Priestia megaterium cells are primed for surviving lethal doses of antibiotics and chemical stress.

Antibiotic resistant infections kill millions worldwide yearly. However, a key factor in recurrent infections is antibiotic persisters. Persisters are not inherently antibiotic-resistant but can withstand antibiotic exposure by entering a non-dividing state. This tolerance often results in prolonged antibiotic usage, increasing the likelihood of developing resistant strains. Here, we show the existence of "primed cells" in the Gram-positive bacterium Priestia megaterium, formerly known as Bacillus megaterium. These cells are pre-adapted to become persisters prior to lethal antibiotic stress. R

Spore heat resistance and specific mineralization.

Spores of Bacillus megaterium ATCC 19213, Bacillus subtilis niger and Bacillus stearothermophilus ATCC 7953 were converted to fully demineralized, but viable, H forms by controlled acid titration. H forms were more heat sensitive than were native forms, but z values were greater for killing of H spores than those for native spores. Therefore, the differences in heat sensitivity between native and H forms decreased with increasing killing temperature. The increase in heat sensitivity associated with demineralization did not appear to be due to damage to cortex lytic enzymes of the germination s

Organomercurials removal by heterogeneous merB genes harboring bacterial strains.

Organomercury lyase (MerB) is a key enzyme in bacterial detoxification and bioremediation of organomercurials. However, the merB gene is often considered as an ancillary component of the mer operon because there is zero to three merB genes in different mer operons identified so far. In this study, organomercurials' removal abilities of native mercury-resistant bacteria that have one or multiple merB genes were examined. Each heterogeneous merB genes from these bacteria was further cloned into Escherichia coli to investigate the substrate specificity of each MerB enzyme. The merB1 gene from Bac

Process optimization of struvite recovered from slaughterhouse wastewater and its fertilizing efficacy in amendment of biofertilizer.

The intensive discharge of slaughterhouse waste into water bodies increases Nitrogen (N), Phosphorus (P) in the wastewater and leads to various environmental problems. On the other hand, the increasing treatment effort after the extraction of these valuable nutrients in the commercial fertilizer reduces the dependence on scarce phosphate resources. The viable solution is to recover N, P as struvite (magnesium ammonium phosphate) from nutrient rich waste water as a small scale treatment unit application. The main parameters that have a significant impact on the process, including pH, Mg: P rati

Bacterial Lipopeptides Are Effective against Pear Fire Blight.

Fire blight, a devastating disease caused by Erwinia amylovora, poses a significant threat to pear and apple trees in Xinjiang province, China. In an effort to combat this pathogen, we isolated 10 bacteria from various components of apple and crabapple trees and conducted screenings to assess their ability to inhibit E. amylovora in vitro. Through biochemical tests and partial 16S rRNA gene sequencing, we identified two promising strains, Priestia megaterium strain H1 and Bacillus subtilis strain I2. These strains were then evaluated for their efficacy in biocontrol under controlled laboratory

Structural and Biochemical Characterization of a Copper-Binding Mutant of the Organomercurial Lyase MerB: Insight into the Key Role of the Active Site Aspartic Acid in Hg-Carbon Bond Cleavage and Metal Binding Specificity.

In bacterial resistance to mercury, the organomercurial lyase (MerB) plays a key role in the detoxification pathway through its ability to cleave Hg-carbon bonds. Two cysteines (C96 and C159; Escherichia coli MerB numbering) and an aspartic acid (D99) have been identified as the key catalytic residues, and these three residues are conserved in all but four known MerB variants, where the aspartic acid is replaced with a serine. To understand the role of the active site serine, we characterized the structure and metal binding properties of an E. coli MerB mutant with a serine substituted for D99

Large-scale screening of nasal swabs for Bacillus anthracis: descriptive summary and discussion of the National Institutes of Health's experience.

