Methylmercury (MeHg) is a potent neurotoxicant. Any source of environmental mercury represents a potential risk for human MeHg poisoning, because the methylation of inorganic mercury to MeHg in waterways results ultimately in its accumulation in the sea food chain, which represents the most prevalent source for human consumption. A small amount of MeHg accumulates in the central nervous system (CNS), particularly in astrocytes. Astrocytic swelling, excitatory amino acid (EAA) release and uptake inhibition, as well as EAA transporter expression inhibition are known sequelae of MeHg exposure. He

The high expression of CD98 was reported in some normal tissues, including blood brain barrier, activated lymphocytes, the basal layer of skin, proximal tubles of kidney, placenta, testis and a wide variety of tumors. The CD98 complex consists of an 80-85kD heavy chain (4F2hc/FRP-1) and a 40-45kD light chain. CD98hc, 4F2hc, and FRP-1 are the same glycosylated protein each other and define antigenicity of CD98. LAT1, the sodium-independent L-type amino acid transporter 1, has been identified as a light chain of the CD98 heterodimer from C6 glioma cells. LAT1 also corresponds to TA1, an oncofeta

Evidence obtained from studies on the mechanisms of action of dopaminergic neurotoxins suggests that glial cells may play an important role in neurodegenerative processes. A possible link between the function of glial cells and nerve cell damage could relate to the ability of astrocytes to convert innocuous compounds into toxic metabolites. Indeed, a mechanism of metabolic activation has been demonstrated in the MPTP (1-methyl-4-phenyl-1,2,3, 6-tetrahydropyridine) model of dopaminergic toxicity. The fully oxidized pyridinium metabolite (MPP+) is the ultimate mediator of MPTP neurotoxicity and

Between 1957 and 1980, 54 children less than 20 years of age with a diagnosis of glioblastoma multiforme or malignant astrocytoma were treated. All patients had a minimum follow-up period of 5 years. Twenty-seven patients had glioblastoma multiforme and 27 had malignant astrocytoma. The median age was 8 years for glioblastoma multiforme patients and 10 years for malignant astrocytoma patients. All patients received radiation therapy, but two died of postoperative complications and did not complete the treatment. The Kernohan Grading System was more useful in distinguishing glioblastoma multifo

Brain metastasis is increasingly understood as a disease of reciprocal adaptation between metastatic cells and the brain microenvironment. Among host cells, astrocytes have emerged as central regulators of metastatic colonization, survival, immune remodeling, and treatment resistance. Recent studies are re-defining astrocytes to explore the functional implications of their cellular heterogeneity. Evidence in brain metastasis indicates that astrocyte responses are heterogeneous across anatomical space, stage of colonization, and tumor type. This framework helps explaining how distinct astrocyte

Accumulating evidence suggests dysfunction of cerebellar-cerebral circuits in depression. However, the potential cellular and molecular alterations associated with depression in the cerebellum remain largely uncharacterized. While postmortem findings in the cerebral cortex indicate astrocyte dysregulation in depressed individuals who died by suicide (DS), the extent to which depression potentially alters cerebellar astrocytes is not well understood. In this study, two canonical astrocyte markers, glial fibrillary acidic protein (GFAP) and aldehyde Dehydrogenase-1 Family member L1 (ALDH1L1) wer

Refractory epilepsy is the main neurological manifestation of Alpers' syndrome, a severe childhood-onset mitochondrial disease caused by bi-allelic pathogenic variants in the mitochondrial DNA (mtDNA) polymerase gamma gene (POLG). The pathophysiological mechanisms underpinning neuronal hyperexcitabilty leading to seizures in Alpers' syndrome remain unknown. However, pathological changes to reactive astrocytes are hypothesised to exacerbate neural dysfunction and seizure-associated cortical activity in POLG-related disease. Therefore, we sought to phenotypically characterise astrocytic patholog

Glioblastoma multiforme, the most common type of primary brain tumour, remains an unsolved clinical problem. A great deal of work has been done in an effort to understand the biology and genetics of glioblastoma multiforme, but clinically effective treatments remain elusive. It is well known that malignant gliomas develop resistance to chemo- and radiotherapy. In this review we evaluated the literature data regarding therapeutic progress for the treatment of astrocytic tumours, focusing our attention on new frontiers for glioblastoma. The research studies performed in in vitro and in vivo mode

In a recent study in Science, Chen et al. engineer astrocytes with anti-amyloid chimeric antigen receptors (CARs) to enable sustained, antigen-directed clearance of amyloid-β (Aβ) in vivo, revealing how CAR design can shape both pathology and glial responses.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by loss of nigral dopaminergic neurons and misfolded α‑synuclein (α‑Syn) aggregation. However, increasing evidence indicates that astrocytes occupy a central position in the multifactorial pathogenesis of PD. As the most abundant glial cells in the Central Nervous System (CNS), astrocytes maintain neural homeostasis via neurotransmitter clearance, ion balance, metabolic support, synaptic regulation, and blood-brain barrier (BBB) integrity. In early PD, astrocytes exert neuroprotective effects; with disease progr

