Nerol induces mitochondrial apoptosis and autophagy in Botrytis cinerea and its protection on postharvest cherry tomato.

Botrytis cinerea, the second prevalent fungal pathogen worldwide, threatens a wide range of plants through its aggressive gray mold infection. This study investigates the antifungal effect of nerol against B. cinerea. Its minimum inhibitory concentration is 0.25 mL/L. Colony diameter reduced by 88.86 % at 48 h. Nerol exposure induced ROS accumulation, increasing malondialdehyde levels, which disrupted membrane integrity. It further induced mitochondrial dysfunction by suppressing the enzyme activities of succinic dehydrogenase and citrate synthase, reducing membrane potential, and promoting cy

Jasmonic acid and nitric oxide orchestrate a hierarchical melatonin cascade for Botrytis cinerea resistance in tomato.

Tomato gray mold, caused by Botrytis cinerea (B. cinerea), poses a major challenge to tomato production. In plants, B. cinerea resistance is positively regulated by melatonin; however, the regulatory mechanism of melatonin biosynthesis during B. cinerea infection is not known. Here, we established the working model of jasmonic acid (JA) and nitric oxide (NO) on melatonin biosynthesis in the state of B. cinerea infection. NO responded to B. cinerea infection earlier than JA. In the early stage of infection, tomato caffeic acid O-methyltransferase 2 (SlCOMT2) was S-nitrosylated by NO at Cys344,

Ultra-Structural Alterations in Botrytis cinerea-The Causal Agent of Gray Mold-Treated with Salt Solutions.

Potassium bicarbonate (PB), calcium chelate (CCh), and sodium silicate (SSi) have been extensively used as antifungal generally recognized as safe (GRAS) compounds against plant pathogenic fungi. In this research, in in vitro tests, the radial growth, conidial germination, and germ tube elongation of Botrytis cinerea was completely inhibited at 0.3% of PB, SSi, and CCh. In in vivo tests, application of PB, SSi, and CCh completely inhibited the occurrence of gray mold incidence of inoculated 'Italia' grape berries at concentrations of 1.0, 0.8, and 0.8%, respectively. In order to investigate th

Observations on the ultrastructure of the choanoflagellate Codosiga botrytis (Ehr.) Saville-Kent with special reference to the flagellar apparatus.

The ultrastructure of the choanoflagellate Codosiga botrytis is described with particular reference to the flagellar appendages, the flagellar rootlet system, the transition zone, the basal body and accessory centrioles, and the stalk. The controversial early reports of flagellar appendages in this species have been confirmed and they have been detected in 2 further species, Salpingoeca frequentissima and Monosiga sp. The appendages consist of a delicate bilateral vane 2 mum wide on either side of the axis, composed of extremely fine overlapping or interwoven fibrils. The flagellar root system

Oligomycin-producing Streptomyces sp. newly isolated from Swiss soils efficiently protect Arabidopsis thaliana against Botrytis cinerea.

UNLABELLED: Botrytis cinerea is a necrotrophic phytopathogen able to attack more than 200 different plant species causing strong yield losses worldwide. Many synthetic fungicides have been developed to control this disease, resulting in the rise of fungicide-resistance B. cinerea strains. The aim of this study was to identify Streptomyces strains showing antagonistic activity against B. cinerea to contribute to plant protection in an environmentally friendly way. We isolated 15 Actinomycete strains from 9 different Swiss soils. The culture filtrates of three isolates showing antifungal activit

Detoxification Mechanism of 8,8-Dimethyl-3-[(R-phenyl)amino]-1,4,5(8H)-naphthalentrione Derivatives by Botrytis cinerea.

The effect of 8,8-dimethyl-3-[(R-phenyl)amino]-1,4,5(8H)-naphthalentrione derivatives (compounds 1⁻13) on the mycelial growth of Botrytis cinerea was evaluated. The fungitoxic effect depended on the substituent and its position in the aromatic ring. Compounds substituted with halogens in meta and/or para positions (compounds 3, 4, 5 and 7), methyl (compounds 8 and 9), methoxyl (compounds 10 and 11), or ethoxy-carbonyl groups (compound 12) presented higher antifungal activity than compound 1, which had an unsubstituted aromatic ring. In addition, compounds with halogens in the ortho position, s

Design and Synthesis of Geranylated Benzaldehydes: Evaluation of Antifungal Activities Against Botrytis cinerea and Phytophthora cinnamomi.

