Emerging roles of neutrophil-borne S100A8/A9 in cardiovascular inflammation.

Elevated neutrophil count is associated with higher risk of major adverse cardiac events including myocardial infarction and early development of heart failure. Neutrophils contribute to cardiac damage through a number of mechanisms, including attraction of other immune cells and release of inflammatory mediators. Recently, a number of independent studies have reported a causal role for neutrophil-derived alarmins (i.e. S100A8/A9) in inducing inflammation and cardiac injury following myocardial infarction (MI). Furthermore, a positive correlation between serum S100A8/A9 levels and major advers

Differential expression of the calcium-binding proteins MRP8 and MRP14 in granulomatous conditions: an immunohistochemical study.

MRP14 and MRP8 are well-characterized calcium-binding proteins present in myeloid cells and mononuclear phagocytes. These antigens can easily be visualized in paraffin-embedded tissue, making use of monospecific polyclonal antibodies. This study evaluates MRP14 and MRP8 expression in mononuclear phagocytes in various granulomatous conditions. MRP14 is strongly expressed in all granulomatous conditions. MRP8 is variably expressed. Mononuclear phagocytes in granulomas of foreign body type, cat-scratch disease and erythema nodosum strongly express MRP8. In contrast, MRP8 expression is weak or abs

S100-alarmins, antenatal corticosteroids and the risk of late-onset sepsis in preterm infants: A prospective cohort study.

OBJECTIVES: Antenatal corticosteroids (aCS) are an important measure improving the outcome of preterm infants. Their influence on late-onset sepsis (LOS) risk remains inconclusive. The alarmin S100A8/A9 protects from LOS by regulating innate immune responses. We examined whether aCS impact on postnatal S100A8/A9 serum-levels and consequently on LOS risk. STUDY DESIGN: In a prospective birth-cohort study of 162 preterm infants born before 32 gestational weeks, we determined postnatal S100A8/A9 serum-levels in relation to the timing of aCS and LOS incidence. RESULTS: aCS administration within

S100A8+S100A9+ transitional macrophages are associated with pulmonary fibrosis progression by integrating immunometabolism and fibrogenic crosstalk.

BACKGROUND: Idiopathic pulmonary fibrosis (IPF) is a progressive interstitial lung disease with limited therapeutic options. Although macrophage heterogeneity has been implicated in pathogenesis, the contribution of transitional macrophage states remains poorly understood. METHODS: We integrated single-cell RNA-seq from 93 human lung specimens (44 IPF, 49 controls) and analyzed peripheral blood single-cell datasets (31 IPF, 17 controls). We examined differentiation trajectories and ligand-receptor networks and assessed metabolic programs. Functional relevance was evaluated in a bleomycin mous

Spatial atlas of upper tract urothelial carcinoma identifies SLC14A1 + and S100A8/9+ tumor cells as key determinants of malignant progression.

Upper tract urothelial carcinoma (UTUC) is characterized by a high incidence of muscle invasion and distinct molecular heterogeneity, with tumor microenvironment (TME) heterogeneity playing a pivotal role in its malignant progression. However, the spatial distribution patterns of functional cell populations in UTUC and their regulatory mechanisms driving high-grade (HG) and muscle-invasive (MI) progression remain largely unelucidated. Herein, we integrated single-cell RNA sequencing (scRNA-seq) and spatial transcriptome sequencing (ST-seq) to systematically characterize the cellular landscape,

Prostaglandins prevent long-lasting pain in a mouse model of chronic postsurgical pain.

BACKGROUND: Chronic postsurgical pain (CPSP) affects 5-85% of patients, depending on the surgery type, with minimal treatment options. A previous study showed that immune response inhibition by dexamethasone (DEXA) or NSAIDs in inflammatory pain assays in mice leads to pain prolongation, which can be reversed by injection of neutrophils or neutrophil-released S100A8/A9 proteins. Here we tested whether DEXA or NSAIDs similarly lead to prolonged pain in the mouse paw incisional assay, and if prostaglandin (PG) receptor agonists can prevent and resolve this pain through upregulation of S100A8/A9

Inhibition of S100A8/A9 ameliorates neuroinflammation by blocking NET formation following traumatic brain injury.

Traumatic brain injury (TBI) triggers a robust inflammatory response that is closely linked to worsened clinical outcomes. S100A8/A9, also known as calprotectin or myeloid-related protein-8/14 (MRP8/14), is an alarmin primarily secreted by activated neutrophils with potent pro-inflammatory property. In this study, we explored the roles of S100A8/A9 in modulating neuroinflammation and influencing TBI outcomes, delving into the underlying mechanisms. S100A8/A9-enriched neutrophils were present in the injured brain tissue of TBI patients, and elevated plasma levels of S100A8/A9 were correlated wi

Calprotectin (S100A8/A9) is not associated with ultrasound-detected synovitis in a longitudinal study of patients with psoriatic arthritis treated with biological disease-modifying anti-rheumatic drugs.

