Targeting TOMM40 unleashes immunogenic mitophagy to facilitate antigen presentation and immunotherapy sensitization.

BACKGROUND: Mitophagy is a mitochondrial quality control process that maintains cellular homeostasis in cancer, yet whether its dysregulation can be exploited to induce tumor immunogenicity remains unclear. METHODS: We integrated pancancer single-cell transcriptomic analyses with genetic perturbation strategies in hepatocellular carcinoma models, including CRISPR/Cas9-mediated gene depletion, in vivo syngeneic tumor systems, and RNA-based lipid nanoparticle delivery. Mechanistic investigations combined mitochondrial functional assays, imaging-based mitophagy analysis, flow cytometry, and tran

HLA-Shuttle: A system for enhancing antigen presentation in immunologically cold tumors.

Epitopic peptides presented by class-I human leukocyte antigen (HLA-I) proteins provide the basis of immune surveillance by T cells. Conversely, reduced surface HLA-I expression is a hallmark of immune evasion by latent viral infections and cancer, which confounds the identification of peptide antigens and neoantigens. Here, we outline a system (HLA-Shuttle) for in vitro manipulation of cells with engineered components of the HLA-I processing machinery to confer a continuum of chaperoning activity throughout their trafficking pathway. HLA-Shuttle restores antigen presentation in immunologicall

Perforin-2 enhances antigen-specific CTL immune response by promoting cross presentation.

Antigen cross-presentation is essential for initiating CD8+ cytotoxic T lymphocyte (CTL) response. Perforin-2 (P2), a pore-forming protein constitutively expressed in dendritic cells (DCs), has been implicated in endocytic cargo escape, but its role in cross-presentation remains poorly defined. Here, we show that loss of P2 markedly impairs DC-mediated cross-presentation of both soluble and particulate antigens, leading to weakened antigen-specific CD8+ T cell responses. Additionally, P2-/- mice exhibited defective endogenous CTL responses and diminished anti-tumor immunity in melanoma models.

Phosphorylation shapes antigen presentation and immune recognition: mechanistic insights and translational perspectives.

Phosphorylation is a reversible post-translational modification that dynamically regulates immune responses by reshaping the antigen profiles. Phosphoproteomics and immunopeptidomics studies reveal that phosphorylated peptides (phosphopeptides) are naturally processed and presented by MHC class I and II molecules and can elicit robust T-cell responses. These epitopes influence proteasome cleavage, TAP transport, and MHC loading, defining the immunopeptidome under physiological and pathological states. Structurally, phosphate groups enhance MHC binding and immunogenicity, positioning phosphoryl

Humanized FcεRI Expressed on Mouse Eosinophils Mediates IgE-Facilitated Eosinophil Antigen Presentation.

High-affinity IgE receptors (FcεRI) are expressed on human blood eosinophils and may be upregulated on eosinophils at sites of allergic inflammation including atopic dermatitis and allergic asthma. FcεRI engagement, however, fails to elicit "effector" responses from eosinophils. Thus, a functional role for FcεRI on eosinophils has been uncertain. We evaluated the role of FcεRI in enhancing eosinophil antigen presentation in vivo by using humanized FcεRI α chain (hFcεRIα) transgenic mice. Eosinophils from hFcεRIα transgenic mice expressed humanized FcεRIα, with higher levels of eosinophils from

Antigen Presentation by Extracellular Vesicles in the Acute Phase of Corneal Transplantation.

PURPOSE: Corneal transplantation is the most frequently performed solid organ transplantation procedure worldwide. We aimed to investigate the role of extracellular vesicles in allorecognition and the rejection of allografts in corneal transplantation using male C57BL/6 (B6 and IAb) and BALB/c (IAd) mice. METHODS: The mice were aged 8 to 12 weeks. We used transmission electron microscopy to visualize the drainage of extracellular vesicles derived from the graft into the cervical lymph nodes using gold colloids as markers in a mouse model of corneal transplantation. RESULTS: Lymph node cells

Immunoevasion strategies for African swine fever virus: Modulation of antigen presentation pathways.

