Antifungal effect and mechanism of chitosan against the rice sheath blight pathogen, Rhizoctonia solani.

The antifungal properties and mechanism of three types of chitosan against the rice sheath blight pathogen, Rhizoctonia solani, were evaluated. Each chitosan had strong antifungal activity against R. solani and protected rice seedlings from sheath blight, in particular, two types of acid-soluble chitosan caused a 60-91 % inhibition in mycelial growth, 31-84 % inhibition of disease incidence, and 66-91 % inhibition in lesion length. The mechanism of chitosan in protection of rice from R. solani pathogen was attributed to direct destruction of the mycelium, evidenced by scanning and transmission

Progressive liver fibrosis in late-onset argininosuccinate lyase deficiency.

A 4-month-old boy, with late-onset argininosuccinate lyase (ASL) deficiency with hepatomegaly, was treated by protein restricted diet and arginine supplementation; he was followed for 3 years. Hepatomegaly and mild liver dysfunction persisted without significant hyperammonemia. He maintained normal psychomotor development to the age of 12 months, but, at 3 years of age, his developmental status is in the borderline normal range. Liver biopsy performed at 12 months of age demonstrated swollen and pale hepatocytes with abnormal glycogen deposition and mild periportal fibrosis. A subsequent liver

Clinical, biochemical and genetic characteristics of patients with argininosuccinate lyase deficiency from a single center cohort in China.

BACKGROUND: Argininosuccinate lyase deficiency (ASLD) is a rare autosomal recessive urea cycle disorder (UCD) resulting from mutations in the ASL gene. Previous studies of ASLD in patients from China have predominantly been limited to individual case reports, lacking comprehensive cohort study. This study aimed to systematically evaluate the clinical features, biochemical abnormalities, and genetic mutations of Chinese ASLD patients, thereby enhancing the understanding of the unique characteristics of this population. METHODS: We conducted a retrospective analysis of clinical and genetic data

Homocysteine over-accumulation as the effect of potato leaves exposure to biotic stress.

Homocysteine (Hcy) is a naturally occurring intermediate metabolite formed during methionine metabolism. It has been well documented that its excess can be extremely toxic to mammalian, yeast and bacterial cells. In spite of the metabolic value of Hcy known for decades, the role of this amino acid in the plant response to stress has not been recognized yet. In the presented study, using potato plant (Solanum tuberosum L.) and Phytophthora infestans as a model system, the presence and tissue localization of Hcy in leaves was examined by an immunohistochemical method. The over-production of Hcy

Herbivore-induced volatile production by Arabidopsis thaliana leads to attraction of the parasitoid Cotesia rubecula: chemical, behavioral, and gene-expression analysis.

Many plant species defend themselves against herbivorous insects indirectly by producing volatiles in response to herbivory. These volatiles attract carnivorous enemies of the herbivores. Research on the model plant Arabidopsis thaliana (L.) Heynh. has contributed considerably to the unraveling of signal transduction pathways involved in direct plant defense mechanisms against pathogens. Here, we demonstrate that Arabidopsis is also a good candidate for studying signal transduction pathways involved in indirect defense mechanisms by showing that: (1) Adult females of Cotesia rubecula, a specia

Dual porphyria in double heterozygotes with porphobilinogen deaminase and uroporphyrinogen decarboxylase deficiencies.

A coexistent dual deficiency of porphobilinogen deaminase (PBG-D; EC 4.3.1.8) and uroporphyrinogen decarboxylase (EC 4.1.1.37) in erythrocytes was recognized in five individuals, four males and one female. Clinically, the female and one male were diagnosed as suffering from acute intermittent porphyria (AIP), and the other two males were diagnosed as having porphyria cutanea tarda (PCT). Biochemically, the excretion pattern of urinary and fecal heme precursors exhibited a complex constellation with signs characteristic for both AIP and PCT. A coexistent dual enzyme deficiency of PBG-D and URO-

Variations in erythrocyte uroporphyrinogen I synthetase activity in non porphyrias.