A series of ten geranylated benzaldehydes (11-20) was designed, synthesized via Suzuki cross-coupling for the C-C series and SN2 substitution for the C-O derivatives, and structurally characterized. The antifungal activity of all compounds against the phytopathogens Botrytis cinerea and Phytophthora cinnamomi, two important plant pathogens associated with substantial agricultural and economic losses worldwide, was evaluated in vitro using a mycelial growth inhibition assay. Dose-response treatment of the data indicates that (E)-2-((3,7-dimethylocta-2,6-dien-1-yl)oxy)-5-hydroxybenzaldehyde (19)

LaZAT9 and LaZAT10 confer resistance to Botrytis elliptica in lily by modulating ROS via glutathione metabolism.

Gray mold caused by Botrytis elliptica (B. elliptica) is a devastating disease in lily production. It is essential to identify resistant germplasm and elucidate the underlying resistance mechanisms for effective disease control in lily. In this study, we evaluated the resistance of different lily germplasms to gray mold and selected the highly resistant Lilium 'Golden Stone' (LGS) and the highly susceptible Lilium leichtlinii var. maximowiczii (LLM). After infection with B. elliptica, LGS exhibited significantly increased antioxidant enzyme activities and marked enrichment of the glutathione m

Fungicide Sensitivity Profiles of Botrytis Isolates from Blueberry and Red Raspberry Fields in Washington and Oregon.

Botrytis spp. cause major economic losses for small fruit growers in Washington and Oregon, the top blueberry- and red raspberry-producing states in the United States. Pre- and postharvest fruit rots are conventionally managed through repeated within-season applications of fungicides from various Fungicide Resistance Action Committee (FRAC) groups. As such, monitoring fungicide resistance profiles of Botrytis spp. from small fruit fields is crucial. To this end, in 2022 and 2023, a total of 708 and 305 Botrytis isolates were obtained from blueberry (WA and OR) and red raspberry (WA) fields, re

A Novel Phenamacril Derivative Targeting Myosin I for the Control of Gray Mold Caused by Botrytis cinerea.

Gray mold caused by Botrytis cinerea severely impacts global crop productivity. In this study, we identified B33 [(Z)-3-amino-N-(3-chloro-4-fluorobenzyl)-2-cyano-3-phenylacrylamide], a novel phenamacril derivative bearing a distinctive substitution pattern where the ester group is replaced by an amino group. As an unprecedented small-molecule inhibitor targeting B. cinerea myosin I (BcMyoI), it represents an unexplored antifungal mode of action for gray mold disease management. Combined assays including phenotypic screening, ATPase inhibition, and microscale thermophoresis (MST) confirmed that

Loss of virulence of Botrytis cinerea mutants defective in phytotoxin production is restored by modifying inoculation medium.

Botrydial, botcinic acid, and their derivatives are major phytotoxic metabolites produced by the necrotrophic fungal pathogen Botrytis cinerea. These phytotoxins are able to induce programmed cell death in the host and thereby promote plant susceptibility to B. cinerea. We observed that a ∆bot2∆boa6 double-mutant strain, which synthesizes neither botrydial nor botcinic acid, was almost avirulent on tomato leaves when the disease assay was performed using synthetic minimal Gamborg B5 medium. However, the virulence of this mutant was restored when the inoculation medium was supplemented with yea

Sensitivity of Botrytis cinerea From Nectarine/Cherry in China to Six Fungicides and Characterization of Resistant Isolates.

Botrytis cinerea, the causal agent of gray mold, can result in considerable preharvest and postharvest losses in many economically valuable plant species. Fungicides were widely used to minimize such losses, but fungicide resistances were detected frequently. In the present study, we collected 164 isolates from nectarine and cherry in China and tested the sensitivity to six fungicides. Among the tested isolates, 71 (43.3%) were resistant to azoxystrobin, 14 (8.5%) to cyprodinil, 7 (4.3%) to boscalid, 4 (2.4%) to carbendazim, 1 (0.6%) to iprodione, and no isolates were found to be resistant to

Arabidopsis WRKY33 is a key transcriptional regulator of hormonal and metabolic responses toward Botrytis cinerea infection.

