Inhibition of mitochondrial function: An alternative explanation for the antipyretic and hypothermic actions of acetaminophen.

AIMS: Acetaminophen is the medication of choice when treating fever because of its limited anti-inflammatory effects. However at overdose it can cause mitochondrial dysfunction and damage, often associated with metabolism to N-acetyl-p-benzoquinone imine (NAPQI). What has never been investigated is whether the inhibition of mitochondrial function, particularly fatty acid uptake and oxidation could be the key to its antipyretic and hypothermic properties. METHODS: Mitochondrial function and fatty acid oxidation (FAO) was determined by measuring oxygen consumption rate (OCR) in isolated mitocho

Immunomodulatory effects of pharmaceutical opioids and antipyretic analgesics: Mechanisms and relevance to infection.

Understanding how pharmaceutical opioids and antipyretic analgesics interact with the immune system potentially has major clinical implications for management of patients with infectious diseases and surgical and critical care patients. An electronic search was carried out on MEDLINE, EMBASE, PsycINFO, CENTRAL and the Cochrane library to identify reports describing the immunomodulatory effects of opioid analgesics and antipyretic analgesics, and their effects in infectious diseases. In adaptive immunity, nonsteroidal anti-inflammatory drugs have divergent effects: augmenting cell-mediated immu

Toxicological Property of Acetaminophen: The Dark Side of a Safe Antipyretic/Analgesic Drug?

Acetaminophen (paracetamol, N-acetyl-p-aminophenol; APAP) is the most popular analgesic/antipyretic agent in the world. APAP has been regarded as a safer drug compared with non-steroidal anti-inflammatory drugs (NSAIDs) particularly in terms of lower risks of renal dysfunction, gastrointestinal injury, and asthma/bronchospasm induction, even in high-risk patients such as the elderly, children, and pregnant women. On the other hand, the recent increasing use of APAP has raised concerns about its toxicity. In this article, we review recent pharmacological and toxicological findings about APAP fr

A peripheral mechanism of fever: differential sensitivity to the antipyretic action of methyl scopolamine.

The organophosphate pesticide (OP) chlorpyrifos leads to an acute period of hypothermia followed by a delayed fever in the rat. Methyl scopolamine, a peripheral muscarinic antagonist, is thought to have little effect on body temperature of the rat because it does not cross the blood brain barrier. However, administration of methyl scopolamine (1 mg/kg, i.p.) during the period of chlorpyrifos-induced fever results in a rapid recovery of core temperature. This indicates a peripheral cholinergic pathway is operative in the febrile response to chlorpyrifos and possibly other modes of fever. In thi

Analgesic and antipyretic activities of an aqueous extract and of the flavone linarin of Buddleia cordata.

The dried aqueous extract of leaves of Buddleia cordata (loganiaceae) and its main flavonoid glycoside, linarin, have been evaluated for analgesic and antipyretic effects in mice and rats, respectively. Both the extract and linarin exerted significant and dose-dependent analgesic and antipyretic activities, the first being obtained against a chemical stimulus (writhing a test in mice) and the second being obtained by a pyretogenic stimulus (yeast-induced hyperthermia test). Furthermore, the response of the animals in the hot plate test was modified by linarin and an aqueous extract. These acti

Analgesic, antipyretic and ulcerogenic activity of Nyctanthes arbor tristis leaf extract.

The leaves of Nyctanthes arbor tristis, besides being used in the treatment of sciatica and arthritis, are advocated for various kinds of fevers and painful conditions by the Ayurvedic physicians. In the present study, the water-soluble portion of an ethanol extract of the leaves was screened for analgesic, antipyretic and ulcerogenic activities. The extract exhibited significant aspirin-like antinociceptive activity but failed to produce morphine-like analgesia. It was also found to possess antipyretic activity against brewer's yeast-induced pyrexia in rats. The extract also produced gastric

Some biological properties of flurbiprofen, an anti-inflammatory, analgesic and antipyretic agent.

