Ebselen attenuates cadmium-induced testicular damage in mice.

This study was designed to examine if ebselen, an organoselenium compound with antioxidant and glutathione peroxidase-mimetic properties, attenuates testicular injury caused by intraperitoneal administration of CdCl(2). A number of toxicological parameters were evaluated in the testes of mice, such as delta-aminolevulinic acid dehydratase (delta-ALA-D) activity, lipid peroxidation, ascorbic acid levels and alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities. Ebselen attenuated lipid peroxidation levels altered by CdCl(2). delta-ALA-D activity inhibited by the highest

Physiological and Biochemical Responses of Nitraria tangutorum to Long-Term Saline Irrigation in an Arid Coal-Mining Restoration Area.

Saline irrigation is a major constraint on vegetation establishment in arid coal-mining landscapes. Nitraria tangutorum Bobrov is a xerohalophytic shrub established in the Dananhu mine-restoration area of Hami, Xinjiang, but its physiological responses to long-term saline irrigation remain insufficiently characterized. In this field study, approximately two-year-old plants were maintained for approximately two growing seasons under freshwater drip irrigation or irrigation water containing 8 or 12 g L-1 total dissolved solids. Unlike short-term controlled salinity assays that evaluate individua

Hydrogen Sulfide Mitigates Chilling Injury of Postharvest Banana Fruits by Regulating γ-Aminobutyric Acid Shunt Pathway and Ascorbate-Glutathione Cycle.

This study aimed to determine the effect of hydrogen sulfide on chilling injury (CI) of banana (Musa spp.) during cold storage (7°C). It was observed that hydrogen sulfide application (2 mmol L-1) markedly reduced the CI index and showed significantly higher chlorophyll contents, along with suppressed chlorophyll peroxidase and chlorophyllase enzyme activity. The treated banana fruits exhibited substantially higher peel lightness (L*), along with significantly a lower browning degree and soluble quinone content. The treated bananas had substantially a higher endogenous hydrogen sulfide content

Critical role of an endogenous gastric peroxidase in controlling oxidative damage in H. pylori-mediated and nonmediated gastric ulcer.

The objective of the present study is to delineate the mechanism of oxidative damage in human gastric ulcerated mucosa despite the presence of some antioxidant enzymes. We report for the first time the critical role of an endogenous peroxidase, a major H(2)O(2) metabolizing enzyme, in controlling oxidative damage in gastric mucosa. Human gastric mucosa contains a highly active peroxidase in addition to the myeloperoxidase contributed by neutrophil. In both non-Helicobacter pylori (H. pylori)- and H. pylori-mediated gastric ulcer, when myeloperoxidase level increases due to neutrophil accumulat

Ascorbic acid treatment, similarly to fluoxetine, reverses depressive-like behavior and brain oxidative damage induced by chronic unpredictable stress.

Reactive oxygen species (ROS) have been shown to play a role in the pathophysiology of depression. Taking into account that experimental chronic unpredictable stress (CUS) induces depressive-like behavior and that ascorbic acid has antidepressant-like effect in animals, the objective of this study was to investigate the influence of ascorbic acid on depressive-like behavior induced by CUS paradigm, serum corticosterone levels and markers of oxidative stress in cerebral cortex and hippocampus of mice. Animals were submitted to CUS procedure during 14 days. From the 8th to the 14th day mice rece

Ascorbate promotes carbon tetrachloride-induced hepatic injury in senescence marker protein 30-deficient mice by enhancing inflammation.

The genetic deletion of the senescence marker protein 30 (SMP30) gene results in ascorbate deficiency and the premature aging processes in mice. Apparent liver injury of SMP30(-/-) mice was less severe than those of wild type (WT) mice, upon chronic CCl(4) injection. The purpose of this study was to investigate the pathophysiology underlying the mild CCl(4) toxicity in SMP30(-/-) mice. Along with the lower level of serum alanine aminotransferase, the livers of SMP30(-/-) mice revealed a lesser glycogen depletion, a decrease in c-Jun N-terminal kinase (JNK)-mediated inflammatory signaling in pa

Photosystem II photochemistry, photoinhibition, and the xanthophyll cycle in heat-stressed rice leaves.

