A single meal elicits regional changes in bombesin-like peptide levels in the gut and brain.

Several studies have demonstrated that exogenous bombesin (BN) elicits a potent satiety effect when administered centrally or systemically. It has been suggested that BN-like peptides may play a physiological role in the control of food intake. The objective of the present study was to determine whether the levels of endogenous BN-like peptides change in response to a meal. All rats were food deprived overnight for a 12 h period. Half the animals were then allowed to feed for 35 min (postprandial group) and the remainder formed the preprandial group. Regions of the brain (hypothalamus, cerebel

Differential effect of bombesin on intraluminal and intravascular release of gastric gastrin and somatostatin in anaesthetized rats.

The aim of the present investigation was to study how bombesin, gastrin-17, cholecystokinin-8 (CCK-8) and electrical vagal stimulation influence the release of gastrin and somatostatin into the gastric lumen. Bombesin (3 and 30 nmol kg-1 h-1), gastrin-17 and CCK-8 (10 nmol kg-1h-1) were infused i.v. and vagal stimulations at 5 V, 2 ms, 5 Hz were performed in anaesthetized rats, in which the stomach was perfused with a dextran solution (pH approximately 6 or approximately 1.5). pH, gastrin and somatostatin levels were measured in the perfusate after having passed the stomach. In addition, blood

Identification of a receptor for peptides of the bombesin family in Swiss 3T3 cells by affinity cross-linking.

The cross-linking agent ethylene glycol-bis(succinimidyl succinate) was used to covalently link 125I-labeled gastrin releasing peptide (125I-GRP) to an Mr 75,000-85,000 surface protein in Swiss 3T3 cells that displays many characteristics of a specific receptor for peptides of the bombesin family. This protein was not present in other cell lines which do not exhibit receptors for bombesin-like peptides. Unlabeled GRP competed for affinity labeling of the Mr 75,000-85,000 protein in a concentration-dependent manner, and other bombesin-related peptides also inhibited the cross-linking of 125I-GR

Effect of atropine and somatostatin on bombesin-stimulated plasma gastrin, cholecystokinin and pancreatic polypeptide in man.

Infusion of the neuropeptide bombesin stimulates the secretion of several gastrointestinal hormones by an unknown mechanism. We have investigated the effects of atropine (15 ng/kg as bolus followed by 2.5 ng/kg X 30 min) and somatostatin (125 micrograms as i.v. bolus followed by 62.5 micrograms/30 min) on the stimulation of 3 hormones (gastrin, cholecystokinin and pancreatic polypeptide) by 60 pmol/kg X 20 min bombesin in 6 healthy volunteers. Plasma samples for measurement of hormones by sensitive and specific radioimmunoassays were obtained at -5, 0, 2.5, 5, 7.5, 10, 15, 20, 25 and 30 min. B

Intracisternal bombesin induced inhibition of gastric secretion is not mediated through prostaglandin or opioid pathways.

Neuromedin B, a mammalian decapeptide which shares homology with the carboxyl terminal fragment of ranatensin R and bombesin, injected intracisternally (880 pmol) did not modify gastric acid secretion stimulated either by pylorus ligation in conscious rats or by pentagastrin infusion in urethane-anesthetized rats. Intracisternal bombesin (300 pmol) further enhanced PGE2-induced inhibition of gastric acid secretion and suppressed prostaglandin-induced stimulation of gastric secretory volume. Gastric response to intracisternal bombesin was not modified by indomethacin (10 mg/kg) or naloxone pret

Effects of inhibitors of cyclic nucleotide phosphodiesterase on actions of cholecystokinin, bombesin, and carbachol on pancreatic acini.

In dispersed acini from guinea pig pancreas two inhibitors of cyclic nucleotide phosphodiesterase, Ro 20-1724 and 3-isobutyl-1-methylxanthine (IBMX), augmented the increase in amylase secretion caused by supramaximal concentrations of cholecystokinin but did not alter the stimulation of enzyme secretion caused by bombesin. The augmentations of the action of cholecystokinin caused by Ro 20-1724 or IBMX could be reproduced by 8-bromo-cAMP. When tested alone or with theophylline, cholecystokinin did not alter cAMP in pancreatic acini; however, with Ro 20-1724 or IBMX, concentrations of cholecysto

Activation of Bombesin Receptor Subtype-3 Promotes Antigen-Presenting Action in Human Bronchial Epithelial Cells.

BACKGROUND: Bombesin receptor subtype-3 (BRS-3) is a member of the bombesin-like peptide receptor family. Our previous studies demonstrated that activation of the human BRS-3 plays a protective role in oxidation-injured human bronchial epithelial cells (HBEC). The present study was designed to determine the role of BRS-3 activation in the antigen-presenting action of HBEC and the corresponding proliferation and differentiation of T cells. MATERIALS AND METHODS: In vivo, an asthma animal model was established and the expression and distribution of BRS-3 were analyzed by immunocytochemistry. In

BB2 bombesin receptor-expressing spinal neurons transmit herpes-associated itch by BB2 receptor-independent signaling.

Although spinal neurons expressing BB2 bombesin receptors are suggested to be involved in itch transmission, their role in pathological itch remains unknown. Because itch is often observed in patients with herpes zoster, we examined the role of BB2 receptor-expressing spinal neurons in herpes-associated itch in mice. Transdermal inoculation of human herpes virus 1 on the midflank produced herpes zoster-like skin lesions and caused the mice to scratch (itch-related behavior) and lick (pain-related behavior) the affected skin. Ablation of BB2 receptor-expressing spinal neurons by intrathecal tre

Abnormal expression of pulmonary bombesin-like peptide immunostaining cells in infants with congenital diaphragmatic hernia.

