Different affinity of monoclonal antibodies for conserved neutralizing epitopes on two strains of rubella virus.

There is, apparently, only one serological type of rubella virus (RV) in the population, although several isolates exist with different characteristics. Some authors failed to detect significant differences among RV strains by neutralization, hemagglutination inhibition, and enzyme immunoassay using polyclonal and monoclonal antibodies, but differences in growth, plaque morphology, and temperature sensitivity between vaccine and wild-type strains were shown by Chantler et al. (3) With the purpose of analyzing the possible differences among several strains of RV, we studied the affinity constan

Production of a monoclonal antibody specific for Mycobacterium avium and immunological activity of the affinity-purified antigen.

Three hybridomas which secrete antibodies to Mycobacterium avium were obtained by the fusion of p3u1 myeloma cells with spleen cells of mice immunized with M. avium culture sonicate. The reactivity of these monoclonal antibodies was determined in 16 species of mycobacteria by an enzyme-linked immunosorbent assay. An antibody, designated Avi-3, reacted only with M. avium and not with the reference strains of the other 15 species of mycobacteria tested, including M. intracellulare, M. paratuberculosis, and M. lepraemurium. Specificity of the antibody was confirmed by assay, using a specific DNA

Relief of pain in mice by an antibody with high affinity for cell adhesion molecule 1 on nerves.

AIMS: Cell adhesion molecule 1 (CADM1) is a member of the immunoglobulin superfamily and is abundantly expressed on nerve fibers. Recently, the anti-CADM1 ectodomain antibody 3E1 has proven useful as a drug delivery vector for CADM1-expressing cells in vitro. When injected subcutaneously into mice, whether 3E1 accumulates on nerve fibers and serves as an analgesic was examined. MAIN METHODS: Injected 3E1 was detected by immunohistochemistry and double immunofluorescence. Analgesic effects were verified by a formalin-induced chemical-inflammatory pain test and video-recorded behavior analysis

Purification of antibodies to O antigen of Salmonella Typhimurium from human serum by affinity chromatography.

Nontyphoidal Salmonellae (NTS) are a common cause of bacteraemia in children and HIV-infected adults in Sub-Saharan Africa. We have previously shown that antibodies play a key role in both bactericidal and cellular mechanisms of immunity to NTS, but found that high concentrations of antibody to Salmonella Typhimurium O antigen (OAg) in the serum of some HIV-infected African adults is associated with impaired killing of NTS. To further investigate the function of antibodies to the OAg of NTS, we developed a method to purify these antibodies from human serum by affinity chromatography. Purified

Antibodies as drug carriers III: design of oligonucleotides with enhanced binding affinity for immunoglobulin G.

PURPOSE: To understand the structural requirements in designing epitope-bearing oligonucleotides with high antibody-binding affinity. METHODS: Binding affinity (KA) and stoichiometry (n) of dinitrophenyl (DNP)-derivatized model 27-mer oligonucleotides (ODNs), GGG(AAA)7GGG, to monoclonal anti-trinitrophenyl (TNP) antibodies were determined using isothermal titration calorimetry (ITC). Structural variations were made in the ODNs to assess the effects of antigenic valence, epitope density, inter-epitope linker length, and linker flexibility. Binding isotherms were fitted with a single binding-si

IgG subclass distribution and relative functional affinity of thyroid microsomal antibodies in postpartum thyroiditis.

An association between the development of postpartum hypothyroidism and high levels of IgG1 subclass microsomal (M) antibodies has been reported. Using an assay designed to detect reasonable levels of all the four IgG subclasses, we found no differences in the proportion of each IgG subclass in M antibodies of patients with postpartum hyperthyroidism or hypothyroidism compared with control postpartum patients with M antibodies but no thyroid dysfunction. However the total amount of M antibody of each IgG subclass was elevated above the controls in the patients with thyroid dysfunction. The rel

Identification of Fasciola hepatica gut-associated glycoproteins as potential vaccine candidates by lectin-affinity chromatography, flukicidal monoclonal antibodies, and affinity-enrichment mass spectrometry.

A significant economic impact to the agriculture and declining drug efficacy due to anthelmintic resistance lead to the search for novel intervention approaches against the zoonotic liver fluke Fasciola hepatica. A cost-effective strategy is to induce protective immunity by vaccination. However, adaptation of the parasite to evade the host immune response hampers classical vaccine development. Therefore, an ideal protective epitope would not be subjected to this evolutionary selection. Antigens, which are not exposed to the host's immune system during a natural parasite infection, but which co

Strategy for the characterization of autoantigens in autoimmune diseases. Investigation of the target antigens of antimitochondrial antibodies by radioimmunoassay, immunoblotting, monoclonal antibodies and affinity chromatography.

We present a strategy to characterize specific antigen/autoantibody systems using polyclonal sera from patients as a probe and crude antigenic preparations such as mitoplasts. Sera from patients with primary biliary cirrhosis (PBC) are characterized by at least one of two specific subtypes of antimitochondrial antibodies (AMA), anti-p48 or anti-p62. Immunoblotting of such sera with mitoplast preparations derived from human, rat and rabbit livers revealed three proteins of approximately 27, 48 and 68 kDa as target antigens. On the basis of the molecular weight of these antigens we were able to

High-affinity optimization potential of the virus neutralizing antibody with twin cysteine-stabilized complementarity-determining region 3.