To investigate how high light affects the responses of photosynthesis to heat stress, the effects of high temperature (25-42.5 degrees C) either in the dark or in the light (1000 micromol m(-2) s(-1)) on photosystem II (PSII) photochemistry and the xanthophyll cycle were investigated in rice plants. At temperatures higher than 35 degrees C, there was a decrease in the CO(2) assimilation rate, and this decrease was greater in the light than in the dark. The maximal efficiency of PSII photochemistry (F(v)/F(m)) showed no significant change in the dark, but did show a significant decrease in the

Hexavalent chromium damages chamomile plants by alteration of antioxidants and its uptake is prevented by calcium.

Toxicity of low (3μM) and high (60 and 120μM) concentrations of hexavalent chromium/Cr(VI) in chamomile plants was studied. Fluorescence staining confirmed reduction of Cr(VI) to Cr(III). Cr was mainly accumulated in the roots with translocation factor <0.007. Notwithstanding this, both shoots and roots revealed increase in oxidative stress and depletion of glutathione, total thiols, ascorbic acid and activities of glutathione reductase and partially ascorbate peroxidase mainly at 120μM Cr. Though some protective mechanisms were detected (elevation of nitric oxide, enhancement of GPX activity

Purification and characterization of a homodimeric catalase-peroxidase from the cyanobacterium Anacystis nidulans.

Cytosolic extracts of the cyanobacterium Anacystis nidulans exhibit both catalase and o-dianisidine peroxidase activity. Native polyacrylamide gel electrophoresis demonstrates one distinct enzyme, which has been purified to essential homogeneity and found to be composed of two identical subunits of equal size (80.5 kDa). The isoelectric point is at pH 4.7. It is a very efficient catalase with a broad pH optimum between 6.5 and 7.5 and a Km for H2O2 of 4.3 mM, a calculated turnover number of 7200 s(-1), and an overall-rate constant of 3.5 x 10(6) M(-1) s(-1). The behaviour of this protoheme-enz

Comparative study on the inhibitory effects of antioxidant vitamins and radon on carbon tetrachloride-induced hepatopathy.

We have previously reported that radon inhalation activates anti-oxidative functions and inhibits carbon tetrachloride (CCl(4))-induced hepatopathy. It has also been reported that antioxidant vitamins can inhibit CCl(4)-induced hepatopathy. In the current study, we examined the comparative efficacy of treatment with radon, ascorbic acid and α-tocopherol on CCl(4)-induced hepatopathy. Mice were subjected to intraperitoneal injection of CCl(4) after inhaling approximately 1000 or 2000 Bq/m(3) radon for 24 h, or immediately after intraperitoneal injection of ascorbic acid (100, 300, or 500 mg/kg

Strigolactone decreases cadmium concentrations by regulating cadmium localization and glyoxalase defense system: Effects on nodules organic acids and soybean yield.

Источник пометил публикацию как отозванную

To decrease environmental and human health risks associated with crop and soil contamination, alternative solutions are still needed. The information on strigolactones (SLs)-mediated elicitation of abiotic stress signaling and triggering physiological alterations is scarce in the plant. To unravel the same, soybean plants were subjected to cadmium (Cd) stress (20 mg kg-1), presence or absence of foliar applied SL (GR24) at the concentration of 10 μM. Excess Cd accumulation causes reduced growth (-52% shoot and +24% root), yield (-35%), physio-biochemical markers, organic acid production, and g

Hydrogen Sulfide and Silicon Together Alleviate Chromium (VI) Toxicity by Modulating Morpho-Physiological and Key Antioxidant Defense Systems in Chickpea (Cicer arietinum L.) Varieties.

Extensive use of chromium (Cr) in anthropogenic activities leads to Cr toxicity in plants causing serious threat to the environment. Cr toxicity impairs plant growth, development, and metabolism. In the present study, we explored the effect of NaHS [a hydrogen sulfide; (H2S), donor] and silicon (Si), alone or in combination, on two chickpea (Cicer arietinum) varieties (Pusa 2085 and Pusa Green 112), in pot conditions under Cr stress. Cr stress increased accumulation of Cr reduction of the plasma membrane (PM) H+-ATPase activity and decreased in photosynthetic pigments, essential minerals, rela

Surface spraying of anthocyanin through antioxidant defense and subcellular sequestration to decrease Cd accumulation in rice (Oryza sativa L.) grains in a lead-zinc mine area.