In October 2001, a letter containing a large number of anthrax spores was sent through the Brentwood post office in Washington, D.C., to a United States Senate office on Capitol Hill, resulting in contamination in both places. Several thousand people who worked at these sites were screened for spore exposure by collecting nasal swab samples. We describe here a screening protocol which we, as a level A laboratory, used on very short notice to process a large number of specimens (3,936 swabs) in order to report preliminary results as quickly as possible. Six isolates from our screening met preli

Characterization of native plant growth promoting rhizobacteria and their anti-oomycete potential against Phytophthora capsici affecting chilli pepper (Capsicum annum L.).

Phytophthora capsici is a notorious fungus which infects many crop plants at their early and late growth stages. In the present study, twelve P. capsici isolates were morphologically characterized, and based on pathogenicity assays; two highly virulent isolates causing post-emergence damping-off on locally cultivated chilli pepper were screened. Two P. capsici isolates, HydPak1 (MF322868) and HydPk2 (MF322869) were identified based on internal transcribed spacer (ITS) sequence homology. Plant growth promoting rhizobacteria (PGPR) play a significant role in disease suppression and plant growth

Functionalized poly(3-hydroxybutyric acid) bodies as new in vitro biocatalysts.

Cytochromes P450 play a key role in the drug and steroid metabolism in the human body. This leads to a high interest in this class of proteins. Mammalian cytochromes P450 are rather delicate. Due to their localization in the mitochondrial or microsomal membrane, they tend to aggregate during expression and purification and to convert to an inactive form so that they have to be purified and stored in complex buffers. The complex buffers and low storage temperatures, however, limit the feasibility of fast, automated screening of the corresponding cytochrome P450-effector interactions, which are

Synthesis, characterization, biological activities, and computational studies of pyrazolyl-thiazole derivatives of thiophene.

This study reports the synthesis, characterization, and biological evaluation of a series of pyrazolyl-thiazole derivatives of thiophene. Seven compounds were synthesized and characterized using NMR spectroscopy and mass spectrometry. The antimicrobial activities of these derivatives were evaluated against various bacterial (Escherichia coli, Bacillus subtilis, Bacillus megaterium, Staphylococcus aureus) and fungal strains (Aspergillus niger, Aspergillus oryzae, Rhizopus, Candida albicans), demonstrating significant inhibition zones and low minimum inhibitory concentrations (MIC). In addition,

Efficiency of corncob-derived biochar and plant growth-promoting rhizobacteria as growth promoters for Coffea arabica L. seedlings.

Corncob-derived biochar and plant growth-promoting rhizobacteria (PGPR) can improve soil fertility and plant performance, but their co-application in Coffea arabica L. nurseries remains poorly characterised. This study evaluated corncob-derived biochar combined with Bacillus megaterium S2-4a1 under well-watered greenhouse conditions. A completely randomised design with six replications was maintained for 180 days and included five treatments: control (T1), S2-4a1 alone (T2), and S2-4a1 combined with 1, 2, or 3% (w/w) biochar (T3-T5). Lack of efficient killing of purified dormant spores of Bacillales and Clostridiales species by glycerol monolaurate in a non-aqueous gel.

Inactivation of Bacillales and Clostridiales spores is of interest, since some cause food spoilage and human diseases. A recent publication (mSphere 3: e00597-1, 2018) reported that glycerol monolaurate (GML) in a non-aqueous gel (GMLg) effectively killed spores of Bacillus subtilis, Bacillus cereus and Clostridioides difficile, and Bacillus anthracis spores to a lesser extent. We now show that (i) the B. subtilis spores prepared as in the prior work were impure; (ii) if spore viability was measured by diluting spores 1/10 in GMLg, serially diluting incubations 10-fold and spotting aliquots on

Metal solubilization from metal-containing solid materials by cyanogenic Chromobacterium violaceum.

