Comparative genomics of the gut commensal Bifidobacterium bifidum reveals adaptation to carbohydrate utilization.

Bifidobacterium bifidum is one of the most abundant members of the gut microbiota at the early stage of life. The established association of the bacterium with the human gut confers health benefits. Such successful persistence of B. bifidum necessitates metabolic adaptation to the host-derived carbohydrates, a process which is poorly understood. The current study focuses on revealing the genomic-based phylogeny (phylogenomics) of B. bifidum and utilizing comparative genomics to decipher the glycolytic abilities of bifidobacterial strains isolated from different human body niches (feces, human

Probiotic Bifidobacterium animalis subsp. lactis consumption slows down orthodontic tooth movement in mice.

INTRODUCTION: Probiotics are live microorganisms that, when consumed in appropriate amount, can provide health benefits. Although many studies have shown positive results with the use of probiotics in bone loss control, as in periodontal disease, the effect of probiotics on a mechanical force-induced alveolar bone resorption is still unknown. Therefore, this study aimed to investigate the impact of the specific probiotic Bifidobacterium animalis subsp. lactis on bone remodeling induced by orthodontic tooth movement. METHODS: For this study, thirty C57BL6/J male mice were used and divided into

Bifidobacterium dentium Fortifies the Intestinal Mucus Layer via Autophagy and Calcium Signaling Pathways.

Much remains unknown about how the intestinal microbiome interfaces with the protective intestinal mucus layer. Bifidobacterium species colonize the intestinal mucus layer and can modulate mucus production by goblet cells. However, select Bifidobacterium strains can also degrade protective glycans on mucin proteins. We hypothesized that the human-derived species Bifidobacterium dentium would increase intestinal mucus synthesis and expulsion, without extensive degradation of mucin glycans. In silico data revealed that B. dentium lacked the enzymes necessary to extensively degrade mucin glycans.

Two membrane proteins from Bifidobacterium breve UCC2003 constitute an ABC-type multidrug transporter.

Intrinsic resistance to drugs is one of the main determining factors in bacterial survival in the intestinal ecosystem. This is mediated by, among others, multidrug resistance (MDR) transporters, membrane proteins which extrude noxious compounds with very different chemical structures and cellular targets. Two genes from Bifidobacterium breve encoding hypothetical membrane proteins with a high homology with members of the ATP-binding cassette (ABC) family of multidrug efflux transporters, were expressed separately and jointly in Lactococcus lactis. Cells co-expressing both proteins exhibited e

Restoration of Gut Microbiota Vertical Transmission by Bifidobacterium Mitigates Pollutant-Induced Intergenerational Obesity.

Mass production and widespread distribution of environmental pollutants enhance human susceptibility to disease, which may be related to gut microbial communities. However, the link between the transgenerational toxic effects of environmental pollutants and animal gut microbial communities remains obscure. Here, we show that environmental dose p,p'-DDE (a typical environmental pollutant) exposure prepregnancy reshapes offspring metabolic homeostasis through vertical transmission of the maternal gut microbiota, inducing the development of obesity. In addition, by taking cross-sectional temporal

Comparative transcriptional network analysis reveals ecological specialization in infant-associated species of Bifidobacterium.

UNLABELLED: Bifidobacterium species are important microorganisms in the infant gut microbiota displaying substantial genomic diversity, yet how this variability is reflected in transcriptional regulatory architecture remains an open question. Here, we integrated pangenome analysis with transcription factor binding site inference and network-based approaches to compare regulatory architectures across Bifidobacterium longum subsp. infantis, Bifidobacterium breve, and B. longum subsp. longum using over 300 high-quality genomes. All three species exhibited open pangenomes, with regulatory interact

Potential mechanisms of Bifidobacterium-mediated synergistic antibiotic eradication of drug-resistant Helicobacter pylori.

The rising prevalence of antibiotic-resistant Helicobacter pylori has become a major obstacle to the success of conventional eradication therapies, including triple and quadruple regimens combining antibiotics with proton pump inhibitors. There is an urgent need for safe and effective ad-junctive strategies to restore antibiotic efficacy. Probiotics, particularly Bifidobacterium species, have gained increasing attention for their potential to enhance H. pylori eradication and mitigate treatment-associated adverse effects. This review provides a comprehensive synthesis of the key molecular mech

WT1-Targeted Oral Bifidobacterium longum Vaccine Enhances Checkpoint Blockade Efficacy in Pancreatic Cancer.