As an important class of flavonoids, anthocyanin has been used to enhance plant-defensive mechanisms against heavy metal stress. However, there are few available reports regarding surface spraying of anthocyanin for reduction of Cd poisoning in rice and its practical applications in paddy fields. After rice growing, measurements were taken of rice growth, pigments, the antioxidant system, thiol compounds, and distribution of Cd in rice tissues. The results showed that surface spraying anthocyanin could promote rice growth, and relative to the control, total chlorophyll significantly increased

Differential alterations of antioxidant defenses as bioindicators of mercury and cadmium toxicity in alfalfa.

Several physiological parameters related to oxidative stress, which is a characteristic of plants exposed to toxic metals, were studied in 3-week-old alfalfa plants treated with cadmium (Cd) or mercury (Hg) at doses of 0, 3, 10 and 30 microM for 7d. The concentrations of biothiols, glutathione (GSH), homoglutathione (hGSH) and phytochelatins (PCs) increased dramatically in metals-treated plants, in particular in the presence of Cd. This was accompanied by a remarkable up-regulation of gamma-glutamyl cysteine synthetase gene, probably in response to the higher demand for GSH|hGSH needed for PC

Smartphone-assisted fast detection of Total antioxidant capacity in fruit and vegetable food based on the excellent peroxidase-like property of Fe-MOF.

The total antioxidant capacity (TAC) has received increasing attention for the evaluation of food quality in recent years. In this work, we reported on a novel Fe-MOF material, which exhibited excellent peroxidase-like properties. Based on this property, smartphone-assisted fast detection of TAC in fruit and vegetable food was developed. The TAC determination strategy, which was described by vitamin C (AA), glutathione (GSH) and L-cysteine (L-Cys), manifested a very fast detection efficiency in just 10 s. Notably, the Fe-MOF paper-based sensor exhibited the LOD (lowest detection limit) for AA,

Enzymes of oxygen metabolism and lipid peroxidation on erythrocytes from copper-deficient rats.

1. Copper deficiency in the rat results in a high decrease of erythrocyte superoxide dismutase activity (by 70%), an increase of glutathione peroxidase activity (by 17%) and glucose 6-phosphate dehydrogenase activity (by 40%) and no change in catalase activity. 2. Ascorbate (30 nM) and copper (10 and 50 nmol/mg protein) enhance about 2-fold the lipid peroxidation of erythrocyte membranes from copper-deficient rats. 3. The osmotic stability of copper-deficient rat erythrocytes is higher compared with that of the controls.

Effects of exogenous taurine on growth, photosynthesis, oxidative stress, antioxidant enzymes and nutrient accumulation by Trifolium alexandrinum plants under manganese stress.

Plants essentially require manganese (Mn) for their normal metabolic functioning. However, excess Mn in the cellular environment is detrimental to plant growth, development, and physio-biochemical functions. Taurine (TAU) is an amino acid with potent antioxidant and anti-inflammatory properties in animals and humans. However, no previous study has investigated the potential of TAU in plant metal stress tolerance. The current study provides some novel insights into the effect of TAU in modulating the defense system of Trifolium alexandrinum plants under Mn toxicity. Manganese toxicity resulted

Role of Fucoxanthin towards Cadmium-induced renal impairment with the antioxidant and anti-lipid peroxide activities.

Kidney damages caused by cadmium are considered to be one of the most dangerous consequences for the human body. This study aimed to investigate the protective effects of fucoxanthin supplementation on mice models subjected to cadmium-induced kidney damage. The mice treated with cadmium chloride (CdCl2) were observed to have significantly reduced the cross-section area of glomeruli. Cadmium exposure has also caused the damage of the structural integrity of mitochondria and increased blood urea nitrogen (BUN), kidney injury molecule 1 (KIM1), and neutrophil gelatinase associated lipocalin (NGAL
