Enhanced harm detection following maternal separation: Transgenerational transmission and reversibility by inhaled amiloride.

Early-life adversities are associated with altered defensive responses. Here, we demonstrate that the repeated cross-fostering (RCF) paradigm of early maternal separation is associated with enhancements of distinct homeostatic reactions: hyperventilation in response to hypercapnia and nociceptive sensitivity, among the first generation of RCF-exposed animals, as well as among two successive generations of their normally reared offspring, through matrilineal transmission. Parallel enhancements of acid-sensing ion channel 1 (ASIC1), ASIC2, and ASIC3 messenger RNA transcripts were detected transg

Effect of amiloride on experimental acid-induced heartburn in non-erosive reflux disease.

BACKGROUND: Acid-sensing ion channels (ASICs) are esophageal nociceptors that are candidates to mediate heartburn in non-erosive reflux disease (NERD). Amiloride, a diuretic, is known to inhibit ASICs. For this reason, we sought a role for ASICs in mediating heartburn by determining whether amiloride could block heartburn in NERD induced by esophageal acid perfusion. METHODS: In a randomized double-blind crossover study, we perfused the esophagus with amiloride or (saline) placebo prior to eliciting acid-induced heartburn in patients with a history of proton pump inhibitor-responsive NERD. Liddle's disease: abnormal regulation of amiloride-sensitive Na+ channels by beta-subunit mutation.

Liddle's disease is an autosomal dominant genetic disorder characterized by severe low renin hypertension ("pseudoaldosteronism") that has been genetically linked to a locus on chromosome 16 encoding the beta-subunit of an amiloride-sensitive Na+ channel (ASSC) (15). Peripheral blood lymphocytes (PBL) express ASSC that are functionally indistinguishable from those expressed by Na(+)-reabsorbing renal epithelial cells (3, 5). The amiloride-sensitive Na+ conductance in PBL from affected and unaffected individuals from the original Liddle's pedigree was examined using whole cell patch clamp. Typi

Hypothyroidism affects the expression of electrogenic amiloride-sensitive sodium transport in rat colon.

BACKGROUND & AIMS: Both hyperthyroidism and hypothyroidism as well as hyperaldosteronism have been associated with changes of epithelial transport. The aim of this study was to evaluate the role of thyroid hormones in the regulation of electrogenic amiloride-sensitive sodium transport by aldosterone in the distal colon of immature and adult rats. METHODS: The changes in amiloride-sensitive short-circuit current (Isc) and Na+, K(+)-adenosine triphosphatase (ATPase) activity were measured in suckling, weanling, and adult euthyroid and methimazole-induced hypothyroid rats. RESULTS: The developm

Amiloride differentially modulates ANP binding in human thyroid cells and bovine endothelial cells.

The diuretic and sodium channel inhibitor, amiloride, has been shown to increase atrial natriuretic peptide (ANP) binding several fold in certain cell types, but in other tissues it causes only marginal increases in specific ANP binding. In the present report we compare the effects of amiloride on ANP binding in bovine endothelial cells and human thyroid-derived cells, two cell types which differ in their predominant ANP receptor subtype. We found that amiloride (10(-3) M) increased specific [125I]ANP binding to 750% above control in endothelial cells, but among several thyroid cultures tested

Amiloride alleviates morphine tolerance by suppressing ASIC3-dependent neuroinflammation in the spinal cord.

BACKGROUND: The use of morphine in clinical medicine is severely constrained by tolerance. Therefore, it is essential to examine pharmacological therapies that suppress the development of morphine tolerance. Amiloride suppressed the expression of inflammatory cytokines by inhibiting microglial activation. Microglia play a crucial role in the establishment of morphine tolerance. Thus, we anticipated that amiloride might suppress the development of morphine tolerance. During this investigation, we assessed the impact of amiloride on mouse morphine tolerance. METHODS: Mice received morphine (10

Amiloride ameliorates muscle wasting in cancer cachexia through inhibiting tumor-derived exosome release.