The activity of erythrocyte uroporphyrinogen I synthetase has been measured in various cases of non-porphyric affections. The results indicate a diminution of this activity in some of the studied cases of chronic renal insufficiency and chronic polyarthritis. This modulation in activity mitigates against its use as a diagnostic criterion of acute intermittent porphyria. On the other hand, an increase in urosynthetase activity has been noted in acute and especially chronic hepatic affections. This increase seems to be connected with the severity of the hepatic affection. The relationship is ill

Modulation of Enhanced Antioxidant Activity by Hydrogen Sulfide Antagonization of Ethylene in Tomato Fruit Ripening.

Ethylene (C2H4) and hydrogen sulfide (H2S) play important physiological roles in regulating fruit ripening and senescence. The mechanism of H2S in ethylene-induced tomato fruit ripening and senescence is still unknown. Here, we show that exogenous H2S reduced the production of superoxide anion (·O2-), malondialdehyde (MDA), and H2O2 in tomato fruit. Further, additional H2S was found to induce the activities of guaiacol peroxidase, catalase, ascorbate peroxidase, and superoxide dismutase compared with C2H4 treatment alone, whereas the activities of lipoxygenase, polyphenol oxidase, and phenylal

Application of silica nanoparticles in maize to enhance fungal resistance.

In this study, maize treated with nanosilica (20-40 nm) is screened for resistance against phytopathogens such as Fusarium oxysporum and Aspergillus niger and compared with that of bulk silica. The resistivity is measured for disease index and expression of plant responsive compounds such as total phenols, phenylalanine ammonia lyase, peroxidase and polyphenol oxidase. The results indicate that nanosilica-treated plant shows a higher expression of phenolic compounds (2056 and 743 mg/ml) and a lower expression of stress-responsive enzymes against both the fungi. Maize expresses more resistance

Structure of the Oxygen, Pyridoxal Phosphate-Dependent Capuramycin Biosynthetic Protein Cap15.

Pyridoxal phosphate-dependent enzymes able to use oxygen as a co-substrate have emerged in multiple protein families. Here, we use crystallography to solve the 2.40 Å resolution crystal structure of Cap15, a nucleoside biosynthetic enzyme that catalyzes the oxidative decarboxylation of glycyl uridine. Our structural study captures the internal aldimine, pinpointing the active site lysine as K230 and showing the site of phosphate binding. Our docking studies reveal how Cap15 is able to catalyze a stereoselective deprotonation reaction, and bioinformatic analysis reveals active site residues tha

Phenolic Compounds of the Medicinal Plants in an Anthropogenically Transformed Environment.

In this article, the impact of an anthropogenically transformed environment on the content of pharmaceutically valuable biologically active compounds in medicinal plants is analyzed. The studied biologically active substances included phenolic compounds (flavonoids, anthocyanins, tannins, and phenolic acids). The number of transmissible forms of heavy metals (HMs), including cadmium, lead, and mercury, were discharged from factories that are present in the soil. Plants uptake these toxic metals from the soil. HM causes changes in the activity of the several enzymes such as phenylalanine ammoni

Exogenous methyl jasmonate enhances phytochemicals and delays senescence in harvested strawberries by modulating GABA shunt pathway.

In order to illustrate the relationship between methyl jasmonate (MeJA) and gamma aminobutyric acid (GABA) in maintaining the quality and improving the postharvest life of strawberry fruit, the harvested fruit were treated with MeJA for 16 h at 20 °C and stored at 3 ± 0.5 °C for 12 days. MeJA enhanced the expression levels of GABA shunt pathway-related genes, including glutamate decarboxylase, GABA transaminase, and succinic semialdehyde dehydrogenase, leading to an increase in GABA accumulation. Treated fruit showed higher levels of total acids, anthocyanins, total phenolics, antioxidants, an

Effect of photodynamic treatments on quality and antioxidant properties of fresh-cut potatoes.