The Arabidopsis (Arabidopsis thaliana) transcription factor WRKY33 is essential for defense toward the necrotrophic fungus Botrytis cinerea. Here, we aimed at identifying early transcriptional responses mediated by WRKY33. Global expression profiling on susceptible wrky33 and resistant wild-type plants uncovered massive differential transcriptional reprogramming upon B. cinerea infection. Subsequent detailed kinetic analyses revealed that loss of WRKY33 function results in inappropriate activation of the salicylic acid (SA)-related host response and elevated SA levels post infection and in the

Modulators of membrane drug transporters potentiate the activity of the DMI fungicide oxpoconazole against Botrytis cinerea.

Modulators known to reduce multidrug resistance in tumour cells were tested for their potency to synergize the fungitoxic activity of the fungicide oxpoconazole, a sterol demethylation inhibitor (DMI), against Botrytis cinerea Pers. Chlorpromazine, a phenothiazine compound known as a calmodulin antagonist, appeared the most potent compound. Tacrolimus, a macrolide compound with immunosuppressive activity, was also active. The synergism of chlorpromazine negatively correlated with the sensitivity of the parent strain and mutants of B. cinerea. The synergism was highest in a mutant that overexpr

The Antagonistic Effect of 2-Phenylethanol Produced by Serratia liquefaciens Strain JS-239 Against Botrytis cinerea.

Botrytis cinerea is a necrotrophic fungal pathogen responsible for gray mold diseases in economically significant crops. Although conventional fungicides are commonly used to control these diseases, they pose environmental hazards, such as soil and water contamination, harm to nontarget organisms, and ecosystem disruption. Additionally, overuse promotes the development of fungicide-resistant strains, reducing their efficacy. Consequently, finding sustainable alternatives is essential for agriculture. In this study, strain JS-239 was isolated from the Northern Unit of Kettle Moraine State Fores

Identification of Volatile Organic Compounds as Natural Antifungal Agents Against Botrytis cinerea in Grape-Based Systems.

Botrytis cinerea Pers., the causal agent of grey mould, causes major economic losses in viticulture by reducing grape and wine quality and yield. Antagonistic yeasts that release bioactive volatile organic compounds (VOCs) represent a sustainable alternative to synthetic fungicides. Here, VOCs produced by Pichia guilliermondii strain ZIM624 were identified and assessed for antifungal activity against B. cinerea. 65 VOCs-including higher alcohols, volatile phenols, esters, and terpenes-were detected using two newly developed and validated analytical methods combining automated headspace solid-p

Armadillo repeat only gene VlARO confers resistance to Botrytis cinerea by promoting guaijaverin accumulation in grapevine.

The ARM repeat only gene VlARO positively regulates grape resistance to gray mold by promoting the guaijaverin accumulation, which directly suppresses the mycelial growth of Botrytis cinerea. Gray mold, caused by the necrotrophic fungus Botrytis cinerea, severely threatens global grapevine production. Here, we identified VlARO, a nuclear-localized gene from the highly resistant grapevine variety 'Beta', as a positive regulator of gray mold resistance. Although VlARO contains Armadillo repeats, it lacks a canonical U-box domain, thus distinguishing it from typical plant U-box E3 ubiquitin ligas

Gadolinium Protects Arabidopsis thaliana against Botrytis cinerea through the Activation of JA/ET-Induced Defense Responses.

Plant food production is severely affected by fungi; to cope with this problem, farmers use synthetic fungicides. However, the need to reduce fungicide application has led to a search for alternatives, such as biostimulants. Rare-earth elements (REEs) are widely used as biostimulants, but their mode of action and their potential as an alternative to synthetic fungicides have not been fully studied. Here, the biostimulant effect of gadolinium (Gd) is explored using the plant-pathosystem Arabidopsis thaliana-Botrytis cinerea. We determine that Gd induces local, systemic, and long-lasting plant d
