Associations between nucleus accumbens structural connectivity, brain function, and initiation of binge drinking.

Adolescent alcohol use is associated with increased risk for alcohol use disorders later in life; therefore, identifying biomarkers for initiation of heavy alcohol use, such as individual differences in the development of white-matter microstructure, may inform prevention strategies that improve public health. This prospective cohort study included 40 adolescents, ages 14 and 15, without substantial history of alcohol or drug use at baseline. Fractional anisotropy (FA), an index of white-matter microstructure, was assessed in pathways connecting the nucleus accumbens (NAcc) to the rest of the

Adolescent but not adult ethanol binge drinking modulates cocaine withdrawal symptoms in mice.

BACKGROUND: Ethanol (EtOH) binge drinking is an increasingly common behavior among teenagers that induces long-lasting neurobehavioral alterations in adulthood. An early history of EtOH abuse during adolescence is highly correlated with cocaine addiction in adulthood. Abstinence of cocaine abuse can cause psychiatric symptoms, such as anxiety, psychosis, depression, and cognitive impairments. This study assessed the consequences of adolescent exposure to EtOH on the behavioral alterations promoted by cocaine withdrawal in adulthood. METHODS: We pretreated juvenile (34-47 days old) or adult (6

Association of moderate alcohol use and binge drinking during pregnancy with neonatal health.

BACKGROUND: Heavy drinking and smoking during pregnancy are known to have a negative impact on the unborn child. However, the impact of low-to-moderate alcohol consumption and binge drinking has been debated recently. The aim of this study was to examine the relationship of moderate prenatal drinking and binge drinking with birthweight, being small for gestational age (SGA) at birth, preterm birth, and neonatal asphyxia. METHODS: Moderate alcohol drinking, binge drinking, and several possible confounders were assessed in 1,258 pregnant women; information on neonatal health was obtained at bir

Activation of σ-receptors induces binge-like drinking in Sardinian alcohol-preferring rats.

Sigma (σ) receptors have been implicated in the behavioral and motivational effects of alcohol and psychostimulants. Sigma receptor antagonists reduce the reinforcing effects of alcohol and excessive alcohol intake in both genetic (alcohol-preferring rats) and environmental (chronic alcohol-induced) models of alcoholism. The present study tested the hypothesis that pharmacological activation of σ-receptors facilitates ethanol reinforcement and induces excessive, binge-like ethanol intake. The effects of repeated subcutaneous treatment with the selective σ-receptor agonist 1,3-di-(2-tolyl)guani

College students' readiness to reduce binge drinking: criterion validity of a brief measure.

We assessed the criterion-related validity of a contemplation ladder used to determine college students' readiness to reduce binge drinking. 1356 students in halls of residence in Dunedin, New Zealand completed a self-report survey including the contemplation ladder, AUDIT questionnaire, and other alcohol-related measures. Two groups of binge drinkers were identified, one reporting bingeing more than twice per week (n=645), and one bingeing 1-2 times per week (n=237). A third group did not report binge drinking (n=474). A higher readiness to change binge drinking was associated with more frequ

Drug use, binge drinking and attempted suicide among homeless and potentially homeless youth.

In order to assess the need for drug-related services for at-risk youth, a survey was conducted among young people aged 12-17 years who, owing to severe family discord, were currently living away from home (homeless) or had experienced periods away from home in the past 12 months (potentially homeless). Prevalence of use and of potentially harmful levels of use of alcohol and other licit and illicit drugs were higher than in a comparative population. Of the 155 people interviewed, 54% reported past physical abuse, 28% reported past sexual abuse, and 73% had a family alcohol or other drug histo

Sex specific modulation of LC neuronal activity in a binge drinking mouse model.