Infants with congenital diaphragmatic hernia (CDH) have a high neonatal mortality and morbidity owing to lung hypoplasia and persistent pulmonary hypertension. Pulmonary neuroendocrine cells produce bombesin-like peptide (BLP), a peptide with growth factor-like properties involved in lung development. We examined the expression of BLP immunostaining in pulmonary neuroendocrine cells (PNEC), and in clusters of these cells called neuroepithelial bodies (NEB), in the lungs of three groups of infants: patients with CDH, newborns with lung hypoplasia due to other causes, and control subjects withou

Targeted chemotherapy with cytotoxic bombesin analogue AN-215 inhibits growth of experimental human prostate cancers.

We developed a powerful cytotoxic analogue of bombesin AN-215, in which the bombesin (BN)-like carrier peptide is conjugated to 2-pyrrolino doxorubicin (AN-201). Human prostate cancers express high levels of receptors for BN/gastrin releasing peptide (GRP) that can be used for targeted chemotherapy. The effects of targeted chemotherapy with cytotoxic BN analogue AN-215 were evaluated in nude mice bearing subcutaneous xenografts of DU-145, LuCaP-35, MDA-PCa-2b and intraosseous implants of C4-2 human prostate cancers. Intraosseous growth of C4-2 tumors was monitored by serum PSA. BN/GRP receptor

Signal transduction events involved in bombesin-stimulated gastrin release from human G cells in culture.

The mechanism of action of bombesin on human antral gastrin cells in culture was evaluated by modulating internal and external calcium levels and intracellular enzyme activities. Increasing extracellular calcium increased basal gastrin release and had an additive effect on bombesin-stimulated gastrin release. Removing extracellular calcium had no effect on bombesin-stimulated gastrin release. Inhibiting the activities of phospholipase C by neomycin and protein kinase C by staurosporine had no effect on basal release but decreased bombesin-stimulated gastrin release by up to 50%. Gastric acid secretion, intraduodenal pH and gastroenteropancreatic hormone release to bombesin in antrocolic transposition dogs.

The effects of bombesin on gastric acid secretion from the Heidenhain pouch (HP), intraduodenal pH and gastroenteropancreatic (GEP) hormone levels were compared between 4 dogs with antrocolic transposition (ACT) and 4 non-ACT dogs. Bombesin, infused i.v. at a dose of 0.5 microgram/kg for one hr, produced a significant increase in gastric acid secretion from HP and a decrease in intraduodenal pH in both groups. However, these changes were more marked in ACT dogs. The intravenous infusion of bombesin also produced, in both groups, an increase in the plasma levels of gastrin, secretin, motilin, i

Bombesin-, calcitonin-, and serotonin-immunoreactive pulmonary neuroendocrine cells in acute and chronic neonatal lung disease.

Bombesin-, calcitonin-, and serotonin-immunoreactive pulmonary neuroendocrine cells (PNEC) were quantitated in the lungs of infants dying of hyaline membrane disease (HMD) and bronchopulmonary dysplasia (BPD). When compared with the lungs of age-matched control infants dying of noncardiopulmonary causes, infants who had HMD demonstrated a decrease in the numbers of bombesin-, calcitonin-, and serotonin-immunoreactive PNECs, while infants who had BPD demonstrated an increase. This difference was most pronounced at 2 months of age, when infants with BPD had a threefold increase in bombesin-, a t

Biodistribution of the Bombesin Receptor-Targeted Radiopharmaceutical Precursor BBN/C1-C2 in a Prostate Cancer Model.

UNLABELLED: Gastrin-releasing peptide receptor (GRPR) is a G protein-coupled receptor expressed in the central nervous system, the gastrointestinal tract, the pancreas, and the adrenal cortical tissue regulating their physiological functions. In addition to normal tissues, GRPR is overexpressed in many solid cancers. At present, several radiolabeled ligands targeting GRPR have been introduced into clinical practice for cancer diagnostics and radioligand therapy. However, there were found the high uptake of radiopharmaceuticals in normal organs and low bioavailability of the drugs caused by the

Development of bombesin-like and histamine-like innervation in the bullfrog (Rana catesbeiana) central nervous system.

Amphibians rely exclusively on behavioral thermoregulation to maintain body temperature within species- and developmental stage-specific critical limits. Several members of the bombesin family of peptides and histamine are included in a class of neurochemicals that have potent thermoregulatory effects in ectothermic and endothermic vertebrate species and may be involved in behavioral thermoregulation in amphibians. Because amphibians respond to environmental temperature cues differently in larval versus adult animals, we used immunocytochemistry to study developmental changes in bombesin-like

Promoting effect of bombesin on the cell proliferation in the rat endocrine pancreas during the early postnatal period.

The present work investigated whether orally administered bombesin influences cell proliferation in the endocrine pancreatic islets of rats during the suckling period and after weaning. Four series of pups were given bombesin diluted in milk (20 micrograms/kg, 3 times daily) or milk alone for 5 days during either the first, second, third or fourth postnatal week of life. Oral administration was used because bombesin-like peptides have been identified in the breast milk of mammals. 45 min before death, animals were given a single [3H]thymidine pulse injection. Plasma gastrin responses to bombesin and antral gastrin concentrations in patients with the intestinal type of gastric cancer.

Mucosal atrophy of the gastric antrum (type B atrophic gastritis) is generally accepted as predisposing to the development of the intestinal type of gastric cancer. Since bombesin stimulates gastrin release selectively from the antral mucosa, the response can be used as a marker for antral mucosal atrophy. In this study we have investigated bombesin-stimulated plasma gastrin responses in 21 patients with the intestinal type of gastric cancer and we have compared the results with 12 patients with the diffuse type of gastric cancer, 17 patients with benign gastric ulcer, and 30 dyspeptic patient