2-(2-Fluoro-4-biphenyl)propionic acid (flurbiprofen) possesses peripheral analgesic, anti-inflammatory and antipyretic properties. It does not possess glucocorticoid or adrenocortical-stimulating properties. It is a highly potent agent which in acute pharmacological test systems produced a significant pharmacological effect in single oral doses varying from 0.04 to 0.47 mg/kg. The peak plasma concentrations attained after these doses were generally of the order of 1 to 3 mug/ml. Doses of 0.33 mg/kg/day, which gave peak plasma concentrations of 0.6 mug/ml, produced a significant inhibition of r

Neuropharmacological and Antipyretic Potentials of Streblus asper Leaves: Integrated In Vivo and In Silico Approaches.

Streblus asper (Moraceae) is traditionally used for neurological and febrile disorders, but its pharmacological basis remains unclear. This study evaluated the S. asper leaf methanolic extract (SAL-ME) for anxiolytic, antidepressant, sedative, and antipyretic activities using Swiss albino mice and in silico docking analyses. Behavioral assays included the elevated plus maze, hole-board, forced swim, tail suspension, hole cross, and open field tests, while brewer's yeast-induced pyrexia was used to assess antipyretic activity. SAL-ME (200 and 400 mg/kg) produced dose-dependent effects, signific

Antipyretic, anti-inflammatory and analgesic activities of Periplaneta americana extract and underlying mechanisms.

Periplaneta americana is a common traditional Chinese medicinal material which has been used to treat arthritis, fever, aches, pains, and inflammation of the extremities for several hundred years. However, little scientific data exists in literature to support its use. The purpose of this study was to evaluate the antipyretic, anti-inflammatory and analgesic activities of Periplaneta americana extract (PAE) and explore its underlying mechanism. The antipyretic, anti-inflammatory and analgesic activities were evaluated by LPS-induced fever, carrageenan-induced paw edema, abdominal writhing, hot

Emerging CAM Ziziphus nummularia with in vivo sedative-hypnotic, antipyretic and analgesic attributes.

Ziziphus nummularia from Rhamnaceae family is traditionally used for sedative-hypnotic, antipyretic and analgesic purposes; however, scientific validations are lacking. This in vivo study was undertaken to verify the above ameliorative properties of Z. nummularia root methanolic extract. Various fractions of the extract were assayed on Balb/c mice by open field, Brewer's yeast-induced hyperthermia and acetic acid-induced writhing experiments. The significance of the outcomes was analyzed with statistical tests. Various fractions of the extract exhibited marked dose-dependent (p < 0.05) sedativ

Antipyretic activity of gingyo-san, a traditional medicine, in influenza virus-infected mice.

Gingyo-san is composed of 10 crude drugs and used as a traditional antipyretic medicine for the treatment of the common cold and influenza virus infection. In a murine intranasal influenza infection model, fever produced by the infection has been demonstrated to be reduced by suppressing interferon-induced interleukin (IL)-1 alpha production. Thus, we focused on the serum level of IL-1 alpha which produces such novel antipyretic activity, and evaluated the relationship between defervescence and the suppression of IL-1 alpha production by Gingyo-san in influenza virus-infected mice. Fever was p

Antipyretic effectiveness of acetaminophen in febrile seizures: ongoing prophylaxis versus sporadic usage.

A controlled clinical study compared the antipyretic effectiveness of acetaminophen administered at regular 4h intervals (group 1, n = 53) versus sporadic usage contingent upon a body temperature above 37.9 degrees C (group 2, n = 51) in 104 children presenting with simple febrile convulsions. The incidence of febrile episodes or temperature values were similar in spite of significantly larger amounts of acetaminophen administered to patients in group 1. Four and 4 children in groups 1 and 2, respectively, had a second episode of febrile seizures, in all of them within the first 24h of admissi

Comparative evaluation of antipyretic activity of ibuprofen and aspirin in children with pyrexia of varied aetiology.

The antipyretic activity of ibuprofen and aspirin was compared in sixteen children with pyrexia due to upper respiratory tract infection and in twelve with fever due to other causes. All 28 children received ibuprofen (7 mg/kg of body-weight) and aspirin (15 mg/kg of body-weight) in a single dose on 2 consecutive days in a crossover manner. Rectal temperature was recorded prior to and at regular intervals up to 8 hours after drug administration. Analysis of the results indicate that ibuprofen and aspirin effectively lower temperature and the two drugs are comparable in their antipyretic activi
