Atmospheric transport and wet deposition have contributed to the worldwide distribution of per- and polyfluoroalkyl substances (PFAS) in terrestrial and aquatic ecosystems, even in remote areas far from known industrial sources. However, little is known regarding the impact of cloud and precipitation formation dynamics on PFAS transport and wet deposition, nor the range of variability in PFAS concentrations within a closely distributed monitoring network. Precipitation samples were collected from a network of 25 stations in a focused geographic region (the Commonwealth of Massachusetts, USA) f

Perfluorooctane sulfonic acid (PFOS) is member of a class of molecules with fluorinated carbon chains known as polyfluoroalkyls. PFOS have been used to produce a variety of industry and comsumer uses. However, a significant concern is that it accumulates in the environment, including in animals and humans, and that it is a potential immunosuppressant. Here we analyze immune homeostasis in mice following chronic exposure to PFOS at levels up to those historically found in PFOS manufacturing workers. Mice were exposed to 0.15, 1.5, 15, or 50 µg /kg of PFOS for 28 days, after which, B cells, T ce

Human exposures to perfluoroalkyl and polyfluoroalkyl substances (PFAS) have been linked to several diseases associated with adverse health outcomes. Animal studies have been conducted, though these may not be sufficient due to the inherent differences in metabolic processes between humans and rodents. Acquiring relevant data on the health effects of short-chain PFAS can be achieved through methods supported by in vitro human cell-based models. Specifically, cytotoxicity assays are the crucial first step to providing meaningful information used for determining safety and providing baseline inf

Considerable human data have shown that the exposure to perfluorooctane sulfonate (PFOS) correlates to the risk of metabolic diseases, however the underlying effects are not clearly elucidated. In this study, we investigated the impacts of PFOS treatment, using in-vivo, ex-vivo and in-vitro approaches, on pancreatic β-cell functions. Mice were oral-gavage with 1 and 5 μg PFOS/g body weight/day for 21 days. The animals showed a significant increase in liver triglycerides, accompanied by a reduction of triglycerides in blood sera and glycogen in livers and muscles. Histological examination of pan

Humans are exposed to complex mixtures of per- and polyfluoroalkyl substances (PFAS). However, human health risk assessment of PFAS currently relies on animal toxicity data derived from individual substance exposure, which may not adequately predict the risk from combined exposure due to possible interactions that can influence the overall risk. Long-chain perfluoroalkyl acids (PFAAs), particularly perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) are recognised as global emerging contaminants of concern due to their ubiquitous distribution in all environmental media, wildlife

A systematic examination was conducted for the first time of a wide carbon-chain range of per- and polyfluoroalkyl compounds (PFASs) in 83 samples of edible vegetable and animal oils from Guiyang, China. The occurrence and levels of 18 PFASs in seven types of edible oil were analysed. Analytes were determined based on a simplified, sensitive and reliable solvent extraction and liquid chromatography-tandem mass spectrometry method. Ten target PFASs were found and the most frequently detected compounds with maximum concentration were PFOS (88%, 1.93 ng g-1), PFNA (55.4%; 6.76 ng g-1), PFHxS (39.

Per- and polyfluoroalkyl substances (PFAS) are a large group of synthetic surfactants of over 12,000 compounds that are incorporated into numerous products for their chemical and physical properties. Studies have associated PFAS with adverse health effects. Although there is a high potential for dermal exposure, these studies are lacking. The present study evaluated the systemic and immunotoxicity of subchronic 28- or 10-days of dermal exposure, respectively, to PFHpS (0.3125-2.5% or 7.82-62.5 mg/kg/dose) or PFOS (0.5% or 12.5 mg/kg/dose) in a murine model. Elevated levels of PFHpS were detect

BACKGROUND: Previous studies have investigated the associations between perfluoroalkyl acids (PFAAs) in women and time to pregnancy (TTP). Inconsistent results may be explained by differences in conditioning on parity.
OBJECTIVES: We used causal directed acyclic graphs to illustrate potential confounding related to previous pregnancies and exposure measurement error due to differences in the interpregnancy interval in pregnancy-based studies that include parous women. We exemplified the potential importance of these issues using data from the Danish National Birth Cohort.
METHODS: We used di

Direct discharge of untreated sewage to surface waters is a common practice in many parts of the world. However, relatively little is known about the behaviour of synthetic organic pollutants under these conditions. This paper describes a sampling campaign designed to track changes in water quality in a surface water system in Vientiane (Lao PDR) receiving significant quantities of untreated waste water. The study was based on following in-channel transport using a fluorescent tracer injected as a pulse, with a focus on the anionic surfactant linear alkylbenzene sulphonate (LAS) and ammonia. W

Prenatal exposure to per- and polyfluoroalkyl substances (PFAS) may disrupt maternal fatty acid (FA) metabolism; however, compartment-specific responses, nonmonotonic dose-response relationships (NMDRs), and mixture effects remain insufficiently characterized. We quantified 14 plasma PFAS and comprehensively profiled FAs, FA-class distributions, health-related indices, and product-to-precursor ratios in plasma (n = 192) and red blood cells (RBCs; n = 119) from pregnant women in China. Associations were assessed using multiple linear regression (MLR), restricted cubic splines (RCS), and Bayesia