This study evaluated the feasibility of curcumin based photodynamic sterilization technology (PDT) applied to fresh-cut potato slices. Potato samples with 30 μmol L-1 curcumin solution were exposed to 420 nm light emitting diodes (LED) at a total dose of 0.7 kJ cm-2. Results showed that PDT inactivated 2.43 log CFU mL-1 of Escherichia coli (BL 21) and 3.18 log CFU mL-1 of Staphylococcus aureus and maintained the color, texture, weight as well as total solid content of treated potatoes. Additionally, loss of phenols and flavonoids was significantly prevented, increasing the total antioxidant ca

Arbuscular mycorrhizal fungal inoculation increases phenolic synthesis in clover roots via hydrogen peroxide, salicylic acid and nitric oxide signaling pathways.

Arbuscular mycorrhizal fungi can increase the host resistance to pathogens via promoted phenolic synthesis, however, the signaling pathway responsible for it still remains unclear. In this study, in order to reveal the signaling molecules involved in this process, we inoculated Trifolium repense L. with an arbuscular mycorrhizal fungus (AMF), Glomus mosseae, and monitored the contents of phenolics and signaling molecules (hydrogen peroxide (H(2)O(2)), salicylic acid (SA), and nitric oxide (NO)) in roots, measured the activities of l-phenylalanine ammonia-lyase (PAL) and nitric oxide synthase (

Assimilation of exogenous cyanide cross talk in Oryza sativa L. to the key nodes in nitrogen metabolism.

Exogenous cyanide (CN-) effects on nitrogen (N) uptake, transport, and assimilation in rice seedlings were investigated at the biochemical and molecular levels. Seedlings were treated with either a 2-d or 4-d supply of potassium cyanide (KCN) in the nutrient solution containing nitrate (NO3-) or ammonium (NH4+). Although a KCN-induced increase was recorded in the activity of β-cyanoalanine synthase (β-CAS) in rice tissues of both NH4+-fed and NO3--fed seedlings, the former showed a significantly greater assimilation rate for CN- than the latter. The addition of KCN decreased NO3- uptake and as

Influence of sodium nitrilotriacetate (NTA) and citric acid on phenolic and organic acids in Brassica juncea grown in excess of cadmium.

Brassica juncea cv. 426308 was grown in soils containing 150 mg Cd(2+)kg(-1) soil. After 38 days, the soil was amended with two rates of citric acid or NTA (10 and 20 mmol kg(-1) soil). Control soil was not amended with chelates. Plants were harvested during growth, immediately before and seven days after chelate addition. Shoot composition of organic and phenolic acids and shoot Cd(2+) concentration were determined. Cadmium concentration remained constant during the growth and increased following NTA and citric acid amendments depending on chelate type and concentration. The highest increment

Quantitation of phenylbutyrate metabolites by UPLC-MS/MS demonstrates inverse correlation of phenylacetate:phenylacetylglutamine ratio with plasma glutamine levels.

Urea cycle disorders (UCDs) are genetic conditions characterized by nitrogen accumulation in the form of ammonia and caused by defects in the enzymes required to convert ammonia to urea for excretion. UCDs include a spectrum of enzyme deficiencies, namely n-acetylglutamate synthase deficiency (NAGS), carbamoyl phosphate synthetase I deficiency (CPS1), ornithine transcarbamylase deficiency (OTC), argininosuccinate lyase deficiency (ASL), citrullinemia type I (ASS1), and argininemia (ARG). Currently, sodium phenylbutyrate and glycerol phenylbutyrate are primary medications used to treat patients

Clinical experience with glycerol phenylbutyrate in 20 patients with urea cycle disorders at a UK paediatric centre.

In urea cycle disorders (UCDs) ammonia scavenger drugs, usually sodium-based, have been the mainstay of treatment. Increasingly, glycerol phenylbutyrate (GPB, Ravicti®) is being used but scant real-world data exist regarding clinical outcomes. A retrospective study of UCD patients initiated on or switched to GPB was performed at a UK centre. Data on population characteristics, treatment aspects, laboratory measurements, and clinical outcomes were collected before and after patients started GPB with a sub-group analysis undertaken for patients with ≥12 months of data before and after starting G
