BACKGROUND: Human Bocavirus 2 (HBoV2) is an emerging enteric virus frequently detected in wastewater, yet its environmental fate, persistence, and treatment remain poorly understood. Improved characterization of HBoV2 is important for advancing wastewater-based epidemiology and understanding viral transmission dynamics. METHODS AND RESULTS: A year-long wastewater surveillance study was conducted from December 2023 to December 2024 in a small urban wastewater treatment system. Viral concentrations were quantified using digital PCR (dPCR), decay kinetics were assessed using controlled mesocosm e

IntroductionDromedary camel bocavirus (DBoV) poses a threat to camel health, yet no vaccine is available.MethodsUsing immunoinformatics, B-cell, CTL, and HTL epitopes from VP1/VP2 were predicted. Four multi-epitope constructs (DBoV-A1 to A4) were designed with different adjuvants and evaluated in silico; DBoV-A2 and A4 were expressed, purified, and tested in BALB/c mice.ResultsDBoV-A2 and A4 showed strong in silico binding to TLR9 and stable molecular dynamics. In mice, both induced significant IgG, IgM, IgA responses, Th1/Th2 cytokine secretion, and CD4⁺ T cell expansion without organ toxicit

Increasing evidence suggests that human bocavirus (HBoV) is associated with respiratory symptoms in the absence of other identifiable pathogens and may even precipitate severe lower respiratory tract infections. However, only a few studies of severe human bocavirus infections in pediatric patients have been reported. The aim of the current study was to collect and analyze clinical data from children diagnosed with mild pneumonia and severe pneumonia, with an emphasis on those testing positive for HBoV from June 2009 to June 2019. Among the 799 HBoV-positive children included in the study, appr

In this study, we present four cases of Human Bocavirus (HBoV) infection in children aged between 1 month and 4 years. Among these cases, two siblings were hospitalized with similar symptoms. Among the four pediatric cases of patients with HBoV infection, three were associated with acute respiratory failure and spontaneous pneumothorax, and two of these presented with subcutaneous emphysema. The presented patients were young children, aged between 1 month and 4 years, two of whom were siblings, suggesting a possible intrafamilial transmission of HBoV1 infection. These cases highlight the impor

A toddler, thriving well, developmentally normal, and fully immunized, presented with fever, cough, and cold for a day, followed by breathing difficulty. Although the child was not ill upon admission, he had a fever and was breathing rapidly. On examination, visible sub-costal retractions and wheezing in both lungs were noted. He required Intensive Care Unit (ICU) management for a brief period, with oxygen supplementation, round-the-clock nebulization, and other supportive care. Initially, he was diagnosed with a wheeze-associated lower respiratory tract infection, as his chest X-ray showed bi

Human bocavirus (HBoV) 1 is considered an important pathogen that mainly affects infants aged 6-24 months, but preventing viral transmission in resource-limited regions through rapid and affordable on-site diagnosis of individuals with early infection of HBoV1 remains somewhat challenging. Herein, we present a novel faster, lower cost, reliable method for the detection of HBoV1, which integrates a recombinase polymerase amplification (RPA) assay with the CRISPR/Cas12a system, designated the RPA-Cas12a-fluorescence assay. The RPA-Cas12a-fluorescence system can specifically detect target gene le

Pseudomembranous laryngotracheobronchitis is rarely reported yet potentially life-threatening infectious cause of airway obstruction in children. The causative organisms of this condition are often considered to promote bacterial superinfection following viral infection. We report a case of pseudomembranous laryngotracheobronchitis in a patient caused by human bocavirus 1 and Mycoplasma pneumoniae (M. pneumoniae). A 2-year-old child was admitted to our hospital presenting with cough, hoarseness, and labored breathing. Computed tomography of the chest revealed atelectasis of the right middle lo

Human bocavirus 1 (HBoV1), a nonenveloped single-stranded DNA parvovirus, causes mild to life-threatening respiratory tract infections, acute otitis media, and encephalitis in young children. HBoV1 often persists in nasopharyngeal secretions for months, hampering diagnosis. It has also been shown to persist in pediatric palatine and adenoid tonsils, which suggests that lymphoid organs are reservoirs for virus spread; however, the tissue site and host cells remain unknown. Our aim was to determine, in healthy nonviremic children with preexisting HBoV1 immunity, the adenotonsillar persistence si

Human bocavirus (HBoV) is a novel parvovirus, often associated with respiratory tract diseases in children. This study explored the epidemiological characteristics and molecular evolution of HBoV-1 in southeastern China. Nasopharyngeal aspirates were collected from children admitted to hospital with acute respiratory tract infections. HBoV-1 was detected using real-time reverse transcription polymerase chain reaction and further characterized by complete genome sequences analysis. Among the 3,022 recruited children, 386 (12.77%) were HBoV-1-positive and 300 (77.72%) had co-detection with other

Wild boar populations are increasingly recognized as important hosts in the ecology of swine viruses, yet data from Croatia remain limited. This study aimed to establish baseline information on the presence of porcine bocavirus (PBoV) and porcine cytomegalovirus (PCMV) in Croatian wild boar within the framework of the national African swine fever (ASF) surveillance program. Spleen and blood samples from 184 ASF-negative wild boar collected across 11 counties were tested using real-time PCR. PCMV DNA was detected in 16 animals (8.69%), with similar detection frequencies in spleen (7.69%) and bl

Plastic bronchitis (PB) is a rare and severe respiratory disease characterized by the formation of branching mucus casts, resulting in airway obstruction. PB can be divided into two types. Type 1 PB is primarily caused by inflammatory casts that result from allergic diseases. In type 2 PB, mucinous casts are produced in association with congenital heart disease. PB is also associated with viral respiratory infections, particularly the influenza A (H1N1) pdm09 virus, which is the most common pathogen affecting pediatric patients. Herein, we report a case of severe type 1 PB caused by human boca

Human bocavirus 1 (HBoV1) infects well-differentiated (polarized) human airway epithelium (HAE) cultured at an air-liquid interface (ALI). In the present study, we applied next-generation RNA sequencing to investigate the genome-wide transcription profile of HBoV1, including viral mRNA and small RNA transcripts, in HBoV1-infected HAE cells. We identified novel transcription start and termination sites and confirmed the previously identified splicing events. Importantly, an additional proximal polyadenylation site (pA)p2 and a new distal polyadenylation site (pA)dREH lying on the right-hand hai

Wastewater-based epidemiology (WBE) represents a valuable tool for assessing viral circulation at the community level. Human bocavirus (HBoV), a member of the Parvoviridae family with four genotypes (HBoV1-4), has been detected in respiratory and enteric samples, although its environmental epidemiology remains poorly characterized. This study aimed to investigate the presence and diversity of HBoV in wastewater samples from Córdoba, Argentina, during 2020-2021. A total of 84 raw sewage samples collected at the city's wastewater treatment plant were analyzed by nested PCR and sequencing of the
