BACKGROUND: Divya-Herbal-Peya (DHP) is a plant-based decoction containing fourteen herbs in precise quantities; usually prescribed by the practitioners in Ayurveda to alleviate stress and minimize the exasperating symptoms of recurring infections. Our study aims to provide an experimental validation to the immunomodulatory properties of DHP.
METHODS: Physico-chemical analysis of DHP was performed to evaluate the presence of secondary metabolites. The phytochemicals were then identified and quantitated through HPTLC, UHPLC, and GC-MS techniques. To address the scientific rationale behind DHP,

Myxozoans are economically important cnidarian endoparasites. Members of this group have been traditionally characterized by a morphology-based taxonomic system. Because myxozoans possess few morphological characters, these data are routinely accompanied by biological traits (host/organ/tissue specificity) and molecular data when describing or identifying myxozoan species. In the present study, a species of Myxobolus was collected from the fins of yellow catfish Tachysurus fulvidraco Richardson, 1846, which was consistent in spore morphology and host/organ specificity with Chinese records of M

Regeneration programs enable animals to restore damaged or lost tissues, and the range of stimuli for these programs is incompletely understood. Here, we used zebrafish, a vertebrate species with exceptional regenerative capacity, to identify chemically induced mutations that alter regeneration-associated gene activation. Transgenic zebrafish with a permissive promoter and EGFP cassette inserted in the vicinity of the pro-regenerative factor gene fgf20a were mutagenized, and larvae homozygous for ENU-induced mutations were assessed for disruptions in fgf20a-directed reporter gene expression fo

Sophora flavescens Ait. (SFA), an extensively utilized herb for the treatment of fevers, inflammatory disorders, ulcers and skin diseases related to bur, contains oxymatrine (OMT) as its principal active constituent. OMT exerts regulatory effects over inflammation, oxidative stress and apoptosis. Neutrophils, critical regulators of the inflammation response, have not been thoroughly elucidated regarding the protective properties and underlying mechanisms of OMT-mediated anti-inflammation. This study was aim to explore the protective effect of OMT on neutrophils under inflammatory conditions an

The midbrain locomotor region (MLR) in the Atlantic stingray, Dasyatis sabina, was identified and characterized. Stimulation (50-100 microA, 60 Hz) of the midbrain in decerebrated, paralyzed animals was used to elicit locomotion monitored as alternating activity in nerves innervating an antagonist pair of elevator and depressor muscles. Effective sites for evoking locomotion in the midbrain included parts of several nuclei: the caudal portion of the interstitial nucleus of the medial longitudinal fasciculus and the caudomedial parts of the cuneiform and subcuneiform nuclei. This region did not

In vertebrates, vitamin A (VA) is crucial for development, tissue homeostasis, vision, and immunity. Retinal, a form of VA, can be produced via enzymatic cleavage of β-carotene by beta-carotene oxygenase 1 (bco1) and bco1-like (bco1l), but the developmental and tissue-specific functions of these genes are poorly understood. While bco1 is found across vertebrate taxa, bco1l is a paralog of bco1 that we discover to have evolved in the ray-finned fishes, the most abundant, speciose, and commercially important group of fishes. We investigated the function of bco1l in ray-finned Siamese fighting fi

The degradation of elastin in aged skin results in the loss of elasticity, the formation of wrinkles, and even a reduced ability for wound healing after injury. However, previous studies about elastin material have been limited by high immunogenicity of animal-derived elastin, the use of single elastin peptide sequences, and residual crosslinking agents. In this study, we designed a multipotent anti-aging recombinant human elastin (rElastin) and utilized it to counteract caudal fin shrinkage in zebrafish and D-galactose-induced intrinsic skin aging in mice. The rElastin demonstrated thermoresp

The mangrove killifish, Kryptolebias marmoratus, can reproduce with self-fertilisation, offering a unique and useful genetic tool for generation of genetic mutants and quick identification of mutated genes. From an ENU-mutated mangrove killifish line R228, we have isolated a novel mutant line, no-fin-ray/nfr in which homozygous mutant of adult fish fin ray development is largely reduced. Illumina RNAseq with 3 embryos each from mutants, siblings and the parental WT strain Hon9 (only 9 embryos as total) identified a mutation in the edaradd in a highly conserved C-terminal death domain. Edaradd

BACKGROUND: Previous studies found that Polygonatum sibiricum polysaccharide (PSP) exhibited anti-inflammatory and anti-oxidative effects on type 2 diabetes (T2DM). However, the roles and detailed mechanisms of PSP on the anti-diabetic osteoporosis are still poorly defined. The purpose of this study was to identify the detailed mechanisms of PSP-induced anti-diabetic osteoporosis. METHODS: The roles of PSP on diabetic osteoporosis were evaluated on a T2DM zebrafish model. Furthermore, the changes of related physiological indicators and metabolites in adult zebrafish (Danio rerio) were investig