Neuronal axons have traditionally been considered to be the primary mediators of functional connectivity among brain regions. However, the role of astrocyte-mediated communication has been largely underappreciated. Astrocytes communicate with one another through gap junctions, but the extent and specificity of this communication remain poorly understood. Astrocyte gap junctions are necessary for memory formation1,2, synaptic plasticity3-5, coordination of neuronal signalling6, and closing the visual and motor critical periods7,8. These findings indicate that this form of communication is essen

Astrocytes are the most abundant glial cells in the brain and an integrative component of the neural network. Studies have shown that ethanol altered expression of an astrocyte marker, i.e., glial fibrillary acidic protein (GFAP), in two key corticolimbic regions, the medial prefrontal cortex (mPFC) and nucleus accumbens (NAc). These regions comprise anatomically and functionally different subregions, i.e., the prelimbic (PL) and infralimbic (IL) cortex of the mPFC, the shell and core subregions of the NAc. However, ethanol effects on GFAP expression within these subregions remain largely unkn

BACKGROUND: Astrocytes participate in both neuropathological and protective processes following traumatic brain injury (TBI) and undergo various characteristic changes, including phenotypic transformation, transcriptional reprogramming, and functional diversification.
METHODS: A comprehensive literature review was conducted in PubMed using key terms "astrocytes" and "traumatic brain injury", and we integrated this existing evidence from transcriptomic analyses to mechanistic studies to elucidate the role and underlying mechanisms of astrocytes in TBI pathophysiology.
RESULTS: Transcriptomic

Parkinson's disease (PD) is characterized by motor symptoms that are mainly attributed to the progressive loss of dopaminergic neurons of the substantia nigra (SN). It is also characterized by abnormal inclusion vesicles, termed Lewy bodies (LBs), enriched with α-synuclein aggregates that may induce inflammation and neurotoxicity. The possibility that factors involved in other neurodegenerative diseases also affect PD-related pathologies, such as α-synuclein uptake, was examined. The apoe4 allele is a major genetic risk factor for Alzheimer's disease (AD) and has also been suggested to be invo

BACKGROUND: Neuropathic pain (NP) is a debilitating chronic pain condition caused by injury or disease of the somatosensory nervous system. Accumulating evidence indicates that astrocytes play a central role in neuroinflammatory regulation and synaptic remodeling, thereby critically influencing the initiation and persistence of NP. However, a comprehensive overview of research trends and knowledge structures in this field is still lacking.
METHODS: The analysis was conducted based on publications retrieved from the Web of Science Core Collection (WoSCC) and Scopus, covering the period from 20

Fructose has recently gained substantial interest in the context of diseases that are connected with sucrose-rich diets. Several studies have shown that astrocytes are able to metabolize fructose to some extent but less efficiently than glucose. To study whether and how fructose can fuel astrocytic energy metabolism, we used cultured primary astrocytes as model. These cells consumed fructose in a time- and concentration-dependent manner that followed apparent Michaelis-Menten type kinetics with a KM value of 2.8 ± 0.9 mM and a VMax value of 325 ± 32 nmol/(mg × h). Although the specific consump

Astrocytes are a diverse and morphologically complex class of glial cells restricted to the central nervous system which have been implicated in the modulation of neuronal activity. The cerebellum is involved in planning movements and motor learning. Within the cerebellum three deep cerebellar nuclei (dentate, interposed and fastigial) provide the sole neuronal output. The fastigial nucleus participates in saccadic and vestibular function, and recent evidence disclosed neuronal projections to cognitive, affective, and motor areas. However, thus far there are no reliable descriptions of the dis

Prolonged high-fat diet (HFD) exposure is associated with hyperphagia, excess caloric intake and weight gain. After initial exposure to a HFD, a brief (24-48 h) period of hyperphagia is followed by the regulation of caloric intake and restoration of energy balance within an acute (3-5 day) period. Previous studies have demonstrated this occurs via a vagally mediated signalling cascade that increases glutamatergic transmission via activation of NMDA receptors located on gastric-projecting neurons of the dorsal motor nucleus of the vagus (DMV). The present study used electrophysiological recordi

Astrocytes play an important role in the central nervous system (CNS). Ion channels in these cells not only function in ion transport, and maintain water/ion metabolism homeostasis, but also participate in physiological processes of neurons and glial cells by regulating signaling pathways. Increasing evidence indicates the ion channel proteins of astrocytes, such as aquaporins (AQPs), transient receptor potential (TRP) channels, adenosine triphosphate (ATP)-sensitive potassium (K-ATP) channels, and P2X7 receptors (P2X7R), are strongly associated with oxidative stress, neuroinflammation and cha
