Hierarchical Reactivity of Enzyme-Mediated Phosphorus Recycling from Organic Mixtures by Aspergillus niger Phytase.

Biological recycling of inorganic phosphorus (Pi) from organic phosphorus (Po) compounds by phosphatase-type enzymes, including phytases, is an important contributor to the pool of bioavailable P to plants and microorganisms. However, studies of mixed-substrate reactions with these enzymes are lacking. Here, we explore the reactivity of a phytase extract from the fungus Aspergillus niger toward a heterogeneous mixture containing, in addition to phytate, different structures of environmentally relevant Po compounds such as ribonucleotides and sugar phosphates. Using a high-resolution liquid chr

Vacuolar H(+)-ATPase plays a key role in cell wall biosynthesis of Aspergillus niger.

The identification of suitable targets is crucial for the discovery and development of new antifungals. Since the fungal cell wall is an essential organelle, the identification of genes involved in cell wall biosynthesis is expected to help discover new antifungal targets. From our previously obtained collection of cell wall mutants with a constitutively active cell wall stress response pathway, we selected a thermosensitive, osmotic-remediable mutant with decreased resistance to SDS for complementation analysis. The phenotypes of this mutant were complemented by a gene encoding a protein with

Crystallization and preliminary X-ray analysis of arabinofuranosidase C from Aspergillus niger strain 3M43.

Crystals of arabinofuranosidase C purified from Aspergillus niger strain 3M43 have been obtained by vapour diffusion. The crystal belongs to the space group P2(1) with cell parameters a = 44.28, b = 71.99, c = 45.27 A and beta = 105.98 degrees with one molecule in the asymmetric unit. The X-ray diffraction pattern of these crystals extends to at least 2.20 A resolution with the use of synchrotron radiation. These crystals are stable on exposure to radiation and are suitable for structure determination.

Optimization of glucose oxidase production by Aspergillus niger using genetic- and process-engineering techniques.

Wild-type Aspergillus niger NRRL-3 was transformed with multiple copies of the glucose oxidase structural gene (god). The gene was placed under the control of the gpdA promoter of A. nidulans. For more efficient secretion the alpha-amylase signal peptide from A. oryzae was inserted in front of god. Compared to the wild type, the recombinant strain NRRL-3 (GOD3-18) produced up to four times more extracellular glucose oxidase under identical culture conditions. Addition of yeast extract (2 gl-1) to a mineral salts medium containing only glucose as carbon source increased volumetric and specific

Manganese deficiency leads to elevated amino acid pools in citric acid accumulating Aspergillus niger.

Free amino acid pools have been investigated in a citric acid accumulating strain of Aspergillus niger during batch growth under manganese sufficient and deficient conditions by means of an improved chromatographic method. Studies on the mycelial content of several nitrogenous compounds under manganese sufficient and deficient conditions showed that manganese deficiency resulted in lower amino acid pool sizes during trophophase and considerable accumulation during idiophase, and in a reduction of the protein and nucleic acid contents. Addition of cycloheximide to mycelia grown with sufficient

Enhancement in the nutritional profile and hepatoprotective activity of pine pollen via Aspergillus niger fermentation.

Fermentation with Aspergillus niger significantly enhances the hepatoprotective effects of pine pollen. This study evaluated the effects of Aspergillus niger-fermented pine pollen (FPP) against alcoholic fatty liver disease (AFLD) using ethanol-induced HepG2 cells and an AFLD mouse model. In AFLD mice, FPP markedly reduced hepatic triglyceride levels from 1.47 ± 0.13 to 0.90 ± 0.11 mg/g and serum alanine aminotransferase (ALT) from 2.27 ± 0.63 to 1.02 ± 0.15 U/mL, indicating significant alleviation of lipid accumulation and liver injury. Mechanistically, FPP suppressed lipogenesis by downregul

Intracellular Aspergillus niger conidia in alveolar macrophages: A diagnostic clue in a lung transplant recipient.

Bronchoalveolar lavage (BAL) is a basic diagnostic method for the detection of fungal infections in lung transplant recipients. Aspergillus species are frequently identified, typically by the presence of septate hyphae; however, the visualization of conidia in cytologic preparations is rare. Aspergillosis caused by Aspergillus niger is an uncommon but recognized infectious complication in this patient population. We report on the case of a 60-year-old lung transplant recipient who underwent routine surveillance bronchoscopy eight weeks post-transplantation in August 2025. A substantial amount o

Mutations in the Second Alternative Oxidase Gene: A New Approach to Group Aspergillus niger Strains.

Alternative oxidase is a terminal oxidase in the branched mitochondrial electron transport chain of most fungi including Aspergillus niger (subgenus Circumdati, section Nigri). A second, paralogous aox gene (aoxB) is extant in some A. niger isolates but also present in two divergent species of the subgenus Nidulantes-A. calidoustus and A. implicatus-as well as in Penicillium swiecickii. Black aspergilli are cosmopolitan opportunistic fungi that can cause diverse mycoses and acute aspergillosis in immunocompromised individuals. Amongst the approximately 75 genome-sequenced A. niger strains, aox

Colorimetric Detection of Metals Using CdS-NPs Synthesized by an Organic Extract of Aspergillus niger.

The use of cadmium sulfide nanoparticles (CdS-NPs) synthesized by fungi presents highly stable chemical and optical characteristics; this makes them a promising alternative for development of colorimetric methods for metal detection. Moreover, application of CdS-NPs is challenging due to the biological material used to carry out synthesis and coating is highly diverse; therefore, it is necessary to evaluate if such components are present in the biological material. Thus, the objective of this work was to detect metallic ions in synthetic water samples using CdS-NPs synthesized by the extract o

Invasive Aspergillus niger Is the Sole Etiological Agent for CSOM : A Clinical Case from Nepal.

