Autotrophic nitrogen removal after ureolytic phosphate precipitation to remove both endogenous and exogenous nitrogen.

Anaerobic digestion yields effluents rich in ammonium and phosphate and poor in biodegradable organic carbon, thereby making them less suitable for conventional biological nitrogen and phosphorus removal. In addition, the demand for fertilizers is increasing, energy prices are rising and global phosphate reserves are declining. This requires both changes in wastewater treatment technologies and implementation of new processes. In this contribution a description is given of the combination of a ureolytic phosphate precipitation (UPP) and an autotrophic nitrogen removal (ANR) process on the anae

Acidophilic heterotrophs: basic aspects and technological applications.

Acidophiles comprise a group of microorganisms adapted to live in acidic environments. Despite acidophiles are usually associated with an autotrophic metabolism, more than 80 microorganisms capable of utilizing organic matter have been isolated from natural and man-made environments. The ability to reduce soluble and insoluble iron compounds has been described for many of these species and may be harnessed to develop new or improved mining processes when oxidative bioleaching is ineffective. Similarly, as these microorganisms grow in highly acidic media and the chances of contamination are red

Field-scale application of spent sulfidic caustic as a source of alternative electron donor for autotrophic denitrification.

Biological reuse of spent sulfidic caustic (SSC) originating from oil refineries is a promising method for the petrochemical industry because of low handling cost. SSC typically contains high concentrations of sulfur, with the most dominant sulfur compounds being sulfide (S(2-)). SSC is also characterized by a high pH and elevated alkalinity up to 5-15% by weight. Because of these characteristics, SSC can be used for denitrification of NO3(-)-N in the biological nitrogen removal process as both the electron donor and buffering agent in sulfur-utilizing autotrophic denitrification. In this stud

Mixotrophic partial denitrification based on ferrous iron and sodium acetate: performance, microbial community, and potential mechanisms.

Partial denitrification (PD) was configured as a nitrite supplier for anaerobic ammonium oxidation (ANAMMOX). The mixotrophic PD (MPD) system using ferrous iron and sodium acetate as co-electron donors was operated at autotrophic-to-heterotrophic (A/H) ratios of 0.07 (Phase I), 0.21 (Phase II), and 0.85 (Phase III). Stable nitrite accumulation rates (NAR) of 78.0 %, 70.8 %, and 59.6 % were achieved in Phases I-III, respectively, while the autotrophic contribution increased from 21.8 % to 76.1 % with increasing A/H. Thauera, Longilinea and Thermomonas were the predominant denitrifiers, whereas

Novel two stage bio-oxidation and chlorination process for high strength hazardous coal carbonization effluent.

Effluent generated from coal carbonization to coke was characterized with high organic content, phenols, ammonium nitrogen, and cyanides. A full scale effluent treatment plant (ETP) working on the principle of single stage carbon-nitrogen bio-oxidation process (SSCNBP) revealed competition between heterotrophic and autotrophic bacteria in the bio-degradation and nitrification process. The effluent was pretreated in a stripper and further combined with other streams to treat in the SSCNBP. Laboratory studies were carried on process and stripped effluents in a bench scale model of ammonia stripp

Engineering Cupriavidus necator for L-threonine production.

L-threonine plays pivotal roles in the medicine, food, and feed industries. In this work, C. necator H16 was engineered for heterotrophic and autotrophic production of L-threonine. Blocking poly-hydroxybutyrate biosynthesis redirected the carbon flux toward L-threonine, resulting in a 6.8-fold increase in L-threonine titer relative to the wild type (from 5.56 mg/L to 38.03 mg/L). Deletion of genes involved in L-threonine catabolism further minimized L-threonine loss. Moreover, strategies including sufficient precursor supply, screening of key enzymes, relief of feedback inhibition, and efficie

Pyrite autotrophic denitrification in a biological contactor for low-strength nitrate-contaminated water treatment.