African swine fever virus (ASFV), a highly lethal pathogen, poses a catastrophic threat to global pork production. While some commercial vaccines exist, their availability remains regional limitations. This review investigates the mechanistic interplay between ASFV and antigen-presenting cells (APCs), with a particular focus on viral evasion strategies that disrupt antigen presentation pathways. Although current understanding of ASFV immunology has largely focused on monocyte/macrophage infection, we provide a systematic analysis of ASFV-mediated immunosuppression across various APCs, includin

Massively parallel immunopeptidome by DNA sequencing provides insights into cancer antigen presentation.

Human leukocyte antigens (HLAs) are encoded by the most polymorphic genes in the human genome. HLA class I alleles control antigen presentation for T cell recognition, which is pivotal for autoimmunity, infectious diseases and cancer. Current knowledge of HLA-bound peptides is limited, skewed and falls short of population-wide HLA binding profiles for high-value targets. Here we present ESCAPE-seq (enhanced single-chain antigen presentation sequencing), a massively parallel platform for comprehensive screening of class I HLA-peptide combinations for antigen presentation via deep DNA sequencing

Functional analysis of antigen presentation by enteric glial cells during intestinal inflammation.

The Enteric Nervous System is composed of a vastly interconnected network of neurons and glial cells that coordinate to regulate homeostatic gut function including intestinal motility, nutrient sensing, and mucosal barrier immunity. Enteric Glial Cells (EGCs) are a heterogeneous cell population located throughout the gastrointestinal tract and have well described roles in regulating intestinal immune responses. Enteric Glial Cells have been suggested to act as nonconventional antigen presenting cells via the Major Histocompatibility Complex II (MHC II), though this has not been confirmed funct

TFEB regulates dendritic cell antigen presentation to modulate immune balance in asthma.

OBJECTIVE: Asthma stands as one of the most prevalent chronic respiratory conditions in children, with its pathogenesis tied to the actived antigen presentation by dendritic cells (DCs) and the imbalance within T cell subgroups. This study seeks to investigate the role of the transcription factor EB (TFEB) in modulating the antigen presentation process of DCs and its impact on the differentiation of T cell subgroups. METHODS: Bone marrow dendritic cells (BMDCs) were activated using house dust mites (HDM) and underwent RNA sequencing (RNA-seq) to pinpoint differentially expressed genes. TFEB m

Antigen presentation induced variation in ovalbumin sensitization between chicken and duck species.

The difference in the allergenicity of chicken ovalbumin (C-OVA) and duck ovalbumin (D-OVA) can be related to their differences in antigen presentation. This study explored the differences in uptake between C-OVA and D-OVA through fluorescence dye-labeling, DC antigen presentation, and the immune response of T cells by using C-OVA and D-OVA allergic animal and cell models. The ileum DCs of mice in the C-C group took up more C-OVA than that of D-D and C-D groups through in vivo imaging. Furthermore, C-OVA induced the maturation of DCs in mice in the C-C group as shown in the up-regulation of th

Cowpox virus employs a two-pronged strategy to outflank MHCI antigen presentation.

Smallpox decimated humanity for thousands of years before being eradicated by vaccination, a success facilitated by the fact that humans are the only host of variola virus. In contrast, other orthopoxviruses such as cowpox virus can infect a variety of mammalian species, although its dominant reservoir appears to be rodents. This difference in host specificity suggests that cowpox may have developed promiscuous immune evasion strategies to facilitate zoonosis. Recent experiments have established that cowpox can disrupt MHCI antigen presentation during viral infection of both human and murine c

Role of keratinocytes in antigen presentation and polarization of human T lymphocytes.

Allergic contact dermatitis is a T-cell-mediated inflammatory skin disease in which interaction between skin keratinocytes and migrating T lymphocytes may play a critical part. In this study, the role of keratinocytes as allergen-/antigen-presenting cells (APCs) leading to activation of T lymphocytes is investigated using a human epidermal cell line A431. It is known that cultured cells do not express human leucocyte antigen (HLA) and hence can be used as APCs independent of HLA profile of both APCs and T cells from human volunteers. This cell line responded to common allergens and irritants b

Antigen presentation and cytotoxic T lymphocyte killing studied in individual, living cells.