Multiple pieces of evidence have shown that prenatal exposure to perfluoroalkyl and polyfluoroalkyl substances (PFASs) is closely related to adverse birth outcomes for infants. However, difficult access to human samples limits our understanding of PFASs transport and metabolism across the human placental barrier, as well as the accurate assessment of fetal PFASs exposure. Herein, we assess fetal exposure to 28 PFASs based on paired serum, placenta, and meconium samples. Overall, 21 PFASs were identified first to be exposed to the fetus prenatally and to be metabolized and excreted by the fetus

BACKGROUND: Long-chained poly- and perfluoroalkyl substances (PFAS) have been used in pesticide formulations but their potential impact on human PFAS exposure has not been addressed.
OBJECTIVES: To investigate if occupationally pesticide exposed female greenhouse workers in Denmark had higher serum concentrations of PFAS than a comparable background population.
METHODS: Serum samples collected between 1996 and 2001 from 181 pregnant greenhouse workers and a contemporary urban population of 48 pregnant women were analyzed for eight PFAS: perfluorooctane sulfonate (PFOS), perfluorohexane sulfo

Perfluorobutanesulfonate (PFBS), an alternative to perfluorooctanesulfonate (PFOS), has raised many health concerns. However, PFBS toxicity in the mammalian gut remains unclear. C57BL/6 mice were exposed to 10 μg/L and 500 μg/L PFBS or 500 μg/L PFOS in their water supply for 28 days. PFBS toxicity in the ileum and colon was explored and compared to that of PFOS. Biochemical analysis showed that tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) levels increased in the ileum exposed to 10 μg/L PFBS, whereas no significant changes were observed in those levels in the colon. Catalase (CAT

The global incidence and mortality rates of liver cancer are rising, necessitating research into environmental and lifestyle risk factors. Early onset liver cancer, diagnosed before the age of 50, is becoming prevalent, suggesting the potential influence of emerging environmental exposures. Per- and polyfluoroalkyl substances (PFAS) are ubiquitous environmental contaminants that have been linked to liver toxicity in humans and shown to cause hepatotoxic effects in animals, but their role in liver cancer remains unclear. This systematic review synthesizes evidence from animal and epidemiologica

Perfluorooctanesulfonic acid (PFOS) poses a potential threat to human health owing to its exceptional environmental persistence and bioaccumulation. Biochar exhibits considerable potential for PFOS removal, and metal/heteroatom doping or co-doping has been shown to effectively enhance its efficiency. In this study, Fe-N co-doped biochar was fabricated from agricultural rice straw waste, using potassium ferrate and melamine as precursors for Fe and N, respectively. The PFOS removal performance of the as-prepared biochar in solution was systematically evaluated by investigating the effects of in

Historical production of per- and polyfluoroalkyl substances (PFAS) via electrochemical fluorination has resulted in complex mixtures of linear (L) and branched (Br) isomers, yet most environmental studies still treat them as single compounds. Emerging research highlights that isomer-specific differences critically shape PFAS environmental fate, bioaccumulation, and toxicity. These distinctions are particularly critical for aquatic organisms, which experience continuous exposure to PFAS and serve as sentinels of ecosystem health. A comprehensive review of literature from January 2000 to Decemb

Assessing the transport and fate of the sulfonate-containing per- and polyfluoroalkyl substances (PFAS) in groundwater requires a molecular-scale understanding of their interactions with minerals, particularly environmentally abundant iron oxides such as hematite. Here, we systematically investigated the adsorption of perfluorooctane sulfonate (PFOS) and its chlorinated ether alternative, 6:2 chlorinated polyfluoroalkyl ether sulfonate (6:2 Cl-PFESA) on hematite surfaces using batch experiments, in situ spectroscopy, surface complexation modeling, and density functional theory (DFT) calculatio

BACKGROUND: Although per- and polyfluoroalkyl substances (PFAS) have been linked to chronic liver diseases, the specific cellular and molecular mechanisms by which different PFAS contribute to human liver dysfunction remain unclear. This study aims to elucidate those mechanisms.
METHODS: We exposed a multi-donor human liver spheroid model composed of multiple cell types to 20 µM of PFHxS, PFOA, PFOS, or PFNA for seven days, followed by single-cell RNA sequencing and lipid staining.
RESULTS: PFAS impacted liver spheroids in a compound- and sex-specific manner. PFOA and PFHxS increased lipid a

PFOS perturbs normal energy metabolism such as lipid metabolism; however, despite being a highly metabolic organ, skeletal muscle toxicity has not been widely studied. Skeletal muscle, a healthy body's largest metabolic organ, presents a target for PFOS-induced metabolic toxicity due to its high mitochondrial content. Therefore, we aimed to evaluate the response of skeletal muscle to PFOS in vivo and compare the responses to liver. Mice were treated with 0, 1, or 10 mg/kg/day PFOS for 21-days and gastrocnemius and liver collected. All mice exhibited a dose-dependent decrease in bodyweight foll

Information about perfluoroalkyl substances (PFASs) like perfluoroalkyl carboxylic acids (PFCAs) and sulfonates (PFSs) in settled dust from emerging and developing countries is still limited, partly due to the lack of efficient analytical methods. In this study, a reliable, simple, and cost-effective analytical procedure was developed and validated to determine 12 PFCAs and 4 PFSs in dust samples. The samples were ultrasonicated with methanol, followed by a dispersive sorbent clean-up step with graphitized carbon and quantification by liquid chromatography with tandem mass spectrometry (LC-MS/
