MEK/ERK-Mediated Transcriptional Repression of Metabolic Genes Confers Host Defense Against Gallid alphaherpesvirus 1 Infection via Modulating Fos Nuclear Translocation.

Infectious laryngotracheitis virus (ILTV), formally known as Gallid alphaherpesvirus 1, represents a prominent alphaherpesvirus that poses a significant threat to the global poultry industry. Current routine vaccination strategies fail to eliminate latent infection. Host-pathogen interaction networks have become a key focus in antiviral research. Our previous study demonstrated that activation of the MEK/ERK signaling pathway upon ILTV infection restricts host cellular metabolic activity to mount protective host antiviral responses, yet the underlying molecular mechanism remains unclear. Cellular transcription factor TFII-I represses adenovirus gene expression.

To successfully replicate, viruses must overcome innate cellular antiviral responses. With human adenovirus (HAdV), a key viral repressor of cellular antiviral responses is the early protein E4ORF3. E4ORF3 relocalizes numerous cellular antiviral proteins, particularly those involved in the interferon (IFN) and DNA damage response (DDR) pathways, to sequester them from viral replication sites in the nucleus. E4ORF3 also directs SUMO modification of a subset of its targets, some of which are subsequently targeted for proteasomal degradation. We previously identified TFII-I, a cellular transcript

Naturally Occurring Mutations in HIV-1 CRF01_AE Capsid Affect Viral Sensitivity to Restriction Factors.

TRIM5α and MxB are known as restriction factors that inhibit the early step of intracellular HIV-1 replication cycle. Both factors are believed to interact with the incoming virus core to suppress HIV-1 infection. The extreme diversity of HIV-1 is thought to be a consequence of its propensity to mutate to escape immune responses and host restriction factors. We recently determined the capsid sequences for 144 HIV-1 CRF01_AE viruses obtained in Thailand from 2005 to 2011. In this study, we further analyzed the amino acid variations among the capsid sequences of 204 HIV-1 CRF01_AE obtained in Th

Exosomes Transport Anti-Human Immunodeficiency Virus Factors from Human Cervical Epithelial Cells to Macrophages.

The female reproductive tract (FRT) is a major site of HIV sexual transmission. As the outermost layer of cells in the FRT, the human cervical epithelial cells (HCEs) have direct contact with HIV or infected cells. Our early work showed that supernatant (SN) from TLR3-activated HCEs contain the antiviral factors that could potently inhibit HIV replication in macrophages. However, it remains to be determined how HCEs transport the anti-HIV factors to macrophages. This follow-up study examined the role of exosomes in HCE-mediated anti-HIV activity. We found that TLR3 activation of HCEs resulted

PLSCR1 suppresses SARS-CoV-2 infection by downregulating cell surface ACE2.

UNLABELLED: Type I interferons exert their antiviral effects against SARS-CoV-2 by inducing the expression of interferon-stimulated genes (ISGs), including but not limited to LY6E, CH25H, IFITM2/3, and IFIH1. However, the antiviral effect and underlying mechanisms of action of most ISGs in SARS-CoV-2 infection are not yet fully understood. By screening 109 ISG-knockout cell lines, we identify that phospholipid scramblase 1 (PLSCR1), an interferon-inducible protein, acts as a crucial restriction factor against SARS-CoV-2 infection. Cells lacking PLSCR1 are highly susceptible to SARS-CoV-2 infec

Non-coding RNA-mediated tug-of-war between emerging viruses and host antiviral defense: lessons from animal viruses.

Emerging animal viruses pose persistent threats to livestock breeding and public health, underscoring the need for a deeper understanding of virus-host interactions. Nevertheless, the regulatory roles and conserved mechanisms of host non-coding RNAs (ncRNAs) during animal virus infections remain fragmented, and ncRNA-mediated mechanisms for emerging animal viruses are particularly poorly characterized, with no systematic collation or theoretical summarization available. Studies indicate that ncRNAs, including miRNAs, lncRNAs, and circRNAs, have emerged as important regulators of the interplay

Oxidized phospholipids associated with lipoprotein(a) contribute to hypofibrinolysis in severe aortic stenosis.