OBJECTIVES: Calprotectin (S100A8/A9) is an established inflammatory marker in rheumatoid arthritis (RA), but its role in psoriatic arthritis (PsA) is less studied. This study evaluated plasma calprotectin as a biomarker of inflammatory activity in PsA by assessing its association with ultrasound-detected synovitis before and during treatment with a biological disease-modifying anti-rheumatic drug (bDMARD). The potential of S100A12, vascular endothelial growth factor, interleukin-6 (IL-6), IL-17A, IL-23, and C-X-C motif chemokine ligand 10 (CXCL10) was also explored. METHOD: Forty-three PsA pa

Neutrophil proteins as potential biomarkers for a sputum-based tuberculosis screening test.

INTRODUCTION: The development of a rapid and affordable assay to screen participants for Q12 additional testing could streamline TB screening in resource-limited settings and for community-wide health screens. Sputum remains the primary testing sample, making it potentially ideal for a screening testing. Neutrophils are highly expanded in sputum from individuals with pulmonary TB with high specificity and have potential as a biomarker for TB. METHODS: Three neutrophil associated proteins, neutrophil gelatinase associated-lipocalin (NGAL), the protein heterodimer S100A8/A9 and the protein deat

Major Neutrophil-Derived Soluble Mediators Associate With Baseline Lung Pathology and Post-Treatment Recovery in Tuberculosis Patients.

BACKGROUND: The inflammatory response to Mycobacterium tuberculosis results in variable degrees of lung pathology during active TB (ATB) with central involvement of neutrophils. Little is known about neutrophil-derived mediators and their role in disease severity at baseline and recovery upon TB treatment initiation. METHODS: 107 adults with confirmed pulmonary TB were categorised based on lung pathology at baseline and following successful therapy using chest X-ray scores (Ralph scores) and GeneXpert bacterial load (Ct values). Plasma, sputum, and antigen-stimulated levels of MMP1, MMP3, MMP

Targeted proteomics reveals serum amyloid A variants and alarmins S100A8-S100A9 as key plasma biomarkers of rheumatoid arthritis.

Serum amyloid A (SAA) and S100 (S100A8, S100A9 and S100A12) proteins were previously identified as biomarkers of interest for rheumatoid arthritis (RA). Among SAA family, two closely related isoforms (SAA-1 and SAA-2) are linked to the acute-phase of inflammation. They respectively exist under the form of three (α, β, and γ) and two (α and β) allelic variants. We developed a single run quantitative method for these protein variants and investigated their clinical relevance in the context of RA. The method was developed and validated according to regulations before being applied on plasma comin

NFκB1 is a suppressor of neutrophil-driven hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) develops on the background of chronic hepatitis. Leukocytes found within the HCC microenvironment are implicated as regulators of tumour growth. We show that diethylnitrosamine (DEN)-induced murine HCC is attenuated by antibody-mediated depletion of hepatic neutrophils, the latter stimulating hepatocellular ROS and telomere DNA damage. We additionally report a previously unappreciated tumour suppressor function for hepatocellular nfkb1 operating via p50:p50 dimers and the co-repressor HDAC1. These anti-inflammatory proteins combine to transcriptionally repress he

S100A8/A9-activated IFNγ+ NK cells trigger β-cell necroptosis in hepatitis B virus-associated liver cirrhosis.

BACKGROUND AND AIMS: Hepatitis B virus (HBV)-associated liver cirrhosis (LC), a common condition with high incidence and mortality rates, is often associated with diabetes mellitus (DM). However, the molecular mechanisms underlying impaired glucose regulation during HBV-associated LC remain unclear. METHODS: Data from 63 patients with LC and 62 patients with LC-associated DM were analysed. Co-culture of NK cells and islet β cell lines were used to study the glucose regulation mechanism. A mouse model of LC was used to verify the effect of S100A8/A9 on the glucose regulation. RESULTS: Higher

Proteomic analysis of lipopolysaccharide-treated submandibular gland in rat.

We investigated changes in the protein profile of submandibular gland (SMG) with inflammation induced by exposure to lipopolysaccharide (LPS) with the aim of identifying potential molecular markers of injured gland. Lipopolysaccharide (2.5µg) was directly administered into rat SMG unilaterally by retrograde ductal injection. At 12hr after treatment, the gland was excised and the proteins identified by two-dimensional difference gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight mass spectrometry. Many proteins in the LPS-treated gland showed a marked change comp

Chronic skin and systemic inflammation modulated by S100A8 and S100A9 complexes.

Increased expression of the homodimeric S100A8 (A8) and S100A9 (A9) alarmins and their Calprotectin (CP) antimicrobial hetero-complex has been reported in Inflammatory Skin Diseases (ISDs) such as Atopic Dermatitis (AD), but the functional consequences of this increase are not known. We evaluated the cell- and tissue-specific functions of A8 and A9 in the local and the extra-cutaneous manifestations of ISD using genetically engineered mouse models. The genes encoding for the A9 or A8 proteins were inactivated in epidermal cells or neutrophils in the JunB∆ep genetic mouse model for AD. Overall,