INTRODUCTION: The optimization of neutralizing monoclonal antibodies (NMAbs) is crucial to counter viral evolution. The structural stability of the heavy-chain complementarity-determining region 3 (H3 CDR) significantly influences affinity maturation potential, yet its impact on computational optimization remains unclear. METHODS: This study employed an artificial intelligence (AI) model to optimize two categories of SARS-CoV-2 NMAbs: one featuring a conformationally stabilized H3 CDR via a twin cysteine motif, and another with flexible H3 CDR loops. Optimized antibody derivatives were evalua

Local antibody feedback enforces a checkpoint on affinity maturation in the germinal center and promotes epitope spreading.

Circulating antibodies from previous immune encounters impact subsequent humoral responses. Here, we investigated how local epitope-specific competition shapes ongoing germinal center (GC) responses by delivering an mRNA-LNP-encoded membrane-bound immunogen displaying three conserved HIV-1 envelope (Env) epitopes to mouse models bearing B cell receptors (BCRs) of defined affinities. High-affinity B cells exhibited shorter GC residency than lower-affinity counterparts. B cells engaged GC reactions at equivalent rates in the presence or absence of clonal lineages binding the same epitope with si

Phys-AbGAT: a physics-informed multi-task graph attention network for robust antibody-antigen binding affinity prediction.

MOTIVATION: Accurate prediction of antibody-antigen binding affinity is essential for therapeutic antibody design. Existing methods often model the dissociation constant (KD) and Gibbs free energy (ΔG) independently, overlooking their thermodynamic relationship and complex paratope-epitope spatial interactions. RESULTS: We introduce Phys-AbGAT, a physics-informed multi-task graph attention network that represents the antibody-antigen interface as a spatial graph. It integrates ESM2 evolutionary embeddings with radial basis functions and employs a thermodynamically motivated auxiliary loss to

Directed evolution of a stem-helix-targeting antibody enables MERS-CoV cross-neutralization through enhanced binding affinity.

Broadly neutralizing antibodies (bnAbs) targeting conserved regions of the betacoronavirus spike are important for pan-betacoronavirus protection and pandemic preparedness. Here, we report on the isolation of a human monoclonal antibody, CC65.1, from a SARS-CoV-2 convalescent donor that targets the conserved S2 stem helix region. CC65.1 neutralizes various sarbecoviruses, including SARS-CoV-2, and binds the MERS-CoV spike but lacks MERS-CoV-neutralizing activity due to insufficient binding affinity. We utilized directed evolution to enhance the binding affinity of CC65.1 for the MERS-CoV S2 st

Affinity targeting of therapeutic proteins to the bone surface-local delivery of sclerostin-neutralizing antibody enhances efficacy.

Currently available biotherapeutics for the treatment of osteoporosis lack explicit mechanisms for bone localization, potentially limiting efficacy and inducing off-target toxicities. While various strategies have been explored for targeting the bone surface, critical aspects remain poorly understood, including the optimal affinity ligand, the role of binding avidity and circulation time, and, most importantly, whether or not this strategy can enhance the functional activity of clinically relevant protein therapeutics. To investigate, we generated fluorescent proteins (eg, mCherry) with site-s

Severely exacerbated neuromyelitis optica rat model with extensive astrocytopathy by high affinity anti-aquaporin-4 monoclonal antibody.

INTRODUCTION: Neuromyelitis optica (NMO), an autoimmune astrocytopathic disease associated with anti-aquaporin-4 (AQP4) antibody, is characterized by extensive necrotic lesions preferentially involving the optic nerves and spinal cord. However, previous in-vivo experimental models injecting human anti-AQP4 antibodies only resulted in mild spinal cord lesions compared to NMO autopsied cases. Here, we investigated whether the formation of severe NMO-like lesions occurs in Lewis rats in the context of experimental autoimmune encephalomyelitis (EAE), intraperitoneally injecting incremental doses o

High affinity binding of long-chain polysialic acid to antibody, and modulation by divalent cations and polyamines.

Long-chain polysialic acid (PSA) is expressed on the vertebrate neural cell adhesion molecule (NCAM) during neuronal plasticity. Its structural similarity to the capsular PSAs of some pathogenic bacteria has hampered the development of polysaccharide vaccines against meningitis. The antibodies formed during immunization require a long epitope for binding, and cross-react with host tissue PSA. The nature of the epitope and possible external effectors involved are still unclear. We have evaluated the interaction of PSA with its antibody mAb735 by surface plasmon resonance. The influences of PSA

Computational rescue of antibody binding to monkeypox virus E8L: from electrostatic repulsion to salt bridge-enhanced affinity.

The ongoing Mpox (monkeypox) outbreak underscores the need for effective therapeutic antibodies. Antibody VACV-66 binds strongly to the vaccinia virus D8 protein but shows markedly reduced cross-reactivity to the MPXV homolog E8L, despite >95% sequence identity. In this study, we combined AlphaFold3 modeling, molecular dynamics simulations, and ASGB-IE free energy calculations to investigate this mechanism and rationally design high-affinity antibody mutations targeting E8L. Our calculations revealed that the single N18D mutation in E8L (where N18 in D8 corresponds to D18 in E8L) introduces el

Novel fully human high-affinity anti-TREM2 antibody shows efficacy in clinically relevant Alzheimer´s mouse model.

BACKGROUND: New drugs to treat Alzheimer´s disease (AD) are urgently needed. Human triggering receptor expressed on myeloid cells 2 (hTREM2) is a validated drug target which is genetically associated with AD. Existing anti-hTREM2 antibodies were raised in animal immune systems, and subsequently humanized, which may incur immunological complications upon repeated preventive or therapeutic applications in vivo in AD patients. In addition, anti-hTREM2 antibodies should be optimized for both, efficacy and safety. METHODS: A novel fully human monoclonal brain-targeting anti-hTREM2 antibody M07-TFN