Tripartite motif-containing (TRIM) E3 ligases are a recently identified family of proteins with potent antiviral activity in mammalian cells. The prototype TRIM E3 ligase, TRIM5α was initially identified as a species-specific antiviral restriction factor but subsequent studies suggest some antiviral activity by several TRIM E3 ligases in human cells. However, the mechanisms of antiviral activity by these proteins and their transcriptional, translational and post-translational regulation are poorly understood. Furthermore, the contribution of TRIM E3 ligases to relative resistance or viral cont

[Progress in the Development of Animal Models Supporting Human Adenovirus Infection].

Human adenoviruses cause respiratory diseases, conjunctivitis, gastroenteritis and even severe pneumonia. Specific antiviral drugs and vaccines are still unavailable. Animal models that support adenovirus infection and pathogenesis are critical for the study of these viruses and the development of prophylactic and therapeutic strategies. However, the receptors of some human adenoviruses have not yet been identified, most human respiratory adenoviruses cannot infect rodents, and human adenoviruses cannot replicate in rodents due to host restrictions. These factors hamper the establishment of ani

Galectin-9 is a restrictor of infection with multiple enteroviruses.

Enteroviruses (EVs), encompassing over 200 sero/genotypes, cause diseases ranging from mild illness to severe systemic infections. Galectin-9 (Gal-9) is an immunomodulatory lectin implicated in various viral infections, yet its role in EVs remains unexplored. Here, we identify Gal-9 as a novel host restriction factor against a broad spectrum of EVs, including EV-A71, EV-D68, and Coxsackievirus B2. LGALS9 deficiency increased viral replication in cells and pathogenicity in a mouse model of EV-A71 infection, while its overexpression inhibited viral replication. Mechanistically, Gal-9 specificall

CD26 acts as a host restriction factor to inhibit Influenza A virus (H1N1) infection.

The frequent mutations and emerging drug resistance of Influenza A virus (IAV) pose a persistent global health challenge. The identification of host restriction factors that can inhibit viral infection is crucial for the development of novel antiviral therapies. CD26 acts as a host restriction factor in inhibiting H1N1 infection and the underlying mechanisms remain unclear. Our findings collectively demonstrate that CD26 inhibits H1N1 infection. In vitro, soluble CD26 (2.5 μg/ml) reduces H1N1 titers, specifically suppresses IL-6 upregulation in H1N1-infected A549 cells via NF-κB/IRF3/STAT1 sig

The naturally attenuated Kunjin strain of West Nile virus shows enhanced sensitivity to the host type I interferon response.

The host determinants that contribute to attenuation of the naturally occurring nonpathogenic strain of West Nile virus (WNV), the Kunjin strain (WNV(KUN)), remain unknown. Here, we show that compared to a highly pathogenic North American strain, WNV(KUN) exhibited an enhanced sensitivity to the antiviral effects of type I interferon. Our studies establish that the virulence of WNV(KUN) can be restored in cells and mice deficient in specific interferon regulatory factors (IRFs) or the common type I interferon receptor. Thus, WNV(KUN) is attenuated primarily through its enhanced restriction by

Interferon signaling in Peromyscus leucopus confers a potent and specific restriction to vector-borne flaviviruses.

Tick-borne flaviviruses (TBFVs), including Powassan virus and tick-borne encephalitis virus cause encephalitis or hemorrhagic fevers in humans with case-fatality rates ranging from 1-30%. Despite severe disease in humans, TBFV infection of natural rodent hosts has little noticeable effect. Currently, the basis for resistance to disease is not known. We hypothesize that the coevolution of flaviviruses with their respective hosts has shaped the evolution of potent antiviral factors that suppress virus replication and protect the host from lethal infection. In the current study, we compared virus

A CRISPR activation screen identifies SPART as a pan-orthoflavivirus restriction factor.

Orthoflaviviruses, including Zika (ZIKV), dengue, Japanese encephalitis, and West Nile viruses, cause diverse clinical syndromes and threaten human health. Identifying factors that inhibit orthoflavivirus infection could lead to antiviral countermeasures. Here, we conducted a genome-wide CRISPR activation screen and identified the host gene SPART (Spartin/SPG20) as a restriction factor against ZIKV and other orthoflaviviruses. SPART interacts with and disrupts the endosomal localization of Itchy E3-ubiquitin ligase (ITCH), which we determine ubiquitinates the ZIKV capsid, thereby triggering un

Activation of TLR3/interferon signaling pathway by bluetongue virus results in HIV inhibition in macrophages.