A novel pyrite autotrophic denitrification biological contactor (PAD-BC) process was developed for efficient nitrogen removal in water treatment, utilizing pyrite-loaded rope packings as both biofilm carrier and electron donor. Autotrophic denitrification rates with pyrite sizes of 0.55-0.05 mm, loadings of 2.5-12.5 g/cm, and nitrate concentrations of 7-28 mg/L followed first-order kinetic model, with rate constants of 0.402-0.501, 0.078-0.487, and 0.445-0.484 /d, respectively. Increased pyrite loading and nitrate concentration enhanced nitrite accumulation and sulfate production. The PAD-BC r

Key role of hydrogen in regulating hydrogenases and the reductive TCA cycle in a thermophilic, autotrophic sulfur-reducing bacterium.

UNLABELLED: Many thermophilic autotrophs in deep-sea hydrothermal vents are anaerobes that require H2 for growth. However, our understanding of how non-methanogenic thermophilic hydrogenotrophs adapt to low H2 conditions is nascent. In this study, the thermophilic autotrophic thiosulfate-reducing bacterium Desulfurobacterium thermolithotrophum HR11 was grown at 72°C in a chemostat at 2 and 43 µM aqueous H2 concentrations to quantify changes in growth and H2S production kinetics and in its proteome in response to H2 availability. At 2 μM H2, cultures showed a decrease in specific growth rate, m

Efficient nitrite accumulation in partial sulfide autotrophic denitrification (PSAD) system: insights of S/N ratio, pH and temperature.

To provide the necessary nitrite for the Anaerobic Ammonium Oxidation (ANAMMOX) process, the effect of nitrite accumulation in the partial sulfide autotrophic denitrification (PSAD) process was investigated using an SBR reactor. The results revealed that the effectiveness of nitrate removal was unsatisfactory when the S/N ratio (mol/mol) fell below 0.6. The optimal conditions for nitrate removal and nitrite accumulation were achieved within the S/N ratio range of 0.7-0.8, resulting in an average Nitrate Removal Efficiency (NRE) of 95.84%±4.89% and a Nitrite Accumulation Rate (NAR) of 75.31%±6.

Nitrate removal in iron sulfide-driven autotrophic denitrification biofilter: Biochemical and chemical transformation pathways and its underlying microbial mechanism.

Iron sulfides-based autotrophic denitrification (IAD) is effective for treating nitrate-contaminated wastewater. However, the complex nitrate transformation pathways coupled with sulfur and iron cycles in IADs are still unclear. In this study, two columns (abiotic vs biotic) with iron sulfides (FeS) as the packing materials were constructed and operated continuously. In the abiotic column, FeS chemically reduced nitrate to ammonium under the ambient condition; this chemical reduction reaction pathway was spontaneous and has been overlooked in IAD reactors. In the biotic column (IAD biofilter),

Growth of Dehalococcoides mccartyi species in an autotrophic consortium producing limited acetate.

The dechlorinating Dehalococcoides mccartyi species requires acetate as carbon source, but little is known on its growth under acetate limiting conditions. In this study, we observed growth and dechlorination of a D. mccartyi-containing mixed consortium in a fixed-carbon-free medium with trichloroethene in the aqueous phase and H2/CO2 in the headspace. Around 4 mM formate was produced by day 40, while acetate was constantly below 0.05 mM. Microbial community analysis of the consortium revealed dominance by D. mccartyi and Desulfovibrio sp. (57 and 22% 16S rRNA gene copies, respectively). From

Measurement and modeling of multiple substrate oxidation by methanotrophs at 20 degrees C.

Earlier experiments have shown that when Methylosinus trichosporium OB3b was grown at 30 degrees C, greater growth and degradation of chlorinated ethenes was observed under particulate methane monooxygenase (pMMO)-expressing conditions than sMMO-expressing conditions. The effect of temperature on the growth and ability of methanotrophs to degrade chlorinated ethenes, however, has not been examined, particularly temperatures more representative of groundwater systems. Thus, experiments were performed at 20 degrees C to examine the effect of mixtures of trichloroethylene, trans-dichloroethylene

Advancements and challenges of partial denitrification/anammox technology: Implementation strategies for plug-flow municipal wastewater treatment.