Interactions between individual, living fibroblasts and cytotoxic T lymphocyte (CTL) clones were analyzed by using video-enhanced differential interference contrast and fluorescence microscopy in a multimode configuration. Fibroblasts expressing known major histocompatibility complex I alleles (MC57: H-2b; Balb: H-2d) were sensitized for killing by incubating or microinjecting them with peptide fragments of lymphocytic choriomeningitis virus. Previous determination of the CTL clones' specificity for these peptides and MHC-I alleles enabled us to study CTL killing of fibroblasts, and nonlethal

The transfusion effect is influenced by the nature of MHC antigen presentation.

We designed a study to determine whether the mode of presentation of major histocompatibility antigens is important for the ability of donor-specific blood transfusions to prolong organ allograft survival. Donor BN rat whole blood, isolated RBC, RBC ghosts, RBC membrane fragments, or whole blood lysates were administered to Lewis rat recipients 7 days before heterotopic allotransplantation of BN hearts. Only allogeneic whole blood or RBC significantly prolonged cardiac allograft survival in this histoincompatible donor-recipient pair. Whole blood lysates or RBC ghosts and membrane fragments tr

Macrophage MR1 antigen presentation promotes MAIT cell immunity and lung microbiota modulation.

Mucosal-associated invariant T cells (MAIT cells) mediate tissue homeostasis and antimicrobial immunity. However, the cells that express major histocompatibility complex (MHC) class I-related protein 1 (MR1) and present microbial vitamin B-derived antigens (VitBAg) to MAIT cells remain unknown. We found that MR1 expression varied across tissues and cell types. Macrophages from the lung and peritoneal cavity expressed the highest levels of MR1 and were the most efficient at capturing and presenting VitBAg to MAIT cells. Expression of MR1 in macrophages was regulated transcriptionally and induce

Viral miRNA antagomirs rescue antigen presentation pathway in ocular HSV-1 infection.

Herpes simplex virus type 1 (HSV-1) establishes lifelong latency by deploying multiple mechanisms to evade host immunity. However, the role of virus-encoded microRNAs (v-miRs) in modulating antiviral immune responses remains incompletely understood. We previously showed that topical inhibition of the HSV-1 v-miRs miR-H6-3p and miR-H27 in infected animals reduces viral titers and restores immune homeostasis. Here, using a murine model of corneal HSV-1 infection, we examined whether in vivo inhibition of these v-miRs restores expression of genes involved in antigen processing and presentation. T

Lymphodepleting chemotherapy potentiates neoantigen-directed T cell therapy by enhancing antigen presentation.

Adoptive cell therapy (ACT) targeting tumor-specific antigens holds promise for solid tumors, but limited neoantigen presentation remains a key barrier to efficacy. Here, we identify and characterize a T cell receptor (TCR), T104, for the KRAS.G12V mutation, a prevalent neoantigen in colorectal, lung, and pancreatic cancers. TCR-T104 selectively recognizes and kills KRAS.G12V-expressing tumor cells. Combining T cell therapy with lymphodepleting chemotherapy significantly enhances tumor cell killing, particularly by TCR-T cells, tumor-infiltrating lymphocytes (TILs), and T cell engager antibodi

Cholangiocarcinoma PDHA1 succinylation suppresses macrophage antigen presentation via alpha-ketoglutaric acid accumulation.

Gemcitabine combined with cisplatin is the first-line chemotherapy for advanced cholangiocarcinoma, but drug resistance remains a challenge, leading to unsatisfactory therapeutic effect. Here, we elucidate the possibility of chemotherapy regimens sensitized by inhibiting succinylation in patients with cholangiocarcinoma from the perspective of post-translational modification. Our omics analysis reveals that succinylation of PDHA1 lysine 83, a key enzyme in the tricarboxylic acid cycle, alters PDH enzyme activity, modulates metabolic flux, and leads to alpha-ketoglutaric acid accumulation in th
