Preclinical evaluation of cannabidiolic acid as a neuroprotective agent in TDP-43 transgenic mice, an experimental model of amyotrophic lateral sclerosis.

Plant-derived cannabinoids, including Δ9-THC, cannabinol, and Sativex-like combinations, have shown neuroprotection in preclinical ALS models. However, minor phytocannabinoids like cannabidiolic acid (CBDA) remain unexplored. This study evaluated the neuroprotective effects of CBDA, cannabidivarin, CBD, Δ9-THC, and Δ9-tetrahydrocannabidivarin in Prp-hTDP-43(A315T) transgenic male mice from early symptomatic (day 65) to advanced stages (day 90). CBDA proved the most effective, improving motor coordination (rotarod test) and reducing neuronal cell death, gliosis, microglial reactivity, and pro-i

Modeling the liquid-liquid chromatography separation of cannabinoids from hemp extracts.

The separation of cannabinoids from hemp materials is nowadays one of the most promising industrial applications of liquid-liquid chromatography (LLC). Despite various experimental research efforts to purify cannabinoids, there are currently few works on process modeling. Thus, this study aimed to explore a straightforward approach to model the LLC separation of cannabinoids from two hemp extracts with different compositions. The feed materials were simplified to mixtures of preselected key components (i.e., cannabidiol, tetrahydrocannabinol, cannabigerol, and cannabinol). The elution profiles

Origin of the Different Binding Affinities of (9R)- and (9S)-Hexahydrocannabinol (HHC) for the CB1 and CB2 Cannabinoid Receptors.

Hexahydrocannabinols (HHCs) are emerging cannabinoids that have become available for recreational use and were recently classified as Schedule II under an international treaty. Although often advertised for having desirable effects, recent studies have shown that commercial products typically contain variable amounts of two epimers, (9R)-HHC and (9S)-HHC. In turn, these epimers have been shown to have different binding affinities to the CB1 and CB2 receptors. We report a computational study that interrogates the origins of these differing affinities. Molecular docking and molecular dynamics si

Hexahydrocannabinol: pharmacokinetics, systemic toxicity, and acute behavioral effects in Wistar rats.

BACKGROUND: Hexahydrocannabinol (HHC) is a new psychoactive substance known for its mind-altering effects and temporary legal status. It is widely used in parts of the Europe and United Kingdom as a legal alternative to ∆9-tetrahydrocannabinol, yet little research has explored its effects and safety. This study examined how HHC is processed in the body, its toxicity, and its impact on behavior in male Wistar rats. METHODS: A 1:1 mixture of (9R)-HHC and (9S)-HHC was administered via intragastric gavage at doses of 1, 5, and 10 mg/kg. Behavioral effects were assessed using the open field test a

In Vitro Inhibition of Carboxylesterase 1 by Major Cannabinoids and Selected Metabolites.

The escalating use of medical cannabis and significant recreational use of cannabis in recent years has led to a higher potential for metabolic interactions between cannabis or one or more of its components and concurrently used medications. Although there have been a significant number of in vitro and in vivo assessments of the effects of cannabis on cytochrome P450 and UDP-glucuronosyltransferase enzyme systems, there is limited information regarding the effects of cannabis on the major hepatic esterase, carboxylesterase 1 (CES1). In this study, we investigated the in vitro inhibitory effect

The effects of pH and temperature on the in vitro bindings of delta-9-tetrahydrocannabinol and other cannabinoids to bovine serum albumin.

Albumin is a major carrier of drugs and fatty acids in biological fluids. These protein-drug complexes serve to solubilize, transport these compounds to sites of action, and have been associated with increased half-life for these compounds. The authors are interested in the pH and temperature effects of the binding of delta-9-tetrahydrocannabinol to albumin. Ultrafiltration techniques were used in the separation of free to bound compounds. Cannabinoids bind to bovine serum albumin rapidly. The cannabinoid binding sites are more sensitive to temperature changes (37-47 degrees C) than changes in

Chemical profiling of Cannabis sativa from eleven Tanzanian regions.

The aim of this research was to investigate the chemical profiles of Cannabis sativa from 11 Tanzanian regions using preliminary tests as well as instrumental analyses with GC-MS and LC-MS. Generally, all the seized samples tested positive for the presence of (Δ9-THC. The preliminary test with Duquenois method followed by chloroform addition revealed the presence of Δ9-tetrahydrocannabinol (Δ9-THC) in all the samples. GC-MS analyses of the samples revealed the presence of nine cannabinoids including Δ9-THC, Δ8-THC, cannabidivarol, cannabidiol, Δ9-tetrahydrocannabivarin (Δ9-THCV), cannabichrome

Metal-Free Synthesis of Pharmaceutically Important Biaryls by Photosplicing.

Many pharmaceuticals feature biaryl motifs that are crucial for their binding to the target. Yet, benchmark methods for selective cross-couplings rely on highly toxic heavy metal catalysts, which are unfavorable in the synthesis of pharmaceuticals. Metal-free coupling reactions, on the other hand, may require harsh conditions and lack selectivity. We report a novel, metal-free cross-coupling reaction that involves the tethering of two phenyl groups by a temporary, traceless sulfonamide linker that directs a photochemical aryl fusion into a single coupling product. The perfect regio- and chemos

Recent applications of capillary electrophoresis-electrospray ionisation-mass spectrometry in drug analysis.

A critical review of applications for the period 2000-2004, taken from the Web of Knowledge database, of the technique capillary electrophoresis-electrospray ionisation-mass spectrometry (CE-ESI-MS) in drug analysis is presented. The review is concerned with molecules of mass less than 500 Da, chosen according to selected structural classes in which they give ESI signals primarily as [M+H](+) ions although other ions, such as [M-H](-), [M+Na](+), and [M+NH(4)](+), are also reported. These structural classes are drugs with amine-containing side chains, drugs with N-containing saturated ring str

Cannabis sativa extracts reduce inclusion formation in a cell model of alpha-synuclein aggregation.

