Structural basis of T-cell specificity and activation by the bacterial superantigen TSST-1.

Superantigens (SAGs) bind simultaneously to major histocompatibility complex (MHC) and T-cell receptor (TCR) molecules, resulting in the massive release of inflammatory cytokines that can lead to toxic shock syndrome (TSS) and death. A major causative agent of TSS is toxic shock syndrome toxin-1 (TSST-1), which is unique relative to other bacterial SAGs owing to its structural divergence and its stringent TCR specificity. Here, we report the crystal structure of TSST-1 in complex with an affinity-matured variant of its wild-type TCR ligand, human T-cell receptor beta chain variable domain 2.1.

Structure of HrcQB-C, a conserved component of the bacterial type III secretion systems.

Type III secretion systems enable plant and animal bacterial pathogens to deliver virulence proteins into the cytosol of eukaryotic host cells, causing a broad spectrum of diseases including bacteremia, septicemia, typhoid fever, and bubonic plague in mammals, and localized lesions, systemic wilting, and blights in plants. In addition, type III secretion systems are also required for biogenesis of the bacterial flagellum. The HrcQ(B) protein, a component of the secretion apparatus of Pseudomonas syringae with homologues in all type III systems, has a variable N-terminal and a conserved C-termi

Cnidarian-algal partnerships structure bacterial communities during strobilation in Cassiopea xamachana.

Cnidarian-algal (Symbiodiniaceae) symbioses rely on complex interactions among the cnidarian host, algal symbionts, and associated bacterial communities. In the upside-down jellyfish Cassiopea xamachana, the polyp-to-medusa transition (strobilation) requires the establishment of symbiosis with Symbiodiniaceae algal partners, yet bacterial community dynamics during this developmental process remain unknown. Here, we experimentally induced symbiosis in aposymbiotic polyps using four algal treatments: xenic Symbiodinium microadriaticum (native symbiont), xenic Breviolum minutum, antibiotic-treate

Structure-based classification of bacterial chitooligosaccharide-specific outer membrane porins (ChiPs).

Chitoporins (ChiPs) are chitooligosaccharide-specific outer membrane porins found in various Gram-negative bacteria and are critical for marine species that use chitin as a carbon and nitrogen source. Vibrio spp. ChiPs, exemplified by VhChiP from Vibrio harveyi, are trimeric OmpC-like 16-stranded β-barrels with a long pore-confining loop L3 that interacts exclusively with long chain chitooligosaccharides. Here, using crystallography and electrophysiology, we show that two atypical ChiPs, Escherichia coli ChiP (EcChiP) and Serratia marcescens ChiP (SmChiP), are monomeric 18-stranded β-barrels w

Resistome profiling and bacterial community structure of semi-urban gutter ecosystems of India.

Environmental factors contribute to antimicrobial resistance, a global health threat. Contaminated gutter water in urban areas spreads resistant bacteria, disrupting ecosystems and promoting biofilm formation, causing widespread concern. This study aimed to evaluate antibiotic-resistant bacterial populations across six gutter ecosystems in Roorkee, Uttarakhand, India during summer against different classes of antibiotics, identify presence of beta-lactamase, and explores total bacterial communities, and predicting metabolic pathways through 16S rRNA based metagenomic approach of V3 region. Bacterial Autoimmune Drug Metabolism Transforms an Immunomodulator into Structurally and Functionally Divergent Antibiotics.

Tapinarof is a stilbene drug that is used to treat psoriasis and atopic dermatitis, and is thought to function through regulation of the AhR and Nrf2 signaling pathways, which have also been linked to inflammatory bowel diseases. It is produced by the gammaproteobacterial Photorhabdus genus, which thus represents a model to probe tapinarof structural and functional transformations. We show that Photorhabdus transforms tapinarof into novel drug metabolism products that kill inflammatory bacteria, and that a cupin enzyme contributes to the conversion of tapinarof and related dietary stilbenes in

Total bacterial load, inflammation, and structural lung disease in paediatric cystic fibrosis.

BACKGROUND: Cystic fibrosis (CF) is characterised by reduced airway clearance, microbial accumulation, inflammation, and lung function decline. Certain bacterial species may contribute disproportionately to worsening lung disease. However, the relative importance of these microorganisms compared to the absolute abundance of all bacteria is uncertain. We aimed to identify the characteristics of lower airway microbiology that best reflect CF airway inflammation and disease in children. METHODS: Analysis was performed on bronchoalveolar lavage (BAL) fluid from 78 participants of the Australasian

Evolutionary pathways to antibiotic resistance are dependent upon environmental structure and bacterial lifestyle.

Bacterial populations vary in their stress tolerance and population structure depending upon whether growth occurs in well-mixed or structured environments. We hypothesized that evolution in biofilms would generate greater genetic diversity than well-mixed environments and lead to different pathways of antibiotic resistance. We used experimental evolution and whole genome sequencing to test how the biofilm lifestyle influenced the rate, genetic mechanisms, and pleiotropic effects of resistance to ciprofloxacin in Acinetobacter baumannii populations. Both evolutionary dynamics and the identitie

Delafloxacin meglumine for the treatment of acute bacterial skin and skin structure infections (ABSSSI).

Delafloxacin meglumine (Baxdela, WQ-3034, ABT-492, RX-3341; Melinta Therapeutics) was approved by the U.S. Food and Drug Administration (FDA) in June 2017 for the treatment of acute bacterial skin and skin structure infections on the basis of data from two phase III trials. Delafloxacin is a broad-spectrum anionic fluoroquinolone and its distinct chemical structure increases its potency in acidic environments. It is known to inhibit DNA replication and repair by targeting DNA gyrase and topoisomerase IV. Delafloxacin is administered via both oral and parenteral routes. It has potent activity a

The effects of ibuprofen on activated sludge: Shift in bacterial community structure and resistance to ciprofloxacin.

