Preparation of open tubular molecule imprinted polymer capillary columns with various templates by a generalized procedure and their chiral and non-chiral separation performance in CEC.

A generalized preparation procedure of open tubular (OT) molecule imprinted polymer (MIP) columns is proposed for a number of templates with acidic functionality such as profen drugs and others. The template (S-enantiomer) was mixed with methacrylic acid, ethylene glycol dimethacrylate and 4-styrenesulfonic acid, dissolved in a porogen mixture of ACN/2-propanol (9/1), and incubated in a pretreated and silanized fused silica capillary by the thermal non-covalent polymerization procedure. The whole preparation procedure was exactly the same for all the MIP capillaries except for the selection of

Morphological regulation of a flower-like hollow β-cyclodextrin covalent organic frameworks for enhanced hydrophilicity and chiral separation.

BACKGROUND: The development of chiral drug analysis methods is the foundation for evaluating drug efficacy and safety. Chiral covalent organic frameworks (CCOFs) show great potential as the chiral stationary phases (CSPs) due to their structural tunability and high stability. However, the inherent hydrophobicity of CCOFs limits their application in capillary electrochromatography (CEC). RESULTS: This study reported the precise morphological regulation of 6-deoxy-6-mercapto-β-cyclodextrin covalent organic frameworks (SH-β-CD COFs). By controlling synthesis parameters, two distinct structures w

Enantioseparation of amino acid amides on an (S)-N-(3,5-dinitrobenzoyl)leucine-N-phenyl-N-propylamide-bonded silica in normal phase CEC.

(S)-N-(3,5-Dinitrobenzoyl)leucine-N-phenyl-N-propylamide-bonded silica was used as a chiral stationary phase for separation of enantiomers of some racemic pi-acidic and pi-basic amino acid amides in normal phase capillary LC (CLC) and CEC. For generation of EOF in CEC, different amounts of water were added to n-hexane-isopropanol (IPA) eluents. Influences of added water and composition of polar modifier on retention, enantioselectivity and resolution were studied. Generally better separation was obtained in the eluents of lower water content. Shorter retention and better enantioselectivities w

Single-step approach for fabrication of vancomycin-bonded silica monolith as chiral stationary phase.

A vancomycin-bonded silica monolithic column for capillary electrochromatography (CEC) was prepared by a single-step in situ sol-gel approach. This sol-gel process incorporates a synthetic sol-gel precursor which contains a macrocyclic antibiotic, vancomycin, to form a porous silica network inside a fused-silica capillary. To avoid degradation of vancomycin during the column fabrication, a mild step was adopted into the sol-gel process. The performance of the vancomycin chiral stationary phase was investigated by CEC in both the reversed-phase mode and the normal-phase mode. The vancomycin chi

UV-polymerized butyl methacrylate monoliths with embedded carboxylic single-walled carbon nanotubes for CEC applications.

The preparation of polymeric monoliths with embedded carboxy-modified single-walled carbon nanotubes (c-SWNTs) and their use for capillary electrochromatography (CEC) is described. Carbon nanotube composites were obtained by preparing a polymerization mixture in the presence of increasing c-SWNT concentrations, followed by UV initiation. The novel stationary phases were studied by optical microscopy, scanning electron microscopy (SEM), transmission electron microscopy (TEM) and Raman spectroscopy. Using short UV-polymerization times, the optimized porogenic solvent (a binary mixture of 1,4-but

In situ functionalization of N-acryloxysuccinimide-based monolith for reversed-phase electrochromatography.

Capillary electrochromatography (CEC) monolithic columns were prepared following a two-step synthetic pathway based on (i) UV-induced in situ radical polymerization of N-acryloxysuccinimide (NAS) and ethylene dimethacrylate (EDMA) and (ii) in situ functionalization of the NAS-containing monolithic matrix with various alkylamines. The first synthetic step was performed using toluene as a porogenic solvent. The successful grafting of the alkylamines onto the reactive matrix was confirmed on the basis of qualitative analysis of Raman spectra recorded before and after the chemical modification ste

Facile preparation of polysaccharide-coated capillaries using a room temperature ionic liquid for chiral separations.

In this study, the dissolution of polysaccharides into an ionic liquid was investigated and applied as a coating onto the capillary walls of a fused-silica capillary in open-tubular CEC. The coating was evaluated by examining the chiral separation of two analytes (thiopental, sotalol) with three cellulose derivatives (cellulose acetate, cellulose acetate phthalate, and cellulose acetate butyrate). Baseline separation of thiopental enantiomers was achieved by use of each polysaccharide coating (Rs: 7.0, 8.1, 7.1), while sotalol provided partial resolution (Rs: 0.7, 1.0, 0.9). In addition, repro

Integration of pillar array columns into a gradient elution system for pressure-driven liquid chromatography.