This review systematically summarizes the latest advances, key challenges, and engineering application strategies of partial denitrification/anammox (PD/A) technology, with a particular focus on plug-flow biosystems. The plug-flow process is widely used, yet its effluent often fails standards due to low C/N ratios. To address this carbon deficiency, PD/A leverages its own low-carbon metabolic feature to offer a promising upgrade pathway toward compliant discharge. Here, this review highlights latest strategies developed to enhance nitrogen removal efficiency (NRE) in plug-flow PD/A systems, in

Simultaneous nitrogen and sulfur removal through synergy of sulfammox, anammox and sulfur-driven autotrophic denitrification in a modified bioreactor enhanced by activated carbon.

Anaerobic ammonium (NH4+ - N) oxidation coupled with sulfate (SO42-) reduction (sulfammox) is a new pathway for the autotrophic removal of nitrogen and sulfur from wastewater. Sulfammox was achieved in a modified up-flow anaerobic bioreactor filled with granular activated carbon. After 70 days of operation, the NH4+ - N removal efficiency almost reached 70%, with activated carbon adsorption and biological reaction accounting for 26% and 74%, respectively. Ammonium hydrosulfide (NH4SH) was found in sulfammox by X-ray diffraction analysis for the first time, which confirmed that hydrogen sulfide

Pesticide Mixtures Cause Short-Term, Reversible Effects on the Function of Autotrophic Periphyton Assemblages.

In a laboratory experiment we investigated the effects of pesticide mixtures on the structure and function of freshwater biofilms, with focus on their photoautotrophic component. We identified 6 herbicides and 1 fungicide commonly found in Swedish streams at relatively high concentrations and created 3 ternary mixtures that were tested in concentration series ranging from observed environmental concentrations to up to 100 times higher. Biofilms were exposed to these pesticide mixtures for 8 d and then allowed to recover for another 12 d. Our results show a rapid and consistent inhibition of ph

Reverse-Engineered Gas-Fermenting Acetogen Strains Recover Enhanced Phenotypes From Autotrophic Adaptive Laboratory Evolution.

Gas-fermenting acetogens, such as Clostridium autoethanogenum, have emerged as promising biocatalysts capable of converting CO and CO2-containing gases into fuels and chemicals relevant for a circular economy. However, the functionalities of the majority of genes in acetogens remain uncharacterised, hindering the development of acetogen cell factories through targeted genetic engineering. We previously identified gene targets through adaptive laboratory evolution (ALE) that potentially realise enhanced autotrophic phenotypes in C. autoethanogenum. In this study, we deleted one of the targets-C

Mixotrophic denitrification of waste activated sludge fermentation liquid as an alternative carbon source for nitrogen removal: Reducing N2O emissions and costs.

Heterotrophic-sulfur autotrophic denitrification (HAD) has been proposed to be a prospective nitrogen removal process. In this work, the potential of fermentation liquid (FL) from waste-activated sludge (WAS) as the electron donor for denitrification in the HAD system was explored and compared with other conventional carbon sources. Results showed that when FL was used as a carbon source, over 99% of NO3--N was removed and its removal rate exceeded 14.00 mg N/g MLSS/h, which was significantly higher than that of methanol and propionic acid. The produced sulfate was below the limit value and th

Autotrophic Uranium (VI) bioreduction in groundwater by siderite: Comparison with sulfur and zero-valent iron.

Uranium (U), recognized as a significant health risk in groundwater, has become a key focus in environmental remediation efforts. While numerous electron donors have been investigated for the removal of U(VI) through microbial processes, the potential of abundant and economical Fe(II)-containing minerals remains unexplored. Here, a new inorganic electron donor, siderite (FeCO3) was proposed. Although siderite demonstrates a lesser electron-donating capacity than Fe(0) and S(0), the Siderite-B bioreactor successfully enriched microbes belonging to the Azotobacter genus, which are known for thei

Autotrophic denitrification supported by sphalerite and oyster shells: Chemical and microbiome analysis.

This research evaluated the metal-sulfide mineral, sphalerite, as an electron donor for autotrophic denitrification, with and without oyster shells (OS). Batch reactors containing sphalerite simultaneously removed NO3- and PO43- from groundwater. OS addition minimized NO2- accumulation and removed 100% PO43- in approximately half the time compared with sphalerite alone. Further investigation using domestic wastewater revealed that sphalerite and OS removed NO3- at a rate of 0.76 ± 0.36 mg NO3--N/(L · d), while maintaining consistent PO43- removal (∼97%) over 140 days.
