Comprehensive genome-wide identification, molecular characterisation, and expression profiling of HSP gene families in Bufo gargarizans.

Heat shock proteins (HSPs) play essential roles in cellular stress responses and development. However, these proteins have not been adequately characterised in the amphibian Bufo gargarizans. In this study, we performed genome-wide identification of the HSP family in B. gargarizans and investigated their expression dynamics under cold stress. A total of 56 BgaHSPs were identified, which were classified into six subfamilies (HSP10, HSP20, HSP40, HSP60, HSP70, and HSP90) and unevenly distributed across 10 chromosomes. Structure and motif analyses revealed family-specific conservation and diverge

A NOVEL AMPHIBIAN MYXOSPOREAN, CYSTODISCUS CHONGQINGENSIS N. SP., INFECTING THE ASIATIC TOAD (BUFO GARGARIZANS) FROM CHINA.

In a parasitological survey of Asiatic toads (Bufo gargarizans) from Chongqing, China, a novel myxosporean was encountered. The species, Cystodiscus chongqingensis n. sp., was found in the gallbladder of toads. The elliptoid myxospores were morphologically consistent with a Cystodiscus species but were distinct from other congeners by myxospore dimensions and DNA sequence comparisons. Myxospores were 14.2 ± 0.4 μm long (13.6-14.9 μm) and 11.1 ± 0.5 μm wide (10.4-12.0 μm), and the polar capsules were 4.6 ± 0.3) μm long (4.0-5.1 μm) and 3.4 ± 0.3 μm wide (2.6-3.8 μm). Phylogenetically, this new

Potential mechanism of delayed endochondral ossification and muscle atrophy induced by nitrate exposure in Bufo gargarizans tadpoles.

Nitrate pollution in aquatic ecosystems is a pervasive and growing problem globally, which can pose a substantial threat to amphibian survival. The shift in amphibian locomotive behaviors, specifically from tadpole tail swimming to adult limbs movement, are essential for facilitating physiological adaptations required for terrestrial survival. However, there is a question as to whether nitrate exposure affects amphibian motor systems. For this purpose, morphological, histological, transmission electron microscopic and transcriptomic analyses were performed on the hindlimb bones and muscles of

The structural discrepancy between the small and large gut microbiota of Asiatic toad (Bufo gargarizans) during hibernation.

Hibernating amphibians are suitable for the research on the adaptation of gut microbiota to long-term fasting and cold stresses. However, the previous studies mainly focus on the large or whole gut microbiota but not the small gut microbiota. To test the structural discrepancy between the small and large gut microbiota during hibernation, we performed two independent batches of 16S rRNA gene amplicon sequencing to profile the small and large gut microbiota of hibernating Asiatic toad (Bufo gargarizans) from two wild populations. Both batches of data revealed that Proteobacteria, Bacteroidetes,

Elevational Variation in Organ Size and Brain-Viscera Energetic Trade-Offs in the Minshan Toad (Bufo minshanicus).

Energy trade-offs underlying brain size evolution represent a central topic in evolutionary biology. The Expensive Brain Hypothesis (EBH) posits that the high energetic costs of large brains must be compensated for by increased energy intake or reduced allocation to other costly tissues and functions. We examined the Minshan toad (Bufo minshanicus) along an elevational gradient (2594-3441 m) on the eastern Tibetan Plateau, measuring relative sizes of the brain, five visceral organs, and two reproductive organs, as well as body condition (SMI). Body size and condition significantly decreased wi

De novo sequencing of peptides from the parotid secretion of the cane toad, Bufo marinus (Rhinella marina).

Amphibian skin secretions are well known as a rich source of bioactive peptides. However, little is known about the presence or role of peptides in the highly toxic, parotid secretion of the cane toad or giant toad, Bufo marinus (Rhinella marina), though small molecule bufadienolides, which act as potent cardiotoxins, have been described. In the current study we used RP-HPLC, MALDI-TOF mass spectrometry and tandem mass spectrometry to analyze and determine the first sequences of peptides from the parotid secretion of B. marinus. We show that peptides in the range of 900-2500 Da are indeed pres

New species of Rhabdias (Nematoda: Rhabdiasidae) from Bufo occidentalis (Anura: Bufonidae) from Sierra Madre del Sur, Mexico.

Rhabdias kuzmini n. sp., a parasite of the lungs of Bufo occidentalis, is described and illustrated. This Mexican taxon differs from the related species in the genus by the possession of 4 lips (2 subdorsal and 2 subventral) and 2 lateral pseudolabia, corpus not inflated, a larger barrel-shaped buccal capsule, equatorial vulva, and the presence of a slightly swollen cuticle in the anterior and posterior ends of the body. This is the 16th species described in the Neotropical Realm and the first species of Rhabdias described from endemic anurans in México.

A record of juvenile Paragonimus skrjabini miyazakii from the urinary bladder of the Japanese toad, Bufo japonicus formosus.

