Campylobacter (C.) jejuni is the most important reported cause for bacterial diarrhoea. The infection can be accompanied by fever and abdominal cramps and in rare cases the Guillain-Barré syndrome or reactive arthritis can develop as a post-infection complication. Several biological properties of Cjejuni, e. g. motility and chemotaxis, contribute to the biological fitness of the pathogen. For this, deficiencies in the function of these features clearly reduce the pathogenicity of C. jejuni without being a virulence factor per se. Opposing to this, there are two essential requirements to determ

The Gram-negative bacterium Campylobacter jejuni causes gastroenteritis and Guillain-Barré syndrome in humans. Recent advances in the immunobiology of C. jejuni have been made. This review summarizes C. jejuni-binding innate receptors and highlights the role of innate immunity in autoimmune diseases. This human pathogen produces a variety of glycoconjugates, including human ganglioside-like determinants and multiple activators of Toll-like receptors (TLRs). Furthermore, C. jejuni targets MyD88, NLRP3 inflammasome, TIR-domain-containing adapter-inducing interferon-β (TRIF), sialic acid-binding …

Campylobacter spp. are the most common cause of bacterial gastroenteritis worldwide and have been isolated from a wide number of different hosts and environmental sources. Waterfowl is considered a natural reservoir for this zoonotic bacterium and may act as a potential infection source for human campylobacteriosis. In this study, faecal samples from 924 barnacle geese were tested for the presence of C. jejuni and C. coli. The resulting C. jejuni and C. coli populations were characterized by multilocus sequence typing (MLST), structure analysis by BAPS and phylogenetic analysis based on full g

Campylobacter jejuni is a common cause of gastroenteritis especially in childhood. However, extraintestinal manifestations of C. jejuni such as bacteremia, cholecystitis, pancreatitis and osteomyelitis are rare. Meningitis in newborns caused by this organism is unusually rare. We report a case of neonatal meningitis in a 15-day old term boy caused by Campylobacter jejuni.

CONCLUSIONS: We have constructed the first curated metabolic model for the food-borne pathogen Campylobacter jejuni and have presented the resulting metabolic insights. We have shown that the combination of in silico and in vivo approaches could point to non-redundant, indispensable genes associated with the well characterised shikimate pathway, and also genes of …

AIM: To determine the effect of two physiologically important temperatures on growth and chemotaxis in Campylobacter jejuni.
METHODS AND RESULTS: Growth curves of Camp. jejuni were compared at 37 degrees C and 42 degrees C. Chemotaxis was compared at 37 degrees C and 42 degrees C by the disc and capillary assays. Student's t-test was applied to the results of the capillary assay to assess the significance in the difference between chemotaxis at the two temperatures. Both, the growth rate and chemotactic ability of the isolate, were found to be greater at 37 degrees C.
CONCLUSIONS: Quorum sen…

Mechanisms of drug resistance in Campylobacter jejuni were investigated. Mutant strains 34PEFr, which was resistant to pefloxacin (128-fold increase in the MIC), and 34CTXr, which was resistant to cefotaxime (32-fold increase in the MIC) and which was derived from the susceptible parent 34s, were obtained by serial passages on pefloxacin and cefotaxime gradient plates, respectively. Both mutants showed cross-resistance to erythromycin, chloramphenicol, tetracycline, beta-lactams, and quinolones. While the quinolone resistance of strain PEFr could be explained by a mutation at codon 86 of the g…

An infant mouse model has been used to investigate the colonisation of the intestine by Campylobacter jejuni and the effect of endotoxin (Escherichia coli O26: B6) on the initial stage of this process. Endotoxin injected 1 or 16 h before the bacterial challenge had no effect on the growth of campylobacters but endotoxin injected 4 to 10 h before the bacterial challenge caused a bacteriostatic effect on the growth of campylobacters which lasted for one day. The bacteriostatic effect was evident both in the small intestine and in the distal part of the intestine containing caecum and colon. The …

During a 2-yr period starting on 22 July 1979, Campylobacter jejuni was isolated in 3,008 stool cultures from 2,450 persons--2,430 (43.%) out of 56,439 patients with acute gastroenteritis and 20 out of 200 asymptomatic household contacts. No isolates of C. jejuni were obtained from any of a control group of 222 subjects. In 176 patients with C. jejuni, Salmonella and/or Shigella were isolated concomitantly. All illnesses were mild to severe and all patients, including 61 requiring hospitalization, recovered uneventfully. Patients' ages ranged from 2 wk to 88 yr, with infants and children compr…