Parkinson's disease (PD) is classified as a synucleinopathy due to the accumulation of protein inclusions rich in the alpha-synuclein (aSyn) protein. Identifying effective pharmacological therapies is important to slow the progression and minimize the symptoms of these diseases. Cannabis sativa has a diverse chemical profile depending on its genotype, including several classes of substances, such as cannabinoids, flavonoids, terpenes, and alkaloids. In this study, we evaluated the effects of four C. sativa extracts with different phytocannabinoid chemical profiles in two cellular models that r

Cannabinoids attenuate norepinephrine-induced melatonin biosynthesis in the rat pineal gland by reducing arylalkylamine N-acetyltransferase activity without involvement of cannabinoid receptors.

Cannabinoids modulate neuronal and neuroendocrine circuits by binding to cannabinoid receptors acting upon cAMP/Ca(2+)-mediated intracellular signaling cascades. The rat pineal represents an established model to investigate intracellular signaling processes because a well defined input, the neurotransmitter norepinephrine, is transformed via cAMP/Ca(2+)-dependent mechanisms into an easily detectable output signal, the biosynthesis of melatonin. Here we investigated the impact of cannabinoids on norepinephrine-regulated melatonin biosynthesis in the rat pineal. We demonstrated that treatment of

Inhalation and oral administration of HHC products - Quantification of (9R)-, (9S)-Hexahydrocannabinol and metabolites in plasma and detectability in on-site drug tests for urine and oral fluid.

The semisynthetic cannabinoid Hexahydrocannabinol (HHC) has gained recognition among drug users. A GC-MS/MS method for the detection of (9R)- and (9S)-HHC and their respective carboxy- and hydroxy-metabolites in plasma has been developed and validated. The method was applied to authentic plasma samples obtained from a self-administration experiment. HHC was either inhaled (Vapes, 95 % HHC) or ingested (Jellys, 25 mg HHC). Maximum plasma (9R)-HHC and (9S)-HHC concentrations of 3.8 ng/mL and 2.5 ng/mL were detected 1.16 h after ingestion and approx. 65 ng/mL (9R)-HHC and 21 ng/mL (9S)-HHC were m

Marihuana: a forensic problem.

Early and unfounded stories concerning marihuana led to many sanctions being placed on the drug. They have also brought about the requirement that the drug be proved safe beyond any reasonable doubt for any consideration to be given to removing restrictive sanctions. Some of the misinformation has been dispelled by controlled studies of its effects. Present available research does not support the thesis of permanent damage, either physical or mental. Yet there appears to be no ongoing research that will adequately demonstrate the safety or the dangers of the drug.

Cannabis for sleep in older adults: use has outpaced the science.

Cannabis use among older adults, defined here as those over 50 years of age, has increased rapidly, and sleep disturbance is among the most common reasons for use. Yet, despite growing use of cannabis and individual cannabinoids to facilitate sleep, evidence supporting their efficacy, tolerability and safety in older adults remains limited. Most experimental studies of cannabinoids and sleep have been conducted in younger adults, often using products and formulations that differ substantially from those currently available. The subjective perception of improved sleep, which may drive increased

Investigation into the oral toxicity profile of TruCBN™.

Floraworks TruCBN™ is a proprietary form of cannabinol (CBN), a high-purity CBN isolate processed to be substantially free of THC. CBN is derived from the Cannabis sativa plant, and is unique among cannabinoids, as it is not produced directly through the cannabis plant's metabolic processes; instead, it forms as an oxidative decomposition product of cannabidiol (CBD) and tetrahydrocannabinol (THC). CBN acts as a low affinity partial agonist at both CB1 and CB2 receptors, with a greater affinity for CB2. Therapeutic properties of CBN include potential efficacy in the treatment of ocular disease

Anticancer effect of minor phytocannabinoids in preclinical models of multiple myeloma.

Multiple myeloma (MM) is a blood cancer caused by uncontrolled growth of clonal plasmacells. Bone disease is responsible for the severe complications of MM and is caused by myeloma cells infiltrating the bone marrow and inducing osteoclast activation. To date, no treatment for MM is truly curative since patients relapse and become refractory to all drug classes. Cannabinoids are already used as palliative in cancer patients. Furthermore, their proper anticancer effect was demonstrated in many cancer models in vitro, in vivo, and in clinical trials. Anyway, few information was reported on the e

Spontaneous Pneumothorax Secondary to Bullous Lung Emphysema Positive for Cannabinoids upon Toxicological Examination.

Cannabis can be related to respiratory diseases, but the relationship between smoking marijuana and the development of a pneumothorax has scarcely been investigated. We aimed to analyze, in patients with a history of cannabis smoking abuse submitted to lung apicectomy for a primary spontaneous pneumothorax (PSP), the correlation between the presence of cannabinoids in the resected lung and the detection of bullous emphysema within the same tissue. Patients undergoing lung apicectomy for a PSP were prospectively enrolled, and the correlation between the presence of cannabinoids in the resected

Amino Acid Supplementation as a Biostimulant in Medical Cannabis (Cannabis sativa L.) Plant Nutrition.

There is growing evidence to support the involvement of nutrients and biostimulants in plant secondary metabolism. Therefore, this study evaluated the potential of amino acid-based supplements that can influence different hydroponic nutrient cycles (systems) to enhance the cannabinoid and terpene profiles of medical cannabis plants. The results demonstrate that amino acid biostimulation significantly affected ion levels in different plant tissues (the "ionome"), increasing nitrogen and sulfur content but reducing calcium and iron content in both nutrient cycles. A significantly higher accumula
