Assessing the impact of Cry insecticidal proteins from Bacillus thuringiensis on the midgut of Spodoptera littoralis (Boisduval).

Bacillus thuringiensis (Bt) Cry toxins exert their insecticidal activity by targeting the larval midgut epithelium, yet integration of physiological, ultrastructural, and microbiological responses under near-native conditions remains limited. In this study, we investigated the effects of Cry1Ca on Spodoptera littoralis larvae using an integrative approach combining physiological measurements with high-resolution scanning and transmission electron microscopy (SEM and TEM). Building on previous whole-organism imaging approaches, we analyzed intact larvae without prior dissection, an approach tha

In Vitro Assessment of Bacillus thuringiensis Exopolysaccharides and Their Effects on Gut Microbiota from Ulcerative Colitis In Vitro.

Bacillus thuringiensis exopolysaccharide BPS-2 inhibits malondialdehyde secretion, enhances antioxidant enzyme activities, and significantly improves the antioxidant status of inflammatory cells. In the present study, the apparent morphology and spatial conformation of BPS-2 were analyzed further, and several functional properties were investigated. The results demonstrated that BPS-2 was a polymeric straight-chain polysaccharide with good thermal stability, exhibiting non-Newtonian properties and good antioxidant and anticancer activities. Notably, this study systematically investigated the i

Identification of Aedes aegypti specificity motifs in the N-terminus of the Bacillus thuringiensis Cry2Aa pesticidal protein.

One advantage of using the Cry proteins of Bacillus thuringiensis as pesticides is their relatively narrow spectrum of activity, thus reducing the risk of non-target effects. Understanding the molecular basis of specificity has the potential to help us design improved products against emerging pests, or against pests that have developed resistance to other Cry proteins. Many previous studies have associated specificity with the binding of the Cry protein, particularly through the apical regions of domain II, to particular receptors on the midgut epithelial cells of the host insect. We have pre

Acaricidal effect and histological damage induced by Bacillus thuringiensis protein extracts on the mite Psoroptes cuniculi.

BACKGROUND: The mite Psoroptes cuniculi is a common worldwide ectoparasite and the most frequently found in rabbit farms. It causes significant economic losses on commercial rabbit breeding associated with poor leather quality, reduced conception rates, weight loss, poor growth and death. Several strategies have been proposed for the treatment of mange caused by this mite, ranging from the use of acaricides, entomopathogenic fungi, essential oils and vaccines. However, therapy and control of both human scabies and animal mange are still based mainly on the use of drugs and chemicals such as iv

Binding properties of Bacillus thuringiensis Cry4A toxin to the apical microvilli of larval midgut of Culex pipiens.

Cry4A is a dipteran-specific delta-endotoxin produced by Bacillus thuringiensis, and toxic to Culex pipiens (mosquito) larvae. The immunohistochemical staining of the midgut sections of C. pipiens larvae revealed that Cry4A bound in vitro and in vivo to the microvilli of the epithelial cells of posterior midgut and gastric caecae. The binding of digoxigenin-labeled Cry4A (DIG-Cry4A) to the apical microvilli was almost abolished in the presence of excess unlabeled Cry4A, suggesting that the binding of Cry4A to the microvilli was specific. Several Cry4A-specific binding proteins were detected us

Coleopteran-active cry gene profiling and insecticidal activity of indigenous Bacillus thuringiensis strains against Holotrichia serrata.

White grub (Holotrichia serrata F.) is a destructive soil-dwelling pest causing severe yield losses in several agricultural and horticultural crops. Owing to its subterranean habit, chemical control is often ineffective, necessitating sustainable biological alternatives. This study investigated the distribution of coleopteran-active cry genes among indigenous Bacillus thuringiensis strains and evaluated their insecticidal activity against H. serrata. A total of 100 indigenous B. thuringiensis strains isolated from the Western Ghats of India were screened using gene-specific PCR for 12 coleopte

Efficient and Scalable Process to Produce Novel and Highly Bioactive Purified Cytosolic Crystals from Bacillus thuringiensis.

Bacillus thuringiensis (Bt) is a Gram-positive soil bacterium that is widely and safely applied in the environment as an insecticide for combatting insect pests that damage crops or are disease vectors. Dominant active ingredients made by Bt are insect-killing crystal (Cry) proteins released as crystalline inclusions upon bacterial sporulation. Some Bt Cry proteins, e.g., Cry5B (formally Cry5Ba1), target nematodes (roundworms) and show exceptional promise as anthelmintics (cures for parasitic nematode diseases). We have recently described inactivated bacteria with cytosolic crystal(s) (IBaCC)

Foliar Application of Bacillus thuringiensis Regulates Soil Physicochemical Properties and Microbial Ecological Functions in Tea Plantations.

Bacillus thuringiensis (Bt) is widely employed as a biological control agent against pests in tea plantations, yet its impacts on soil health and microbial ecology remain insufficiently understood. This study investigated the effects of two Bt application regimes, namely moderate-frequency conventional Bt application (Bt1, 3 sprays over 21 days) and high-frequency intensive Bt application (Bt2, 6 sprays over 42 days), on soil physicochemical properties, enzyme activities, and microbial community structure and function in tea soils. The moderate-frequency conventional Bt1 significantly improved

Enhancing UV radiation protection of Bacillus thuringiensis formulations using sulfur quantum dots: synthesis and efficacy evaluation.

