A nocardioform bacterium was isolated from the spleen tissue of an armadillo infected with M. leprae and easily propagated in pure culture in mineral salt medium supplemented with only simple C and N sources (e.g., liquid paraffin, tetradecane, ammonium salts, urea, asparagine, gelatin, xanthin, hypoxanthin etc.). Complex organic substances, e.g., tyrosin, casein, peptone, meat extract, egg proteins, serum, blood, yeast extract as well as medium 199, did not support the growth of this organism. Microscopically, the organism consisted of acid-fast, long, slender rods which originated from long,

The ARM repeat only gene VlARO positively regulates grape resistance to gray mold by promoting the guaijaverin accumulation, which directly suppresses the mycelial growth of Botrytis cinerea. Gray mold, caused by the necrotrophic fungus Botrytis cinerea, severely threatens global grapevine production. Here, we identified VlARO, a nuclear-localized gene from the highly resistant grapevine variety 'Beta', as a positive regulator of gray mold resistance. Although VlARO contains Armadillo repeats, it lacks a canonical U-box domain, thus distinguishing it from typical plant U-box E3 ubiquitin ligas

Nearly half of the genes in the Plasmodium falciparum genome have not yet been functionally investigated. We used homology-based structural modeling to identify multiple copies of Armadillo repeats within one uncharacterized gene expressed during the intraerythrocytic stages, PF3D7_0410600, subsequently referred to as P. falciparum Armadillo-Type Repeat Protein (PfATRP). Soluble recombinant PfATRP was expressed in a bacterial expression system, purified to apparent homogeneity and the identity of the recombinant PfATRP was confirmed by mass spectrometry. Affinity-purified α-PfATRP rabbit antib

Eukaryotic Armadillo (ARM) repeat proteins are multifaceted with prominent roles in cell-cell adhesion, cytoskeletal regulation and intracellular signaling among many others. One such ARM repeat containing protein, ARM Repeats Only (ARO), has recently been demonstrated in both Toxoplasma (TgARO) and Plasmodium (PfARO) parasites to be targeted to the rhoptries during the late asexual stages. TgARO has been implicated to play an important role in rhoptry positioning i.e. directing the rhoptry towards the apical end of the parasite. Here, we report for the first time that PfARO exhibits a DNA bin

Endocytic activity of phagocytic cells from armadillos infected with viruses, parasites or bacteria is unknown. This report shows that eosinophils from armadillos infected with microfilaria act against these helmintic parasites but have deficiencies in their oxygen-dependent bacteriocidal mechanisms and also in endocytic capacity against yeast.

The microniche of the dimorphic fungus Paracoccidiodes brasiliensisremains undefined in spite of the many attempts to isolate it from natural sources. Until recently, knowledge was also scanty concerning the presence of natural infections in animals; however, in the last decade, the fungus has been repeatedly isolated from the nine-banded armadillo, Dasypus novemcinctusin Brazil. A study aimed at determining the presence of infected armadillos in one of the paracoccidioidomycosis endemic areas of Colombia (Manizales, Department of Caldas) was undertaken. Based on the records of paracoccidioido…

The Gran Chaco, Bolivia, has a total of seven species of armadillos with the three-banded (Tolypeutes matacus) and nine-banded (Dasypus novemcinctus) the most commonly hunted by the local Isoseño-Guarani people. Armadillos are known carriers of zoonotic pathogens, including Mycobacterium leprae, Toxoplasma gondii, and Trypanosoma cruzi; thus human handling and consumption of these species may have a significant public health impact. A health assessment that included physical examinations, hematology, plasma biochemical analyses, levels of exposure to selected infectious agents, and endoparasit…

Thirty, nine-banded armadillos weighing between 3 and 5 kilograms trapped from an area endemic for armadillo leprosy were collected at random; killed, autopsied and examined histopathologically. Also, one of the right inguinal lymph nodes was removed under sterile precautions and examined using PCR, direct smear examination, mouse footpad study, culture in laboratory media and histopathology with a view to detecting Mycobacterium leprae. Blood was collected at death and tested for IgM antibodies to PGL-1. According to the PCR study of the inguinal lymph nodes 16 of 30 armadillos (53.3%) had ev…

Sporotrichosis is a globally distributed subcutaneous mycosis caused by dimorphic Sporothrix species commonly found in soil, mosses, and decaying plant matter. The lymphocutaneous manifestation, historically associated with occupational activities and sapronotic transmission, has recently been observed to also occur through animal contact, particularly notable in Brazil. We describe a rare case of lymphocutaneous sporotrichosis with simultaneous pulmonary complications resulting from the scratching of a southern three-banded armadillo, Tolypeutes matacus, primarily inhabiting the arid forests

The number of viral-associated neoplasms reported in wildlife has increased over the last decades, likely because of growing research effort … tly, a molecular screening was carried out to detect the occurrence of this pathogen in other giant armadillos in the same region. Overall, this novel virus was detected in 14.3% (3/21) of the tested giant armadillos. We suggest this herpesvirus, the first in Xenarthra, as a plausible aetiology of the neoplasm. The implications of CiHV-1 for this species are …

