Behavioral responses to methylphenidate and apomorphine in rats exposed neonatally to bisphenol-A.

Neonatal exposure of rats to bisphenol-A, an endocrine disruptor, has recently been proposed as a possible animal model of attention-deficit hyperactivity disorder, because such rats exhibit motor hyperactivity and impaired habituation to a novel environment 4-5 weeks after bisphenol-A pretreatment. To extend the original experiments, the present study additionally analyzed the effects of neonatal exposure to bisphenol-A (20 and 40 microg) on rat habituation to a novel environment and drug-induced behavior particularly at a later stage after bisphenol-A pretreatment. Single intracisternal admi

Other formulations and future considerations for apomorphine for subcutaneous injection therapy.

This manuscript reviews apomorphine administration in formulations other than intermittent bolus injection, and comments on other potential uses for this unique compound. Continuous sc apomorphine therapy has been shown to alter peak-dose dyskinesia thresholds in advancing patients, and in some instances may replace all other anti-parkinson therapies. In general continuous infusion of sc apomorphine at a rate of 4 mg/h is well tolerated, and has been postulated to be equivalent to approximately 600 mg levodopa/day. This therapy is associated with skin complications, particularly nodule formati

Effect of apomorphine on the conflict-induced jumping stereotypy in bank voles.

In conventional laboratory cages, bank voles (Clethrionomys glareolus) develop a jumping up-and-down stereotypy already before the age of one month. Central DA systems are thought to be involved in the expression of these conflict-induced stereotypies (CIS). Stereotypies can also be elicited pharmacologically, most commonly by amphetamine and apomorphine. Hence, administration of apomorphine to jumping bank voles provides the opportunity to compare pharmacologically-induced stereotypies (PHIS) and CIS in that species. A pilot study showed that apomorphine induced stereotyped licking that is qu

Pharmacological characterization of the enhancement of apomorphine-induced gnawing in mice by cocaine.

The present study was designed to provide additional information on the behavioral and pharmacological mechanisms associated with the augmentation of apomorphine-induced gnawing in C57BL/6J mice. (-)-Cocaine enhanced apomorphine-induced gnawing at doses devoid of effects on gnawing when given alone. The effect was stereoselective, with (+)-cocaine devoid of activity in this test. Peripheral synapses may also not be critical to the cocaine enhancement, as cocaine methiodide, a charged species, was also without effect. The local anesthetic actions of cocaine were evaluated with lidocaine, a loca

Effects of naloxone on the behaviors evoked by amphetamine and apomorphine in adult cats.

1. This work was undertaken in order to study whether the opioid system is involved in the modulation of the behaviors induced by two agonists of the dopaminergic system, amphetamine and apomorphine in adult cats. 2. Naloxone, an antagonist of the mu, delta and kappa opioid receptors was administered to twelve female mongrel cats; 0.5, 1.0 and 2.0 mg/kg s.c. were injected in order to analyse its own effect of naloxone. This drug produced NREMs behavior and accordingly the cat showed an overall decrease of its activities. 3. Amphetamine (2.5 mg/kg s.c.) and apomorphine (2.0 mg/kg s.c.) were inj

Naloxone does not prevent apomorphine-induced emesis or hypotension in dogs.

Previous data have shown that apomorphine-induced respiratory depression can be reversed by the opiate antagonist, naloxone. The present study investigates the influence of naloxone on cardiovascular changes and vomiting elicited by apomorphine in dogs. In chloralose-anaesthetized animals, naloxone (0.02 mg/kg i.v.) failed to modify either the decrease in blood pressure and the biphasic changes (bradycardia followed by a long-lasting tachycardia in heart rate or the characteristics (occurrence, latency, duration) of the emesis elicited by apomorphine (200 micrograms/kg i.v.). In contrast, in c

Effects of estradiol benzoate on apomorphine-induced pecking behavior in the pigeon.

Clinical and laboratory observations suggested an interaction between sex hormones and dopaminergic functions. Estrogens are reported to exert dual effects on dopaminergic system(s). In the apomorphine-induced pecking behavior model in the pigeon, acute administration of estradiol benzoate (5, 10, 20 mg/kg) significantly reduced the total pecking counts, indicating an antidopaminergic effect. Other sex steroids, testosterone and progesterone were found to be ineffective. Chronic administration of low dose estradiol benzoate (20 micrograms/kg/day) resulted in an increase in total pecking counts

Effects of naloxone on methamphetamine and apomorphine stereotypy and on haloperidol catalepsy in rats.