Bluetongue virus (BTV), a nonenveloped double-stranded RNA virus, is a potent inducer of type Ι interferons in multiple cell systems. In this study, we report that BTV16 treatment of primary human macrophages induced both type I and III IFN expression, resulting in the production of multiple antiviral factors, including myxovirus resistance protein A, 2',5'-oligoadenylate synthetase, and the IFN-stimulated gene 56. Additionally, BTV-treated macrophages expressed increased HIV restriction factors (apolipoprotein B mRNA-editing enzyme catalytic polypeptide 3 G/F/H) and CC chemokines (macrophage

Uncovering the evolving arms race between host immunity and HIV-1.

Despite an effective combination of antiretroviral therapy, HIV persists as a lifelong infection and global health threat. The human host equips restriction factors and interferon (IFN)-stimulated genes that target every step of the viral life cycle. However, HIV-1 has evolved a coordinated immune evasion strategy using a limited set of accessory proteins with distinct antagonistic functions. This functional division of labor allows HIV-1 to disable key immune pathways and ensure persistence. Here, we explore the molecular interplay between host defenses and HIV-1, organizing antiviral factors

Virus-Host Cell Interplay during Hepatitis E Virus Infection.

The molecular interplay between cellular host factors and viral proteins is a continuous process throughout the viral life cycle determining virus host range and pathogenesis. The hepatitis E virus (HEV) is a long-neglected RNA virus and the major causative agent of acute viral hepatitis in humans worldwide. However, the mechanisms of liver pathology and clinical disease remain poorly understood for HEV infection. This review summarizes our current understanding of HEV-host cell interactions and highlights experimental strategies and techniques to identify novel host components required for th

Promyelocytic leukemia protein is a restriction factor for Junín virus independently of Z matrix protein.

The New World (NW) mammarenavirus Junín (JUNV) is the etiological agent of Argentine hemorrhagic fever, a human endemic disease of Argentina. Promyelocytic leukemia protein (PML) has been reported as a restriction factor for several viruses although the mechanism/s behind PML-mediated antiviral effect may be diverse and are a matter of debate. Previous studies have reported a nuclear to cytoplasm translocation of PML during the murine Old World mammarenavirus lymphocytic choriomeningitis virus (LCMV) infection. This translocation was found to be mediated by the viral Z protein. Here, we show t

ATG13 promotes K63-linked polyubiquitination of MAVS via ASB1 to restrict influenza A virus replication.

Autophagy-associated protein 13 (ATG13) plays a pivotal role in regulating the assembly of the autophagy initiation complex. However, the function and mechanism of action of ATG13 during influenza virus infection remain incompletely understood. This study elucidates that ATG13 restricts influenza A virus (IAV) replication while potentiating the activation of the type I interferon (IFN-I) pathway. Mechanistically, coimmunoprecipitation assays revealed that ATG13 interacts with MAVS and recruits the E3 ubiquitin ligase ASB1 to catalyze K63-linked polyubiquitination at the K348 residue, thereby a

SAMHD1-dependent retroviral control and escape in mice.

SAMHD1 is a host restriction factor for human immunodeficiency virus 1 (HIV-1) in cultured human cells. SAMHD1 mutations cause autoimmune Aicardi-Goutières syndrome and are found in cancers including chronic lymphocytic leukaemia. SAMHD1 is a triphosphohydrolase that depletes the cellular pool of deoxynucleoside triphosphates, thereby preventing reverse transcription of retroviral genomes. However, in vivo evidence for SAMHD1's antiviral activity has been lacking. We generated Samhd1 null mice that do not develop autoimmune disease despite displaying a type I interferon signature in spleen, ma

A non-canonical role for the autophagy machinery in anti-retroviral signaling mediated by TRIM5α.

TRIM5α is a key cross-species barrier to retroviral infection, with certain TRIM5 alleles conferring increased risk of HIV-1 infection in humans. TRIM5α is best known as a species-specific restriction factor that directly inhibits the viral life cycle. Additionally, it is also a pattern-recognition receptor (PRR) that activates inflammatory signaling. How TRIM5α carries out its multi-faceted actions in antiviral defense remains incompletely understood. Here, we show that proteins required for autophagy, a cellular self-digestion pathway, play an important role in TRIM5α's function as a PRR. Ge