Two antigens were tested and compared in relation to the 48-h Fernandez reaction. They were obtained from standard human and from standard armadillo lepromin. All the tests were negative in patients with lepromatous leprosy and highly positive in those with tuberculoid leprosy and in lepromin-positive contacts. There was total agreement in all tests done with the two types of antigen. The antigenic component has the following basic properties: it precipitates with 80% saturated ammonium sulfate; it is not destroyed by autoclaving or by treatment with 0.4% phenol; it is non-dialysable; and it i

The aim of this work is to compare the taxonomic diversity of parasite species of the large hairy armadillo Chaetophractus villosus in its native range and in another recently introduced population (Tierra del Fuego island), and to evaluate whether the isolation of the latter determines a decrease in its parasitic diversity. Forty specimens from Buenos Aires and Tierra del Fuego Provinces were collected and examined for helminths. Eleven parasite species were found in the native population, and only one species was present in Tierra del Fuego (Trichohelix tuberculata). This may be explained be

The natural habitat of Paracoccidioides brasiliensis remains undefined but the repeated demonstration of infection by this fungus in the nine-banded armadillo Dasypus novemcinctus has opened interesting research avenues. We report here the isolation of this fungus from the spleen of a naked-tailed armadillo Cabassous centralis (Miller 1899) captured in a coffee farm localized in the Colombian endemic area for paracoccidioidomycosis. This particular isolate was identified by its dimorphism and also by comparison of the PbGP43 gene and ribosomal internal transcribed spacer regions (ITS) with rec

This study is the first to detect Wnt/armadillo signaling pathway activity in the germarium of Fz3-RFP transgenic Drosophila melanogaster females orally exposure to sublethal concentrations of the keto-enol insecticides Movento® 240 SC (0, 11.2, 22.4, 37.3 mg/L) and Envidor® 240 SC (0, 12.3, 24.6, 41.1 mg/L) for 72 h. Both insecticides induced irregular overexpression of receptor Fz3 in regions 1 and 2a of the germarium, accompanied by morphological changes, tissue disorganization, and reduced cell number compared to controls. In wild-type females, oxidative stress was evidenced by increased M

Hypokalemia of unknown cause can often be challenging to diagnose. Although armadillo repeat-containing 5 (ARMC5) gene mutations are primarily associated with primary bilateral macronodular adrenal hyperplasia and Cushing's syndrome, their potential role in other endocrine disorders remains largely unexplored. A 50-year-old man presented with limb weakness and persistent hypokalemia. Comprehensive screening tests, including imaging and endocrinological evaluations, ruled out primary aldosteronism, Cushing's syndrome, and other common causes of hypokalemia. Genetic analysis revealed a heterozyg

The anatomical, histological, histochemical and ultrastructural characteristics of the Harderian gland of the armadillo Chaetophractus villosus were described. The gland is the largest structure in the bony orbit. It is situated in the anteroventral region of the orbit. Obvious structural differences are not observed between males and females. The gland is compound-branched tubulo-alveolar, being characterized by a single layer of columnar cells surrounded by myoepithelial cells. It possesses a single excretory duct opened into the inner canthus. All glandular cells show yellow-green autofluor

The anatomical, histological, histochemical and ultrastructural characteristics of the lacrimal gland (LG) and nictitans gland (NG) of the armadillo Chaetophractus villosus were described. The histochemical and histological features of both glands in male and female adult animals were compared. The tissues were processed with conventional techniques for light and transmission electron microscopy. Fixed specimens were submitted to a battery of tests for glycans, glycosaminglycans, glycoconjugates, proteins, and lipids. The LG of the armadillo may be considered within the set of glandulae lacrim

We studied three different isolates of Paracoccidioides brasiliensis obtained from the mesenteric lymph node (D3LY1), the spleen (D3S1) and the liver (D3LIV1) of the same armadillo (Dasypus novemcinctus). Pulmonal inflammatory area was evaluated by intravenous inoculation of 10(6) yeast cells of each isolates in young, male, ddY mice. Moreover, the partial sequence of GP43kDa gene of P. brasiliensis was analyzed. The lung inflammatory area was greater in animals inoculated with isolate D3S1. The partial sequence of GP43kDa gene indicated that isolate D3S1 is different from isolates D3LY1 and D

The detection of Mycobacterium leprae in non-human hosts, particularly armadillos, has raised questions about their role in the transmission of leprosy. In the United States, M. leprae strains present in Dasypus novemcinctus armadillos are involved in maintaining the disease. In Brazil, various armadillo species are recognized as naturally infected hosts for M. leprae, and the handling or consumption of armadillo meat are considered as potential risk factors for leprosy. However, genetic evidence confirming or refuting this in Brazil is lacking. To address this gap, Single Nucleotide Polymorph…