We found juveniles of Paragonimus in the urinary bladder of a Japanese toad (Bufo japonicus formosus) captured in Kyoto. These were molecularly identified as Paragonimus skrjabini miyazakii Kamo, Nishida, Hatsushika et Tomimura 1961. This is the first report of P. s. miyazakii found in anuran hosts in Japan, indicating that anurans can be paratenic hosts of P. s. miyazakii, as is also the case for Paragonimus skrjabini skrjabini in China. This finding suggests that definitive hosts of P. s. miyazakii can be infected by eating not only crabs or mammal paratenic hosts, but also anurans.

Probing the effects of hexavalent chromium exposure on histology and fatty acid metabolism in liver of Bufo gargarizans tadpoles.

Hexavalent chromium is one of the major detrimental heavy metal pollutants. B. gargarizans tadpoles were treated with different concentrations of Cr6+ (0, 13, 52, 104, 208, and 416 μg Cr6+ L-1) from Gs 2 to Gs 42. The effect of Cr6+ on histopathological alterations and transcript levels of fatty acid metabolism-related genes as well as fatty acids composition and content in liver were examined. Histopathological changes were observed in liver at 52, 104, 208, and 416 μg Cr6+ L-1. Moreover, RT-qPCR analyses showed the downregulated mRNA levels of the genes related to fatty acid synthesis (SCD,

The effect of soil composition and hydration on the bioavailability and toxicity of cadmium to hibernating juvenile American toads (Bufo americanus).

The soil ecotoxicology literature has focused primarily on a few major taxa, to the neglect of other fossorial organisms such as amphibians. We selected cadmium (Cd) and the American toad (Bufo americanus) as a model contaminant and biological species to assess the impact of soil contamination on amphibian hibernation survival and post-hibernation condition. Soil sand composition (50, 70, 90%) and hydration (100, 150% water holding capacity (WHC)) were manipulated in addition to Cd concentration (0, 56, 165, 483 microg/g) to determine whether these soil properties affect toxicity. Melittin-induced changes in Na and Cl movements across the skin and cornea (in vitro) of the toad Bufo marinus.

Melittin, from bee venom, increases short-circuit current (Isc) across the skin and cornea of toads. In skin this reflects a rise in the influx of Na and is inhibited by meclofenamic acid (inhibits prostaglandin synthetase). In corneas with melittin on the inside the rise in Isc is inhibited by bumetanide (inhibits Cl transport) and meclofenamic acid. Melittin on the tear side of the cornea causes a biphasic change in Isc, and a rise in all undirectional fluxes of Na and Cl. This effect was not changed by bumetanide or meclofenamic acid. Melittin apparently has two types of effects, one mediat

Preliminary pharmacological characterization of the isolated circular strips of gastric muscularis muscle of Bufo marinus: a new preparation.

This paper describes a preliminary pharmacological study of isolated circular strips of gastric muscularis muscle of Bufo marinus under isometric conditions. The stomach of Bufo marinus was surgically removed and the muscularis muscle was isolated and divided into 12 equal parts from the fundus to the pylorus. The preparation contracted to acetylcholine (ACh), DL-muscarine, and K+. Histamine and 5-hydroxytryptamine had no effect, while dopamine caused relaxation. Prostaglandin E2 (PGE2) caused relaxation of the tonic contractions and potentiation of spontaneous contractions. A challenge of the

Antioxidant responses to azinphos methyl and carbaryl during the embryonic development of the toad Rhinella (Bufo) arenarum Hensel.

Amphibian embryos are naturally exposed to prooxidant conditions throughout their development. Environmental exposure to contaminants may affect their capacity to respond to challenging conditions, to progress in a normal ontogenesis, and finally to survive and succeed in completing metamorphosis. We studied the effects of the exposure to two anticholinesterase agents, the carbamate carbaryl and the organophosphate azinphos methyl, on the antioxidant defenses of developing embryos of the toad Rhinella (Bufo) arenarum. Reduced glutathione (GSH) levels were increased early by carbaryl, but were

Nitrate induced hepatic fibrosis in tadpoles of Bufo gargarizans by mediating alterations in toll-like receptor signaling pathways.

The nitrate pollution has become an increasingly serious environmental problem worldwide, and the toxic effects of elevated nitrate levels in the environment on aquatic animals remain to be elucidated. The purpose of the present study was to investigate the mechanisms of liver injury to tadpoles after exposure to nitrate from embryonic to metamorphic climax and to assess the recovery process of liver function after cessation of exposure. In the group with continuous nitrate exposure, the livers and thyroid of tadpoles showed remarkably histological lesions, of this with structural disorganizat

Helminth community structure in tadpoles of northern leopard frogs (Rana pipiens) and Woodhouse's toads (Bufo woodhousii) from Nebraska.

Currently, little information is available on helminth community structure in tadpoles, the ephemeral, feeding, non-reproductive larva stages of anurans. We examined the helminth community structure in tadpoles of northern leopard frogs, Rana pipiens, and Woodhouse's toads, Bufo woodhousii, from a first-order stream in Western Nebraska. Specifically, we were interested in how species-specific difference in size, time to metamorphosis, and habitat partitioning among larval anurans affect their helminth community structure. From June-July 2009, we collected 30 tadpoles of northern leopard frogs,