Pancreatic ductal adenocarcinoma (PDAC) remains largely unresponsive to immune checkpoint inhibitors (ICI) due to its profoundly immunosuppressive tumor microenvironment. To overcome this resistance, we evaluated an oral WT1-targeted cancer vaccine using a recombinant Bifidobacterium longum strain (B. longum 420) engineered to display a partial WT1 antigen. Oral immunization with B. longum 420 activated WT1-associated cellular immunity, increasing IL-12-producing CD45+ cells and Th1/cytotoxic cytokine-producing T cells, and enhancing splenocyte cytotoxic activity against Pan02 cells. Early-Life Antibiotic Exposure Disrupts Bifidobacterium in Infants: A qPCR-Based Cohort Study.

BACKGROUND: Early infancy represents a critical window for establishing the gut microbiome, during which Bifidobacterium species dominate and play essential roles in metabolic, immune and intestinal maturation. Antibiotic exposure during this sensitive period may disrupt microbial development, yet quantitative data on its longitudinal impact remain limited. To evaluate the effect of systemic antibiotic exposure during the first 6 months of life on the abundance of Bifidobacterium using quantitative real-time polymerase chain reaction in a prospective infant cohort. Placental endocrine function is controlled by maternal gut Bifidobacterium in germ-free mice.

BACKGROUND: Recent studies have shown that the maternal gut microbiota can regulate placental growth, particularly the transport region, in association with fetal growth. However, the specific role of certain microorganisms in modulating the hormonal production of the placenta, which is critical for supporting fetal development and maintaining a healthy pregnancy, remains largely unexplored. In this context, the objective of this study is to determine whether the maternal colonisation with the early life gut bacterium Bifidobacterium breve UCC2003 regulates placental endocrine function. Microencapsulated Quercetin and Bifidobacterium animalis Independently Preserve Jejunal Enteric Neurons During Colorectal Carcinogenesis.

BACKGROUND: Colorectal cancer (CRC) is a leading cause of cancer-related deaths, significantly disrupting enteric neurotransmission within the colon. While the effects of CRC on the enteric nervous system (ENS) of the colon are well documented, its impact on the small intestine remains underexplored. This study aims to investigate the influence of colorectal carcinogenesis on the small intestine's ENS and evaluate the individual neuroprotective effects of microencapsulated quercetin and Bifidobacterium animalis. METHODS: Wistar rats were subjected to chemically induced colorectal carcinogenes

Therapeutic Effect of Bifidobacterium Administration on Experimental Autoimmune Myasthenia Gravis in Lewis Rats.

Beneficial effects of probiotics on gut microbiota homeostasis and inflammatory immune responses suggested the investigation of their potential clinical efficacy in experimental models of autoimmune diseases. Indeed, administration of two bifidobacteria and lactobacilli probiotic strains prevented disease manifestations in the Lewis rat model of Myasthenia Gravis (EAMG). Here, we demonstrate the clinical efficacy of therapeutic administration of vital bifidobacteria (i.e., from EAMG onset). The mechanisms involved in immunomodulation were investigated with ex vivo and in vitro experiments. Imp

Dietary Changes Involving Bifidobacterium longum and Other Nutrients Delays Chronic Kidney Disease Progression.

BACKGROUND: Recent studies suggest that prebiotic and/or probiotic treatments ameliorate kidney function in humans and animals by improving the gut environment. However, the gut microbiota and kidney disease interactions remain to be determined. This study investigated whether synbiotics modulate the gut microbiota and ameliorate kidney function using a rat model of chronic kidney disease (CKD). As uremic toxins are associated with CKD-related mineral and bone disorder, the secondary aim was to evaluate the relationship between synbiotics and secondary hyperparathyroidism (SHPT). METHODS: 5/6

Oral immunization of mice using Bifidobacterium longum expressing VP1 protein from enterovirus 71.

Bifidobacterium longum is an attractive candidate for delivering biologically active proteins by the mucosal route due to its non-pathogenic and colonizing properties. Enterovirus 71 (EV71) has aroused widespread attention recently due to several epidemics, and great attention should be paid to the fact that there are currently no effective antiviral drugs or vaccines against EV71 infection. In this report, we described a recombinant B. longum that could be used to develop an oral vaccine against EV71 infection. A VP1 expression vector (pBBADs-VP1) was constructed by amplifying the EV71 VP1 ge