Different cyanogenic bacterial strains (Chromobacterium violaceum, Pseudomonas fluorescens, Bacillus megaterium) were cultivated under cyanide-forming conditions in the presence of metal-containing solids such as nickel powder or electronic scrap. All microorganisms were able to form water-soluble metal cyanides, however, with different efficiencies. C. violaceum was able to mobilize nickel as tetracyanonickelate [Ni(CN)4(2-)] from fine-grained nickel powder. Gold was microbially solubilized as dicyanaoaurate [Au(CN)2-] from electronic waste. Additionally, cyanide-complexed copper was detected

Characterization of biodegradation in a 17th century easel painting and potential for a biological approach.

It is important to characterize the microorganisms involved in biodeterioration processes to understand their effects on cultural assets and to define an efficient strategy for protecting artworks, monuments, and buildings from microbiological recolonization. In this study, we analyzed the microbial communities dwelling on the verso (front) and recto (back) sides of a 17th century easel painting attributed to Carlo Bononi, an Italian artist of the first Baroque period. Cultivable bacteria and fungi colonizing the painting were isolated and identified in order to characterize the microbial comm

In Vitro Anti-Oxidant and Anti-Microbial Potentiality Investigation of Different Fractions of Caryota urens Leaves.

BACKGROUND: Caryota urens is a member of the Arecaceae family and a common plant in the Southeast Asian region. This plant has been reported as an anti-microbial agent in recent years. Thus, we aimed to find out the MIC (minimum inhibitory concentration) against different pathogenic microorganism. METHODS: The leaves of C. urens were extracted and fractioned using different reagents (chloroform, n-hexane and carbon tetrachloride). Disc diffusion method was implemented for the assessment of in vitro anti-microbial potency (500 and 250 µg/disc). RESULT: The entire fraction showed good effect (

Vinasse: from a residue to a high added value biopolymer.

This work aimed to study the feasibility of using vinasse for polyhydroxybutyrate (PHB) production by Bacillus megaterium. To optimize the culture medium, a Box-Behnken design was employed considering carbon (C), nitrogen (N), and phosphorus (Ph) concentrations as independent variables and PHB productivity as the response variable. The productivity decreased when C or N were increased, probably due to the presence of phenolic compounds and the limitation of N for the production of PHB by Bacillus sp. bacteria. An additional experimental design to optimize the C/N ratio and growing conditions (

Bacillus strains exhibit various plant growth promoting traits and their biofilm-forming capability correlates to their salt stress alleviation effect on maize seedlings.

Soil salinity interferes with plant growth and development. Bacillus genus has been used to increase the growth and productivity of a wide variety of crops by alleviating the effects of salt stress. A total of thirty two Bacillus isolates were obtained from maize rhizosphere, and their plant growth-promoting (PGP) traits and biocontrol activities were tested. Bacillus isolates displayed varying degrees of PGP properties-the production of extracellular enzymes, indole acetic acid, hydrogen cyanide, phosphate solubilization, biofilm formation, and antifungal potential against several fungal path

Bacterial Volatile Organic Compounds Mediate a Cooperative Relationship Between Bacteria and Plants Under Heavy Metal Stress.

Bacterial volatile organic compounds (BVOCs) are increasingly recognised as effective signalling molecules that stimulate plant growth and stress responses. Although BVOC production may impose metabolic costs on bacteria, the benefits gained by VOC-emitting bacteria through BVOC-mediated communication with plants remain unclear. In this study, we established Pb-stressed conditions for Bacillus megaterium, Arabidopsis thaliana or both organisms to investigate their reciprocal interactions. Pb-stressed bacteria emitted a distinct VOC profile and exhibited significant reprogramming of sugar and n

MICP mediated by indigenous bacteria isolated from tailings for biocementation for reduction of wind erosion.

In this study, native ureolytic bacteria were isolated from copper tailings soils to perform microbial-induced carbonate precipitation (MICP) tests and evaluate their potential for biocement formation and their contribution to reduce the dispersion of particulate matter into the environment from tailings containing potentially toxic elements. It was possible to isolate a total of 46 bacteria; among them only three showed ureolytic activity: Priestia megaterium T130-1, Paenibacillus sp. T130-13 and Staphylococcus sp. T130-14. Biocement cores were made by mixing tailings with the isolated bacter