The role of proteoglycans (PGs) in regulating growth factor signalling during endochondral ossification, or formation of bone around a cartilage template, is established. However, whether PGs regulate other skeletal processes, like intramembranous ossification or fin regeneration, has not been studied extensively. Given that endochondral ossification in PG-deficient fam20b mutant zebrafish is altered due to increased Bmp signalling, we hypothesized that PGs also normally inhibit Bmp signalling during intramembranous ossification and fin regeneration. To test this hypothesis, the functional rel

Largemouth bass (Micropterus salmoides), a freshwater fish species, is cultivated widely across China. The industry has been greatly affected by various viral diseases. We generated a new immortal cell line from the fin of M. salmoides (MSF). It has been successfully maintained in continuous culture for over 80 passages. Cells multiply well at 28°C using Medium 199 or Leibovitz's L-15 supplemented with 10%-20% fetal bovine serum. MSF cells comprise a mass of epithelialoid cells. Chromosome analysis showed that ploidy at passage 55 was 2n = 58. Amplification and analysis of the sequence of cyto

Present study highlighted the ultramicroscopic (SEM) alterations of the skin, eye, barbel, and fins of spawn of an air-breathing teleost (Clarias batrachus, Linn. 1758) induced by UV-B radiation (280-320 nm) at a dose (@4.07 × 10-20J/photon/m2) under the time-frame of 5, 10 and 15 min/d in the laboratory condition for the periods of 5 and 10 days. Limnological parameters revealed no significant changes throughout the period of experimentation which were measured by PCS Testr 35 Multi-Parameter. Morphometric analysis revealed that during the extended exposure period of 10 days the spawn size an

BACKGROUND: A long, slender body plan characterized by an elongate antorbital region and posterior displacement of the unpaired fins has evolved multiple times within ray-finned fishes, and is associated with ambush predation. The axial skeleton of ray-finned fishes is divided into abdominal and caudal regions, considered to be evolutionary modules. In this study, we test whether the convergent evolution of the ambush predator body plan is associated with predictable, regional changes in the axial skeleton, specifically whether the abdominal region is preferentially lengthened relative to the

As many tropical aquatic ecosystems worldwide, the Doce River estuary (DRE, southeastern Brazil) has increasingly faced multiple anthropogenic threats, including deforestation, mining, species introduction and water management. The 2015 collapse of the Fundão iron ore tailings dam severely changed water properties and increased heavy metal concentration in sediment. However, the potential synergistic effects of the observed threats on fish populations remain largely unknown. After detecting an elevated number of fin lesions in DRE fishes, we used the catfish Genidens genidens (Cuvier, 1829) to

Fishes exhibit an astounding diversity of locomotor behaviors from classic swimming with their body and fins to jumping, flying, walking, and burrowing. Fishes that use their body and caudal fin (BCF) during undulatory swimming have been traditionally divided into modes based on the length of the propulsive body wave and the ratio of head:tail oscillation amplitude: anguilliform, subcarangiform, carangiform, and thunniform. This classification was first proposed based on key morphological traits, such as body stiffness and elongation, to group fishes based on their expected swimming mechanics.

Hoxc6 gene can be described as having roles in axial patterning in early embryogenesis, and in at least some species, having a contribution to limb positioning. In this study, we cloned and characterised Pampus argenteus Hoxc6. The highly conserved HOXC6 protein sequence contains a homeodomain and a low-complexity region. Expression of Hoxc6 mRNA was measured at different developmental stages and in different tissues by real-time PCR (p < 0.05), and was high during eye capsule and brain differentiation stages, but low in 7 and 13-day-old larvae. Hoxc6 mRNA was more abundant in fin tissue than

BACKGROUND: Matrix metalloproteinases 13 (MMP13) is a potent endopeptidase that regulate cell growth, migration, and extracellular matrix remodeling. However, its role in fin regeneration remains unclear.
RESULTS: mmp13a expression is strongly upregulated during blastema formation and persists in the distal blastema. mmp13a knockdown via morpholino electroporation impairs regenerative outgrowth by decreasing cell proliferation, which correlates with a downregulation of fgf10a and sall4 expression in the blastema. Laminin distribution in the basement membrane is also affected in mmp13a MO-inje

BACKGROUND: Translocation of native species and introduction of non-native species are potentially harmful to the existing biota by introducing e.g. diseases, parasites and organisms that may negatively affect the native species. The enemy release hypothesis states that parasite species will be lost from host populations when the host is introduced into new environments.
METHODS: We tested the enemy release hypothesis by comparing 14 native and 29 introduced minnow (Phoxinus phoxinus) populations in Norway with regard to the ectoparasitic Gyrodactylus species community and load (on caudal fin
