Location and morphology of parasympathetic preganglionic neurons in the sacral spinal cord of the cat revealed by retrograde axonal transport of horseradish peroxidase.

The distribution and morphology of preganglionic neurons in the sacral parasympathetic nucleus (SPN) of the cat have been studied with the horseradish peroxidase (HRP) tracing technique. HRP applied to the cut pelvic nerve was identified in cells located ipsilaterally, primarily in the intermediate gray matter. They formed a column approximately 10 mm long, usually contained within two, but occasionally three, sacral segments, S2 contained a majority of cells. In transverse sections the SPN had the appearance of an inverted "L". Cells were medium-sized, oval or spindle-shaped, and transversely

[Change in the activity of sympathetic preganglionic neurons of the lumbar segments of the spinal cord as a result of thermal stimulation of the skin of the abdomen].

The activity of antidromically identified sympathetic preganglionic neurons (SPNs) located in the lateral horn of the L2--L3 spinal cord segments was recorded in anesthetized and immobilized cats. The localization of SPNs with tonic activity and the conduction velocities of their axons were revealed. Two groups of SPNs were shown which increased their firing rates either in cooling or heating of the abdominal skin. Some intermediate neurons possibly conducting thermal information to SPNs, were studied. The data obtained are considered within the limits of segmentary model of the vasomotor ther

The long-term regulation of ganglionic tyrosine hydroxylase by preganglionic nerve activity.

The role of neural activity in the long-term regulation of tyrosine hydroxylase in peripheral adrenergic neurons had been studied by electrically stimulating the preganglionic nerves innervating the superior cervical ganglion. Tyrosin hydroxylase activity was elevated maximally 3 days after stimulation. The magnitude of the increase varied from 25 to 120% depending on the frequency and duration of the stimulation. Dopamine-beta-hydroxylase activity was also increased after preganglionic nerve stimulation, but there was no change in dopa decarboxylase activity or in the ganglion protein content

Cerebellar influence on parasympathetic neurones innervating intra-ocular muscles.

Previous work has suggested that cerebellum may control the size of the pupil and transmission in the pathway mediating the pupillary light reflex. In this work we found that electrical stimulation of the cerebellar nuclei (nucleus fastigius and nucleus interpositus bilaterally) evoked a discharge in the short ciliary nerves which innervate the intraocular muscles. The latency was 5.7 msec, which is short enough for a direct excitatory connexion from cerebellar efferents onto the preganglionic neurones in the Edinger-Westphal nucleus. These neurones are controlling the ciliary muscle of the le

The site of sympathetic deficit in cluster headache.

The pattern of autonomic deficit in the face of cluster headache patients resembles the deficit in patients with a postganglionic sympathetic lesion from some other cause; however, the presence of abnormal cardiac rhythms and bilateral pupillary reflex deficit in some patients with cluster headache suggests that the lesion might compromise central sympathetic drive. To investigate this possibility, the vasomotor and sudomotor startle reflex was investigated in the hands of six cluster headache patients with ocular and thermoregulatory signs of postganglionic sympathetic deficit in the face; fo

Parasympathetic preganglionic cardiac motoneurons labeled after voluntary diving.

A dramatic bradycardia is induced by underwater submersion in vertebrates. The location of parasympathetic preganglionic cardiac motor neurons driving this aspect of the diving response was investigated using cFos immunohistochemistry combined with retrograde transport of cholera toxin subunit B (CTB) to double-label neurons. After pericardial injections of CTB, trained rats voluntarily dove underwater, and their heart rates (HR) dropped immediately to 95 ± 2 bpm, an 80% reduction. After immunohistochemical processing, the vast majority of CTB labeled neurons were located in the reticular form

Ion channel and receptor-mediated regulation of axonal conduction reliability in sympathetic preganglionic neurons.

UNLABELLED: Sympathetic preganglionic neurons (SPNs) provide the sole spinal output to the peripheral sympathetic nervous system. Although sympathetic control is traditionally attributed to synaptic integration within the spinal cord and ganglia, the reliability of spike propagation along SPN axons themselves has received little attention. Here, and in companion papers, we show that axonal conduction in adult mouse thoracic SPNs is highly modifiable and constitutes a critical site of sympathetic gain control. Using an ex vivo preparation preserving intact paravertebral and splanchnic pathways

Characterization of vagal input to the rat esophageal muscle.

There is recent morphological evidence for an interaction of autonomic nerve fibers and extrinsic motor nerves of the rat esophagus. The aim of the present study was to investigate a possible functional role of this autonomic innervation of vagal motor fibers on rat esophageal smooth and striated muscle function in vitro. The entire esophagus with both Nn vagi, including the Nn recurrentes, was dissected and placed in an organ bath with oxygenated Krebs-Ringer buffer. Contractile activity was measured in longitudinal direction with a force transducer. Both Nn vagi were placed on a bipolar plat

Characterization of supraspinal vasomotor pathways and autonomic dysreflexia after spinal cord injury in F344 rats.