Bacillus thuringiensis (Bt) is a widely used microbial insecticide, but its effectiveness is limited due to the degradation of Bt spores and crystals under UV radiation from sunlight. The objective of this study was to develop a novel Bt formulation with improved UV protection by utilizing sulfur quantum dots (SQDs) as stabilizing agents in a Pickering emulsion. The SQDs were comprehensively characterized using FTIR, XRD, TEM, HRTEM, UV, and fluorescence analyses, which confirmed the formation of well-dispersed, spherical SQDs. The microcapsule formulation with SQDs demonstrated superior UV st

Retrotransposon-mediated disruption of a chitin synthase gene confers insect resistance to Bacillus thuringiensis Vip3Aa toxin.

The vegetative insecticidal protein Vip3Aa from Bacillus thuringiensis (Bt) has been produced by transgenic crops to counter pest resistance to the widely used crystalline (Cry) insecticidal proteins from Bt. To proactively manage pest resistance, there is an urgent need to better understand the genetic basis of resistance to Vip3Aa, which has been largely unknown. We discovered that retrotransposon-mediated alternative splicing of a midgut-specific chitin synthase gene was associated with 5,560-fold resistance to Vip3Aa in a laboratory-selected strain of the fall armyworm, a globally importan

Recombinant Beauveria bassiana expressing Bacillus thuringiensis toxin Cyt1Aa: a promising approach for enhancing Aedes mosquito control.

UNLABELLED: The entomopathogenic fungus Beauveria bassiana provides an eco-friendly substitute to chemical insecticides for mosquito control. Nevertheless, its widespread application has been hindered by its comparatively slow efficacy in eliminating mosquitoes. To augment the potency of B. bassiana against Aedes mosquitoes, a novel recombinant strain, Bb-Cyt1Aa, was developed by incorporating the Bacillus thuringiensis toxin gene Cyt1Aa into B. bassiana. The virulence of Bb-Cyt1Aa was evaluated against Aedes aegypti and Aedes albopictus using insect bioassays. Compared to the wild-type (WT) s

Mosquitocidal Potential of Native Bacillus thuringiensis Strain SY49-1 against Disease Vector, Culex pipiens (Diptera: Culicidae).

Mosquitoes are regarded as troublesome organisms worldwide due to their parasitic and pathogenic aspect causing malaria, yellow fever, dengue, West Nile and filariasis. Bacillus thuringiensis (Bt) products have effectively been used for decades in controlling the mosquito mediated diseases and also decreasing the chemical input into the environment. In the present study, biocontrol potential of previously characterized Bt SY49-1 strain was investigated on Culex pipiens larvae. Fourth instars of C. pipiens were subjected to spore/δendotoxins (50, 100, 250 and 500 µg mL-1) of Bt SY49-1 strain, c

Histopathological effects of cypermethrin and Bacillus thuringiensis var. israelensis on midgut of Chironomus calligraphus larvae (Diptera: Chironomidae).

Pesticides are extensively used for the control of agricultural pests and disease vectors, but they also affect non-target organisms. Cypermethrin (CYP) is a synthetic pyrethroid used worldwide. Otherwise, bioinsecticides like Bacillus thuringiensis var. israelensis (Bti) have received great attention as an environmentally benign and desirable alternative. In order to evaluate the toxicity of those pesticides, Chironomus calligraphus was selected due to its high sensitivity to some toxicants. Third and fourth instars larvae were exposed to serial dilutions of CYP and Bti to determine LC50 valu

Cry1Ab protein from Bacillus thuringiensis and MON810 cry1Ab-transgenic maize exerts no adjuvant effect after airway exposure.

The genetically modified (GM) maize event MON810 has been inserted with a processed version of the transgene, cry1Ab, derived from the soil bacterium Bacillus thuringiensis (Bt) to express proteins with insecticidal properties. Such proteins may introduce new allergens and also act as adjuvants that promote allergic responses. While focus has been on safe consumption and hence the oral exposure to GM food and feed, little is known regarding inhalation of pollen and desiccated airborne plant material from GM crops. The aim of this study was to investigate whether plant material from the Cry1Ab-

Biochemical, molecular cytogenetic and correlation analyses of Bacillus thuringiensis sublethal concentration toxicity on Spodoptera frugiperda larval Hemolymph.

Bacillus thuringiensis is considered one of the most environmentally safe tools for controlling various insect pests, including the fall armyworm, Spodoptera frugiperda (J.E. Smith). This pest remains one of the most serious threats to global food security. Although Bacillus thuringiensis var. kurstaki (Btk) is an essential long-term pest control agent, most research has examined its lethal effects only on larval midgut tissue as the primary target organ. Accordingly, this study explored Btk toxicity on the third-instar S. frugiperda larval hemolymph as a secondary consequence using a multifac