Binge drinking substantially elevates the risk of developing alcohol use disorder, with pronounced sex-specific differences observed. However, there is a limited understanding of the underlying sex differences in the neural mechanisms that underpin binge drinking. The present study utilized the Drinking in the Dark (DID) paradigm, in combination with electrophysiology and chemogenetics, to reveal the critical role of the locus coeruleus (LC) in regulating sexually dimorphic binge-like drinking behavior. Electrophysiological analysis revealed that after three weeks of DID modeling, female mice

Factors Associated with Binge Drinking Among Brazilian Graduate Students: A Cross-Sectional Study.

BACKGROUND: Binge drinking is characterized by the excessive consumption of alcohol within a short period of time, resulting in several adverse effects on health and social life. Despite the relevance of this issue, few studies have specifically investigated this behavior among graduate students. This study identified factors associated with binge drinking among Brazilian graduate students. METHODS: A cross-sectional survey was conducted from May to July 2022 using an electronic questionnaire that assessed binge drinking, sociodemographic and academic characteristics, as well as physical and

Binge drinking and veteran status increase risk for suicide planning in U.S. adults.

OBJECTIVE: Suicide is a leading cause of preventable death. Problematic alcohol use and service in the U.S. military confer elevated risk for suicide, yet it is unknown whether their combination further heightens the risk. Extant literature is circumscribed to veterans enrolled in Veterans Affairs care, those with diagnosed alcohol use disorder, or suicide mortality, resulting in notable literature gaps on non-Veterans Affairs-enrolled veterans, nondiagnostic problematic drinking behaviors, and premortality suicide risk. METHOD: To address these gaps, this study included U.S. adults who parti

When the Going Gets Tough: Multimorbidity and Heavy and Binge Drinking Among Adults.

INTRODUCTION: Multimorbidity, the presence of two or more long-term health conditions in the same individual, is an emerging epidemic associated with increased morbidity and mortality. Continued drinking concurrent with alcohol-related chronic conditions, particularly with multimorbidity, is likely to further elevate health risk. This study aimed to examine the associations of multimorbidity among diabetes, hypertension, heart disease, and cancer with drinking, and moderation of these associations by age. METHODS: Logistic regression modeling was performed in 2023 using a nationally represent

Somatostatin signaling modulates binge drinking behavior via the central nucleus of the amygdala.

Somatostatin (SST) is a neuropeptide widely expressed in the central nervous system with dense expression in limbic regions such as the extended amygdala. It has recently gained attention for playing a role in modulating alcohol use disorders and co-morbid neuropsychiatric disorders. However, the role of SST in the central nucleus of the amygdala (CeA), a key region for neuropeptide regulation of alcohol and anxiety related behaviors, in alcohol consumption has not been assessed. In this work we perform an initial examination of the interaction between the CeA SST system and binge ethanol inta

Binge drinking induces an acute burst of markers of hepatic fibrogenesis (PRO-C3).

BACKGROUND & AIMS: Binge drinking is associated with an increased risk of liver disease. Morbidity and mortality of alcohol-related liver disease (ALD) is associated with collagen deposition in the hepatic extracellular matrix (ECM). However, the acute effects of binge drinking on ECM turnover are unknown. We aimed to investigate the effects on hepatic ECM turnover following a binge drinking episode. METHODS: We performed a pathophysiological intervention study with 15 non-alcoholic fatty liver disease (NAFLD) patients, 15 ALD patients and 10 healthy controls. We used 40% ethanol in 9 mg/mL N

Tetracycline derivatives reduce binge alcohol consumption in High Drinking in the Dark mice.

Alcohol use disorders (AUDs) are prevalent, and are characterized by binge-like drinking, defined by patterns of focused drinking where dosages ingested in 2-4 h reach intoxicating blood alcohol levels (BALs). Current medications are few and compliance with the relatively rare prescribed usage is low. Hence, novel and more effective medications are needed. We developed a mouse model of genetic risk for binge drinking (HDID: High Drinking in the Dark mice) by selectively breeding for high BALs after binge drinking. A transcriptional analysis of HDID brain tissue with RNA-Seq implicated neuroin

Pharmacogenetic Manipulation of the Nucleus Accumbens Alters Binge-Like Alcohol Drinking in Mice.

