Discovery of Novel Prebiotic Carbohydrates and Sugar Mimics of BlMsmE, a Solute-Binding Protein of the ABC Transporter from Bacillus licheniformis.

Stachyose is a typical prebiotic that can be utilized by the probiotic strain Bacillus licheniformis. Pioneering X-ray crystallography has determined the structure of stachyose in complex with the solute-binding protein MsmE in B. licheniformis (BlMsmE). The present work describes a combined strategy for the identification of putative BlMsmE-specific ligands, which can be used for the development of prebiotics. After a ligand-based virtual similarity screening of a large ZINC database containing ∼22 M compounds, we identified 3575 ligands. A total of 600 structures for which the Tanimoto coeff

YtnP lactonase from Bacillus licheniformis T2 mitigates virulence and biofilm formation in carbapenem-resistant Acinetobacter baumannii via quorum sensing quenching.

BACKGROUD: Carbapenem-resistant Acinetobacter baumannii (CRAB) has become a predominant nosocomial pathogen worldwide, with biofilm formation and quorum sensing (QS) primarily responsible for its high antimicrobial tolerance and clinical refractory infections. Targeting bacterial virulence rather than conventional bactericidal strategies represents a promising alternative to combat multidrug-resistant CRAB. METHODS: In this study, we isolated a N-acyl homoserine lactonase YtnP from mangrove rhizosphere-derived Bacillus licheniformis T2. We comprehensively biochemically characterized the YtnP

Structural basis and functional mining of active site loop for fine tuning substrate affinity of L-asparaginase from Bacillus licheniformis.

The widespread presence of the environmental contaminant acrylamide (AA) in thermally processed foods has been linked to serious environmental and health challenges. Although the formation of AA can be fundamentally inhibited by L-asparaginase (L-ASNase), its removal efficiency is highly dependent on substrate affinity. However, the structural mechanism by which the affinity of this L-ASNase is regulated remains poorly understood. Herein, a rational design strategy of active site loop (ASL) engineering was initially employed to improve the substrate affinity of L-ASNase from Bacillus lichenifo

Prophylactic administration of overproducing-abscisic acid Bacillus licheniformis attenuated DSS-induced colitis in mice by regulating the gut microbiota and immune activity.

BACKGROUND: Inflammatory bowel disease (IBD) involves the complex interplay among the mucosal barrier, microbiota, immunity and genetic factors. There are currently no satisfactory treatments for IBD. Administration of the probiotic Bacillus licheniformis (Bl) can improves colitis by regulating the gut microbiota. The phytohormone abscisic acid (ABA) treatment has favorable effects on immunity, as well as on inflammatory diseases like colitis. We hypothesized that the expression of an additional cyp gene by the Bl to increase its ABA production would enhance its effects. RESULTS: In this stud