A gradient elution system for pressure-driven liquid chromatography (LC) on a chip was developed for carrying out faster and more efficient chemical analyses. Through computational fluid dynamics simulations and an experimental study, we found that the use of a cross-Tesla structure with a 3 mm mixing length was effective for mixing two liquids. A gradient elution system using a cross-Tesla mixer was fabricated on a 20 mm × 20 mm silicon chip with a separation channel of pillar array columns and a sample injection channel. A mixed solution of water and fluorescein in methanol was delivered to

Screening for biomarkers in plasma from patients with gangrenous and phlegmonous appendicitis using CE and CEC in combination with MS.

Today a high degree of "false" appendicitis diagnoses are occurring. In this study, a screening experiment of biomarkers of two different kinds of appendicitis, gangrenous and phlegmonous, were conducted with CE and CEC coupled to MS. Plasma samples were obtained from patients pre- and post-surgery. A large amount of data was generated to be able to compare them, and chemometrics tools were utilized to visualize the differences. Indicative patterns were found for both pre- and post-surgery of the two types of inflammation as well as between them. The divergences were traced back to the MS peak

A bioinspired approach for the modulation of electroosmotic flow and protein-surface interactions in capillary electrophoresis using silylated amino-amides blocks and covalent grafting.

We explore a bioinspired approach to design tailored functionalized capillary electrophoresis (CE) surfaces based on covalent grafting for biomolecules analysis. First, the approach aims to overcome well-known common obstacles in CE protein analysis affecting considerably the CE performance (asymmetry, resolution, and repeatability) such as the unspecific adsorption on fused silica surface and the lack of control of electroosmotic flow (EOF). Then, our approach, which relies on new amino-amide mimic hybrid precursors synthesized by silylation of amino-amides (Si-AA) derivatives with 3-isocyana

Separation of acetylcholinesterase inhibitors using polymeric surfactants in polyelectrolyte multilayer coatings.

The main objective of this study is the use of polymeric surfactants in polyelectrolyte multilayer (PEM) coatings for the separation of the pharmaceutical substances acetylcholinesterase inhibitors (AChEIs). AChEIs are used for the treatment of Alzheimer's Disease and Myasthenia Gravis. In the open-tubular capillary electrochromatography (OT-CEC) mode, the PEM coating is evaluated using nine AChEIs. Optimal conditions are established by altering several experimental parameters such as the pH of the background electrolyte (BGE), the anionic polymer for the PEM coating, the concentration of NaCl

CEC-ESI ion trap MS of multiple drugs of abuse.

This article describes a method for the separation and determination of nine drugs of abuse in human urine, including amphetamines, cocaine, codeine, heroin and morphine. This method was based on SPE on a strong cation exchange cartridge followed by CEC-MS. The CEC experiments were performed in fused silica capillaries (100 microm x 30 cm) packed with a 3 mum cyano derivatized silica stationary phase. A laboratory-made liquid junction interface was used for CEC-MS coupling. The outlet capillary column was connected with an emitter tip that was positioned in front of the MS orifice. A stable el

Rapid determination of sterols in vegetable oils by CEC using methacrylate ester-based monolithic columns.

A method for the determination of sterols in vegetable oils by CEC with UV-Vis detection, using methacrylate ester-based monolithic columns, has been developed. To prepare the columns, polymerization mixtures containing monomers of different hydrophobicities were tried. The influence of composition of polymerization mixture was optimized in terms of porogenic solvent, monomers/porogens and monomer/crosslinker ratios. The composition of the mobile phase was also studied. The optimum monolith was obtained with lauryl methacrylate monomer at 60:40% (wt:wt) lauryl methacrylate/ethylene dimethacryl

Construction of chiral crown ethers in robust covalent organic frameworks for electrochromatographic enantioseparation.

Capillary electrochromatography (CEC) is a rapidly emerging separation technique that merges the high separation efficiency of capillary electrophoresis with the exceptional selectivity of liquid chromatography. However, it remains a synthetic challenge to design functional chiral stationary phases (CSPs) with high chemical stability against acid and base in CEC enantioseparation. Here we demonstrate that incorporating chiral crown ethers into stable covalent organic frameworks (COFs) enables efficient and stable separations of racemates by CEC. This facilitates the crafting of two three-dimen

Enantioselectivity of mass spectrometry: challenges and promises.

With the fast growing market of pure enantiomer drugs and bioactive molecules, new chiral-selective analytical tools have been instigated including the use of mass spectrometry (MS). Even though MS is one of the best analytical tools that has efficiently been used in several pharmaceutical and biological applications, traditionally MS is considered as a "chiral-blind" technique. This limitation is due to the MS inability to differentiate between two enantiomers of a chiral molecule based merely on their masses. Several approaches have been explored to assess the potential role of MS in chiral