BACKGROUND: Chronic alcohol intake leads to long-lasting changes in reward- and stress-related neuronal circuitry. The nucleus accumbens (NAc) is an integral component of this circuitry. Here, we investigate the effects of DREADDs (Designer Receptors Exclusively Activated by Designer Drugs) on neuronal activity in the NAc and binge-like drinking. METHODS: C57BL/6J mice were stereotaxically injected with AAV2 hSyn-HA hM3Dq, -hM4Di, or -eGFP bilaterally into NAc [core + shell, core or shell]. We measured clozapine-n-oxide (CNO)-induced changes in NAc activity and assessed binge-like ethanol (Et

What colleges are doing about student binge drinking. A survey of college administrators.

In 1999, the Harvard School of Public Health College Alcohol Study surveyed 734 US college administrators to learn what colleges were doing to prevent binge drinking. Respondents rated the severity of student alcohol-abuse problems and described prevention efforts and institutional investments in prevention infrastructure. Prevention practices were widespread in the areas of general education about alcohol, use of policy controls to limit access to alcohol, restricting advertising at home-game sporting events, and allocation of living space to alcohol-free dormitories. Programming was less pre

Aspirin pretreatment reduces ethanol withdrawal severity in a mouse model of binge drinking.

Nonsteroidal antiinflammatory drugs (NSAIDs) such as aspirin, ibuprofen, and indomethacin, which inhibit prostaglandin (PG) synthesis, have a pronounced effect on a broad range of ethanol (EtOH) actions. Given this, it is somewhat surprising that NSAID treatment has not been found to alter major signs of ethanol withdrawal. To date, the only effect found has been indirect, that is, NSAID treatment reduces the efficacy of PG precursor administration in the treatment of ethanol withdrawal via the inhibition of PG formation. However, in those studies reporting negative results NSAID administratio

Greater inhibition of female rat binge alcohol intake by adrenergic receptor blockers using a novel Two-Shot rat binge drinking model.

Binge drinking (BD) contributes strongly to the harms of alcohol use disorder. Most rodent models do not result in binge-level blood alcohol concentrations (BACs), and to better understand individual and sex differences in neurobiological mechanisms related to BD, the use of outbred rat strains would be valuable. Here, we developed a novel BD model where after 3+ months of intermittent access to 20% alcohol Wistar rats drank, twice a week, with two 5-minute intake (what we called Two-shot) separated by a 10-minute break. Our findings showed during Two-Shot that most animals reached ≥ 80mg% BAC

Alcohol consumption during binge-eating episodes and associations with drinking patterns and alcohol problems among adults with binge-spectrum eating disorders.

Individuals with binge-spectrum eating disorders (BSEDs) clinically report drinking alcohol during binge-eating episodes, but no empirical research has yet been conducted. Drinking alcohol during binge-eating episodes may exacerbate overconsumption of food and feelings of guilt or sadness in response to binge eating. Characterizing this co-occurring behavior is therefore an important first step and contribution to knowledge about a potential contributor to binge eating. The current study aimed to characterize alcohol use during binge-eating episodes among adults with BSEDs and determine the re

Cross-Sensitization between Binge Eating and Binge Drinking in a Novel C57BL/6NJ Murine Model of Disease Comorbidity Requires PDE4B Activation.

There is a high rate of comorbidity between binge eating (BE) and binge drinking (BD) behaviors, suggesting a common neuropathology. Recently, phosphodiesterase 4B (PDE4B) was identified as a pleiotropic gene associated with comorbid alcohol use disorder (AUD) and anorexia nervosa with BE in a genome-wide association study, implicating PDE4B as a potential contributor to shared genetic risk between these disorders. Here, we developed a novel mouse model of comorbid BE and BD in C57BL/6NJ mice in which mice underwent 10 d of BE, followed by 10 d of BD. Females exhibited cross-sensitization from