Pretreatment with the opiate antagonist naloxone, at 1.25-5 mg/kg, increased the intensity of methamphetamine stereotypy, had no effect (over a range of 0.3125-5 mg/kg) on apomorphine stereotypy, and antagonized haloperidol catalepsy in rats at 1.25-5 mg/kg. It is suggested that naloxone, by blocking the opiate receptors located on the nigro-striatal and mesolimbic dopamine (DA) nerve terminals, releases the DA systems from endogenous inhibition, presumably caused by endogenous opiate systems, and thereby potentiates methamphetamine stereotypy and antagonizes haloperidol catalepsy. However, th

Effect of some beta-carbolines on phenylethylamine and apomorphine stereotypies in rats.

Tetrahydro-beta-carbolines (THBC:s) have recently been shown to occur in the human body and their role in mental diseases has been discussed. The effect of THBC:s and some other beta-carbolines (25 mg/kg IP) were studied on the stereotypies caused by apomorphine (APO, 2 mg/kg IP) and phenylethylamine (PEA, 50 mg/kg IP) in rats. These effects of dopaminergic drugs like apomorphine as well as of phenylethylamine have sometimes been used as animal models of paranoid schizophrenia. Dose effect relationships were studied from ther most potent substances. All beta-carbolines studied significantly in

Influence of intraventricularly applied apomorphine on somato-vegetative reflexes in cats.

To obtain information about the dopaminergic neurotransmission processes taking part in the somatovegetative reflexes the effects of intraventricularly given apomorphine were studied in lightly anaesthetized cats. The level of blood pressure and the reflexogenic hypertension due to central sciatic nerve stimulation were not changed when apomorphine was applied into the lateral and third but was inhibited when given into the fourth ventricle. The nictitating membrane contractions elicited by sciatic nerve stimulation were inhibited by apomorphine when introduced into each part of the ventricula

Catalepsy induced by morphine or haloperidol: effects of apomorphine and anticholinergic drugs.

To investigate the extent of cholinergic involvement in opiate-induced catalepsy, the effects of three anticholinergic drugs were studied on morphine-induced catalepsy. Haloperidol-induced catalepsy was also examined. Maximum catalepsy in rats was obtained with 30 mg/kg morphine or 3 mg/kg haloperidol. The anticholinergic drugs atropine, benztropine, and scopolamine were unable to antagonize morphine-induced catalepsy, yet readily antagonized haloperidol-induced catalepsy. Low doses of apomorphine (7.5 mg/kg), on the other hand, readily antagonized morphine catalepsy, but 13-fold higher doses

Assessing Eligibility for Apomorphine Therapy in Parkinson's Disease: The Telemonitoring Potential.

BACKGROUND: Although there are established guidelines for the selection of suitable candidates, objective and timely referral for the use of apomorphine remains challenging. OBJECTIVES: This research examined how telemonitoring may facilitate referrals for continuous subcutaneous apomorphine infusion in Parkinson's disease (PD). METHODS: A Blind Rater and a multi-disciplinary team (MDT) including an expert PD nurse, neurologist, and geriatrician determined apomorphine pump treatment eligibility for 20 patients with PD. The MDT considered telemonitoring data and physical examination to determ

Hyperphosphorylated Tau Inflicts Intracellular Stress Responses that Are Mitigated by Apomorphine.

Abnormal phosphorylation of the microtubule-binding protein tau in the brain is a key pathological marker for Alzheimer's disease and additional neurodegenerative tauopathies. However, how hyperphosphorylated tau causes cellular dysfunction or death that underlies neurodegeneration remains an unsolved question critical for the understanding of disease mechanism and the design of efficacious drugs. Using a recombinant hyperphosphorylated tau protein (p-tau) synthesized by the PIMAX approach, we examined how cells responded to the cytotoxic tau and explored means to enhance cellular resistance t

Depletion of apomorphine induced behavioral sensitization in rats treated with escitalopram.

Apomorphine is a classical psychostimulant and used throughout the world as prescribed medicine. Despite of their therapeutic effects, use of psychostimulants is restricted because some psychosis and impulse control disorders are the consequence of their long term use. Studies suggest that serotonin (5-Hydroxytryptamine; 5-HT) has a critical role in psychosis and drug abuse. Center of the present article is to assess the impacts of serotonin in the easing of misuse potential; the sensitization prompted by recommended psychostimulant apomorphine. We researched that whether Escitalopram can weak