BACKGROUND: Cancer cachexia (CAC) reduces patient survival and quality of life. Developments of efficient therapeutic strategies are required for the CAC treatments. This long-term process could be shortened by the drug-repositioning approach which exploits old drugs approved for non-cachexia disease. Amiloride, a diuretic drug, is clinically used for treatments of hypertension and edema due to heart failure. Here, we explored the effects of the amiloride treatment for ameliorating muscle wasting in murine models of cancer cachexia. METHODS: The CT26 and LLC tumor cells were subcutaneously in

Evaluation of second generation amiloride analogs as therapy for cystic fibrosis lung disease.

Epithelial sodium channel (ENaC) blockers have been proposed as a therapy to restore mucus clearance (MC) in cystic fibrosis (CF) airways. The therapeutic effects of the first generation ENaC blocker, amiloride, in CF patients, however, were minimal. Because the failure of amiloride reflected both its low potency and short duration of action on airway surfaces, we investigated whether the increased potency of benzamil and phenamil would produce more favorable pharmacodynamic properties. In vitro potency, maximal efficacy, rate of recovery from maximal block of ENaC, and rate of drug absorption

Amiloride-sensitive sodium transport of the rat distal colon during early postnatal development.

To evaluate developmental changes in colonic sodium transport, the sensitivity of the transepithelial potential and short-circuit current to amiloride was investigated. The amiloride-sensitive short-circuit current (IscNa), which represents the electrogenic sodium transport through Na+ channels, rose significantly from day 5, reached a peak on day 10, and entirely disappeared after weaning. The maximum rate of electrogenic, amiloride-sensitive sodium transport was 12.0 microEq/cm2 X h. The IscNa was suppressed by adrenalectomy and/or premature weaning but not by a mineralocorticoid antagonist,

Cortical collecting tubule potassium secretion: effect of amiloride, ouabain, and luminal sodium concentration.

The present study examined the effect of sodium transport inhibition by amiloride, ouabain, and luminal sodium removal on potassium secretion in isolated cortical collecting tubules from adrenalectomized and DOCA-stimulated rabbits. Collecting tubules from adrenalectomized rabbits had a mean potassium secretion of 3.62 +/- 0.37 pmoles X mm-1 X min-1, which significantly decreased to 1.52 +/- 0.21 pmoles X mm-1 X min-1 after addition of amiloride, but no additional effect was observed after the addition of ouabain. The transepithelial voltage (VT) became less positive after exposure to amilorid

Amiloride-sensitive salt and fluid absorption in small intestine of sodium-depleted rats.

Secondary hyperaldosteronism produced by Na+ depletion was associated with increases in salt and fluid absorption in both the small intestine and the distal colon but not in the cecum and the proximal colon. Because these changes had not been documented for the small intestine, this study focused on the regulation of this tissue. Increased NaCl and water absorption was expressed in vitro by increases in short-circuit current and transepithelial potential and in vivo by increased fluid absorption and a decreased luminal content of Na+ and water. For example, the short-circuit current in the ile

Interactions of SARS-CoV-2 envelope protein with amilorides correlate with antiviral activity.

SARS-CoV-2 is the novel coronavirus that is the causative agent of COVID-19, a sometimes-lethal respiratory infection responsible for a world-wide pandemic. The envelope (E) protein, one of four structural proteins encoded in the viral genome, is a 75-residue integral membrane protein whose transmembrane domain exhibits ion channel activity and whose cytoplasmic domain participates in protein-protein interactions. These activities contribute to several aspects of the viral replication-cycle, including virion assembly, budding, release, and pathogenesis. Here, we describe the structure and dyna

Effect of amiloride on endoplasmic reticulum stress response in the injured spinal cord of rats.

After traumatic spinal cord injury (SCI), endoplasmic reticulum (ER) stress exacerbates secondary injury, leading to expansion of demyelination and reduced remyelination due to oligodendrocyte precursor cell (OPC) apoptosis. Although recent studies have revealed that amiloride controls ER stress and leads to improvement in several neurological disorders including SCI, its mechanism is not completely understood. Here, we used a rat SCI model to assess the effects of amiloride on functional recovery, secondary damage expansion, ER stress-induced cell death and OPC survival. Hindlimb function in

Amiloride inhibits the initiation of Coxsackievirus and poliovirus RNA replication by inhibiting VPg uridylylation.