Cardiovascular dysfunction usually occurs after high thoracic and cervical spinal cord injury (SCI). The disruption of supraspinal vasomotor pathways (SVPs) results in the loss of bulbospinal regulation of sympathetic preganglionic neurons, leading to hypotension and compensatory tachycardia at rest. Episodic autonomic dysreflexia can develop upon sensory stimulation below the level of injury. In rodents, the precise spatial distribution of SVPs in the spinal cord originating from the rostral ventrolateral medulla (RVLM) has not been fully defined. To facilitate future studies of axon regenera

Intraspinal Plasticity Associated With the Development of Autonomic Dysreflexia After Complete Spinal Cord Injury.

Traumatic spinal cord injury (SCI) leads to disruption of sensory, motor and autonomic function, and triggers structural, physiological and biochemical changes that cause reorganization of existing circuits that affect functional recovery. Propriospinal neurons (PN) appear to be very plastic within the inhibitory microenvironment of the injured spinal cord by forming compensatory circuits that aid in relaying information across the lesion site and, thus, are being investigated for their potential to promote locomotor recovery after experimental SCI. Yet the role of PN plasticity in autonomic d

[Metasympathetic system and neuroendocrine elements of the human cecal appendix. Immunohistochemical study].

This study was carried out to demonstrate the neuroanatomical bases of pain in non inflamed appendices. Fifty human cecal appendices were examined by means of an immunohistochemical method utilizing antibodies for S100 protein, Neuron Specific Enolase, Chromogranin A and Serotonin. In all samples histological analysis did not demonstrate inflammatory alterations. The neuroendocrine pattern was investigated. Results show numerous nerve fibers and ganglions around the muscle layers and within the submucosa and the lamina propria. Large neuroendocrine cell population shows immunohistochemical sta

Oculosympathetic paresis. Analysis of 100 hospitalized patients.

Of 100 consecutive, adult, hospitalized inpatients with acquired unilateral oculosympathetic palsy, in 63 the central neuron was involved-usually due to strokes; in 21, tumor or trauma affected the preganglionic neuron; and in 13, postganglionic damage was sustained from a variety of causes. The preponderance of first neuron involvement by stroke is in part a reflection of patient sampling, but emphasizes the fact that central causes of Horner's syndrome are common.

CNS cell groups projecting to the submandibular parasympathetic preganglionic neurons in the rat: a retrograde transneuronal viral cell body labeling study.

The retrograde transneuronal viral tracing method was used to study the CNS nuclei that innervate the parasympathetic preganglionic neurons controlling the submandibular gland in the rat. A genetically engineered beta-galactosidase expressing Bartha strain of pseudorabies virus (PRV) was injected into the submandibular gland of rats. After 4 days, PRV infected tissues were reacted with the Bluo-Gal substrate (halogenated indolyl-beta-D-galactoside) and labeled cell bodies were identified throughout the brain. In the medulla oblongata, cell body labeling was seen in the superior salivatory nucl

Response of adrenal efferent nerve activity to noxious stimulation of the skin.

The effect of pinching the various segmental skin areas (neck, forepaw, lower chest, abdomen, thigh, hindpaw) on adrenal sympathetic efferent nerve activity which contributes to secretion of adrenal medullary hormones was examined in anesthetized rats. In the spinal cord-intact animal, pinching the lower chest, abdomen, forepaw or hindpaw produced a large reflex increase in efferent nerve activity. The response to pinching neck or thigh was small. In the spinalized animal, pinching the lower chest or abdomen produced a great response, but pinching the other skin areas including forepaw, hindpa

Dynamics of the remodeling of neural connections in the superior cervical ganglion in rats after dosed compression of the preganglionic trunk.

The aim of the present work was to obtain a quantitative evaluation of post-traumatic regeneration of nerve cells in the superior cervical ganglion (SCG) by measuring the ratio of the number of neurons (N) in the ganglion to the number of preganglionic myelinated fibers (F) in the cervical sympathetic trunk (N/F). This was addressed using light and electron microscopy. Studies were performed using white male rats divided into three groups: intact (aged seven months), age controls (aged 19 months), and experimental - one year after compression of the sympathetic trunk at the base of the SCG per

Brainstem origin of preganglionic cardiac motoneurons in the muskrat.

The muskrat, and aquatic rodent with a brisk and reliable diving response, shows a remarkable bradycardia after nasal stimulation. However, the medullary origin of cardiac preganglionic motoneurons is unknown in this species. We injected fat pads near the base of the heart of muskrats with a WGA-HRP solution to label retrogradely preganglionic parasympathetic neurons that project to the cardiac plexi. Results showed that the preponderance of labeled neurons was in ventrolateral parts of the medulla from 1.5 mm caudal to the obex to 2.0 mm rostral. Eighty-nine percent of the labeled neurons wer

Intermittent Horner's syndrome on alternate sides: a hint for locating spinal lesions.

A patient with a cervical intramedullary ependymoma is described who exhibited a Horner's syndrome on alternate sides. Pharmacological study demonstrated damage to the pre-ganglionic sympathetic neurons. Disregarding cases with the Shy-Drager syndrome, three comparable case reports have been found in the literature where the fluctuating expression of Horner's syndrome, alternating from one side to the other, was attributable to a lesion of the cervical spinal cord. In contrast to the present patient, the pharmacological responses of these patients indicated a lesion of the central sympathetic