Campylobacter jejuni (C. jejuni) is a prevalent zoonotic pathogen that poses a significant threat to global public health, causing gastroenteritis with symptoms ranging from mild diarrhea to severe complications, and is widely distributed among various animal hosts, including poultry, livestock, and wild birds. Among these, wild birds, characterized by their exceptional species diversity, high mobility, and long-distance migratory behavior, are capable of disseminating infectious agents across geographic regions and ecological settings. However, wild birds are increasingly recognized as potent…

Campylobacter jejuni is a typical zoonotic bacterium, colonizing the gut of many bird species as commensal. In humans, C. jejuni is a major foodborne pathogen. Infection of humans causes campylobacteriosis in the small intestine, constituting a main source of bacteria-dependent gastroenteritis cases worldwide. In particular, the ingestion of under-cooked rooster meat, raw milk and contaminated water, as well as cross-contamination of ready-to-eat food after handling raw chicken meat, are responsible for the majority of C. jejuni infections. As a consequence, infected individuals may acquire wa…

Lysophospholipids are an important class of lipids in both prokaryotic and eukaryotic organisms. These lipids typically constitute a very small proportion (<1%) of the bacterial lipidome but can constitute 20%-45% of the Campylobacter jejuni lipidome under stress conditions. It is thus of importance to include these lipids in model C. jejuni membrane simulations for an accurate representation of the lipidic complexity of these systems. Here, we present atomistic models for four lysophospholipids from the C. jejuni lipidome, each derived from existing phospholipid models. Herein, we use molecul…

OBJECTIVE: Guillain-Barré syndrome (GBS) is a postinfectious neuropathy most frequently caused by Campylobacter jejuni. Lipo-oligosaccharides (LOS), expressed by C. jejuni induce antibodies that cross-react with self-glycolipids in peripheral nerves, causing neuropathy. Less than 1 in 1,000 persons infected with C. jejuni develop GBS, and the factors that determine GBS susceptibility are poorly understood. We hypothesized that these persons have a high intrinsic dendritic cell (DC) response to C. jejuni LOS through Toll-like receptor 4 (TLR4) activation.
METHODS: Intrinsic DC responsiveness t…

Campylobacter enteritis has emerged as one of the most common forms of human diarrheal illness. In this study we have investigated the virulence potential of five new C. jejuni chicken isolates (RO14, RO19, RO24, RO29 and RO37) originated from private households in the rural regions of Banat and Transylvania in Romania. Following isolation and in vitro virulence assay, on HCT-8 cells, our results show that all the C. jejuni chicken isolates overcome the virulence abilities of the highly virulent strain C. jejuni 81-176. Motility, an important virulence factor was significantly improved in all

Campylobacter jejuni continues to be the leading cause of bacterial food-borne illness worldwide, so improvements to current methods used for bacterial detection and disease prevention are needed. We describe here the genome and proteome of C. jejuni bacteriophage NCTC 12673 and the exploitation of its receptor-binding protein for specific bacterial detection. Remarkably, the 135-kb Myoviridae genome of NCTC 12673 differs greatly from any other proteobacterial phage genome described (including C. jejuni phages CP220 and CPt10) and instead shows closest homology to the cyanobacterial T4-related…

The lipooligosaccharide (LOS) molecules of Campylobacter jejuni are involved in virulence and induction of the Guillain-Barré syndrome (GBS). This study analysed the transcription of the LOS synthesis genes from the GBS-inducing C. jejuni strain HB 93-13 under microaerobic conditions. Fourteen consecutive genes Cj1132c, waaC, htrB, wlaNC, wlaND, cgtA, cgtB, cstII, neuB, neuC, neuA, wlaVA, wlaQA, and waaF were included. The results of rapid amplification of cDNA ends and single-stranded ligation of complementary ends showed initiation sites with potential promoter regions on both DNA strands in…

Campylobacter spp. continue to be the greatest cause of bacterial gastrointestinal infections in humans worldwide. They encounter many stresses in the host intestinal tract, on foods and in the environment. However, in common with other enteric bacteria, they have developed survival mechanisms to overcome these stresses. Many of the survival mechanisms used by Campylobacter spp. differ from those used by other bacteria, such as Escherichia coli and Salmonella spp. This review summarizes the mechanisms by which Campylobacter spp. adapt to stress conditions and thereby increase their ability to