The mechanism of amiloride inhibition of Coxsackievirus B3 (CVB3) and poliovirus type 1 (PV1) RNA replication was investigated using membrane-associated RNA replication complexes. Amiloride was shown to inhibit viral RNA replication and VPgpUpU synthesis. However, the drug had no effect on polymerase elongation activity during either (-) strand or (+) strand synthesis. These findings indicated that amiloride inhibited the initiation of RNA synthesis by inhibiting VPg uridylylation. In addition, in silico binding studies showed that amiloride docks in the VPg binding site on the back of the vir

Effects of amiloride and bumetanide on hyperpolarization after movement across the distal colon epithelium.

The aim of the paper was to define which transepithelial ion transport pathways were activated during stimulation by movement across the colonic epithelium. The experiments were performed with Ussing method on 241 specimens of isolated distal colon wall from 51 rabbits. The tissue was stimulated by a flux from peristaltic pump which caused transient hyperpolarization of the isolated tissue. In the control, the transepithelial potential difference (PD) and its changes during stimulation (dPD) were -3.16 +/- 1.81 mV and -2.16 +/- 0.99 mV, respectively. Preincubation of the tissue in the presence

Mutations in the extracellular loop of alpha-rENaC alter sensitivity to amiloride and reactive species.

We studied the effects of two mutations of the extracellular loop of the alpha-subunit of the (ENaC) on amiloride-sensitive current in Xenopus laevis oocytes and the inhibition of this current by 3-morpholinosydnonimine (SIN-1). Injection of oocytes with wild-type (wt) alpha-,beta-,gamma-rENaC cRNA (8.3 ng/subunit) resulted 48-72 h later in inward Na(+) currents (-5.5 +/- 0.8 microA; means +/- SE at -100 mV; n = 21), which were completely inhibited by amiloride. Oocytes injected with either alpha(Y279A)- or alpha(Y283A)- and beta-,gamma-rENaC cRNAs had significantly lower Na(+) currents. Furth

Effect of inhaled amiloride on the bronchial response to methacholine and cold air hyperventilation challenges.

Inhaled amiloride has been recently demonstrated to have an effect on the decline of pulmonary function in patients with cystic fibrosis. Other diuretics have been demonstrated to provide protection against bronchoconstriction in asthmatic subjects. We report on the effect of inhaled amiloride on cold air hyperventilation challenge (CAHC) and methacholine challenge in asthmatics. We studied nine subjects with mild-moderate asthma in a double-blind, placebo-controlled, crossover study. Our results showed amiloride did not significantly protect against the bronchoconstriction induced by CAHC. In

Effect of amiloride on sodium transport in the proximal, distal, and entire human colon in vivo.

In vitro studies under short-circuited conditions suggest that amiloride, a diuretic agent which is thought to block apical membrane sodium entry, has significant effects on sodium absorption by the human colon. To evaluate this in vivo, we studied the effect of amiloride applied in concentrations of 10(-5) and 10(-4) M on sodium transport and potential difference (PD) in human colon during steady-state perfusion. Net sodium absorption was reduced 25% by amiloride and chloride absorption by 15%; potassium and bicarbonate secretion rates were enhanced. In other studies the colon was divided int

Kinetics of the effect of amiloride on the permeability of the apical membrane of rabbit descending colon to sodium.

The effects of the addition of graded concentrations of amiloride, (A)m, to the mucosal bathing solution on the permeability of the apical membrane of rabbit descending colon to Na (PmNa) were determined when the Na activity in the mucosal bathing solution, (Na)m, was 18, 32 or 100 mM. PmNa was obtained from current-voltage relations determined on tissues bathed with a high-K serosal solution before and after the addition of a maximally inhibitory concentration of amiloride to the mucosal solution as described by Turnheim et al.