Aspergillus causing chronic suppurative otitis media (CSOM) is rare in immunocompetent people; however, it can occur as a significant opportunistic pathogen in immunocompromised patients. Here, in our study, a 53-year-old diabetic patient having a history of CSOM visited the Department of Otorhinolaryngology-Head and Neck Surgery (ENT-HNS), Tribhuvan University and Teaching Hospital (TUTH), Nepal, in March 2016. Although he was on medication with an antibacterial ear drop from the last 10 days, his right ear was presented with otorrhea, pruritus, otalgia, aural fullness, hearing impairment, an

Microbiological and immunological characteristics of a lethal pulmonary Aspergillus niger infection in a non-neutropenic patient.

Invasive pulmonary aspergillosis is increasingly described in non-neutropenic patients, such as patients with COPD receiving corticosteroids and the critically ill. Here, we present a case of a lethal pulmonary Aspergillus niger infection in a COPD patient. Immunological tests showed an impaired innate and adaptive immune response to Aspergillus. A history of COPD, unresponsiveness to antibiotics and especially a suggestive CT-scan should trigger the clinician to consider diseases caused by Aspergillus.

Aspergillus niger PA2: a novel strain for extracellular biotransformation of L-tyrosine into L-DOPA.

L-DOPA (3,4-dihydroxyphenyl-L-alanine), an amino acid derivative is the most widely used drug of choice for the treatment of Parkinson's disease and other neurologic injuries. The present study deals with the elevated biochemical transformation of L-tyrosine to L-DOPA by Aspergillus niger PA2, a potent tyrosinase producer, isolated from decomposed food wastes. This appears to be the first report on A. niger as a notable extracellular tyrosinase producer. The extracellular tyrosinase activity produced remarkably higher levels of L-DOPA, i.e. 2.44 mg mL(-1) when the media was supplemented with 5

The antifungal efficacy of nano-metals supported TiO₂ and ozone on the resistant Aspergillus niger spore.

Recently, antimicrobial efficacy of nano-metals has been extensively investigated. However, most of the related studies focused on the bactericidal effectiveness. Molds, especially their spores, are more resistant than bacteria, and can build a high concentration in houses due to dampness. Therefore, a comprehensive evaluation of the antifungal effectiveness of nano-metals is necessary. In this study, the nano-metals (Ag, Cu and Ni) supported catalysts were successfully prepared by the incipient wetness impregnation method, while the titanium dioxide (Degussa (Evonik) P25) nanoparticle was ser

Optimization of a natural medium for cellulase by a marine Aspergillus niger using response surface methodology.

The components of a natural medium were optimized to produce cellulase from a marine Aspergillus niger under solid state fermentation conditions by response surface methodology. Eichhornia crassipes and natural seawater were used as a major substrate and a source of mineral salts, respectively. Mineral salts of natural seawater could increase cellulase production. Raw corn cob and raw rice straw showed a significant positive effect on cellulase production. The optimum natural medium consisted of 76.9 % E. crassipes (w/w), 8.9 % raw corn cob (w/w), 3.5 % raw rice straw (w/w), 10.7 % raw wheat b

[Effect of local anesthetics on the transport of mannose in the microsomal membranes of Aspergillus niger van Tieghem].

The influence of various local anaesthetics has been studied on the mannose transfer enzymic system which is localized in Aspergillus niger van Tieghem microsomal membranes. The n-alkanes and tertiary amines do not seem to react on the first step of the reaction: i.e. the polyprenylphosphate-mannose biosynthesis. However two amines, the dibucaine and to a lesser extent the tetracaine, inhibit the mannose transfer from the polyprenylphosphate-mannose to the endogenous proteins.

Bioreactor as the root cause of the "manganese effect" during Aspergillus niger citric acid fermentations.

High-yield citric acid production by the filamentous Ascomycete fungus Aspergillus niger requires a combination of extreme nutritional conditions, of which maintaining a low manganese (II) ion concentration (<5 μg L-1) is a key feature. Technical-scale production of citric acid predominantly uses stainless-steel tank fermenters, but glass bioreactors used for strain improvement and manufacturing process development also contain stainless steel components, in which manganese is an essential alloying element. We show here that during citric acid fermentations manganese (II) ions were leaching fr

Primary Cutaneous Aspergillosis in Immunocompetent Patients Due to Aspergillus niger: A Report of 4 Cases.

Primary cutaneous aspergillosis is a cutaneous fungal infection due to the direct inoculation of spores of Aspergillus species into the disrupted skin. Primary cutaneous aspergillosis presents with a variety of localized cutaneous lesions, such as erythematous macules, papules, plaques, or nodules that can progress to necrosis, erosion, ulceration, or fistulization. Many species of Aspergillus can cause the disease, and one of them is Aspergillus niger that rarely affects immunocompetent patients and that has peculiar characteristics on the histopathological examination. We present a series of

Efficient removal of methylene blue by sodium alginate microspheres immobilizing Aspergillus niger and Fe3O4/biochar.

The extensive discharge of synthetic dyes from textile and pharmaceutical industries has created an urgent demand for eco-friendly and cost-effective purification technologies. In this study, a magnetic bio-based hybrid microgel adsorbent (SAFBCA) was fabricated by incorporating Fe3O4/biochar (FBC) and Aspergillus niger (A. niger) into a sodium alginate (SA) hydrogel matrix. Physicochemical characterization confirmed the formation of porous microspheres with a moderate specific surface area (8.22 m2 g⁻1) and sufficient magnetic responsiveness (9.31 emu g⁻1) for facile recovery. The adsorbent a
