak electrical stimulation (-0.278 V vs. Standard hydrogen electrode). BES with dissolved RM anthraquinone-2,6-disulfonate (AQDS 0.5 mmol/L, T-A) was used as a comparison. The results showed that dechlorination of 2,4,6-TCP co

Effects of exposure to per- and polyfluoroalkyl substances on vaccine antibodies: A systematic review and meta-analysis based on epidemiological studies.

Vaccines are essential for children to defend against infection. Per- and polyfluoroalkyl substances (PFAS) are emerging contaminants with the characteristics of persistence and bioaccumulation. PFAS exposure can affect the function of the nervous, endocrine, and immune system of animals and humans. We aimed to conduct a systematic review and meta-analysis of the epidemiological studies investigating potential relationships between PFAS exposure and vaccine antibody levels, and assessed whether PFAS would affect vaccine response in healthy children. A literature search was conducted in PubMed,

Occurrence and fate of legacy and novel per- and polyfluoroalkyl substances (PFASs) in freshwater after an industrial fire of unknown chemical stockpiles.

An industrial warehouse illegally storing a large quantity of unknown chemical and industrial waste ignited in an urban area in Melbourne, Australia. The multiday fire required firefighters to use large amounts of fluorine-free foam that carried contaminated firewater runoff into an adjacent freshwater creek. In this study, the occurrence and fate of 42 per- and polyfluoroalkyl substances (PFASs) was determined from triplicate surface water samples (n = 45) from five locations (upstream, point-source, downstream; 8 km) over three sampling campaigns from 2018 to 2020. Out of the 42 target PFASs

Perfluorooctane sulfonate effects on the reproductive axis in adult male rats.

Perfluorooctane sulfonate (PFOS) is a neurotoxic agent and it can disrupt the endocrine system activity. This work was undertaken to evaluate the possible effects of PFOS exposure on the hypothalamic-pituitary-testicular axis (HPT) in adult male rats, and to evaluate the possible morphological alterations induced by PFOS in the endocrine tissues of this axis. Adult male rats were orally treated with 0.5; 1.0; 3.0 and 6.0 mg of PFOS/kg/day for 28 days. After PFOS exposure, hypothalamic noradrenaline concentration increased in the anterior hypothalamus and in the median eminence, not changing in

[Determination of seven perfluoroalkyl and polyfluoroalkyl substances in serum of pregnant women and evaluation of neonatal neurobehavior based on high performance liquid chromatography-tandem mass spectrometry].

Perfluoroalkyl and polyfluoroalkyl substances (PFASs) have been extensively used as synthetic fluorine-containing compounds in various consumer products, including surfactants, cookware, lubricants, clothing, and food packaging, since the 1950s. Evidence has shown that PFASs cross the placental barrier and interfere with fetal thyroid hormone homeostasis, which is crucial for fetal growth and neurobehavioral development in children aged 2-9 years. However, no epidemiological data on the association between prenatal PFAS exposure and neonatal neurobehavioral development are available. In this s

An overview of the impact of PFAS on animals, humans, and the environment using a One Health approach.

Per- and polyfluoroalkyl substances (PFAS) are a group of synthetic chemicals characterized by a fluorinated carbon chain that confers unique physicochemical properties. Widely used in industrial and consumer products, including textiles, food packaging, and firefighting foams, PFAS are highly persistent in the environment, earning them the designation of "forever chemicals." Their stability contributes to their widespread diffusion across different environmental compartments (water, soil, air) and multiple exposure pathways (e.g., diet). These lead to PFAS bioaccumulation and biomagnification

Predictors of Serum Per- and Polyfluoroalkyl Substances (PFAS) Concentrations in a Rural Agricultural Community Affected by Biosolids Application.

Human exposure pathways to PFAS from contaminated biosolids applied as fertilizers are not well understood. In the Maine Biosolids Study, we examined predictors of serum PFAS concentrations in adults (N = 144) living near sites of historical agricultural biosolids application. All participants had PFAS concentrations measured in their private drinking well water by the Maine Department of Environmental Protection from 2020 to 2023. In late 2023, we measured serum PFAS concentrations and administered a survey on water-related, occupational/recreational, residential, local food intake, and socio

Performance of a Sequencing Biofilter Coupled with a Dual-Media Granular Activated Carbon Filter for PFAS Mitigation in Landfill Leachate.

The performance of a sequencing batch biofilter granular reactor (SBBGR), followed by a dual media granular activated carbon (GAC) column, was evaluated in terms of its ability to remove selected per- and polyfluoroalkyl substances (PFAS) from landfill leachate. The results show that the SBBGR achieved an overall reduction of 51%, with the preferential removal of long-chain PFAS, while short-chain PFAS were only partially removed. Subsequent GAC treatment exhibited compound-specific breakthrough behavior, which was governed by chain length. Short-chain PFAS (e.g., perfluorobutanoic acid) exhib

Integrated thermal and phytoremediation of agricultural soils impacted by PFAS.

Per- and polyfluoroalkyl substances (PFAS) are persistent synthetic compounds that have contaminated millions of hectares of agricultural land through decades of biosolids application. Conventional remediation approaches, such as thermal destruction or excavation, are prohibitively expensive, carbon intensive, and leave affected farmland unfit for agriculture. Here, we present a potential scalable remediation strategy that combines phytoremediation, biochar production, and enhanced weathering to simultaneously remove PFAS from soil, immobilize residual contamination, and achieve durable carbon