Ibuprofen (IBP) is ranked at the 4th place among 57 pharmaceutical compounds according to the number of citations in prioritization documents. The response of microbial community of activated sludge to IBP was studied at the concentrations of 50-5000mg/L. Batch incubation was performed in an OxiTop® device for 21days. The reduction of biological oxygen demand depended on the IBP concentration and varied in the range from 321 to 107mg O2/L. Massive DNA sequencing analysis of the activated sludge revealed that Proteobacteria became more dominant when grown in the presence of IBP. Microbial diver

[Influence of methane and trichloroethylene domestication on bacterial community structure in landfill cover soil.].

The biological oxidation capacity and change of microbial community structure of typical landfill cover soil cultured in situ by methane and trichloroethylene (TCE) were studied. The rates of CH4 oxidation and TCE degradation were 0.20-0.87 Μmol·g-1 soil·h-1 and 0.009-0.013 mg·L-1·h-1 among these landfill cover soils, respectively. The activity of CH4 oxidation in Shandong landfill cover soil was higher than that in landfill cover soils sampled from Guangdong, Shanghai and Chongqing. The α-diversity and microbial community structure before and after co-metabolism degradation were then investig

Exploring structure and interactions of the bacterial adaptor protein YjbH by crosslinking mass spectrometry.

Adaptor proteins assist proteases in degrading specific proteins under appropriate conditions. The adaptor protein YjbH promotes the degradation of an important global transcriptional regulator Spx, which controls the expression of hundreds of genes and operons in response to thiol-specific oxidative stress in Bacillus subtilis. Under normal growth conditions, the transcription factor is bound to the adaptor protein and therefore degraded by the AAA+ protease ClpXP. If this binding is alleviated during stress, the transcription factor accumulates and turns on genes encoding stress-alleviating

Structure of a bacterial type III secretion system in contact with a host membrane in situ.

Many bacterial pathogens of animals and plants use a conserved type III secretion system (T3SS) to inject virulence effector proteins directly into eukaryotic cells to subvert host functions. Contact with host membranes is critical for T3SS activation, yet little is known about T3SS architecture in this state or the conformational changes that drive effector translocation. Here we use cryo-electron tomography and sub-tomogram averaging to derive the intact structure of the primordial Chlamydia trachomatis T3SS in the presence and absence of host membrane contact. Comparison of the averaged str

Current and emerging drugs for acute bacterial skin and skin structure infections: an update.

INTRODUCTION: Antibiotic resistance, the emergence of multidrug-resistant bacteria and the exponential rise of methicillin-resistant Staphylococcus aureus (MRSA) acute bacterial skin and skin structure infections (ABSSSI) pose a great threat to the public health. In order to combat these threats, development of new antibiotics is vital and measures to decrease antibiotic resistance should be prioritized. AREAS COVERED: This manuscript provides an overview of the current FDA-approved antibiotics and the antibiotics in development for MRSA ABSSSI. EXPERT OPINION: A great need exists currently

Structural alterations of faecal and mucosa-associated bacterial communities in irritable bowel syndrome.

Irritable bowel syndrome (IBS) is the most common functional gastrointestinal disorder in western countries. Previous studies on IBS, mostly based on faecal samples, suggest alterations in the intestinal microbiota. However, no consensus has been reached regarding the association between specific bacteria and IBS. We explore the alterations of intestinal bacterial communities in IBS using massive sequencing of amplified 16S rRNA genes. Mucosal biopsies of the ascending and descending colon and faeces from 16 IBS patients and 9 healthy controls were analysed. Strong inter-individual variation w

Crystal structure and specific binding mode of sisomicin to the bacterial ribosomal decoding site.

Sisomicin with an unsaturated sugar ring I displays better antibacterial activity than other structurally related aminoglycosides, such as gentamicin, tobramycin, and amikacin. In the present study, we have confirmed by X-ray analyses that the binding mode of sisomicin is basically similar but not identical to that of the related compounds having saturated ring I. A remarkable difference is found in the stacking interaction between ring I and G1491. While the typical saturated ring I with a chair conformation stacks on G1491 through CH/π interactions, the unsaturated ring I of sisomicin with a

Structure based discovery of small molecule suppressors targeting bacterial lysozyme inhibitors.

The production of lysozyme inhibitors, competitively binding to the lysozyme active site, is a bacterial strategy to prevent the lytic activity of host lysozymes. Therefore, suppression of the lysozyme-inhibitor interaction is an interesting new approach for drug development since restoration of the bacterial lysozyme sensitivity will support bacterial clearance from the infected sites. Using molecular modelling techniques the interaction of the Salmonella PliC inhibitor with c-type lysozyme was studied and a protein-protein interaction based pharmacophore model was created. This model was use

Liquid-free ion-conductive elastomers with bioinspired oriented structures based on bacterial cellulose for dual-mode humidity sensing and energy harvesting.

A detection system that integrates flexibility, dual-modal humidity sensing, and self-powering capability is crucial for real-time monitoring of human holistic health. However, the disordered ion transport pathway restricts the versatility and application range of liquid-free ion conductive elastomer (ICEs). This study prepared an ICE with a biomimetic-oriented structure using bacterial cellulose, in which bacterial cellulose serves as the orientation skeleton to provide a fast ion transport channel, thereby integrating bimodal humidity response and self-powering capability.
